Whole Genome Sequencing for Determining the Source of Mycobacterium bovis Infections in Livestock Herds and Wildlife in New Zealand.
The ability to DNA fingerprint Mycobacterium bovis isolates helped to define the role of wildlife in the persistence of bovine tuberculosis in New Zealand. DNA fingerprinting results currently help to guide wildlife control measures and also aid in tracing the source of infections that result from movement of livestock. During the last 5 years we have developed the ability to distinguish New Zealand (NZ) M. bovis isolates by comparing the sequences of whole genome sequenced (WGS) M. bovis samples. WGS provides much higher resolution than our other established typing methods and greatly improves the definition of the regional localization of NZ M. bovis types. Three outbreak investigations are described and results demonstrate how WGS analysis has led to the confirmation of epidemiological sourcing of infection, to better definition of new sources of infection by ruling out other possible sources, and has revealed probable wildlife infection in an area considered to be free of infected wildlife. The routine use of WGS analyses for sourcing new M. bovis infections will be an important component of the strategy employed to eradicate bovine TB from NZ livestock and wildlife
Effect of thyroid hormone therapy on fatigability in older adults with subclinical hypothyroidism: a nested study within a randomized placebo-controlled trial
Background:
Fatigue often triggers screening for and treatment of subclinical hypothyroidism. However, data on the impact of levothyroxine on fatigue is limited and previous studies might not have captured all aspects of fatigue.
Method:
This study is nested within the randomized, placebo-controlled, multicenter TRUST trial, including community-dwelling participants aged ≥65 and older, with persistent subclinical hypothyroidism (TSH 4.60–19.99 mIU/L, normal free thyroxine levels) from Switzerland and Ireland. Interventions consisted of daily levothyroxine starting with 50 μg (25 μg if weight <50 kg or known coronary heart diseases) together with dose adjustments to achieve a normal TSH and mock titration in the placebo group. Main outcome was the change in physical and mental fatigability using the Pittsburgh Fatigability Scale over 1 year, assessed through multivariable linear regression with adjustment for country, sex, and levothyroxine starting dose.
Results:
Among 230 participants, the mean ± standard deviation (SD) TSH was 6.2 ± 1.9 mIU/L at baseline and decreased to 3.1 ± 1.3 with LT4 (n = 119) versus 5.3 ± 2.3 with placebo (n = 111, p < .001) after 1 year. After adjustment we found no between-group difference at 1 year on perceived physical (0.2; 95% CI −1.8 to 2.1; p = .88), or mental fatigability (−1.0; 95% CI −2.8 to 0.8; p = .26). In participants with higher fatigability at baseline (≥15 points for the physical score [n = 88] or ≥13 points for the mental score [n = 41]), the adjusted between-group differences at 1 year were 0.4 (95% CI −3.6 to 2.8, p = .79) and −2.2 (95% CI −8.8 to 4.5, p = .51).
Conclusions:
Levothyroxine in older adults with mild subclinical hypothyroidism provides no change in physical or mental fatigability
Fuelling the nuclear ring of NGC 1097
Galactic bars can drive cold gas inflows towards the centres of galaxies. The
gas transport happens primarily through the so-called bar ``dust lanes'', which
connect the galactic disc at kpc scales to the nuclear rings at hundreds of pc
scales much like two gigantic galactic rivers. Once in the ring, the gas can
fuel star formation activity, galactic outflows, and central supermassive black
holes. Measuring the mass inflow rates is therefore important to understanding
the mass/energy budget and evolution of galactic nuclei. In this work, we use
CO datacubes from the PHANGS-ALMA survey and a simple geometrical method to
measure the bar-driven mass inflow rate onto the nuclear ring of the barred
galaxy NGC~1097. The method assumes that the gas velocity in the bar lanes is
parallel to the lanes in the frame co-rotating with the bar, and allows one to
derive the inflow rates from sufficiently sensitive and resolved
position-position-velocity diagrams if the bar pattern speed and galaxy
orientations are known. We find an inflow rate of averaged over a time span of 40 Myr, which varies by a
factor of a few over timescales of 10 Myr. Most of the inflow appears to
be consumed by star formation in the ring which is currently occurring at a
rate of -, suggesting that the
inflow is causally controlling the star formation rate in the ring as a

Kinematic analysis of the super-extended HI disk of the nearby spiral galaxy M 83

