Monitoring and the Risk Governance of Repository Development and Staged Closure:Exploratory Engagement Activity in Three European Countries.

    Get PDF
This report is the product of research activity within the EC Seventh Framework Programme “Monitoring Developments for Safe Repository Operation and Staged Closure” (MoDeRn) Project. This project aims to further develop understanding of the role of monitoring in staged implementation of geological disposal to a level of description that is closer to the actual implementation of monitoring. It focuses on monitoring conducted to confirm the basis of the long term safety case and on monitoring conducted to inform on options available to manage the stepwise disposal process from construction to closure (including e.g. the option of waste retrieval). This report investigates the potential of citizen stakeholder engagement in the identification of monitoring objectives and the development of monitoring strategies for geological disposal of high level waste (HLW) or spent nuclear fuel (SNF). It builds on an earlier MoDeRn report describing monitoring the safe disposal of radioactive waste as a socio-technical activity (Bergmans, Elam, Simmons and Sundqvist 2012)

Wood construction under cold climate:Part one: Impact of cold temperatures on the shear strength of different adhesives glued wood joints of Norway spruce and Scots pine

As wood constructions increasingly use engineered wood products worldwide, concerns arise about the integrity of the wood and adhesives system. The glueline stability is a crucial issue for engineered wood application, especially under cold climate. In this study, Norway spruce (Picea abies) and Scots pine (Pinus sylvestris) joints (150mm x 20mm x 10mm) were bonded with seven commercially available resins (PUR, PVAc, EPI, MF, MUF1, PRF and MUF2) and tested at six temperatures (20, -20, -30, -40, -50 and -60 °C), respectively. Generally, for both species, temperature changes significantly affected shear strength of wood joints. As temperature decreased, the shear strength decreased. PUR resin resulted in the strongest shear strength at all temperatures tested. MF resin responded to temperature changes in a similar ways as the PUR resin. The shear strength of wood joints with EPI resins was sensitive to temperature change. MUF, PRF and PVAc resins demonstrated different characters with Norway spruce and Scot pine. At room temperature, all types of adhesive showed relative stability, in terms of shear strength variation. While at low temperature, the shear strength varied considerably. More specimens need to be tested in further work to more completely present the issue. The EN 301 and EN 302 may need to be specified based on wood species.Godkänd; 2014; 20141112 (aliwan

Different induction mechanisms of mRNA for inducible nitric oxide synthase in rat smooth muscle cells in culture and in aortic strips

    Get PDF
AbstractThe expression of mRNA for the inducible form of nitric oxide synthase, (iNOS), was studied in rat aortic smooth muscle cells, (SMCs) in cell culture and in strips of rat aorta by reverse transcriptase coupled to the polymerase chain reaction. iNOS mRNA expression was weak in cultured SMCs when exposed to either interferon-Îł (IFNÎł) or lipopolysaccharide (LPS), but the combination LPS + IFNÎł enhanced the expression. In aortic strips LPS alone induced a pronounced expression, with no further increase by IFNÎł. Cycloheximide potentiated the expression of iNOS mRNA in SMCs in culture stimulated with LPS + IFNÎł but attenuated the response in aortic strips. The results indicate different cellular signaling pathways for the induction of iNOS mRNA by LPS and/or IFNÎł, in cultured SMCs and in rat aortic strips

Use of venous-thrombotic-embolic prophylaxis in patients undergoing surgery for renal tumors: a questionnaire survey in the Nordic countries (The NORENCA-2 study)

    Get PDF
Purpose: To examine the variation in venous thromboembolism prophylactic treatment (VTEP) among renal cancer patients undergoing surgery. Materials and methods: An Internet-based questionnaire on renal tumor management before and after surgery was mailed to all Nordic departments of urology. The questions focused on the use of VTEP and were subdivided into different surgical modalities. Results: Questionnaires were mailed to 91 institutions (response rate 53%). None of the centers used VTEP before surgery, unless the patient had a vena caval tumor thrombus. Overall, the VTEP utilized during hospitalization for patients undergoing renal surgery included early mobilization (45%), compression stockings (52%) and low-molecular-weight heparin (89%). In patients undergoing open radical Nx, 80% of institutions used VTEP during their hospitalization (23% compression stockings and 94% low-molecular-weight heparin). After leaving the hospital, the proportion and type of VTEP received varied considerably across institutions. The most common interval, used in 60% of the institutions, was for a period of 4 weeks. The restriction to the Nordic countries was a limitation and, therefore, may not reflect the practice patterns elsewhere. It is a survey study and, therefore, cannot measure the behaviors of those institutions that did not participate. Conclusion: We found variation in the type and duration of VTEP use for each type of local intervention for renal cancer. These widely disparate variations in care strongly argue for the establishment of national and international guidelines regarding VTEP in renal surgery.Peer reviewe

