    EEG–fMRI mapping of asymmetrical delta activity in a patient with refractory epilepsy is concordant with the epileptogenic region determined by intracranial EEG

    We studied a patient with refractory focal epilepsy using continuous EEG-correlated fMRI. Seizures were characterized by head turning to the left and clonic jerking of the left arm, suggesting a right frontal epileptogenic region. Interictal EEG showed occasional runs of independent nonlateralized slow activity in the delta band with right frontocentral dominance and had no lateralizing value. Ictal scalp EEG had no lateralizing value. Ictal scalp EEG suggested right-sided central slow activity preceding some seizures. Structural 3-T MRI showed no abnormality. There was no clear epileptiform abnormality during simultaneous EEG–fMRI. We therefore modeled asymmetrical EEG delta activity at 1–3 Hz near frontocentral electrode positions. Significant blood oxygen level-dependent (BOLD) signal changes in the right superior frontal gyrus correlated with right frontal oscillations at 1–3 Hz but not at 4–7 Hz and with neither of the two frequency bands when derived from contralateral or posterior electrode positions, which served as controls. Motor fMRI activations with a finger-tapping paradigm were asymmetrical: they were more anterior for the left hand compared with the right and were near the aforementioned EEG-correlated signal changes. A right frontocentral perirolandic seizure onset was identified with a subdural grid recording, and electric stimulation of the adjacent contact produced motor responses in the left arm and after discharges. The fMRI localization of the left hand motor and the detected BOLD activation associated with modeled slow activity suggest a role for localization of the epileptogenic region with EEG–fMRI even in the absence of clear interictal discharges

    A Prospective Controlled Trial of Routine Opt-Out HIV Testing in a Men's Jail

    Approximately 10 million Americans enter jails annually. The Centers for Disease Control and Prevention now recommends routine opt-out HIV testing in these settings. The logistics for performing routine opt-out HIV testing within jails, however, remain controversial. The objective of this study was to evaluate the optimal time to routinely HIV test newly incarcerated jail detainees using an opt-out strategy.This prospective, controlled trial of routine opt-out HIV testing was conducted among 298 newly incarcerated male inmates in an urban men's jail in New Haven, Connecticut. 298 sequential entrants to the men's jail over a three week period in March and April 2008 were assigned to be offered routine opt-out HIV testing at one of three points after incarceration: immediate (same day, n = 103), early (next day, n = 98), or delayed (7 days, n = 97). The primary outcome was the proportion of men in each group consenting to testing.Routine opt-out HIV testing was significantly higher for the early (53%: AOR = 2.6; 95% CI = 1.5 to 4.7) and immediate (45%: AOR = 2.3; 95% CI = 1.3 to 4.0) testing groups compared to the delayed (33%) testing group. The immediate and early testing groups, however, did not significantly differ (p = 0.67). In multivariate analyses, factors significantly associated with routine opt-out HIV testing were assignment to the 'early' testing group (p = 0.0003) and low (bond > or = $5,000, immigration or federal charges or pre-sentencing > 30 days) likelihood of early release (p = 0.04). Two subjects received preliminary positive results and one of them was subsequently confirmed HIV seropositive.In this men's jail where attrition was high, routine opt-out HIV testing was not only feasible, but resulted in the highest rates of HIV testing when performed within 24 hours of incarceration.ClinicalTrials.gov NCT00624247

