    Histological and Morphometric Examination of the Testes of Boars and Male Pigs Immunocastrated with Improvac®

    Background: Surgical castration of male piglets is frequently performed to prevent development of an unpleasant taste and flavor in meat of mature males, called boar taint. Disadvantages of this method are primarily related to the lower feed efficiency of surgically castrated male pigs, production of less lean meat and more fat on the carcasses than boars, as well as ethical issues associated with the procedure itself. Immunocastration is a method of immunization against gonadotropinreleasing hormone (GnRH) and represents an alternative to avoid boar taint. This study aimed to examine the influence of immunocastration on histological and morphometric parameters of male pig testes.Materials, Methods & Results: A total of 60 crossbred male pigs selected from 30 litters were assigned to two experimental groups: boars (n = 30) and immunocastrated males (n = 30). Two doses (2 mL per dose) of the Improvac® vaccine were given to pigs scheduled for immunocastration. The first injection was given at 10 weeks of age, and the second injection at 20 weeks of age. All pigs were fed the same commercial diet ad libitum, group-housed (15 pigs/pen) and slaughtered at 25 weeks of age. After slaughter, testicle weights were measured and samples of testicular tissue were taken for histological examination. Immunocastrated males had lower testicle weights than boars (P < 0.01). Furthermore, structural damage to the germinal epithelium was visible in the seminiferous tubules of vaccinated pigs. In the testicular interstitium of these animals, the number of Leydig cells were reduced; almost all of these cells exhibited different degrees of abnormality. Moreover, the diameter of the seminiferous tubules and thickness of the germinal epithelium were greater (P < 0.01), and the percentage of germinal epithelium in the seminiferous tubules as well as percentage of blood vessels in the interstitium were higher in boars as compared to immunocastrated males (P < 0.01). Seminiferous tubule lumen diameter was greater (P < 0.01), and percentage of lumen in the seminiferous tubules as well as percentage of non-blood vessel tissue in the interstitium were higher in immunocastrated males than boars (P < 0.01).Discussion: Vaccination against GnRH, as used in this study, reduced the synthesis of follicle-stimulating hormone (FSH) and luteinizing hormone (LH), which led to decreased spermatogenesis and androgen production. The smaller diameter of the seminiferous tubules as well as germinal epithelium thickness, and lower percentage of germinal epithelium in the seminiferous tubules in immunocastrated males could be explained by reduced FSH synthesis, and subsequent reduced spermatogenesis. The described testicular malformations in immunocastrated animals caused their lower weight, and greater diameter of the lumen as well as a higher lumen percentage of the seminiferous tubules. The vaccine reduced LH synthesis, so the Leydig cells of immunocastrated animals had lower activity than the same cells in boars. The percentage of blood vessels in the interstitium of boar testes was higher, which was related to the blood supply required for normal function of Leydig cells. It was concluded that differences existed in the structure of the seminiferous tubules and interstitium between boars and immunocastrated males , which were reflected in values of the examined histological and morphometric parameters. The development of these differences was related to the inhibition of GnRH synthesis and secretion

    Cut Length Distributions of Haylage Particles

    Alfalfa is one of the most important crops for forage production. Traditional method of alfalfa conservation assumes hay preparation. However, nowadays it is also commonly processed in the form of silage and haylage. Physiological effects of forages that are included in diets depend on plant species, stage of maturity, method of preservation and diet composition. Physical characteristics of rations for ruminants are primarily influenced by dietary forage to concentrate ratio, type of forages and concentrates, and mean particle size of feeds. Length distribution of forage particles represents an important parameter for ruminant’s diet formulation, especially for dairy cattle. During silage production, harvest considerations should be focused to obtaining the adequate particle size distribution of the ensiling crop particles. This paper presents results of testing three contemporary types of self-propelled silage harvesters applied in the alfalfa haylage preparation: Claas Jaguar 950, Krone Big X 700 and Krone Big X 500. All machines were adapted with pick-up headers. In the study are analyzed length distributions of chopped alfalfa particles. Resulting frequency distributions of produced haylage are characterised by high mass percentage of the fraction comprehending the largest particles. It is also evident that harvester Class Jaguar 950 achieved the mean chopping length closest to preset value

