    A Look into Mindset, Motivation, and Self- Regulation: How to Foster Motivation and the Growth Mindset in the Classroom

    People are naturally curious beings, and the desire to learn is a natural and innate part of human nature. So why is it that by the time students reach their secondary education, their motivation is at an all time low? As students’ motivation is directly linked to their success and well-being, this problem is troubling. Many researchers have sought to find the cause of this shift, and what they have found is that although there cannot merely be a single determining factor to account for this shift, the vast majority of our schools and classrooms are promoting strategies, systems, and climates that undermine the natural desire to learn. To foster motivation and a growth mindset, teachers and administration alike must reevaluate the ways they attempt to control student behavior to allow students to feel autonomous, competent, and connected to others. However, current research is disconnected from the every day classroom; the practical means for teaching staff to adopt these philosophies is unmistakably absent. Therefore, this project proposes authentic structures, strategies, and lessons that provide teachers with the frameworks needed to promote these ideals within their schools and classrooms, thereby increasing student motivation and success

    Kiosks 21: a new role for information kiosks?

    Discusses and analyses the latest generation of information kiosks, Kiosks 21, which features information provision/promotion, interaction, transaction and relationships. In contrast to their task based predecessors, these kiosks focus on customer service delivery to ‘customers in context’. Five case studies of such kiosks located respectively in an airport, railway station, car rental base, hotel lobby, and shopping mall are analysed to demonstrate the way in which the kiosks are implemented to meet the differing requirements of customers in different contexts. Case studies are analysed in terms of kiosk design and location, user profile, information architecture, interface design, communication, and commerce. A range of areas for research and development are proposed.</p

    Inhibition of hypoxia-induced miR-155 radiosensitizes hypoxic lung cancer cells

    miR-155 is a prominent microRNA (miRNA) that regulates genes involved in immunity and cancer-related pathways. miR-155 is overexpressed in lung cancer, which correlates with poor patient prognosis. It is unclear how miR-155 becomes increased in lung cancers and how this increase contributes to reduced patient survival. Here, we show that hypoxic conditions induce miR-155 expression in lung cancer cells and trigger a corresponding decrease in a validated target, FOXO3A. Furthermore, we find that increased levels of miR-155 radioprotects lung cancer cells, while inhibition of miR-155 radiosensitizes these cells. Moreover, we reveal a therapeutically important link between miR-155 expression, hypoxia, and irradiation by demonstrating that anti-miR-155 molecules also sensitize hypoxic lung cancer cells to irradiation. Our study helps explain how miR-155 becomes elevated in lung cancers, which contain extensive hypoxic microenvironments, and demonstrates that inhibition of miR-155 may have important therapeutic potential as a means to radiosensitize hypoxic lung cancer cells

    Payment of trial participants can be ethically sound: Moving past a flat-rate

    The South African Medicines Control Council (MCC) policy that trial participants be paid a flat-rate of R150 per visit in clinical trials has been criticised in the press1,2; its affordability by non-industry funded trials questioned3; that it is an excessive or inappropriate reward4; or that it neglects critical factors like the design or nature of the study.3, 4 It might also be argued that this amount will act as an “undue inducement” for participants to enrol. In our view the principal problem with the MCC policy is that it violates justice in that participants are paid the same amount but do not do the same things or make the same financial sacrifices.5 It has been argued that it would be more appropriate to have a broad contextualised policy6 and that participants should be paid for their time and expenses.4 However, these recommendations have not been specified in the South African literature. To take these recommendations forward we apply two theoretical models of payment (Wage Payment and Reimbursement models) to operationalise payments for time, inconvenience and expenses (TIE). We recommend that participants be paid for their time at a rate similar to national unskilled labour rates, with increments for inconvenient procedures (set nationally), and that they be refunded their direct expenses. This is operationally complex but ethically sound. Our recommendations for stakeholders include that the NHREC assume control of payment norms and endorses payment for TIE. A new payment approach will struggle to overcome the legacy of a flat rate because of community expectations and researchers who have enjoyed a simple administrative procedure

    Patterns in Crew-Initiated Photography of Earth from ISS - Is Earth Observation a Salutogenic Experience?