    SUMO is a small post-translational modifier, that is attached to lysine residues in target proteins. It acts by altering proteinprotein interactions, protein localisation and protein activity. SUMO chains can also act as substrates for ubiquitination, resulting in proteasome-mediated degradation of the target protein. SUMO is removed from target proteins by one of a number of specific proteases. The processes of sumoylation and desumoylation have well documented roles in DNA metabolism and in the maintenance of chromatin structure. To further analyse the role of this modification, we have purified protein complexes containing the S. pombe SUMO protease, Ulp2. These complexes contain proteins required for ribosome biogenesis, RNA stability and protein synthesis. Here we have focussed on two translation initiation factors that we identified as co-purifying with Ulp2, eIF4G and eIF3h. We demonstrate that eIF4G, but not eIF3h, is sumoylated. This modification is increased under conditions that produce cytoplasmic stress granules. Consistent with this we observe partial co-localisation of eIF4G and SUMO in stressed cells. Using HeLa cells, we demonstrate that human eIF4GI is also sumoylated; in vitro studies indicate that human eIF4GI is modified on K1368 and K1588, that are located in the C-terminal eIF4A- and Mnk-binding sites respectively

    The relationship between adverse childhood experiences, coping using sex, and adult sexual coercion in non-incarcerated, community-based females

    Adverse childhood experiences have been associated with negative outcomes in adulthood, including sexual offending. Using a cross-sectional design, we investigated whether self-reported adverse childhood experiences related to the perpetration of coercive sexual acts among 250 females recruited from the community. Furthermore, we examined whether sexualised coping mediated any potential relationship between childhood experiences and sexual coercion. A Spearman’s rank order correlation revealed no relationship between adverse childhood experiences and sexual coercion. However, adverse childhood experiences were significantly correlated with sexualised coping, which in turn was correlated with sexual coercion. Additionally, there was a significant but small indirect effect of adverse childhood experiences on sexual coercion through sexualised coping. Findings may help researchers to better understand the causal relationship between childhood experiences, sexual coping, and sexual coercion in females

    A method and system for determining range to a target are provided. A beam of electromagnetic energy is transmitted through an aperture in an opaque screen such that a portion of the beam passes through the aperture to generate a region of diffraction that varies as a function of distance from the aperture. An imaging system is focused on a target plane in the region of diffraction with the generated image being compared to known diffraction patterns. Each known diffraction pattern has a unique value associated therewith that is indicative of a distance from the aperture. A match between the generated image and at least one of the known diffraction patterns is indicative of a distance between the aperture and target plane

    The effect of exercise intensity on cognitive performance during short duration treadmill running

    This study examined the effect of short duration, moderate and high-intensity exercise on a Go/NoGo task. Fifteen, habitually active (9 females and 6 males aged 28 ± 5 years) agreed to participate in the study and cognitive performance was measured in three sessions lasting 10 min each, performed at three different exercise intensities: rest, moderate and high. Results indicated significant exercise intensity main effects for reaction time (RT) (p = 0.01), the omission error rate (p = 0.027) and the decision error rate (p = 0.011), with significantly longer RTs during high intensity exercise compared to moderate intensity exercise (p = 0.039) and rest (p = 0.023). Mean ± SE of RT (ms) was 395.8 ± 9.1, 396.3 ± 9.1 and 433.5 ± 16.1 for rest, moderate and high intensity exercise, respectively. This pattern was replicated for the error rate with a significantly higher omission error and decision error rate during high intensity exercise compared to moderate intensity exercise (p = 0.003) and rest (p = 0.001). Mean ± SE of omission errors (%) was 0.88 ± 0.23, 0.8 ± 0.23 and 1.8 ± 0.46% for rest, moderate and high intensity exercise, respectively. Likewise, mean ± SE of decision errors (%) was 0.73 ± 0.24, 0.73 ± 0.21 and 1.8 ± 0.31 for rest, moderate and high intensity exercise, respectively. The present study’s results suggest that 10 min workout at high intensity impairs RT performances in habitually active adults compared to rest or moderate intensity exercise