We study the super-extended HI disk of M83 (∼50 kpc in radius), in particular disc kinematics, rotation and the turbulent nature of the atomic interstellar medium. We define distinct regions in the outer disk (rgal > central optical disk), including ring, southern area, and southern and northern arm. We examine HI gas surface density, velocity dispersion and non-circular motions in the outskirts, which we compare to the inner optical disk. We find an increase of velocity dispersion (σv) towards the pronounced HI ring, indicative of more turbulent HI gas. Additionally, we report over a large galactocentric radius range (until rgal ∼ 50 kpc) that σv is slightly larger than thermal (i.e. > 8 km s-1). We find that a higher star formation rate (as traced by FUV emission) is not always necessarily associated with a higher HI velocity dispersion, suggesting that radial transport could be a dominant driver for the enhanced velocity dispersion. We further find a possible branch that connects the extended HI disk to the dwarf irregular galaxy UGCA365, that deviates from the general direction of the northern arm. Lastly, we compare mass flow rate profiles (based on 2D and 3D tilted ring models) and find evidence for outflowing gas at rgal ∼ 2 kpc, inflowing gas at rgal ~ 5.5 kpc and outflowing gas at rgal ~ 14 kpc. We caution that mass flow rates are highly sensitive to the assumed kinematic disk parameters, in particular, to the inclination.Publisher PDFPeer reviewe

PHANGS-JWST First Results: Spurring on Star Formation: JWST Reveals Localized Star Formation in a Spiral Arm Spur of NGC 628
We combine JWST observations with Atacama Large Millimeter/submillimeter Array CO and Very Large Telescope MUSE Hα data to examine off-spiral arm star formation in the face-on, grand-design spiral galaxy NGC 628. We focus on the northern spiral arm, around a galactocentric radius of 3-4 kpc, and study two spurs. These form an interesting contrast, as one is CO-rich and one CO-poor, and they have a maximum azimuthal offset in MIRI 21 μm and MUSE Hα of around 40° (CO-rich) and 55° (CO-poor) from the spiral arm. The star formation rate is higher in the regions of the spurs near spiral arms, but the star formation efficiency appears relatively constant. Given the spiral pattern speed and rotation curve of this galaxy and assuming material exiting the arms undergoes purely circular motion, these offsets would be reached in 100-150 Myr, significantly longer than the 21 μm and Hα star formation timescales (both < 10 Myr). The invariance of the star formation efficiency in the spurs versus the spiral arms indicates massive star formation is not only triggered in spiral arms, and cannot simply occur in the arms and then drift away from the wave pattern. These early JWST results show that in situ star formation likely occurs in the spurs, and that the observed young stars are not simply the “leftovers” of stellar birth in the spiral arms. The excellent physical resolution and sensitivity that JWST can attain in nearby galaxies will well resolve individual star-forming regions and help us to better understand the earliest phases of star formation
Intravascular Food Reward
Consumption of calorie-containing sugars elicits appetitive behavioral responses and dopamine release in the ventral striatum, even in the absence of sweet-taste transduction machinery. However, it is unclear if such reward-related postingestive effects reflect preabsorptive or postabsorptive events. In support of the importance of postabsorptive glucose detection, we found that, in rat behavioral tests, high concentration glucose solutions administered in the jugular vein were sufficient to condition a side-bias. Additionally, a lower concentration glucose solution conditioned robust behavioral responses when administered in the hepatic-portal, but not the jugular vein. Furthermore, enteric administration of glucose at a concentration that is sufficient to elicit behavioral conditioning resulted in a glycemic profile similar to that observed after administration of the low concentration glucose solution in the hepatic-portal, but not jugular vein. Finally using fast-scan cyclic voltammetry we found that, in accordance with behavioral findings, a low concentration glucose solution caused an increase in spontaneous dopamine release events in the nucleus accumbens shell when administered in the hepatic-portal, but not the jugular vein. These findings demonstrate that the postabsorptive effects of glucose are sufficient for the postingestive behavioral and dopaminergic reward-related responses that result from sugar consumption. Furthermore, glycemia levels in the hepatic-portal venous system contribute more significantly for this effect than systemic glycemia, arguing for the participation of an intra-abdominal visceral sensor for glucose
A united statement of the global chiropractic research community against the pseudoscientific claim that chiropractic care boosts immunity.
BACKGROUND: In the midst of the coronavirus pandemic, the International Chiropractors Association (ICA) posted reports claiming that chiropractic care can impact the immune system. These claims clash with recommendations from the World Health Organization and World Federation of Chiropractic. We discuss the scientific validity of the claims made in these ICA reports. MAIN BODY: We reviewed the two reports posted by the ICA on their website on March 20 and March 28, 2020. We explored the method used to develop the claim that chiropractic adjustments impact the immune system and discuss the scientific merit of that claim. We provide a response to the ICA reports and explain why this claim lacks scientific credibility and is dangerous to the public. More than 150 researchers from 11 countries reviewed and endorsed our response. CONCLUSION: In their reports, the ICA provided no valid clinical scientific evidence that chiropractic care can impact the immune system. We call on regulatory authorities and professional leaders to take robust political and regulatory action against those claiming that chiropractic adjustments have a clinical impact on the immune system
