    Comparative analysis of cytosolic and mitochondrial ATP synthesis in embryonic and postnatal hippocampal neuronal cultures

    ATP in neurons is commonly believed to be synthesized mostly by mitochondria via oxidative phosphorylation. Neuronal mitochondria have been studied primarily in culture, i.e., in neurons isolated either from embryos or from neonatal pups. Although it is generally assumed that both embryonic and postnatal cultured neurons derive their ATP from mitochondrial oxidative phosphorylation, this has never been tested experimentally. We expressed the FRET-based ATP sensor AT1.03 in cultured hippocampal neurons isolated either from E17 to E18 rat embryos or from P1 to P2 rat pups and monitored [ATP]c simultaneously with mitochondrial membrane potential ΔΨm; TMRM) and NAD(P)H autofluorescence. In embryonic neurons, transient glucose deprivation induced a near-complete decrease in [ATP]c, which was partially reversible and was accelerated by inhibition of glycolysis with 2-deoxyglucose. In the absence of glucose, pyruvate did not cause any significant increase in [ATP]c in 84% of embryonic neurons, and inhibition of mitochondrial ATP synthase with oligomycin failed to decrease [ATP]c. Moreover, ΔΨm was significantly reduced by oligomycin, indicating that mitochondria acted as consumers rather than producers of ATP in embryonic neurons. In sharp contrast, in postnatal neurons pyruvate added during glucose deprivation significantly increased [ATP]c (by 54 ± 8%), whereas oligomycin induced a sharp decline in [ATP]c and increased ΔΨm. These signs of oxidative phosphorylation were observed in all tested P1-P2 neurons. Measurement of ΔΨm with the potential-sensitive probe JC-1 revealed that neuronal mitochondrial membrane potential was significantly reduced in embryonic cultures compared to the postnatal ones, possibly due to increased proton permeability of inner mitochondrial membrane. We conclude that, in embryonic, but not postnatal neuronal cultures, ATP synthesis is predominantly glycolytic and the oxidative phosphorylation-mediated synthesis of ATP by mitochondrial F1Fo-ATPase is insignificant. © 2013 Surin, Khiroug, Gorbacheva, Khodorov, Pinelis and Khiroug.Peer reviewe

    The effect of information seeking behavior on online database usage among the undergraduate students

    Every year the library of IIUM subscribes a quite number of online databases with huge expenses but the utilization of these databases is countable. 10 out of 53 subscriptions have been terminated by 2015 to save the cost go in vain. The purpose of this paper is to develop a model based on the ISB elements to identify the reason behind this less usage and to increase the usage of online databases in future. It has undertaken a quantitative approach to identify the elements of ISB among undergraduate (UG) students. The primary data has been collected through questionnaires based on the variables identified in the objective. There are 118 samples participated in this research and the proposed model has been adapted from Savolainen and Thomas D. Wilson. Psychological perception, searching strategies and the information resources have been considered as the independent variable and measured the usage of the online database among UG students as the dependent variable. The results have shown that there are seven elements which have an influence on the usage of the online database among IIUM UG students. Despite the limited number of samples and a specific group of students, the proposed model can enhance the usage of online database system subscribed by IIUM library as well a search engine can be developed to get the usage increased. In that sense, it is recommended to involve more participants from the different level of students and users to get diverse elements of ISB. However, the paper contributes to bridge the gap between the online database usage and the users which is to believe an effective outcome for online database subscription. It is hoped that the proposed model would significantly fill this gap and help the library to increase more users for their expensive databases

    Measurements of light background at large depth in the ocean

    The mean intensity of Cerenkov emission from the products of K(40) decay and bioluminescence was measured at depths to 5 km. The intensity of ocean light background is found to depend upon depth and at the 5 km level is equal on averaged to 300 + or - 60 quanta/sq cms into spatial angle of 2 pi sterradian in transparency window. The amplitudes, duration and number of BL flashes were measured at various depths. The intensive flashes due to BL are shown to be observed rather seldom at depths over 4 km

    Genetic diversity of Plasmodium falciparum isolates from Pahang, Malaysia based on MSP-1 and MSP-2 genes

