Insulin levels are decreased in the cerebrospinal fluid of women with prodomal Alzheimer's disease
Previous studies have failed to reach consensus on insulin levels in cerebrospinal fluid of Alzheimer's disease (AD) patients and on its relation to pathological features. We performed a new analysis in patients at different stages of AD, and investigated the relationship of insulin levels with biochemical disease markers and with cognitive score. We included 99 patients from our Memory Clinic (Karolinska University Hospital, Sweden), including: 27 patients with mild AD, 13 that progressed from mild cognitive impairment (MCI) to AD in two years time, 26 with MCI stable after two years, and 33 with subjective cognitive impairment. Insulin was significantly decreased in the cerebrospinal fluid of both women and men with mild AD. Insulin deficits were seen in women belonging to both MCI groups, suggesting that this occurs earlier than in men. Insulin was positively associated with amyloid-β 1-42 (Aβ1-42) levels and cognitive score. Furthermore, total-tau/(Aβ1-42*insulin) ratio showed strikingly better sensitivity and specificity than the total-tau/Aβ1-42 ratio for early AD diagnosis in women
Impact of chronic stress protocols in learning and memory in rodents: systematic review and meta-analysis
The idea that maladaptive stress impairs cognitive function has been a cornerstone of decades in basic and clinical research. However, disparate findings have reinforced the need to aggregate results from multiple sources in order to confirm the validity of such statement. In this work, a systematic review and meta-analyses were performed to aggregate results from rodent studies investigating the impact of chronic stress on learning and memory. Results obtained from the included studies revealed a significant effect of stress on global cognitive performance. In addition, stressed rodents presented worse consolidation of learned memories, although no significantly differences between groups at the acquisition phase were found. Despite the methodological heterogeneity across studies, these effects were independent of the type of stress, animals' strains or age. However, our findings suggest that stress yields a more detrimental effect on spatial navigation tests' performance. Surprisingly, the vast majority of the selected studies in this field did not report appropriate statistics and were excluded from the quantitative analysis. The 20th International Symposium on HIV and Emerging Infectious Diseases took place in Marseille, France. It had a refreshing European look with reinforced partnerships with the European AIDS Clinical Society and the British HIV Association and with international speakers and participants. Topics included HIV and global health, HIV and hepatitis cure, the microbiome and immunotherapies, clinical research and methodology, as well as chemsex, pre-exposure prophylaxis, sexually transmitted infections and emerging infectious diseases. Novel areas of research were also described, such as electronic technology in order to improve HIV management, and the expert patient
Interactions between age, stress and insulin on cognition: implications for Alzheimer's disease
There is much interest in understanding the mechanisms responsible for interactions among stress, aging, memory and Alzheimer's disease. Glucocorticoid secretion associated with early life stress may contribute to the variability of the aging process and to the development of neuro- and psychopathologies. Maternal separation (MS), a model of early life stress in which rats experience 3 h of daily separation from the dam during the first 3 weeks of life, was used to study the interactions between stress and aging. Young (3 months) MS rats showed an altered hypothalamic-pituitary-adrenal (HPA) axis reactivity, depressive-like behavior in the Porsolt swimming test and cognitive impairments in the Morris water maze and new object recognition test that persisted in aged (18 months) rats. Levels of insulin receptor, phosphorylated insulin receptor and markers of downstream signaling pathways (pAkt, pGSK3 beta, pTau, and pERK1 levels) were significantly decreased in aged rats. There was a significant decrease in pERK2 and in the plasticity marker ARC in MS aged rats compared with single MS or aged rats. It is interesting to note that there was a significant increase in the C99 : C83 ratio, A beta levels, and BACE1 levels the hippocampus of MS aged rats, suggesting that in aged rats subjected to early life stress, there was an increase in the amyloidogenic processing of amyloid precursor protein (APP). These results are integrated in a tentative mechanism through which aging interplay with stress to influence cognition as the basis of Alzheimer disease (AD). The present results may provide the proof-of-concept for the use of glucocorticoid-/insulin-related drugs in the treatment of AD
