    El Instituto Público Maestro Gabriel está ubicado en el Distrito IV, Municipio de Managua, Barrio San Luis y está situado de Inerte 1c al este. En esta Institución educativa fue realizada la investigación por las estudiantes del III año de la carrera de Pedagogía con mención en Administración de la Educación, UNAN – Managua, con el objetivo de diagnosticar las condiciones administrativas del Instituto, en el período del I Semestre del 2017. El presente estudio, es de tipo descriptivo, de corte transversal, con un enfoque cualitativo. La población de la comunidad educativa es de 215 del turno vespertino, y la muestra fue seleccionada de forma aleatoria con un total de 12, lo cual estuvo conformada por el equipo 1 administrativo y 11 docentes. La información se recopiló con la aplicación de entrevistas, guía de análisis documental (legales, académicos y administrativos). Los resultados más relevantes en la investigación, fueron: el director no ha realizado acompañamiento pedagógico a los docentes directamente en el aula, la mayoría de los documentos legales no se encuentran en físico en el instituto, ausencia del organigrama, no se ha elaborado la visión y misión de la institución educativa. Las recomendaciones significativas, fueron: Elaborar la Visión y Misión, Diseñar el POA del Instituto, realizar acompañamiento pedagógico y poseer documentos legales en físico. Producto de los resultados de la investigación, se elaboró para la colegio público en estudio la propuesta de Plan Operativo Anual, 2017, con énfasis en la función administrativa

    Synthesis and defect characterization of hybrid ceria nanostructures as a possible novel therapeutic material towards COVID-19 mitigation

    This study reports the synthesis of hybrid nanostructures composed of cerium dioxide and microcrystalline cellulose prepared by the microwave-assisted hydrothermal route under distinct temperature and pH values. Their structural, morphological and spectroscopic behaviors were investigated by X-Rays Diffraction, Field Emission Gun Scanning Electron Microscopy, High-Resolution Transmission Electron Microscopy, and Fourier-Transform Infrared, Ultraviolet–Visible, Raman and Positron Annihilation Lifetime spectroscopies to evaluate the presence of structural defects and their correlation with the underlying mechanism regarding the biocide activity of the studied material. The samples showed mean crystallite sizes around 10 nm, characterizing the formation of quantum dots unevenly distributed along the cellulose surface with a certain agglomeration degree. The samples presented the characteristic Ce–O vibration close to 450 cm−1 and a second-order mode around 1050 cm−1, which is indicative of distribution of localized energetic levels originated from defective species, essential in the scavenging of reactive oxygen species. Positron spectroscopic studies showed first and second lifetime components ranging between 202–223 ps and 360–373 ps, respectively, revealing the presence of two distinct defective oxygen species, in addition to an increment in the concentration of Ce3+-oxygen vacancy associates as a function of temperature. Therefore, we have successfully synthesized hybrid nanoceria structures with potential multifunctional therapeutic properties to be further evaluated against the COVID-19.Fil: Silva Rosa Rocha, Leandro.     Gestión que realiza el equipo de dirección para el fortalecimiento de la función administrativa del Instituto nacional publico maestro Gabriel, municipio de Managua, distrito IV, realizado en el año 2018

    El presente trabajo investigativo consiste en valorar la incidencia de la gestión que realiza el equipo de dirección en la función administrativa del Instituto Maestro Gabriel, Municipio de Managua, Distrito IV, realizado en el año 2019, identificando las gestiones realizadas por la dirección del centro, y elaborar propuestas de estrategias de gestión administrativa en función de la calidad educativa. El presente estudio, es de tipo descriptivo, de corte transversal, con un enfoque cualitativo. La población de la comunidad educativa es de 99 estudiantes del turno vespertino, y la muestra fue seleccionada de forma aleatoria con un total de 12, lo cual estuvo conformada por el equipo 1 administrativo y 11 docentes. La información se recopiló con la aplicación de entrevistas al director y docente, grupo focal dirigido a estudiantes, guía de observación y guía de análisis documental (legales, académicos y administrativos). Los resultados más relevantes de la investigación, fueron: En el aspecto de la gestión el director no realiza gestiones para el buen funcionamiento del Instituto, y las gestiones educativas realizadas hace 3 años, no logro ejecutarlas, en cambio los estudiantes, entienden como gestión la limpieza de las áreas verdes, juegos deportivos, rehabilitación de la banda rítmica, en relación a las funciones administrativas, se detectó que el equipo directivo ejerce un liderazgo democrático, toma en cuenta a las opiniones de la comunidad educativa para la elaboración y ejecución del plan operativo anual. En relación al proceso de planificación de las gestiones que se realizan en el instituto, no se elaboran proyectos de mejora en el Instituto. Las recomendaciones significativas, fueron: Propuesta en la planificación sobre la gestión educativa, referente a la reparación y mantenimiento de los aires acondicionados, así como la pintura de las paredes y renovación parcial del sistema eléctrico del instituto.     Increased voltage-dependent K+ channel Kv1.3 and Kv1.5 expression correlates with leiomyosarcoma aggressiveness