    To provide for the well-being of crewmembers on future exploration missions, understanding how space station crewmembers handle the inherently stressful isolation and confinement during long-duration missions is important. A recent retrospective survey of previously flown astronauts found that the most commonly reported psychologically enriching aspects of spaceflight had to do with their Perceptions of Earth. Crewmembers onboard the International Space Station (ISS) photograph Earth through the station windows. Some of these photographs are in response to requests from scientists on the ground through the Crew Earth Observations (CEO) payload. Other photographs taken by crewmembers have not been in response to these formal requests. The automatically recorded data from the camera provides a dataset that can be used to test hypotheses about factors correlated with self-initiated crewmember photography. The present study used objective in-flight data to corroborate the previous questionnaire finding and to further investigate the nature of voluntary Earth-Observation activity. We examined the distribution of photographs with respect to time, crew, and subject matter. We also determined whether the frequency fluctuated in conjunction with major mission events such as vehicle dockings, and extra-vehicular activities (EVAs, or spacewalks), relative to the norm for the relevant crew. We also examined the influence of geographic and temporal patterns on frequency of Earth photography activities. We tested the hypotheses that there would be peak photography intensity over locations of personal interest, and on weekends. From December 2001 through October 2005 (Expeditions 4-11) crewmembers took 144,180 photographs of Earth with time and date automatically recorded by the camera. Of the time-stamped photographs, 84.5% were crew-initiated, and not in response to CEO requests. Preliminary analysis indicated some phasing in patterns of photography during the course of a mission (significant quadratic and trimodal models). There was also a small but significant increase in photo activity on the weekends. In contrast, fewer photos were taken during major station events and for a period of time immediately preceding those events. Data on photography patterns presented here represent a relatively objective group-level measure of Earth observing activities on ISS. Crew Earth Observations offers a self-initiated positive activity that may be important in salutogenesis (maintenance of well-being) of astronauts on long-duration missions. Consideration should be given to developing substitute activities for crewmembers in future exploration missions where there will not be the opportunity to look at Earth, such as on long-duration transits to Mars

    Improving identification & management of familial hypercholesterolaemia in primary care: pre- and post-intervention study

    Background and Aims: Familial hypercholesterolaemia (FH) is a major cause of premature heart disease but remains unrecognised in most patients. This study investigated if a systematic primary care-based approach to identify and manage possible FH improves recommended best clinical practice. Methods: Pre- and post-intervention study in six UK general practices (population 45,033) which invited patients with total cholesterol >7.5 mmol/L to be assessed for possible FH. Compliance with national guideline recommendations to identify and manage possible FH (repeat cholesterol; assess family history of heart disease; identify secondary causes and clinical features; reduce total & LDL-cholesterol; statin prescribing; lifestyle advice) was assessed by calculating the absolute difference in measures of care pre- and six months post-intervention. Results: The intervention improved best clinical practice in 118 patients consenting to assessment (of 831 eligible patients): repeat cholesterol test (+75.4%, 95% CI 66.9-82.3); family history of heart disease assessed (+35.6%, 95% CI 27.0-44.2); diagnosis of secondary causes (+7.7%, 95% CI 4.1-13.9), examining clinical features (+6.0%, 95% CI 2.9-11.7). For 32 patients diagnosed with possible FH using Simon-Broome criteria, statin prescribing significantly improved (18.8%, 95% CI 8.9-35.3) with non-significant mean reductions in cholesterol post-intervention (total: -0.16 mmol/L, 95% CI -0.78-0.46; LDL: -0.12 mmol/L, 95% CI -0.81-0.57). Conclusions: Within six months, this simple primary care intervention improved both identification and management of patients with possible FH, in line with national evidence-based guidelines. Replicating and sustaining this approach across the country could lead to substantial improvement in health outcomes for these individuals with very high cardiovascular risk

    Amputation-induced reactive oxygen species are required for successful Xenopus tadpole tail regeneration.

