    Large scale statistical analysis of GEO datasets

    The problem addressed here is that of simultaneous treatment of several gene expression datasets, possibly collected under different experimental conditions and/or platforms. Using robust statistics, a large scale statistical analysis has been conducted over 2020 datasets downloaded from the Gene Expression Omnibus repository. The differences between datasets are compared to the variability inside a given dataset. Evidence that meaningful biological information can be extracted by merging different sources is provided

    Pericentric heterochromatin reprogramming by new histone variants during mouse spermiogenesis

    During male germ cell postmeiotic maturation, dramatic chromatin reorganization occurs, which is driven by completely unknown mechanisms. For the first time, we describe a specific reprogramming of mouse pericentric heterochromatin. Initiated when histones undergo global acetylation in early elongating spermatids, this process leads to the establishment of new DNA packaging structures organizing the pericentric regions in condensing spermatids. Five new histone variants were discovered, which are expressed in late spermiogenic cells. Two of them, which we named H2AL1 and H2AL2, specifically mark the pericentric regions in condensing spermatids and participate in the formation of new nucleoprotein structures. Moreover, our investigations also suggest that TH2B, an already identified testis-specific H2B variant of unknown function, could provide a platform for the structural transitions accompanying the incorporation of these new histone variants

    Identification of a novel BET bromodomain inhibitor-sensitive, gene regulatory circuit that controls Rituximab response and tumour growth in aggressive lymphoid cancers.: CYCLON-induced Rituximab resistance

    International audienceImmuno-chemotherapy elicit high response rates in B-cell non-Hodgkin lymphoma but heterogeneity in response duration is observed, with some patients achieving cure and others showing refractory disease or relapse. Using a transcriptome-powered targeted proteomics screen, we discovered a gene regulatory circuit involving the nuclear factor CYCLON which characterizes aggressive disease and resistance to the anti-CD20 monoclonal antibody, Rituximab, in high-risk B-cell lymphoma. CYCLON knockdown was found to inhibit the aggressivity of MYC-overexpressing tumours in mice and to modulate gene expression programs of biological relevance to lymphoma. Furthermore, CYCLON knockdown increased the sensitivity of human lymphoma B cells to Rituximab in vitro and in vivo. Strikingly, this effect could be mimicked by in vitro treatment of lymphoma B cells with a small molecule inhibitor for BET bromodomain proteins (JQ1). In summary, this work has identified CYCLON as a new MYC cooperating factor that autonomously drives aggressive tumour growth and Rituximab resistance in lymphoma. This resistance mechanism is amenable to next-generation epigenetic therapy by BET bromodomain inhibition, thereby providing a new combination therapy rationale for high-risk lymphoma

Self-hypnosis combined to self-care learning have been used in the past years to treat these symptoms, at the moment of diagnosis, during and/or after the cancer treatments.     Two experiments have been performed to investigate heating by high-intensity laser-generated electrons, in the context of studies of the fast ignitor approach to inertial confinement fusion (ICF). A new spectrometer and layered targets have been used to detect Kα emission from aluminum heated by a fast electron beam. Results show that a temperature of about 40 eV is reached in solid density aluminum up to a depth of about 100 μm

    A transcriptomic analysis of human centromeric and pericentric sequences in normal and tumor cells

    Although there is now evidence that the expression of centromeric (CT) and pericentric (PCT) sequences are key players in major genomic functions, their transcriptional status in human cells is still poorly known. The main reason for this lack of data is the complexity and high level of polymorphism of these repeated sequences, which hampers straightforward analyses by available transcriptomic approaches. Here a transcriptomic macro-array dedicated to the analysis of CT and PCT expression is developed and validated in heat-shocked (HS) HeLa cells. For the first time, the expression status of CT and PCT sequences is analyzed in a series of normal and cancer human cells and tissues demonstrating that they are repressed in all normal tissues except in the testis, where PCT transcripts are found. Moreover, PCT sequences are specifically expressed in HS cells in a Heat-Shock Factor 1 (HSF1)-dependent fashion, and we show here that another independent pathway, involving DNA hypo-methylation, can also trigger their expression. Interestingly, CT and PCT were found illegitimately expressed in somatic cancer samples, whereas PCT were repressed in testis cancer, suggesting that the expression of CT and PCT sequences may represent a good indicator of epigenetic deregulations occurring in response to environmental changes or in cell transformation

    Dans un contexte de mondialisation économique, l’examen des rapports entre arbitrage et droit de la concurrence n’est pas dénué d’ambiguïté et de complexité. Ce sujet a déjà fait couler beaucoup d’encre et pourtant, à l’heure où l’arbitrage connaît un essor sans précédent, des réponses incertaines sont données à bon nombre de questions. Tant la jurisprudence que la doctrine manquent d’uniformité sur le sujet. Ce mémoire vise à faire le point sur l’évolution, les ambitions ainsi que les méfiances relatives à l’arbitrage en tant que mode alternatif de résolution des litiges concernant le droit de la concurrence. Il a notamment pour objectif d’aller un pas plus loin en observant le problème sous un angle comparatif. Pour ce faire, il sera nécessaire d’analyser les problèmes qui s’observent dans la pratique d’un point de vue national, certes, mais il s’agira surtout de prendre en compte les perspectives tant européennes qu’internationales. Ces analyses permettront de déterminer si l’on est sur la voie d’une collaboration constructive dans ce cadre ou, au contraire, si les antinomies dont font preuve ces matières risquent de dresser un obstacle à l’élaboration d’un système cohérent et globalement accepté.Master [120] en droit, Université catholique de Louvain, 201