Frequency of exposure with MamaNatalie simulation and self-perceived competence in the delivery room among student nurses
Abstract onlyTechnology can support and modify education in many ways. Technology can also aid in nursing education in the form of high-fidelity simulations using electronic training manikins. Clinical Instructors can program the manikins to simulate scenarios that student nurses might encounter in clinical practice. Based on the manikin's symptoms, students can take vital signs, monitor changes, and make decisions. These simulations gave them a sense of realism as if they were working with an actual client. The target population and sample size, determined using the stratified random sampling method, comprised 147 respondents from a private university in Iloilo City. This study utilized a quantitative descriptive-correlational research design with a validated and reliable research instrument. The results showed that 78.2% of the respondents were female and 21.8 % were male; 69.4% were aged 21; and 53.7% were in the lower middle class. Notably, 35.4% of respondents reported a frequency of exposure range of 2.30 or below, while 37.4% had a self-perceived competence range of 4.71 or above. Statistical analysis indicated a strong relationship between self-perceived competence and frequency of exposure (Spearman’s rho = + .645), and this strong relationship was statistically significant (p= .002). This suggested that self-perceived competence directly influenced the frequency of exposure of Student Nurses in a private university in Iloilo City. In conclusion, the respondents were predominantly female, mostly 21 years old, and in the lower middle class. The frequency of exposure was found to be less than two simulations and the mean score of 4.5 suggested that the respondents' self-perceived competence was "very high."Includes bibliographical referencesBachelor of Science in Nursin

Destabilization of glacial climate by the radiative impact of Atlantic Meridional overturning circulation disruptions
Unidad de excelencia María de Maeztu MdM-2015-0552During each of the dramatic global warmings that ended the Pleistocene ice ages, the Atlantic Meridional Overturning Circulation (AMOC) was disrupted. It is not clear whether this was a contributing cause or simply an effect of deglaciation. Here we show that in an ensemble of simulations with a global climate model, AMOC disruption causes a consistent and sustained positive radiative imbalance of ~0.4Wm2. The imbalance is accommodated by heat accumulation in the ocean interior, representing an overall planetary warming, subsequently released by deep convection in the North Atlantic when the AMOC resumes. The results suggest a means by which AMOC disruptions could have helped to tip the planet out of stable glaciated states. However, the fact that AMOC disruptions occurred during prior Heinrich Stadials without causing deglaciation shows that other factors, such as ice sheet dynamics, or controls on CO2, were also key for deglaciation
Registered replication report: A large multilab cross-cultural conceptual replication of Turri et al. (2015)
According to the justified true belief (JTB) account of knowledge, people can truly know something only if they have a belief that is both justified and true (i.e., knowledge is JTB). This account was challenged by Gettier, who argued that JTB does not explain knowledge attributions in certain situations, later called “Gettier-type cases,” wherein protagonists are justified in believing something to be true, but their belief was correct only because of luck. Laypeople may not attribute knowledge to protagonists with justified but only luckily true beliefs. Although some research has found evidence for these so-called Gettier intuitions, Turri et al. found no evidence that participants attributed knowledge in a counterfeit-object Gettier-type case differently than in a matched case of JTB. In a large-scale, cross-cultural conceptual replication of Turri and colleagues’ Experiment 1 (N = 4,724) using a within-participants design and three vignettes across 19 geopolitical regions, we did find evidence for Gettier intuitions; participants were 1.86 times more likely to attribute knowledge to protagonists in standard cases of JTB than to protagonists in Gettier-type cases. These results suggest that Gettier intuitions may be detectable across different scenarios and cultural contexts. However, the size of the Gettier intuition effect did vary by vignette, and the Turri et al. vignette produced the smallest effect, which was similar in size to that observed in the original study. Differences across vignettes suggest that epistemic intuitions may also depend on contextual factors unrelated to the criteria of knowledge, such as the characteristics of the protagonist being evaluated.info:eu-repo/semantics/publishedVersio