    Two Large HI Shells in the Outer Galaxy near l=279 degrees

    As part of a survey of HI 21-cm emission in the Southern Milky Way, we have detected two large shells in the interstellar neutral hydrogen near l=279 deg. The center velocities are +36 and +59 km/s, which puts the shells at kinematic distances of 7 and 10 kpc. The larger shell is about 610 pc in diameter and very empty, with density contrast of at least 15 between the middle and the shell walls. It has expansion velocity of about 20 km/s and swept up mass of several million solar masses. The energy indicated by the expansion may be as high as 2.4 X 10^53 ergs. We estimate its age to be 15 to 20 million years. The smaller shell has diameter of about 400 pc, expansion velocity about 10 km/s and swept up mass of about 10^6 solar masses. Morphologically both regions appear to be shells, with high density regions mostly surrounding the voids, although the first appears to have channels of low density which connect with the halo above and below the HI layer. They lie on the edge of the Carina arm, which suggests that they may be expanding horizontally into the interarm region as well as vertically out of the disk. If this interpretation is correct, this is the first detection of an HI chimney which has blown out of both sides of the disk.Comment: 21 pages, 14 jpeg figures, accepted for publication in A

    EXCEDE Technology Development III: First Vacuum Tests

    This paper is the third in the series on the technology development for the EXCEDE (EXoplanetary Circumstellar Environments and Disk Explorer) mission concept, which in 2011 was selected by NASA's Explorer program for technology development (Category III). EXCEDE is a 0.7m space telescope concept designed to achieve raw contrasts of 1e6 at an inner working angle of 1.2 l/D and 1e7 at 2 l/D and beyond. This will allow it to directly detect and spatially resolve low surface brightness circumstellar debris disks as well as image giant planets as close as in the habitable zones of their host stars. In addition to doing fundamental science on debris disks, EXCEDE will also serve as a technological and scientific precursor for any future exo-Earth imaging mission. EXCEDE uses a Starlight Suppression System (SSS) based on the PIAA coronagraph, enabling aggressive performance. We report on our continuing progress of developing the SSS for EXCEDE, and in particular (a) the reconfiguration of our system into a more flight-like layout, with an upstream deformable mirror and an inverse PIAA system, as well as a LOWFS, and (b) testing this system in a vacuum chamber, including IWA, contrast, and stability performance. The results achieved so far are 2.9e-7 contrast between 1.2-2.0 l/D and 9.7e-8 contrast between 2.0-6.0 l/D in monochromatic light; as well as 1.4e-6 between 2.0-6.0 l/D in a 10% band, all with a PIAA coronagraph operating at an inner working angle of 1.2 l/D. This constitutes better contrast than EXCEDE requirements (in those regions) in monochromatic light, and progress towards requirements in broadband light. Even though this technology development is primarily targeted towards EXCEDE, it is also germane to any exoplanet direct imaging space-based telescopes because of the many challenges common to different coronagraph architectures and mission requirements.Comment: 12 pages, 12 figures, to be published in proceedings of SPIE Astronomical Telescopes + Instrumentation (2014

    An integrated Structure-from-Motion and time-lapse technique for quantifying ice-margin dynamics

    Fine resolution topographic data derived from methods such as Structure from Motion (SfM) and Multi-View Stereo (MVS) have the potential to provide detailed observations of geomorphological change, but have thus far been limited by the logistical constraints of conducting repeat surveys in the field. Here, we present the results from an automated time-lapse camera array, deployed around an ice-marginal lake on the western margin of the Greenland ice sheet. Fifteen cameras acquired imagery three-times per day over a 426 day period, yielding a dataset of ~19 000 images. From these data we derived 18 point clouds of the ice-margin across a range of seasons and successfully identified calving events (ranging from 234 to 1475 m2 in area and 815–8725 m3 in volume) induced by ice cliff undercutting at the waterline and the collapse of spalling flakes. Low ambient light levels, locally reflective surfaces and the large survey range hindered analysis of smaller scale ice-margin dynamics. Nevertheless, this study demonstrates that an integrated SfM-MVS and time-lapse approach can be employed to generate long-term 3-D topographic datasets and thus quantify ice-margin dynamics at a fine spatio-temporal scale.
