The centenary of the beginning of the First World War brought forth a steady stream of commemorations and publications. Standing out was Christopher Clark’s The Sleepwalkers, a global bestseller, whose controversial reinterpretation of the crisis of July 1914 and of the responsibility for its violent outcome ignited a widespread academic and public debate.1 Overshadowed by the anniversary of the Great War’s outbreak was the bicentenary of another great war’s conclusion, two hundred years after the end of the Napoleonic wars. In this relatively subdued realm, most of the attention was devoted to the final instance of armed conflict, with official commemorations of, books about, large-scale reenactments of, and multimedia exhibitions displaying the Battle of Waterloo

    A discourse analysis of trainee teacher identity in online discussion forums

    Teacher education involves an identity transformation for trainees from being a student to being a teacher. This discourse analysis examined the online discussion board communications of a cohort of trainee teachers to better understand the situated identities of the trainees and how they were presented online. Their discussion board posts were the primary method of communication during placement periods and, as such, provided insight into how the trainees situated their identities in terms of being a student or being a teacher. During the analysis, the community boundaries, language and culture were explored along with the tutor's power and role in the identity transformation process. This involved looking at the lexis used by the students, the use of pronouns to refer to themselves and others such as teachers and pupils, the types of messages allowed in the community and the effect of the tutor's messages on their communication. The research found that the trainees felt comfortable with teaching but did not feel like teachers during the course. Tutors and school teachers need to develop an awareness of the dual nature of trainees' identities and help promote the transition from student to teacher. In the beginning of the course, trainees should be familiarised with teacher vocabulary and practical concepts in addition to pedagogical theory. Towards the end of the course, trainee identity as teachers could be promoted through the use of authentic assessments that mirror real teacher tasks and requirements

    Comparative effectiveness of less commonly used systemic monotherapies and common combination therapies for moderate to severe psoriasis in the clinical setting.

    BACKGROUND: The effectiveness of psoriasis therapies in real-world settings remains relatively unknown. OBJECTIVE: We sought to compare the effectiveness of less commonly used systemic therapies and commonly used combination therapies for psoriasis. METHODS: This was a multicenter cross-sectional study of 203 patients with plaque psoriasis receiving less common systemic monotherapy (acitretin, cyclosporine, or infliximab) or common combination therapies (adalimumab, etanercept, or infliximab and methotrexate) compared with 168 patients receiving methotrexate evaluated at 1 of 10 US outpatient dermatology sites participating in the Dermatology Clinical Effectiveness Research Network. RESULTS: In adjusted analyses, patients on acitretin (relative response rate 2.01; 95% confidence interval [CI] 1.18-3.41), infliximab (relative response rate 1.93; 95% CI 1.26-2.98), adalimumab and methotrexate (relative response rate 3.04; 95% CI 2.12-4.36), etanercept and methotrexate (relative response rate 2.22; 95% CI 1.25-3.94), and infliximab and methotrexate (relative response rate 1.72; 95% CI 1.10-2.70) were more likely to have clear or almost clear skin compared with patients on methotrexate. There were no differences among treatments when response rate was defined by health-related quality of life. LIMITATIONS: Single time point assessment may result in overestimation of effectiveness. CONCLUSIONS: The efficacy of therapies in clinical trials may overestimate their effectiveness as used in clinical practice. Although physician-reported relative response rates were different among therapies, absolute differences were small and did not correspond to differences in patient-reported outcomes

    Subseasonal variability associated with Asian summer monsoon simulated by 14 IPCC AR4 coupled GCMs

    This study evaluates the subseasonal variability associated with the Asian summer monsoon in 14 coupled general circulation models (GCMs) participating in the Intergovernmental Panel on Climate Change (IPCC) Fourth Assessment Report (AR4). Eight years of each model's twentieth-century climate simulation are analyzed. The authors focus on the three major components of Asian summer monsoon: the Indian summer monsoon (ISM), the western North Pacific summer monsoon (WNPSM), and the East Asian summer monsoon (EASM), together with the two dominant subseasonal modes: the eastward- and northward-propagating boreal summer intraseasonal oscillation (BSIO) and the westward-propagating 12-24-day mode. The results show that current state-of-the-art GCMs still have difficulties and display a wide range of skill in simulating the subseasonal variability associated with Asian summer monsoon. During boreal summer (May-October), most of the models produce reasonable seasonal-mean precipitation over the ISM region, but excessive precipitation over the WNPSM region and insufficient precipitation over the EASM region. In other words, models concentrate their rain too close to the equator in the western Pacific. Most of the models simulate overly weak total subseasonal (2-128 day) variance, as well as too little variance for BSIO and the 12-24-day mode. Only 4-5 models produce spectral peaks in the BSIO and 12-24-day frequency bands; instead, most of the models display too red a spectrum, that is, an overly strong persistence of precipitation. For the seven models with three-dimensional data available, five reproduce the preconditioning of moisture in BSIO but often with a too late starting time, and only three simulate the phase lead of low-level convergence. Interestingly, although models often have difficulty in simulating the eastward propagation of BSIO, they tend to simulate well the northward propagation of BSIO, together with the westward propagation of the 12-24-day mode. The northward propagation in these models is thus not simply a NW-SE-tilted tail protruding off of an eastward-moving deep-tropical intraseasonal oscillation.open444

