    On the role of gallbladder emptying and incretin hormones for nutrient-mediated TSH suppression in patients with type 2 diabetes

    Bile acids are possible candidate agents in newly identified pathways through which energy expenditure may be regulated. Preclinical studies suggest that bile acids activate the enzyme type 2 iodothyronine deiodinase, which deiodinates thyroxine (T(4)) to the biologically active triiodothyronine (T(3)). We aimed to evaluate the influence of bile acid exposure and incretin hormones on thyroid function parameters in patients with type 2 diabetes. Thyroid-stimulating hormone (TSH) and thyroid hormones (total T(3) and free T(4)) were measured in plasma from two human studies: i) 75 g-oral glucose tolerance test (OGTT) and three isocaloric (500 kcal) and isovolaemic (350 ml) liquid meals with increasing fat content with concomitant ultrasonographic evaluation of gallbladder emptying in 15 patients with type 2 diabetes and 15 healthy age, gender and BMI-matched controls (meal-study) and ii) 50 g-OGTT and isoglycaemic intravenous glucose infusions (IIGI) alone or in combination with glucose-dependent insulinotropic polypeptide (GIP), glucagon-like peptide 1 (GLP1) and/or GLP2, in ten patients with type 2 diabetes (IIGI-study). In both studies, TSH levels declined (P<0.01) similarly following all meal and infusion stimuli. T(3) and T(4) concentrations did not change in response to any of the applied stimuli. TSH levels declined independently of the degree of gallbladder emptying (meal-study), route of nutrient administration and infusion of gut hormones. In conclusion, intestinal bile flow and i.v. infusions of the gut hormones, GIP, GLP1 and/or GLP2, do not seem to affect thyroid function parameters. Thus, the presence of a ‘gut–thyroid–pituitary’ axis seems questionable

    Systemic infection by Yersinia enterocolitica in chinchillas (Chinchilla laniger)

    Yersinia enterocolitica é uma bactéria Gram-negativa que causa infecções em diversas espécies de mamíferos. O agente, geralmente, provoca infecções restritas ao intestino e linfonodos mesentéricos, porém a infecção pode se tornar sistêmica ocasionando lesões em outros órgãos como fígado e baço. Neste trabalho descrevem-se dois surtos de infecções sistêmicas causadas pela Yersinia enterocolitica em criatórios comerciais de chinchilas no Rio Grande do Sul (Brasil). Os proprietários relatavam que os animais acometidos apresentavam apatia, anorexia e morte. Foram encaminhados 13 animais para a realização de necropsia. No exame post mortem dos animais observou-se esplenomegalia, hepatomegalia e áreas multifocais esbranquiçadas no fígado, baço, pulmões, rins e intestino. No exame microscópico visualizou-se infiltrado inflamatório de neutrófilos e macrófagos, necrose, deposição de fibrina e ocasionalmente pode ser observado coco-bacilos no centro das áreas de necrose. No cultivo bacteriológico obteve-se o crescimento de Yersinia enterocolitica nos animais provenientes dos dois criatórios. O agente foi isolado de amostras no fígado, baço, intestino e pulmões dos animais necropsiados, além do cultivo de fezes de animais de uma das propriedades acometidas. A yersiniose, portanto, é uma patologia que deve ser investigada em casos de mortalidade de chinchilas.Yersinia enterocolitica is a Gram-negative bacterium, which causes infections in several mammalian species. It is often recognized as an agent causing intestinal and mesenteric lymph nodes lesions. However, Yersinia enterocolitica infection may also become systemic, with lesions in others organs such as liver and spleen. This paper describes outbreaks of systemic infection due to Yersinia enterocolitica in two commercial chinchilla breeders in Rio Grande do Sul (Brazil). Owners reported that affected animals showed apathy, anorexia prior to death. Macroscopic examination performed in 13 animals revealed splenomegaly, hepatomegaly and multifocal whitish pinpoint foci in liver, spleen, lung, kidney and intestine. Microscopically, the affected tissues had infiltration of neutrophils and macrophages, as well as fibrin and necrosis with central areas containing cocobacilli bacteria. Yersinia enterocolitica was isolated from liver, spleen, lung and intestine samples from animals of both breeders, and from feces of chinchillas of one of the breeders. Therefore, yersiniosis is a disease to be investigated in cases of mortality of chinchillas

    High proportion of smaller ranged hummingbird species coincides with ecological specialization across the Americas.

