    Import 03/11/2016Tato bakalářská práce pojednává o tématu automatické detekce velikosti terče zrakového nervu. Práce obsahuje stručný úvod do anatomie oka a komplexní rešerši pojednávající o onemocnění retinopatie nedonošených. Dále bakalářská práce obsahuje komplexní rešerši metod možných pro využití k detekci optického disku. Hlavní částí bakalářské práce je návrh algoritmu pro automatickou detekci velikosti terče zrakového nervu z retinálních snímků pořízených pomocí retinální kamery RetCam3. Jednotlivé kroky navrhovaného algoritmu jsou v textu podrobně popsány. Navrhovaný algoritmus byl otestován na 120 retinálních záznamech a výsledky detekce byly zhodnoceny v závěru.This bachelor thesis is devoted to the theme of automatic detection of optical nerve area. Thesis includes brief introduction of eye anathomy and complex research about the illness rethinopathy of prematurity. Next part of the bachelor thesis is dedicated to complex research of the methodes which can be used for a deteciton of optical nerv area on a retina. The main part of this thesis is dedicated to proposed algorithm for automatic detction of optical nerv area on the images acquired by RetCam3 system. Each step of the proposed algorithm is described in details in the following text.     Human pyroglutamyl peptidases and their involvement in Alzheimer’s disease

    The N-terminally pyroglutamyl-modified β-amyloid (Aβ) peptides are found in abundance in the pathological Alzheimer disease (AD) brain deposits. Such modification not only increases the hydrophobic properties of a given molecule, but also plays a protecting role against proteolytic degradation. This project involved the study of the human type I and type II pyroglutamyl peptidases and their involvement in Aβ processing in AD. Human PcpI has been successfully overexpressed in Escherichia coli strain and purified to homogeneity. The protein displayed significant instability in vitro. To overcome this problem a number of methods were employed such as screening for an optimal protein expression system and buffer composition, site-directed mutagenesis and chemical modification of selected surface residues. This resulted in the selection of the HEPPS buffer system as providing the most stabilising conditions for human PcpI. Improvement in the protein stability enabled initial crystallisation experiments and the identification of favourable conditions for crystal production. Further optimization of this process is needed in order to obtain good quality crystals which are required for structural study. The study on human PcpII involved an extensive screening for optimal expression conditions in bacterial, baculovirus/insect and mammalian systems. The truncated PcpII isoform PcpII/S62-H1024, which lacks the N-terminal transmembrane domain, was successfully expressed and secreted from the HEK 293T cell line using three different pOPIN-based constructs. Moreover, homology modelling of human PcpII catalytic domain was performed, which helped to gain an insight into the three-dimensional structure of the protein and its mode of substrate binding. Lastly, immunohistochemical staining of the human AD brain tissue sections was performed to compare the level and distribution of PcpI and PcpII enzymes between diseased and control cases. The results confirmed that the neurodegenerative conditions lead to the increased synthesis of both enzymes in the cortical AD tissues. Zabývá se předními sloupky automobilu, jejich vliv na viditelnost a zhodnocení šířky vzhledem k aktivní bezpečnosti vozidla. V úvodu práce se zabývám danou problematikou pasivní, aktivní bezpečnosti, karoserii a deformační zónou vozidla. Další část tvoří výběr nejpoužívanějšího vozidla v České republice a zjištění jeho parametrů. Dále řeším základním matematickým odvozením úhlové hodnoty rozhledu způsobené konstrukčními prvky vozidla. Výsledné zjištěné hodnoty se umístí do vybrané křižovatky a ověří dosažené výsledky.Subject of this thesis is to inquire structural parameters of vehicle in regards to sight limitation on intersection. Thesis is in general dealing with vehicle front columns, their effect on sight and evaluation of their width in regards to active safety of vehicle. In the first part I am dealing with passive and active safety issues, vehicle body and vehicle crumple zone. Next part of thesis contains selection of the most used vehicle in Czech republic and detection of its parameters. I am also dealing with basic mathematical derivation of sight angle caused by structural components of vehicle.     Automatic Analysis and Modeling of the Optic Nerve Target from Retinal Records