BMP-7 inhibits TGF-β-induced invasion of breast cancer cells through inhibition of integrin β3 expression

BACKGROUND The transforming growth factor (TGF)-β superfamily comprises cytokines such as TGF-β and Bone Morphogenetic Proteins (BMPs), which have a critical role in a multitude of biological processes. In breast cancer, high levels of TGF-β are associated with poor outcome, whereas inhibition of TGF-β-signaling reduces metastasis. In contrast, BMP-7 inhibits bone metastasis of breast cancer cells. METHODS In this study, we investigated the effect of BMP-7 on TGF-β-induced invasion in a 3 dimensional invasion assay. RESULTS BMP-7 inhibited TGF-β-induced invasion of the metastatic breast cancer cell line MCF10CA1a, but not of its premalignant precursor MCF10AT in a spheroid invasion model. The inhibitory effect appears to be specific for BMP-7, as its closest homolog, BMP-6, did not alter the invasion of MCF10CA1a spheroids. To elucidate the mechanism by which BMP-7 inhibits TGF-β-induced invasion, we analyzed invasion-related genes. BMP-7 inhibited TGF-β-induced expression of integrin ι(v)β(3) in the spheroids. Moreover, targeting of integrins by a chemical inhibitor or knockdown of integrin β(3) negatively affected TGF-β-induced invasion. On the other hand, overexpression of integrin β(3) counteracted the inhibitory effect of BMP7 on TGF-β-induced invasion. CONCLUSION Thus, BMP-7 may exert anti-invasive actions by inhibiting TGF-β-induced expression of integrin β(3).Prostatic carcinom

Genetic risk and a primary role for cell-mediated immune mechanisms in multiple sclerosis.

Multiple sclerosis is a common disease of the central nervous system in which the interplay between inflammatory and neurodegenerative processes typically results in intermittent neurological disturbance followed by progressive accumulation of disability. Epidemiological studies have shown that genetic factors are primarily responsible for the substantially increased frequency of the disease seen in the relatives of affected individuals, and systematic attempts to identify linkage in multiplex families have confirmed that variation within the major histocompatibility complex (MHC) exerts the greatest individual effect on risk. Modestly powered genome-wide association studies (GWAS) have enabled more than 20 additional risk loci to be identified and have shown that multiple variants exerting modest individual effects have a key role in disease susceptibility. Most of the genetic architecture underlying susceptibility to the disease remains to be defined and is anticipated to require the analysis of sample sizes that are beyond the numbers currently available to individual research groups. In a collaborative GWAS involving 9,772 cases of European descent collected by 23 research groups working in 15 different countries, we have replicated almost all of the previously suggested associations and identified at least a further 29 novel susceptibility loci. Within the MHC we have refined the identity of the HLA-DRB1 risk alleles and confirmed that variation in the HLA-A gene underlies the independent protective effect attributable to the class I region. Immunologically relevant genes are significantly overrepresented among those mapping close to the identified loci and particularly implicate T-helper-cell differentiation in the pathogenesis of multiple sclerosis

The participatory turn in radioactive waste management:Deliberation and the social-technical divide

    Get PDF
National policies for long-term management of radioactive waste have for decades been driven by technical experts. The pursuit of these technocratic policies led in many countries to conflict with affected communities. Since the late 1990s, however, there has been a turn to more participatory approaches. This participatory turn reflects widespread acknowledgement in the discourse of policy actors and implementing organisations of the importance of social aspects of radioactive waste management and the need to involve citizens and their representatives in the process. This appears to be an important move towards democratisation of this particular field of technological decision making but, despite these developments, technical aspects are still most often brought into the public arena only after technical experts have defined the ‘problem’ and decided upon a ‘solution’. This maintains a notional divide between the treatment of technical and social aspects of radioactive waste management and raises pressing questions about the kind of choice affected communities are given if they are not able to debate fully the technical options. The article aims to contribute to better understanding and addressing this situation by exploring the complex entanglement of the social and the technical in radioactive waste management policy and practice, analysing the contingent configurations that emerge as sociotechnical combinations. Drawing upon empirical examples from four countries that have taken the participatory turn - Belgium, Slovenia, Sweden and the United Kingdom – the article describes the different ways in which sociotechnical combinations have been constructed, and discusses their implications for future practice
