    This present article aims to share the results from theresearch and implementation of the teaching method called “WQs”,which is based on the theories found mainly in the studies about teachingthrough themes and the technological resources found in the computerand its programs. The research used the qualitative interpretative methodand the implementation was with two teams from High Schools studentsand with students majoring in Executive Secretariat. It was noticed theefficacy from the method for the foreign language teaching through thistechnological tool that has been spread in schools and academicenvironments.O presente artigo tem como objetivo compartilhar osresultados da pesquisa e implementação do método de ensinodenominado WQs, o qual utiliza-se principalmente dos fundamentosteórico-práticos encontrados nos estudos sobre ensino mediante tarefase de recursos tecnológicos encontrados com a utilização do computadore seus programas. A pesquisa utilizou o método qualitativo interpretativoe a implementação deu-se em duas turmas do ensino médio de umcolégio estadual e em turma de graduandos do curso de secretariadoexecutivo. Constatou-se a eficiência do método para o ensino de línguaestrangeira através desta ferramenta tecnológica que vem ocupando cadavez mais espaço no ambiente escolar e acadêmico

    Ciliate Communities Respond via Their Traits to a Wastewater Treatment Plant With a Combined UASB-Activated Sludge System

    Assessing functional diversity of communities is an efficient method to link community composition to ecosystem quality. Still, studies using functional traits of microeukaryote ciliate communities in biological wastewater treatment plants are lacking. The present work explores the functional diversity of the ciliate protist community in a wastewater treatment plant (WWTP) operating with a combined UASB-activated sludge system, and specifically to: 1) investigate the taxonomic and functional composition of the ciliate communities over time; 2) compare taxonomic and functional diversity indices with regard to its applicability in WWPS; 3) assess the relationship between the ciliate community's functional composition and the WWTPs temporal conditions; and 4) investigate the potential use of functional diversity as an indicator of WWTP efficiency. Totally, we recorded 21 ciliate species throughout 37 samplings. The number of species was low compared to other plants. Bacterivorous and flake-forming species were the main functional strategies found in the samples. The correlation between taxonomic and functional richness was significant, indicating a functionally redundant community. There was a correlation between the Simpson and Rao's quadratic entropy indexes suggesting that loss of taxonomic diversity leads to a loss of functional diversity. The homogeneity of the measured physical and chemical data led to functional homogenization and redundancy (homogenous CWM) of the ciliate community. The functional diversity is positively correlated with parameters of removal efficiency, indicating a promising application in WWTPs. Future studies will broaden knowledge on functional diversity in biological wastewater treatment systems, this being a first step with the unprecedented application of this methodology in artificial ecosystems.Peer reviewe

    A loss-of-function NCSTN mutation associated with familial Dowling Degos disease and hidradenitis suppurativa

    Dowling Degos disease (DDD) is a rare autosomal dominant genodermatosis characterized by acquired, slowly progressive reticulated pigmented lesions primarily involving flexural skin areas. Mutations in KRT5, POGLUT-1 and POFUT-1 genes have been associated with DDD, and loss-of-function mutations in PSENEN, a subunit of the gamma-secretase complex, were found in patients presenting with DDD or DDD comorbid with hidradenitis suppurativa (HS). A nonsense mutation in NCSTN, another subunit of the gamma-secretase, was already described in a patient suffering from HS and DDD but whether NCSTN could be considered a novel gene for DDD is still debated. Here, we enrolled a four-generation family with HS and DDD. Through Whole Exome Sequencing (WES) we identified a novel nonsense mutation in the NCSTN gene in all the affected family members. To study the impact of this variant, we isolated outer root sheath cells from patients' hair follicles. We showed that this variant leads to a premature stop codon, activates a nonsense-mediated mRNA decay, and causes NCSTN haploinsufficiency in affected individuals. In fact, cells treated with gentamicin, a readthrough agent, had the NCSTN levels corrected. Moreover, we observed that this haploinsufficiency also affects other subunits of the gamma-secretase complex, possibly causing DDD. Our findings clearly support NCSTN as a novel DDD gene and suggest carefully investigating this co-occurrence in HS patients carrying a mutation in the NCSTN gene

    Vitamin K Supplementation Modulates Bone Metabolism and Ultra-Structure of Ovariectomized Mice