    THE DYEING PROCESS OF KNITTED FABRICS AT DIFFERENT TEMPERATURES USING ULTRASOUND

    The dyeing of knitted fabrics made from 100 % cellulose using on-line procedure vinyl sulfonic reactive dye, with or without ultrasound energy, is carried out in this paper. The impact of temperature has been observed. The dye exhaustion is monitored using the method of absorption spectrophotometry, and the quality control of the coloration is monitored using color measurements. The acting of ultrasound on coloration consistency, as well as on some mechanical characteristics has also been examined. All examples of the ultrasound dyeing process show greater dye exhaustion in comparison to the conventional procedure. In addition, all the samples, which have been dyed with the ultrasound energy at 40°C, are significantly darker and have deeper color in comparison with the referent sample. The temperature has a great influence on kinetic energy of the dye molecules, and therefore on the diffusion processes in the dyeing system. The exhaustion chart indicates that when the temperature is lower the exhaustion degree drops. However, all the samples dyed with the ultrasound energy have bigger exhaustion. Besides that, ultrasound energy contributes to warming up the processing environment, so the additional warm up with the electricity is unnecessary, unlike the conventional way of dyeing. Since the reactive dyes chemically connect themselves with the cellulose substrate and in that way form covalent connection, the dyed fabrics have good washing consistency. Analysis results indicate that the consistencies are identical regardless the applied dyeing procedure. In other words, the dyeing method using the ultrasound energy produces the dyed fabric of the same quality. After analyzing the results of breaking force and elongation at break of knitted fabrics, it is noticeable that there is no degradation of previously mentioned knitted fabrics features (horizontally and vertically) during the ultrasound wave’s activity

    DYEING OF KNITTED MICRO-VISCOSE IN THE PRESENCE OF ULTRASOUND WITH DIFFERENT FREQUENCIES

    In dyeing process, the object is to transport or diffuse dyes and chemicals into the fibre. Various novel processes, including ultrasound, are being introduced and studied as more environmentally friendly alternatives. Encouraging results have been reported for the use of ultrasound energy in dyeing processes of micro-viscose. The recent studies revealed major ultrasound applications advances: savings of processing time, energy, chemicals, as well as environmental protection. Influence of various ultrasound frequencies (40, 200 and 400 kHz) on dyeing of micro-viscose knitted fabrics, by a reactive dye has been reported in this work. A method of reflection spectrophotometry has been employed to record reemission curves of the colored compounds. A software packet has been employed to calculate CIELab colored coordinates. Then, a comparison has been made with samples colored by conventional procedure according to CIELab76 and CMC (2:1) criteria. The use ultrasound in textile dyeing processing offers many potential advantages. The results prove better dye exhaustion by ultrasound and consequently the better fixing. The exhaustion for the bifunctional dye (containing two vinylsulphone groups) reaches 71.75 % without ultrasound, and 83.69 % with 400 kHz ultrasound. The 40 kHz, 150 W ultrasound causes a cavitation of higher intensity, compared to 200 and 400 kHz ultrasounds. In this particular case, destruction of cavitation bubbles is very intensive. That is why a large amount of cavitation energy is being transformed into a heat, yielding the additional bath heating. The ultrasounds with higher frequencies (200 and 400 kHz) cannot use such a strong power. The applied powering this case reaches 0.6 W. The cavitation bubbles are now smaller the cavitation disintegration is not so strong, and the energy loss is much smaller, i.e. a smaller amount of energy has been transformed into a heat. An ultrasound of an equal power, but of higher frequency contributes to the somewhat higher exhaustion and fixing. The ultrasound dyeing produces much obscured colours, compared the standard. The differences are evident and not negligible. The comparison of the samples treated ultrasound of different frequencies during dyeing revealed the higher coulours intensities with the increase of ultrasound frequencies of the equal power (200 and 400 kHz). However, the increase is not so expressed

