    A simple method to determine mating type in Neurospora discreta

    In order to circumvent the problem in assigning the mating types of some N. discreta strains by fluffy testers, a PCR based method was developed. Between the two reference N. discreta strains (FGSC 3228 A and FGSC 4378 a), we observed the false perithecia to be more abundant in the a mating type. When we analyzed the octads from a cross of these two, the false perithecia appear to be associated with a particular morphology and mating type

    Fabrication and investigation of superhydrophobic surface by dip coating

    60-66The present research work is focused on to fabricate and examine the characteristics of a superhydrophobic surface on copper (Cu) substrates by dip coating method. Three different samples of ‘Cu’ are set with chemical etching, mechanical rubbing and the combination of above two methods. The variations in the exteriorsurface morphologies have been examined by profilometer, scanning electron microscope and energy-dispersive X-ray spectroscopy. The adapted samples are immersed in a solution of silver nitrate follow by immersing in a combination of ethanol and perflurodecyltriethoxysilane for 10 min. The presence of leaf and dendrites assemblies have been observed on the surface, which are useful to trap the air between them and the entrapment allows water to roll off from the surface. The outcomes reveal that the modified substrates have a water contact angle (WCA) of 159° in the case of ‘Cu’ using combination of chemical etching and mechanical rubbing followed by immersion coating. The proposed methodology has the advantage of size compatibility and easy scale up for the development of superhydrophobic surface on copper in a cost and time effective manner

    A bioinspired optimization strategy: to minimize the travel segment of the nozzle to accelerate the fused deposition modeling process

    The fused deposition modeling process of digital printing uses a layer-by-layer approach to form a three-dimensional structure. Digital printing takes more time to fabricate a 3D model, and the speed varies depending on the type of 3D printer, material, geometric complexity, and process parameters. A shorter path for the extruder can speed up the printing process. However, the time taken for the extruder during printing (deposition) cannot be reduced, but the time taken for the extruder travel (idle move) can be reduced. In this study, the idle travel of the nozzle is optimized using a bioinspired technique called "ant colony optimization" (ACO) by reducing the travel transitions. The ACO algorithm determines the shortest path of the nozzle to reduce travel and generates the tool paths as G-codes. The proposed method’s G-code is implemented and compared with the G-code generated by the commercial slicer, Cura, in terms of build time. Experiments corroborate this finding: the G-code generated by the ACO algorithm accelerates the FDM process by reducing the travel movements of the nozzle, hence reducing the part build time (printing time) and increasing the strength of the printed object

    Differential actions of glycodelin-A on Th-1 and Th-2 cells: A paracrine mechanism that could produce the Th-2 dominant environment during pregnancy

    Background: The maternalfetal interface has a unique immunological response towards the implanting placenta. It is generally accepted that a T-helper type-2 (Th-2) cytokine prevailing environment is important in pregnancy. The proportion of Th-2 cells in the peripheral blood and decidua is significantly higher in pregnant women in the first trimester than in non-pregnant women. Glycodelin-A (GdA) is a major endocrine-regulated decidual glycoprotein thought to be related to fetomaternal defence. Yet the relationship between its immunoregulatory activities and the shift towards Th-2 cytokine profile during pregnancy is unclear. Methods GdA was immunoaffinity purified from human amniotic fluid. T-helper, T-helper type-1 (Th-1) and Th-2 cells were isolated from the peripheral blood. The viability of these cells was studied by XTT assay. Immunophenotyping of CD4/CD294, cell death and GdA-binding were determined by flow cytometry. The mRNA expression, surface expression and secretion of Fas/Fas ligand (FasL) were determined by quantitative polymerase chain reaction, flow cytometry and ELISA, respectively. The activities of caspase-3, -8 and -9 were measured. The phosphorylation of extracellular signal-regulated kinases (ERK), p38 and, c-Jun N-terminal kinase was determined by western blotting. Results Although GdA bound to both Th-1 and Th-2 cells, it had differential actions on the two cell-types. GdA induced cell death of the Th-1 cells but not the Th-2 cells. The cell death was mediated through activation of caspase -3, -8 and -9 activities. GdA up-regulated the expression of Fas and inhibited ERK activation in the Th-1 cells, which might enhance the vulnerability of the cells to cell death caused by a trophoblast-derived FasL. Conclusions The data suggest that GdA could be an endometrial factor that contributes to the Th-2/Th-1 shift during pregnancy. © 2011 The Author.postprin

    A Fragment of the LG3 Peptide of Endorepellin Is Present in the Urine of Physically Active Mining Workers: A Potential Marker of Physical Activity

    Biomarker analysis has been implemented in sports research in an attempt to monitor the effects of exertion and fatigue in athletes. This study proposed that while such biomarkers may be useful for monitoring injury risk in workers, proteomic approaches might also be utilised to identify novel exertion or injury markers. We found that urinary urea and cortisol levels were significantly elevated in mining workers following a 12 hour overnight shift. These levels failed to return to baseline over 24 h in the more active maintenance crew compared to truck drivers (operators) suggesting a lack of recovery between shifts. Use of a SELDI-TOF MS approach to detect novel exertion or injury markers revealed a spectral feature which was associated with workers in both work categories who were engaged in higher levels of physical activity. This feature was identified as the LG3 peptide, a C-terminal fragment of the anti-angiogenic/anti-tumourigenic protein endorepellin. This finding suggests that urinary LG3 peptide may be a biomarker of physical activity. It is also possible that the activity mediated release of LG3/endorepellin into the circulation may represent a biological mechanism for the known inverse association between physical activity and cancer risk/survival

    The rank reversal problem in multi-criteria decision making : a literature review

    Despite the importance of multicriteria decision-making (MCDM) techniques for constructing effective decision models, there are many criticisms due to the occurrence of a problem called rank reversal. Nevertheless, there is a lack of a systematic literature review on this important subject which involves different methods. This study reviews the pertinent literature on rank reversal, based on 130 related articles published from 1980 to 2015 in international journals, which were gathered and analyzed according to the following perspectives: multicriteria technique, year and journal in which the papers were published, co-authorship network, rank reversal types, and research goal. Thus our survey provides recommendations for future research, besides useful information and knowledge regarding rank reversal in the MCDM field