    Background/Aims: Osteoporosis is a bone metabolic disease that affects mostly post-menopausal women. There has been shown that vitamin K (VK) supplementation during menopause may decrease bone loss as well as risk of bone breaking. Aiming to clarify the beneficial role of VK in bone metabolism during menopause, we investigated mineral metabolism and bone ultrastructure of ovariectomized (OVX) mice. Methods: To determine the effects chronic use of VK in bone structure and mineral metabolism in OVX mice, we used several methods, such as DXA, µCTScan, and SEM as well as biomolecular techniques, such as ELISA and qRT-PCR. In addition, complete analysis of serum hormonal and other molecules associated to bone and lipid metabolism were evaluated overview the effects of VK in menopause murine model. Results: VK treatment significantly affects Pi metabolism independently of OVX, changing Pi plasma, urinary output, balance, and Pi bone mass. Interestingly, VK also increased VLDL in mice independently of castration. In addition, VK increased compact bone mass in OVX mice when we evaluated it by DXA, histomorphometry, µCTScanning. VK increased bone formation markers, osteocalcin, HYP- osteocalcin, and AP whereas it decreased bone resorption markers, such as urinary DPD/creatinine ratio and plasmatic TRAP. Surprisingly, SEM images revealed that VK treatment led to amelioration of microfractures observed in OVX untreated controls. In addition, SHAM operated VK treated mice exhibited higher number of migrating osteoblasts and in situ secretion of AP. OVX led to decreased to in situ secretion of AP that was restored by VK treatment. Moreover, VK treatment increased mRNA expression of bone Calbindin 28KDa independently of OVX. Conclusion: VK treatment in OVX mice exhibited beneficial effects on bone ultrastructure, mostly by altering osteoblastic function and secretion of organic bone matrix. Therefore, VK could be useful to treat osteopenic/osteoporotic patients

    The role of protected areas in the avoidance of anthropogenic conversion in a high pressure region : a matching method analysis in the core region of the brazilian cerrado

    Global efforts to avoid anthropogenic conversion of natural habitat rely heavily on the establishment of protected areas. Studies that evaluate the effectiveness of these areas with a focus on preserving the natural habitat define effectiveness as a measure of the influence of protected areas on total avoided conversion. Changes in the estimated effectiveness are related to local and regional differences, evaluation methods, restriction categories that include the protected areas, and other characteristics. The overall objective of this study was to evaluate the effectiveness of protected areas to prevent the advance of the conversion of natural areas in the core region of the Brazil’s Cerrado Biome, taking into account the influence of the restriction degree, governmental sphere, time since the establishment of the protected area units, and the size of the area on the performance of protected areas. The evaluation was conducted using matching methods and took into account the following two fundamental issues: control of statistical biases caused by the influence of covariates on the likelihood of anthropogenic conversion and the non-randomness of the allocation of protected areas throughout the territory (spatial correlation effect) and the control of statistical bias caused by the influence of auto-correlation and leakage effect. Using a sample design that is not based on ways to control these biases may result in outcomes that underestimate or overestimate the effectiveness of those units. The matching method accounted for a bias reduction in 94–99% of the estimation of the average effect of protected areas on anthropogenic conversion and allowed us to obtain results with a reduced influence of the auto-correlation and leakage effects. Most protected areas had a positive influence on the maintenance of natural habitats, although wide variation in this effectiveness was dependent on the type, restriction, governmental sphere, size and age group of the unit

The total ciliate fauna of the activated sludge of the Picarro Wastewater Treatment Plant (Picarro WWTP) was composed of 36 morphospecies belonging to 33 genera. These included 21 species observed in the activated sludge samples on the day of collection and 15 species found in cultures. The activated sludge of the Picarro WWTP contained a diversified ciliate community composed mainly of indicator organisms. The most frequently occurring morphospecies were Aspidisca cicada, Vorticella spp., Gastronauta aloisi, Acineria uncinata, and Epistylis plicatilis complex. These results showed that satisfactory operating conditions prevailed at the Picarro WWTP.     Checklist of ciliated protozoa from surface water and sediment samples of Atibaia River, Campinas, São Paulo (Southeast Brazil).

    The knowledge of the diversity of ciliated protozoa in an aquatic ecosystem is an important information that can serve as a tool in the environmental quality management. A survey of the ciliated protozoa of the Atibaia River, Campinas, São Paulo was done during 24 months. Surface water and sediment samples were collected monthly from two sampling points. Qualitative analyses of ciliates were performed in vivo and by protargol staining. A total of 66 taxa belonging to 55 genera were identified. This is the first study in Atibaia River and, as this is an impacted water body, these informations can be very useful for conservation