    THE EFFECTS OF AN ERGONOMIC EXERCISE PROGRAM WITH A PILATES BALL ON REDUCING THE RISK OF THE INCIDENCE OF MUSCULOSKELETAL DISORDERS

    Introduction: Very few scientists have studied the influence of rehabilitation exercises on lowering the risks of muscular-skeletal disorder incidence (MSD). Therefore, it is necessary to study the influence of these exercise programmes on the prevention of and decrease in the risk of MSD development among schoolchildren. A special programme is based on the education of schoolchildren on ergonomic risks and their prevention by means of specific exercises with a Pilates ball for each recognized MSD, with the assumption that its sixteen-week application will significantly influence the risk of MSD incidence. Methods: The overall sample consisted of 55 students whose chronological age was 11, of both sexes, which was divided into two sub-samples: the experimental group (EG) of 28 students and the control group (CG) of 27 students. The Spinal Mouse with the appropriate programme support was used for the evaluation of muscular-skeletal disorders of the spine in the sagittal and frontal parts (kyphosis, lordosis, thoracic scoliosis). Results and Discussion: The results, following the experimental programme, have shown significantly lowered values of the kyphotic and thoracic scoliotic curve in the experimental group (EG), whereas the condition of the control group (CG) deteriorated, however not with the statistical significance. As for lordosis, there were no significant changes in either group during the experimental treatment. A variance analysis showed that exercises with a Pilates ball programme had statistically significant effects on decreasing MSD with kyphotic and thoracic scoliotic curves but with no significant effects on lordosis as compared to the school exercise programme. Conclusion: The conclusion drawn from the analysis of the effects of the specially defined ergonomic programme, in the form of specific exercises with a Pilates ball, is that it leads to the decreased risk of MSD incidence among fourth-grade primary school students. Applied experimental actions had a positive influence on lowering the spine MSD in the case of the experimental group, but that there were no significant changes in the case of the control group

    Effects of Acute Administration of D,L-Homocysteine Thiolactone on the Antioxidative Status of Rat Intestine and Liver

    Oxidative stress appears to play a role in the pathogenesis of several inflammatory gastrointestinal diseases. Increased homocysteine levels may play a role in the pathogenesis of Chrons disease and ulcerative colitis. The aim of this study was to examine the influence of homocysteine on the antioxidant status of rat intestine and liver. The levels of thiobarbituric acid reactive substances (TBARS), activity of catalase (CAT) and total antioxidant status (TAS) were investigated in the isolated gut and liver of young male rats in the control group (8 rats) and after 3-hour incubation in high doses of D, L-homocysteine thionolactone (Hcy) (10 mu mol/L) (8 rats). Samples of duodenum, ileum, colon and liver were homogenized in sodium phosphate buffer (1: 10). Homogenates were centrifuged at 10000 for 10 min at 4 degrees C and the supernatant was taken for biochemical assays. Our results showed that high D, L-homocysteine thionolactone concentration reduced enzymatic catalase activity in homogenates of the isolated segments of duodenum (27.04%) p LT 0.01; ileum (37.27%), colon (34.17%) and liver (67.46%) p LT 0.001. Exposition to high D, L-homocysteine thiolactone concentration significantly increased TBARS levels in the duodenum (106.05%), ileum (47.24%), colon (112.75%) and liver (32.07%) (p LT 0.01). Homocysteine also modified the total antioxidant status of homogenates from the duodenum, ileum, colon and liver, increasing by 20.68% (duodenum), 24.74% (ileum), 14.88% (colon) and 19.35% (liver) (p LT 0.001). Homocysteine induced a consistent oxidative stress in rats intestine and liver (reduced activity of catalase and increased level of TBARS), but the elevated activity of TAS in our experiments could be explained as an adaptive response to the generated free radicals which indicates the failure of the total antioxidant defense mechanism to protect the tissues from damage caused by homocysteine