    Voltage-dependent K+ channels (Kv) are involved in the proliferation and differentiation of mammalian cells, since Kv antagonists impair cell cycle progression. Although myofibers are terminally differentiated, some myoblasts may re-enter the cell cycle and proliferate. Since Kv1.3 and Kv1.5 expression is remodeled during tumorigenesis and is involved in smooth muscle proliferation, the purpose of this study was to analyze the expression of Kv1.3 and Kv1.5 in smooth muscle neoplasms. In the present study, we examined human samples of smooth muscle tumors together with healthy speci­mens. Thus, leiomyoma (LM) and leiomyosarcoma (LMS) tumors were analyzed. Results showed that Kv1.3 was poorly expressed in the healthy muscle and indolent LM specimens, whereas aggressive LMS showed high levels of Kv1.3 expression. Kv1.5 staining was correlated with malignancy. The findings show a remodeling of Kv1.3 and Kv1.5 in human smooth muscle sarcoma. A correlation of Kv1.3 and Kv1.5 expression with tumor aggressiveness was observed. Thus, our results indicate Kv1.5 and Kv1.3 as potential tumorigenic targets for aggressive human LMS

    Validation of Cell-Free DNA Collection Tubes for Determination of EGFR Mutation Status in Liquid Biopsy from NSCLC Patients

    Altres ajuts: Roche Farma S.A., Spain.Precision medicine has revolutionized the understanding and treatment of cancer by identifying subsets of patients who are amenable to specific treatments according to their molecular characteristics, as exemplified by epidermal growth factor receptor (EGFR) mutations in non-small cell lung cancer (NSCLC). Although tissue biopsy is the gold standard for determining molecular alterations in tumors, its limitations have prompted the development of new techniques for studying tumor biomarkers in liquid biopsies, such as mutation analysis in cell-free DNA (cfDNA). cfDNA analysis can accurately determine tumor progression and prognosis and more effectively identify appropriate targeted therapies. However, cfDNA is vulnerable, particularly during plasma sample shipping. We compared the cell- and DNA-stabilizing properties of cell-free DNA blood collection tubes (BCTs) with those of the traditional shipping method (frozen plasma) for EGFR mutation testing using the cobas ® EGFR Mutation Test v2 in a prospective cohort of 49 patients from three different Spanish hospitals. In total, 98 NSCLC samples, two from each patient, were studied; five of the 49 cases were considered invalid by cobas ® with one of the two shipping methods analyzed. After excluding these samples, we analyzed 88 samples from 44 patients. Considering the current methodology (frozen plasma) for sending samples as the gold standard, we evaluated the sensitivity and specificity of cfDNA BCT shipment. The global agreement between the two methods was 95.4%, with 100% sensitivity and 94.6% specificity for the cfDNA BCTs. cfDNA BCTs had a positive predictive value of 81.8% and negative predictive value of 100%. cfDNA BCTs have the same sensitivity for EGFR mutation analysis in liquid biopsy as the current methodology and very high specificity. They also have some additional advantages in terms of collection and further shipment. Therefore, cfDNA BCTs can be perfectly incorporated into the routine practice for EGFR mutation determination.     The umbilical cord, preeclampsia and the VEGF family

    Q1Artículo original783-795Introduction The VEGF family has been identified as abnormal in preeclampsia (PE). Hypertensive disorders of pregnancy (HDP) are major contributors to maternal and neonatal morbidity and mortality worldwide; likewise, umbilical cord anatomical abnormalities (UCAA) are linked to poor neonatal outcomes. Based on the relationship described between PE and UCAA and the role of the VEGF family in PE, this study explored VEGF expression in placental and UC tissued from patients with PE and with UCAA. Methods We performed an observational, analytical study on placentas, comparing protein and mRNA expression in four groups: patients with PE, patients with UC abnormalities, patients with both, and patients with none of them. Using immunohistochemistry, we studied VEGF A, VEGF R1 (FLT1), MMP1, and PLGF. With quantitative reverse transcription polymerase chain reaction we described mRNA expression of PLGF, VEGF and sFLT1, and sFLT1/PLGF ratio. Results Forty newborns were included. Sixty-seven percent of mothers and 45% of newborns developed no complications. Immunohistochemistry was performed on UC and placental disc paraffin-embedded tissue; in the latter, the mRNA of the VEGF family was also measured. Statistically significant differences were observed among different expressions in both HDP and UCAA groups. Interestingly, the UCAA group exhibited lower levels of sFLT1 and VEGF-A in comparison with other groups, with significant P-value for sFLT1 (P=0000.1). Conclusion The origin of UCAA abnormalities and their relation with HDP are still unknown. VEGF family alterations could be involved in both. This study provides the first approach related to molecules linked to UCAA

    Integrated thematic geological mapping of the Atlantic Margin of Iberia.