    Ecological communities that experience stable climate conditions have been speculated to preserve more specialized interspecific associations and have higher proportions of smaller ranged species (SRS). Thus, areas with disproportionally large numbers of SRS are expected to coincide geographically with a high degree of community-level ecological specialization, but this suggestion remains poorly supported with empirical evidence. Here, we analysed data for hummingbird resource specialization, range size, contemporary climate, and Late Quaternary climate stability for 46 hummingbird-plant mutualistic networks distributed across the Americas, representing 130 hummingbird species (ca 40% of all hummingbird species). We demonstrate a positive relationship between the proportion of SRS of hummingbirds and community-level specialization, i.e. the division of the floral niche among coexisting hummingbird species. This relationship remained strong even when accounting for climate, furthermore, the effect of SRS on specialization was far stronger than the effect of specialization on SRS, suggesting that climate largely influences specialization through species' range-size dynamics. Irrespective of the exact mechanism involved, our results indicate that communities consisting of higher proportions of SRS may be vulnerable to disturbance not only because of their small geographical ranges, but also because of their high degree of specialization

    Rehabilitation and release of confiscated songbirds into the wild: A pilot study

    Songbirds are currently the most prevalent animals in illegal trafficking in Brazil and other countries, so they are often confiscated, and this poses legal, ethical, and conservation challenges. Returning them to nature requires complex and expensive management, a topic that is sparingly addressed in the literature. Here, we described the processes and costs associated with an attempt to rehabilitate and release confiscated songbirds into the wild. A total of 1,721 songbirds of several species were quarantined, rehabilitated, and released, primarily on two farms located within their typical geographical distribution. Health assessments were performed on samples from 370 birds. Serology revealed no antibodies against Newcastle disease, and Salmonella spp. cultures were negative. Real-time polymerase chain reactions detected M. gallisepticum in samples from seven birds. Atoxoplasma spp. and Acuaria spp. infections, sepsis, and trauma were the top causes of bird death. About 6% of the released birds were recaptured, within an average period of 249 days after release, and at a mean distance of 2,397 meters from the release sites. The majority of these birds were found with free-living mates within or close to fragments of transitional ecoregions with native or cultivated grasslands, and native groves/forests, and shrublands. However, eucalyptus plantations with rich understory regeneration provided a suitable environment for the released forest species to settle, since they were recaptured during the defense of these sites. Over half of the recaptured birds presented behavioral profiles with both dominant and tame traits. Birds with dominant traits are more likely to settle in habitats and face the live decoys during fieldwork, whereas birds with tame characteristics tend to accept close contact with humans. Ultramarine grosbeak (Cyanoloxia brissonii), the least common species among those released, at the release sites showed an almost 2-fold recapture rate in the shortest mean distances from the release sites. This suggests less territory competition, perhaps a major factor of bird re-establishment here. The total per-bird cost was USD 57.     Improved clinical investigation and evaluation of high-risk medical devices: the rationale and objectives of CORE-MD (Coordinating Research and Evidence for Medical Devices)

    : In the European Union (EU) the delivery of health services is a national responsibility but there are concerted actions between member states to protect public health. Approval of pharmaceutical products is the responsibility of the European Medicines Agency, whereas authorizing the placing on the market of medical devices is decentralized to independent 'conformity assessment' organizations called notified bodies. The first legal basis for an EU system of evaluating medical devices and approving their market access was the medical device directives, from the 1990s. Uncertainties about clinical evidence requirements, among other reasons, led to the EU Medical Device Regulation (2017/745) that has applied since May 2021. It provides general principles for clinical investigations but few methodological details-which challenges responsible authorities to set appropriate balances between regulation and innovation, pre- and post-market studies, and clinical trials and real-world evidence. Scientific experts should advise on methods and standards for assessing and approving new high-risk devices, and safety, efficacy, and transparency of evidence should be paramount. The European Commission recently awarded a Horizon 2020 grant to a consortium led by the European Society of Cardiology and the European Federation of National Associations of Orthopaedics and Traumatology, that will review methodologies of clinical investigations, advise on study designs, and develop recommendations for aggregating clinical data from registries and other real-world sources. The CORE-MD project (Coordinating Research and Evidence for Medical Devices) will run until March 2024; here we describe how it may contribute to the development of regulatory science in Europe
