    Feeling Seen: A Pathway to Transformation

    Chronic exposure to racial indignities can engender a subjective sense of invisibility, in which an individual feels that the dominant culture fails to recognize one’s worth, abilities, and talents. The sense of feeling unseen can permeate myriad aspects of the lived experience and negatively impact well-being. Using the case of an African American male in therapy with an African American female psychotherapist, this article presents how implicit and explicit acts of recognition of the patient and acknowledgment of race, integrated into a change-oriented and experiential psychotherapeutic process can facilitate transformational experiences. This case study seeks to highlight the importance of therapeutic alliance and patient perception of therapist empathy, which may contribute to enhanced well-being of African American men seeking psychotherapy

    Developmental and Individual Differences in Visual Word Search

    Neurodevelopmental disorders and increasing age are marked by abnormal functioning of executive functions, especially working memory. Cognitive training programs have been developed to attempt to strengthen the underlying cognitive capacities that foster executive functioning abilities. These interventions have demonstrated variable results in populations of older adults and children. Given the wide range of previous findings, it is possible that a third variable is implicated in performance on these types of interventions. Additionally, given the active role of visual processing in executive function (EF) training programs and its functional relationship with working memory, it is hypothesized that visual processing could co-vary with response to training interventions. Moreover, with the potential clinical impact for such interventions for rural areas, understanding the mechanisms behind these interventions and identifying possible non-responders would be of benefit. Thus, the current study aimed to determine if individual and developmental factors impact performance on a demanding working memory (WM) task in a way that could explain past findings and offer recommendations for treatment prediction. Using an asynchronous, online design, a diverse sample of adult and child participants completed three cognitive tasks, including measures of working memory, and a novel cognitive task. Performance on a digit span task, a measure of WM, was significantly correlated with performance efficacy on a novel visual search task. Children also performed faster than adults on the search task. Individuals from rural areas also performed better than urban individuals on digit span, and the novel search task. The present study demonstrates evidence for the role of individual, developmental, and geographic factors in performance on a novel cognitive task. As such, it is possible that these factors have confounded previous findings; future studies would extend the literature by determining if these factors can be used to predict treatment response and thus treatment appropriateness for various groups

    Increasing Character Strength Knowledge, Interest, and Skill: Preliminary Evidence for a Collaborative and Multimethod Assessment Procedure

    Introduction: The study’s objective was to evaluate whether a qualitative, collaborative, and multimethod assessment protocol increased reports of character strength interest, knowledge, and perceived skills. Methods: Thirty-two participants completed three phases of data collection. Participants were first screened for well-being, which was used as an auxiliary covariate to order participants into experimental conditions. Selected participants were randomly assigned to a control or collaborative and multimethod assessment (card sort × qualitative interview) condition. Participants completed pre- and post-measures of strength interest, knowledge, and perceived skill. In the final phase, second phase participants were invited to report on strength-related outcomes 24 h post-administration using an online survey. Results: A series of 2 (Assessment Condition) × 3 (Time) mixed ANOVAs were analyzed. Results revealed a significant assessment condition by time interaction for strength knowledge and perceived skill. Participants in the collaborative and multimethod assessment condition reported higher strength knowledge and perceived skills compared to control participants. These effects were maintained for 24 h. Conclusion: The findings offer preliminary yet sizable support for using collaborative and multimethod assessment procedures to increase strength knowledge and perceived skill. Because of the qualitative, collaborative, and individualized nature of our assessment protocol, the findings offer a low-cost and contextually bound pathway to increase strength-based outcomes

    Luzp4 defines a new mRNA export pathway in cancer cells

    Cancer testis antigens (CTAs) represented a poorly characterized group of proteins whose expression is normally restricted to testis but are frequently up-regulated in cancer cells. Here we show that one CTA, Luzp4, is an mRNA export adaptor. It associates with the TREX mRNA export complex subunit Uap56 and harbours a Uap56 binding motif, conserved in other mRNA export adaptors. Luzp4 binds the principal mRNA export receptor Nxf1, enhances its RNA binding activity and complements Alyref knockdown in vivo. Whilst Luzp4 is up-regulated in a range of tumours, it appears preferentially expressed in melanoma cells where it is required for growth

    Mind-modelling with corpus stylistics in David Copperfield

    We suggest an innovative approach to literary discourse by using corpus linguistic methods to address research questions from cognitive poetics. In this article, we focus on the way that readers engage in mind-modelling in the process of characterisation. The article sets out our cognitive poetic model of characterisation that emphasises the continuity between literary characterisation and real-life human relationships. The model also aims to deal with the modelling of the author’s mind in line with the modelling of the minds of fictional characters. Crucially, our approach to mind-modelling is text-driven. Therefore we are able to employ corpus linguistic techniques systematically to identify textual patterns that function as cues triggering character information. In this article, we explore our understanding of mind-modelling through the characterisation of Mr. Dick from David Copperfield by Charles Dickens. Using the CLiC tool (Corpus Linguistics in Cheshire) developed for the exploration of 19th-century fiction, we investigate the textual traces in non-quotations around this character, in order to draw out the techniques of characterisation other than speech presentation. We show that Mr. Dick is a thematically and authorially significant character in the novel, and we move towards a rigorous account of the reader’s modelling of authorial intention

    Live Imaging of Mitosomes and Hydrogenosomes by HaloTag Technology

    Hydrogenosomes and mitosomes represent remarkable mitochondrial adaptations in the anaerobic parasitic protists such as Trichomonas vaginalis and Giardia intestinalis, respectively. In order to provide a tool to study these organelles in the live cells, the HaloTag was fused to G. intestinalis IscU and T. vaginalis frataxin and expressed in the mitosomes and hydrogenosomes, respectively. The incubation of the parasites with the fluorescent Halo-ligand resulted in highly specific organellar labeling, allowing live imaging of the organelles. With the array of available ligands the HaloTag technology offers a new tool to study the dynamics of mitochondria-related compartments as well as other cellular components in these intriguing unicellular eukaryotes

    The Minimal Proteome in the Reduced Mitochondrion of the Parasitic Protist Giardia intestinalis

    The mitosomes of Giardia intestinalis are thought to be mitochondria highly-reduced in response to the oxygen-poor niche. We performed a quantitative proteomic assessment of Giardia mitosomes to increase understanding of the function and evolutionary origin of these enigmatic organelles. Mitosome-enriched fractions were obtained from cell homogenate using Optiprep gradient centrifugation. To distinguish mitosomal proteins from contamination, we used a quantitative shot-gun strategy based on isobaric tagging of peptides with iTRAQ and tandem mass spectrometry. Altogether, 638 proteins were identified in mitosome-enriched fractions. Of these, 139 proteins had iTRAQ ratio similar to that of the six known mitosomal markers. Proteins were selected for expression in Giardia to verify their cellular localizations and the mitosomal localization of 20 proteins was confirmed. These proteins include nine components of the FeS cluster assembly machinery, a novel diflavo-protein with NADPH reductase activity, a novel VAMP-associated protein, and a key component of the outer membrane protein translocase. None of the novel mitosomal proteins was predicted by previous genome analyses. The small proteome of the Giardia mitosome reflects the reduction in mitochondrial metabolism, which is limited to the FeS cluster assembly pathway, and a simplicity in the protein import pathway required for organelle biogenesis