    North American monsoon and convectively coupled equatorial waves simulated by IPCC AR4 coupled GCMs

    This study evaluates the fidelity of North American monsoon and associated intraseasonal variability in the Intergovernmental Panel on Climate Change (IPCC) Fourth Assessment Report (AR4) coupled general circulation models (CGCMs). Twenty years of monthly precipitation data from each of the 22 models' twentieth-century climate simulations, together with the available daily precipitation data from 12 of them, are analyzed and compared with Global Precipitation Climatology Project (GPCP) monthly and daily precipitation. The authors focus on the seasonal cycle and horizontal pattern of monsoon precipitation in conjunction with the two dominant convectively coupled equatorial wave modes: the eastward-propagating Madden-Julian oscillation (MJO) and the westward-propagating easterly waves. The results show that the IPCC AR4 CGCMs have significant problems and display a wide range of skill in simulating the North American monsoon and associated intraseasonal variability. Most of the models reproduce the monsoon rainbelt, extending from southeast to northwest, and its gradual northward shift in early summer, but overestimate the precipitation over the core monsoon region throughout the seasonal cycle and fail to reproduce the monsoon retreat in the fall. Additionally, most models simulate good westward propagation of the easterly waves, but relatively poor eastward propagation of the MJO and overly weak variances for both the easterly waves and the MJO. There is a tendency for models without undiluted updrafts in their deep convection scheme to produce better MJO propagation.open221

    Phosphoprotein Associated with Glycosphingolipid-Enriched Microdomains Differentially Modulates Src Kinase Activity in Brain Maturation

    Src family kinases (SFK) control multiple processes during brain development and function. We show here that the phosphoprotein associated with glycosphigolipid-enriched microdomains (PAG)/Csk binding protein (Cbp) modulates SFK activity in the brain. The timing and localization of PAG expression overlap with Fyn and Src, both of which we find associated to PAG. We demonstrate in newborn (P1) mice that PAG negatively regulates Src family kinases (SFK). P1 Pag1-/- mouse brains show decreased recruitment of Csk into lipid rafts, reduced phosphorylation of the inhibitory tyrosines within SFKs, and an increase in SFK activity of >/ = 50%. While in brain of P1 mice, PAG and Csk are highly and ubiquitously expressed, little Csk is found in adult brain suggesting altered modes of SFK regulation. In adult brain Pag1-deficiency has no effect upon Csk-distribution or inhibitory tyrosine phosphorylation, but kinase activity is now reduced (−20–30%), pointing to the development of a compensatory mechanism that may involve PSD93. The distribution of the Csk-homologous kinase CHK is not altered. Importantly, since the activities of Fyn and Src are decreased in adult Pag1-/- mice, thus presenting the reversed phenotype of P1, this provides the first in vivo evidence for a Csk-independent positive regulatory function for PAG in the brain

    The role of Fyn tyrosine kinase in oligodendrocyte development and myelination

    Fyn, a member of the Src family of non-receptor protein tyrosine kinases, is highly expressed in the brain and has been suggested to play an important role in the developing central nervous system (CNS). Recent studies indicate that Fyn is important for normal myelination, but there are still many unanswered questions. In this thesis, I analyzed myelination and oligodendrocyte development in mice that lack detectable Fyn protein (fyn–/– mice). I found a significant myelin deficit in fyn–/– mouse forebrain, but not spinal cord. The myelin deficit in forebrain was most severe at one month of age and persisted beyond one year of age. Myelin content in src–/– mice, yes–/– mice, and lyn –/– mice was normal; thus, Fyn is unique among Src-family kinases in that it is required for normal myelination. Analysis of fyn–/– mouse spinal cord and corpus callosum revealed a normal number of oligodendrocytes in the spinal cord, but a reduced number in corpus callosum. In all areas studied, myelin that was present in fyn–/– mice was morphologically normal. The number of myelinated fibers appeared normal in spinal cord and optic nerve from fyn–/– mice; in contrast, the number of myelinated fibers in the fyn–/– corpus callosum was significantly reduced at one month of age. Therefore, the fyn–/– hypomyelination phenotype was restricted to brain and was due to reduced numbers of oligodendrocytes and myelinated fibers. Finally, I investigated myelin content in mice expressing a catalytically-inactive full-length form of Fyn. These mice were also significantly myelin deficient at one month of age, indicating that the kinase activity of Fyn is essential for normal myelination. To determine whether Fyn is necessary for normal oligodendrocyte development, I analyzed the development of oligodendrocytes in neuron-free glial cell cultures from fyn–/– mice. I demonstrated that fyn–/– oligodendrocytes developed to the stage where they elaborate an extensive network of processes and express the antigenic components of mature oligodendrocytes. However, I observed fewer oligodendroglia in fyn–/– cultures and a reduced number of morphologically mature cells. In addition, I found that, unlike in wild-type cells, insulin-like growth factor failed to stimulate morphological development in fyn–/– oligodendroglia