    This paper synthesizes the geology of the Atlantic Margin off the coast of Iberia and surrounding Abyssal Plains using published thematic mapping freely downloadable from EMODNET-Geology portal at different scales. Selected information was chosen in order to highlight mineral occurrences and natural hazards overlaid on geological and morphological maps. Altogether, this information is published and interpreted here for the first time; nevertheless this exercise can be carried out by anyone interested and allows different visualizations of geological objects. Cross-correlations of geological objects and processes can easily arise. Because all of the information (each piece of data and metadata) in the EMODNET-Geology portal has bibliographic references associated, readers are able to find the original source of information. It is shown that clicking in and out of layers of information (that cannot be found all together in a single scientific paper) allows quick cross-correlation using the EMODNET Geology thematic portal. This allows a free, versatile and quick way of cross-correlating geological objects and processes in vast marine areas and their comparison with onshore geolog

    Design and implementation of a mobile app for the pharmacotherapeutic follow-up of patients diagnosed with immune-mediated inflammatory diseases: eMidCare

    BackgroundPharmacotherapeutic management of immune-mediated inflammatory diseases (IMID) has become more complex due to the development of new treatments, such as biological therapies. Mobile health, especially apps, can provide IMID patients with greater autonomy and facilitate communication with healthcare professionals. Our objective was to design and implement an app for remote monitoring and communication with IMID patients.MethodsA multidisciplinary group was created to design and develop an app for IMID patients in a tertiary hospital. The app functionalities were identified through a focus group with IMID patients and through an observational, descriptive study of available apps for IMID patients at App Store and Play Store platforms. Once the app was designed and developed, we offered the app to IMID patients who initiated a new biological therapy. The inclusion period was from December 2020 to August 2021. We performed an observational, longitudinal study to assess the app’s impact on medication safety, communication, satisfaction, and usability.ResultsWe designed an app (eMidCare®) with the following modules: My Medication, My Questionnaires, Adverse Events, Useful Information, Messages, and Patient Profile. A total of 85 patients were installed with the app. The median (range) follow-up time for app use was 123 (5-270) days. In the My Medication module, 100% of patients registered their biological therapy and 25.9% also used this module to record each dose of medication administered. A total of 82 adverse events (AEs) were registered. Thirty-two percent of the patients registered at least 1 AE. The most frequent AEs were fatigue, injection site reaction, headache, and nausea. Fifty-two percent of patients used the Messages module to communicate with healthcare professionals. The most frequent messages concerned doubts about managing AEs (26.2%) and drug interactions (18.9%). The satisfaction survey yielded a median (range) score of 9.1 (7-10) out of 10.ConclusionsWe developed an app, eMidCare®, which reminds patients to take their medication, enables them to record AEs, and helps them communicate with healthcare professionals. Approximately one-third of the patients registered the administration of the biological therapies and registered at least 1 AE. The most used and most satisfactory functionality was communication with health professionals

    High-Throughput Sequencing of RNA Silencing-Associated Small RNAs in Olive (Olea europaea L.)

    Small RNAs (sRNAs) of 20 to 25 nucleotides (nt) in length maintain genome integrity and control gene expression in a multitude of developmental and physiological processes. Despite RNA silencing has been primarily studied in model plants, the advent of high-throughput sequencing technologies has enabled profiling of the sRNA component of more than 40 plant species. Here, we used deep sequencing and molecular methods to report the first inventory of sRNAs in olive (Olea europaea L.). sRNA libraries prepared from juvenile and adult shoots revealed that the 24-nt class dominates the sRNA transcriptome and atypically accumulates to levels never seen in other plant species, suggesting an active role of heterochromatin silencing in the maintenance and integrity of its large genome. A total of 18 known miRNA families were identified in the libraries. Also, 5 other sRNAs derived from potential hairpin-like precursors remain as plausible miRNA candidates. RNA blots confirmed miRNA expression and suggested tissue- and/or developmental-specific expression patterns. Target mRNAs of conserved miRNAs were computationally predicted among the olive cDNA collection and experimentally validated through endonucleolytic cleavage assays. Finally, we use expression data to uncover genetic components of the miR156, miR172 and miR390/TAS3-derived trans-acting small interfering RNA (tasiRNA) regulatory nodes, suggesting that these interactive networks controlling developmental transitions are fully operational in olive