    Tato diplomová práce pojednává o tématu automatické analýzy a modelování terče zrakového nervu z retinálních záznamů. Algoritmus se skládá z několika kroků. Prvním krokem algoritmu je před-zpracování obrazu. V rámci druhé části algoritmu, věnující se segmentaci obrazu, byly navrhnuty dva algoritmy. Automatický algoritmus pro obrazovou segmentaci využívající umělé inteligence a algoritmus pro asistovanou segmentaci na snímcích majících nízké rozlišení. Další část algoritmu se zabývá detekcí hranic optického disku pomocí metody aktivních kontur. Posledním krokem algorit-mu je extrakce geometrických parametrů optického disku. Práce také obsahuje stručný úvod k one-mocnění retinopatie nedonošených, možnosti diagnostiky, léčby a screeningu. Popis retinální kame-ry Systém RetCam3, pomocí které byly získána pacientská data. Dále také úvod do embryonálního vývoje lidského oka. Diplomová práce obsahuje komplexní rešerši nových metod a trendů využíva-ných pro segmentaci a detekci optického disku z retinální záznamů.This master thesis is devoted to the theme of automatic segmentation and modeling of optic disc from retinal images. The proposed algorithm consists of several steps. First step of the algorithm is image pre-processing. Second part of algorithm is devoted to the image segmentation. For the image segmentation, this master thesis proposed two algorithms. First image segmentation algorithm is fully automatic and uses a swarm intelligence algorithm. The second image segmentation algorithm uses an assisted segmentaion and is determinated for images with low resolution. For the optic disc boundary detection the algorithm uses an active contour method. The last step of algorithm is de-voted to the extraction of optic disc geometrical features. Thesis contains a brief introduction to the illnes rethinopathy of prematurity, possibilities of diagnostic process, medical treatment and scree-ning. Next topic is devoted to the digital screening system RetCam3. Last part of theoretical text was devoted to the embryonic development of human eye. Parametry komunikace jsou navrženy na základě provedených průzkumů v terénu. Trasa je řešena od okružní křižovatky u hypermarketu Lidl po křížení na silnice II/479 směr Šenov. Jsou také řešeny úpravy nových křížení a sjezdy ke stávajícím pozemkům. Práce je provedena formou studie, kde v úvodu je řešena problematika návrhu, a jsou popsány základní poznatky vymezeného území. V další části jsou popsány samotné technické parametry návrhu a je proveden orientační odhad nákladů. Nedílnou součástí této práce je také výkresová část.Subject of this bachelor work is design of interconnection between road II/479 and U Nadrazi St. in Havirov, through U Zavor street. The road parameters are designed on surveys conducted in the field. The route is designed from the roundabout at hypermarket Lidl to crossroads on the road II/479 direction Senov. Adjustments of new crossroads and exits to the existing land are solved as well. Bachelor thesis is carried out as a study where at introduction is solved the design issues and describes the basic knowledge of the assigned area. The next section describes the actual technical parameters of the design and an indicative cost estimate. Trasa byla řešena od křižovatky Mánesova, Astronautů a Matuškova, až po napojení na ulici Dlouhá třída. Dále byly také řešeny úpravy napojení stávajících komunikací, zastávkových ploch a navržení nových parkovacích a odstavných stání. Práce byla provedena formou studie. V úvodu byla řešena problematika návrhu, a jsou popsány základní poznatky vymezeného území. V další části této práce byly popsány samotné technické parametry návrhu. Výsledný návrh byl vyjádřen ve výkresové části, která je nedílnou součástí této práce.The aim of this diploma work was research of the current traffic situation in Studentská and Manesova streets in Havířov. Route was solved from crossroad Mánesova, Astronautů and Matučkova to connection in Dlouhá třida street. Adjustments of connections to current route, bus stop areas and design of new parking were also solved. The work was made in the form of study. At the beginning problems of design were solved and basic knowledge of defined area were described. Technical parameters of design were described in the next section of this work. U radu je dokazana važnost specijalizacije hotelijerstva shodno potrebama i željama turista u vidu hotela kulturnog turizma sa specijaliziranim ambijentom kao što su heritage hoteli, difuzni hoteli, art i design hoteli. Na primjeru Hotela Kurija Janković iz Virovitice, potvrđeno je kako očuvanje i zaštita kulturnog dobra te prenamjena kulturnog zdanja u kategoriju heritage hotela doprinosi stvaranju novog proizvoda i razvoju lokalne zajednice. Tematizacijom u hotelijerstvu u vidu umjetnosti, dizajna, gastronomije, sporta stvara se prepoznatljivost na tržištu te zadovoljavanje sve raznovrsnijih potreba suvremenih turista.The main leading thought for sustainability, which the tourist management and the way of life of a today's tourist aim at, starts and finishes at the recognition and revitalisation of the cultural inheritance and the life tradition of a particilar destination. Nowadays, the hotel industry, as a dominant economic and service industry in tourism, strives for recognisability and brand development in terms of culture, design, food and experience. This paper proves the importance for the hotel industry to specialize according to needs and wishes of tourists in terms of cultural tourism by hotels with a special outlook and premises such as heritage hotels, diffused hotels, art and design hotels. At the example of Kurija Janković Hotel from Virovitica, it is proved how conservation and protection of cultural goods and a transformation of a cultural building into a cathegory of a heritage hotel can contribute to the creation of a new product and to the development of the local community. By theming in the hotel industry, through art, design, gastronomy and sport a recognisability at the market is created and more and more diversified needs of contemporary tourists are satisfied

    The Extent of Volatility Predictability Evaluation of forecasting accuracy dependent on time, distribution and model order

    Masteroppgave økonomi og administrasjon- Universitetet i Agder, 2015This thesis focuses on the accuracy and ability of out-of-sample volatility forecasting over different time horizons. Using data at daily frequency we forecast the future volatility over multiple time horizons (1, 3, 6, 9 and 12 months) and evaluate the goodness of forecasting by comparing the Naïve, ARCH, GARCH, EGARCH and GJR-GARCH models using the MSE and the Predictive Power (P). We include different probability distributions for the error terms in an attempt to improve the models accuracy. The research is conducted using three indices: FTSE 100, S&P 500 and the Hang Seng. We find that the goodness of forecasting accuracy decreases dramatically after the 3 month horizon and the selection of a more representative error distribution improves the accuracy of the short term forecasts. The results also show that the higher order GARCH models, beyond (1,1), do not improve the forecasting accuracy

    OPTICAL NERVE SEGMENTATION USING THE ACTIVE SHAPE METHOD

    The paper deals with the segmentation procedure for optical nerve localization and the consequent determination of geometrical parameters such as optical nerve area, radius and diameter. An extraction of these geometrical parameters is especially important for clinical practice particularly in the case where retinal lesions are present. On the base of the optical nerve extraction, we are capable of comparing it with area of retinal lesions. Via this approach it is possible to track time evaluation of retinal lesions. The proposed algorithm for segmentation of optical nerve area is performed within two main steps. In the first step, the active contour method is used specially for the localization of the optical nerve. This part of the algorithm generates mathematical model of the optical nerve in binary form. Consequently, on the base of this mathematical model of the optical nerve respective geometrical parameters are worked out for future comparison with retinal lesions. Image preprocessing is an integral part of the segmentation procedure, improving the observability of the optical nerve to ensure as relevant detection of the optical nerve as possible