    <p>Abstract</p> <p>Background</p> <p>Malaria is still a public health problem in Malaysia especially in the interior parts of Peninsular Malaysia and the states of Sabah and Sarawak (East Malaysia). This is the first study on the genetic diversity and genotype multiplicity of <it>Plasmodium falciparum </it>in Malaysia.</p> <p>Methods</p> <p>Seventy-five <it>P. falciparum </it>isolates were genotyped by using nested-PCR of <it>MSP-1 </it>(block 2) and <it>MSP-2 </it>(block 3).</p> <p>Results</p> <p><it>MSP-1 </it>and <it>MSP-2 </it>allelic families were identified in 65 blood samples. RO33 was the predominant <it>MSP-1 </it>allelic family identified in 80.0% (52/65) of the samples while K1 family had the least frequency. Of the <it>MSP-2 </it>allelic families, 3D7 showed higher frequency (76.0%) compared to FC27 (20.0%). The multiplicity of <it>P. falciparum </it>infection (MOI) was 1.37 and 1.20 for <it>MSP-1 </it>and <it>MSP-2</it>, respectively. A total of seven alleles were detected; of which three <it>MSP-1 </it>allelic families (RO33, MAD20 and K1) were monomorphic in terms of size while <it>MSP-2 </it>alleles were polymorphic (two 3D7 and two FC27). Heterozygosity (H<sub>E</sub>) was 0.57 and 0.55 for <it>MSP-1 </it>and <it>MSP-2</it>, respectively.</p> <p>Conclusions</p> <p>The study showed that the MOI of <it>P. falciparum </it>is low, reflected the low intensity of malaria transmission in Pahang, Malaysia; RO33 and 3D7 were the most predominant circulating allelic families. The findings showed that <it>P. falciparum </it>has low allelic diversity with a high frequency of alleles. As a result, antimalarial drug efficacy trials based on MSP genotyping should be carefully interpreted.</p

    Prevalence and associated factors of Schistosomiasis among children in Yemen: implications for an effective control programme

    BACKGROUND Schistosomiasis, one of the most prevalent neglected tropical diseases, is a life-threatening public health problem in Yemen especially in rural communities. This cross-sectional study aims to determine the prevalence and associated risk factors of schistosomiasis among children in rural Yemen. METHODS/FINDINGS Urine and faecal samples were collected from 400 children. Urine samples were examined using filtration technique for the presence of Schistosoma haematobium eggs while faecal samples were examined using formalin-ether concentration and Kato Katz techniques for the presence of S. mansoni. Demographic, socioeconomic and environmental information were collected via a validated questionnaire. Overall, 31.8% of the participants were found to be positive for schistosomiasis; 23.8% were infected with S. haematobium and 9.3% were infected with S. mansoni. Moreover, 39.5% of the participants were anaemic whereas 9.5% had hepatosplenomegaly. The prevalence of schistosomiasis was significantly higher among children aged >10 years compared to those aged ≤ 10 years (P<0.05). Multivariate analysis confirmed that presence of other infected family member (P<0.001), low household monthly income (P = 0.003), using unsafe sources for drinking water (P = 0.003), living nearby stream/spring (P = 0.006) and living nearby pool/pond (P = 0.002) were the key factors significantly associated with schistosomiasis among these children. CONCLUSIONS/SIGNIFICANCE This study reveals that schistosomiasis is still highly prevalent in Yemen. These findings support an urgent need to start an integrated, targeted and effective schistosomiasis control programme with a mission to move towards the elimination phase. Besides periodic drug distribution, health education and community mobilisation, provision of clean and safe drinking water, introduction of proper sanitation are imperative among these communities in order to curtail the transmission and morbidity caused by schistosomiasis. Screening and treating other infected family members should also be adopted by the public health authorities in combating this infection in these communities

    Application of multi-criteria in the selection of running systems for regional use of tractors in agriculture

    The basis of the modern machine and tractor fleet of industrial and agricultural tractors are power vehicles with tracked and wheeled operating systems of various design designs. Both of these and other energy products have both their advantages and quite significant disadvantages. The article discusses the summary parametric and factorial characteristics of wheeled and tracked power vehicles used in field and transport work in the Amur region. The criteria for the formation of their traction properties, ways to reduce slipping, reduce the load of the machine-tractor unit on the soil, and the use of agricultural machinery during waterlogging are analysed. The results of a study on the use of mobile energy means in waterlogged conditions during the cultivation of agricultural crops, where their main advantages and disadvantages are reflected, the directions of choosing an energy means for use in agricultural production technologies are substantiated. It has been established that the use of tracked power vehicles with high traction properties, when aggregated with wide-reach machines in the fields of the region, will be more effective during periods of work when there is a large precipitation. At the same time, wheel energy means will also be in demand in the natural and climatic conditions of the Amur region as the most versatile structurally suitable means for performing most agricultural field operations as part of a tractor-transport unit. In this connection, the expediency of equipping the production and industrial complex and means of mechanisation of farms and agricultural enterprises of the region with energy means with various running systems in accordance with the used and scientifically based regional system of machines and technologies is justified
