Novel PI3Kγ Mutation in a 44-Year-Old Man with Chronic Infections and Chronic Pelvic Pain
A 44-year-old man is presented here with 14 years of chronic purulent sinusitis, a chronic fungal rash of the scrotum, and chronic pelvic pain. Treatment with antifungal therapy resulted in symptom improvement, however he was unable to establish an effective long-term treatment regimen, resulting in debilitating symptoms. He had undergone extensive work-up without identifying a clear underlying etiology, although Candida species were cultured from the prostatic fluid. 100 genes involved in the cellular immune response were sequenced and a missense mutation was identified in the Ras-binding domain of PI3Kγ. PI3Kγ is a crucial signaling element in leukotaxis and other leukocyte functions. We hypothesize that his mutation led to his chronic infections and pelvic pain

The Ca and Mg isotope record of the Cryogenian Trezona carbon isotope excursion

JAH acknowledges support from NSF (IES-1410317) and from NSF OCE CAREER Grant (1654571).The Trezona carbon isotope excursion is recorded on five different continents in platform carbonates deposited prior to the end-Cryogenian Marinoan glaciation (>635 Ma) and represents a change in carbon isotope values of 16–18‰. Based on the spatial and temporal reproducibility, the excursion previously has been interpreted as tracking the carbon isotopic composition of dissolved inorganic carbon in the global ocean before the descent into a snowball Earth. However, in modern restricted shallow marine and freshwater settings, carbon isotope values have a similarly large range, which is mostly independent from open ocean chemistry and instead reflects local processes. In this study, we combine calcium, magnesium, and strontium isotope geochemistry with a numerical model of carbonate diagenesis to disentangle the degree to which the Trezona excursion reflects changes in global seawater chemistry versus local shallow-water platform environments. Our analysis demonstrates that the most extreme carbon isotope values (∼-10‰ versus +10‰) are preserved in former platform aragonite that was neomorphosed to calcite during sediment-buffered conditions and record the primary carbon isotope composition of platform-top surface waters. In contrast, the downturn and recovery of the Trezona excursion are recorded in carbonates that were altered during early fluid-buffered diagenesis and commonly are dolomitized. We also find that the nadir of the Trezona excursion is associated with a fractional increase in siliciclastic sediments, whereas the recovery from the excursion correlates with a relative increase in carbonate. This relationship suggests that the extreme negative isotopic shift in platform aragonite occurred in concert with periods of increased input of siliciclastic sediments, changes in water depth, and possibly nutrients to platform environments. Although the process for generating extremely negative carbon isotope values in Neoproterozoic platform carbonates remains enigmatic, we speculate that these excursions reflect kinetic isotope effects associated with CO2 invasion in platform waters during periods of intense primary productivity.Publisher PDFPeer reviewe
Sterile inflammation as a factor in human male infertility: Involvement of Toll like receptor 2, biglycan and peritubular cells
Changes in the wall of seminiferous tubules in men with impaired spermatogenesis imply sterile inflammation of the testis. We tested the hypothesis that the cells forming the wall of seminiferous tubules, human testicular peritubular cells (HTPCs), orchestrate inflammatory events and that Toll like receptors (TLRs) and danger signals from the extracellular matrix (ECM) of this wall are involved. In cultured HTPCs we detected TLRs, including TLR2. A TLR-2 ligand (PAM) augmented interleukin 6 (IL-6), monocyte chemo-attractant protein-1 (MCP-1) and pentraxin 3 (PTX3) in HTPCs. The ECM-derived proteoglycan biglycan (BGN) is secreted by HTPCs and may be a TLR2-ligand at HTPCs. In support, recombinant human BGN increased PTX3, MCP-1 and IL-6 in HTPCs. Variable endogenous BGN levels in HTPCs derived from different men and differences in BGN levels in the tubular wall in infertile men were observed. In testes of a systemic mouse model for male infertility, testicular sterile inflammation and elevated estradiol (E2) levels, BGN was also elevated. Hence we studied the role of E2 in HTPCs and observed that E2 elevated the levels of BGN. The anti-estrogen ICI 182,780 blocked this action. We conclude that TLR2 and BGN contribute to sterile inflammation and infertility in man