    Understanding the molecular mechanisms that promote successful tissue regeneration is critical for continued advancements in regenerative medicine. Vertebrate amphibian tadpoles of the species Xenopus laevis and Xenopus tropicalis have remarkable abilities to regenerate their tails following amputation, through the coordinated activity of numerous growth factor signalling pathways, including the Wnt, Fgf, Bmp, Notch and TGF-β pathways. Little is known, however, about the events that act upstream of these signalling pathways following injury. Here, we show that Xenopus tadpole tail amputation induces a sustained production of reactive oxygen species (ROS) during tail regeneration. Lowering ROS levels, using pharmacological or genetic approaches, reduces the level of cell proliferation and impairs tail regeneration. Genetic rescue experiments restored both ROS production and the initiation of the regenerative response. Sustained increased ROS levels are required for Wnt/β-catenin signalling and the activation of one of its main downstream targets, fgf20 (ref. 7), which, in turn, is essential for proper tail regeneration. These findings demonstrate that injury-induced ROS production is an important regulator of tissue regeneration

    p21-Activated Kinases Are Required for Transformation in a Cell-Based Model of Neurofibromatosis Type 2

    NF2 is an autosomal dominant disease characterized by development of bilateral vestibular schwannomas and other benign tumors in central nervous system. Loss of the NF2 gene product, Merlin, leads to aberrant Schwann cell proliferation, motility, and survival, but the mechanisms by which this tumor suppressor functions remain unclear. One well-defined target of Merlin is the group I family of p21-activated kinases, which are allosterically inhibited by Merlin and which, when activated, stimulate cell cycle progression, motility, and increased survival. Here, we examine the effect of Pak inhibition on cells with diminished Merlin function.Using a specific peptide inhibitor of group I Paks, we show that loss of Pak activity restores normal cell movement in cells lacking Merlin function. In addition, xenografts of such cells form fewer and smaller tumors than do cells without Pak inhibition. However, in tumors, loss of Pak activity does not reduce Erk or Akt activity, two signaling proteins that are thought to mediate Pak function in growth factor pathways.These results suggest that Pak functions in novel signaling pathways in NF2, and may serve as a useful therapeutic target in this disease

    Host Responses to Intestinal Microbial Antigens in Gluten-Sensitive Mice

    BACKGROUND AND AIMS: Excessive uptake of commensal bacterial antigens through a permeable intestinal barrier may influence host responses to specific antigen in a genetically predisposed host. The aim of this study was to investigate whether intestinal barrier dysfunction induced by indomethacin treatment affects the host response to intestinal microbiota in gluten-sensitized HLA-DQ8/HCD4 mice. METHODOLOGY/PRINCIPAL FINDINGS: HLA-DQ8/HCD4 mice were sensitized with gluten, and gavaged with indomethacin plus gluten. Intestinal permeability was assessed by Ussing chamber; epithelial cell (EC) ultra-structure by electron microscopy; RNA expression of genes coding for junctional proteins by Q-real-time PCR; immune response by in-vitro antigen-specific T-cell proliferation and cytokine analysis by cytometric bead array; intestinal microbiota by fluorescence in situ hybridization and analysis of systemic antibodies against intestinal microbiota by surface staining of live bacteria with serum followed by FACS analysis. Indomethacin led to a more pronounced increase in intestinal permeability in gluten-sensitized mice. These changes were accompanied by severe EC damage, decreased E-cadherin RNA level, elevated IFN-gamma in splenocyte culture supernatant, and production of significant IgM antibody against intestinal microbiota. CONCLUSION: Indomethacin potentiates barrier dysfunction and EC injury induced by gluten, affects systemic IFN-gamma production and the host response to intestinal microbiota antigens in HLA-DQ8/HCD4 mice. The results suggest that environmental factors that alter the intestinal barrier may predispose individuals to an increased susceptibility to gluten through a bystander immune activation to intestinal microbiota