Spectroscopic Target Selection in the Sloan Digital Sky Survey: The Quasar Sample
We describe the algorithm for selecting quasar candidates for optical
spectroscopy in the Sloan Digital Sky Survey. Quasar candidates are selected
via their non-stellar colors in "ugriz" broad-band photometry, and by matching
unresolved sources to the FIRST radio catalogs. The automated algorithm is
sensitive to quasars at all redshifts lower than z=5.8. Extended sources are
also targeted as low-redshift quasar candidates in order to investigate the
evolution of Active Galactic Nuclei (AGN) at the faint end of the luminosity
function. Nearly 95% of previously known quasars are recovered (based on 1540
quasars in 446 square degrees). The overall completeness, estimated from
simulated quasars, is expected to be over 90%, whereas the overall efficiency
(quasars:quasar candidates) is better than 65%. The selection algorithm targets
ultraviolet excess quasars to i^*=19.1 and higher-redshift (z>3) quasars to
i^*=20.2, yielding approximately 18 candidates per square degree. In addition
to selecting ``normal'' quasars, the design of the algorithm makes it sensitive
to atypical AGN such as Broad Absorption Line quasars and heavily reddened

Gemini Near Infrared Spectrograph -- Distant Quasar Survey: Augmented Spectroscopic Catalog and a Prescription for Correcting UV-Based Quasar Redshifts
Quasars at most often have redshifts measured from rest-frame
ultraviolet emission lines. One of the most common such lines, C IV
, shows blueshifts up to , and in
rare cases even higher. This blueshifting results in highly uncertain redshifts
when compared to redshift determinations from rest-frame optical emission
lines, e.g., from the narrow [O III] feature. We present
spectroscopic measurements for 260 sources at
having
mag from the Gemini Near Infrared
Spectrograph - Distant Quasar Survey (GNIRS-DQS) catalog, augmenting the
previous iteration which contained 226 of the 260 sources whose measurements
are improved upon in this work. We obtain reliable systemic redshifts based on
[O III] for a subset of 121 sources which we use to calibrate
prescriptions for correcting UV-based redshifts. These prescriptions are based
on a regression analysis involving C IV full-width-at-half-maximum intensity
and equivalent width, along with the UV continuum luminosity at a rest-frame
wavelength of 1350 A. Applying these corrections can improve the accuracy and
the precision in the C IV-based redshift by up to
and , respectively, which correspond to
Mpc and Mpc in comoving distance at . Our prescriptions
also improve the accuracy of the best available multi-feature redshift
determination algorithm by , indicating that the
spectroscopic properties of the C IV emission line can provide robust redshift
estimates for high-redshift quasars. We discuss the prospects of our
prescriptions for cosmological and quasar studies utilizing upcoming large

The Paradox of Power in CSR: A Case Study on Implementation
Purpose Although current literature assumes positive outcomes for stakeholders resulting from an increase in power associated with CSR, this research suggests that this increase can lead to conflict within organizations, resulting in almost complete inactivity on CSR.
Methods A single in-depth case study, focusing on power as an embedded concept.
Results Empirical evidence is used to demonstrate how some actors use CSR to improve their own positions within an organization. Resource dependence theory is used to highlight why this may be a more significant concern for CSR.
Conclusions Increasing power for CSR has the potential to offer actors associated with it increased personal power, and thus can attract opportunistic actors with little interest in realizing the benefits of CSR for the company and its stakeholders. Thus power can be an impediment to furthering CSR strategy and activities at the individual and organizational level

The clustering of galaxies in the SDSS-III Baryon Oscillation Spectroscopic Survey : measuring DA and H at z = 0.57 from the baryon acoustic peak in the Data Release 9 spectroscopic Galaxy sample
We present measurements of the angular diameter distance to and Hubble parameter at z = 0.57 from the measurement of the baryon acoustic peak in the correlation of galaxies from the Sloan Digital Sky Survey III Baryon Oscillation Spectroscopic Survey. Our analysis is based on a sample from Data Release 9 of 264 283 galaxies over 3275 square degrees in the redshift range 0.43 < z < 0.70. We use two different methods to provide robust measurement of the acoustic peak position across and along the line of sight in order to measure the cosmological distance scale. We find DA(0.57) = 1408 ± 45 Mpc and H(0.57) = 92.9 ± 7.8 km s−1 Mpc−1 for our fiducial value of the sound horizon. These results from the anisotropic fitting are fully consistent with the analysis of the spherically averaged acoustic peak position presented in Anderson et al. Our distance measurements are a close match to the predictions of the standard cosmological model featuring a cosmological constant and zero spatial curvature.Publisher PDFPeer reviewe
