    Effects of Mountain Rivers Cascade Hydropower Stations on Water Ecosystems

As the main way of water resources and hydropower development, the construction of cascade hydropower stations, while meeting the requirements of water resources utilization for social development, has also brought adverse effects on river ecosystems. Therefore, the impact of the construction of cascade hydropower stations on mountainous river ecosystems, where the minimum ecological flow of rivers must be ensured and reviewed. In addition, this paper proposed the deficiencies and outlooks for cascade hydropower stations based on previous research results

Guangxi is one of the provinces with relatively rich water resources in China, while the ecological water network exists deficiencies and faces challenges. The current situation and defects of ecological water network in Guangxi province will be discussed. By studying the experience of the establishing and the preserve of ecological water network in various regions at home and abroad, some suggestions and targeted measures will be mentioned for a better ecological water network in Guangxi

    Untargeted Safety Pharmacology Screen of Blood-Activating and Stasis-Removing Patent Chinese Herbal Medicines Identified Nonherbal Ingredients as a Cause of Organ Damage in Experimental Models

    Blood activation and stasis removal from circulation is a central principle for treatment of syndromes related to cerebral and cardiovascular diseases in Chinese herbal medicine. However, blood-activating and stasis-removing patent Chinese herbal medicine (BASR-pCHM) widely used with or without prescription in China and elsewhere are highly variable in composition and manufacture standard, making their safety assessment a challenging task. We proposed that an integrated evaluation of multiple toxicity parameters of BASR-pCHM would provide critical reference and guidelines for their safe clinical application. Examination of standardized extracts from 58 compound BASR-pCHM in vivo in VEGFR2-luc mice and in vitro in cardiac, renal, and hepatic cells identified Naoluotong capsule (NLTC) as a potent organ/cell damage inducer. Composition analysis revealed that NLTC was the one that contained nonherbal ingredients among the BASR-pCHM collection. In vivo and in vitro experiments confirmed that NLTC, as well as its chemical supplement tolperisone hydrochloride, caused organ and cell damage by reducing cell viability, mitochondrial mass/activity, while the NLTC herbal components did not. Taken together, our study showed that safety evaluation of patent herbal medicines already on market is still necessary and urgently needed. In addition, chemical/herbal interactions should be considered as an important contributor of potential toxicity when evaluating the safety of herbal medicine

    Near-atomic, non-icosahedrally averaged structure of giant virus Paramecium bursaria chlorella virus 1

    Giant viruses are a large group of viruses that infect many eukaryotes. Although components that do not obey the overall icosahedral symmetry of their capsids have been observed and found to play critical roles in the viral life cycles, identities and high-resolution structures of these components remain unknown. Here, by determining a near-atomic-resolution, five-fold averaged structure of Parameciumbursaria chlorella virus 1, we unexpectedly found the viral capsid possesses up to five major capsid protein variants and a penton protein variant. These variants create varied capsidmicroenvironments for the associations of fibers, a vesicle, and previously unresolved minor capsid proteins. Our structure reveals the identities and atomic models of the capsid components that do not obey the overall icosahedral symmetry and leads to a model for how these components are assembled and initiate capsid assembly, and this model might be applicable to many other giant viruses

We have independently developed a novel statistical time delay dynamic model on the basis of the distribution models from CCDC. Based only on the numbers of confirmed cases in different regions in China, the model can clearly reveal that the containment of the epidemic highly depends on early and effective isolation. We apply the model on the epidemic in Japan and conclude that there could be a rapid outbreak in Japan if no effective quarantine measures are carried out immediately

    The potency of the fs260 connexin43 mutant to impair keratinocyte differentiation is distinct from other disease-linked connexin43 mutants

    Although there are currently 62 mutants of Cx43 (connexin43) that can cause ODDD (oculodentodigital dysplasia), only two mutants have also been reported to cause palmar plantar hyperkeratosis. To determine how mutants of Cx43 can lead to this skin disease, REKs (rat epidermal keratinocytes) were engineered to express an ODDD-associated Cx43 mutant always linked to skin disease (fs260), an ODDD-linked Cx43 mutant which has been reported to sometimes cause skin disease (fs230), Cx43 mutants which cause ODDD only (G21R, G138R), a mouse Cx43 mutant linked to ODDD (G60S), a non-disease-linked truncated Cx43 mutant that is trapped in the endoplasmic reticulum (Δ244*) or full-length Cx43. When grown in organotypic cultures, of all the mutants investigated, only the fs260-expressing REKs consistently developed a thinner stratum corneum and expressed lower levels of Cx43, Cx26 and loricrin in comparison with REKs overexpressing wild-type Cx43. REKs expressing the fs260 mutant also developed a larger organotypic vital layer after acetone-induced injury and exhibited characteristics of parakeratosis. Collectively, our results suggest that the increased skin disease burden exhibited in ODDD patients harbouring the fs260 mutant is probably due to multiple additive effects cause by the mutant during epidermal differentiation

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    In the beginning of the 20th century, there were 87 million elderly people aged 65 and older. However, by the years 2030 and 2050, elderly people in China will rapidly growth to 243–252 million and 352–398 million, respectively. (Zeng, Y., 2010) The population aging transition will take place in China with this staggering rapidity, compared to European societies. Predictably, as for this aging pressure, Chinese society still needs more time to react to it. Ageing is not only a challenge for the society but also a big challenge for design. Along with the increasing numbers of the elderly, they cannot be ignored by design any more. The discussion here aspires to move the design mindset beyond accessibility or in other words ‘Design for disability’, and introduce ‘Design for capability’ as a process of social innovation. In the paper, the design research will focus on meta-design, as well as participatory design and social innovation as auxiliary research, for designing a ‘seed’ as a change agent. The ‘seed’, as a meta-design solution, can be described as ‘a shared design endeavor aimed at sustaining emergence, evolution and adaptation’ (Giaccardi, E., 2005). It offers a framework for both designers and users to change original mindsets in the practice. (Giaccardi, E., 2005) Furthermore, the following question will be carried through the whole research: how to reposition ourselves as designers on the intersection of meta-design, design for social innovation and participatory design? What is the design approach to generate tools that can encourage inactive elderly citizens as ‘passive receivers’ to transform as ‘active participants’? How can the tools studied here contribute in a synergic relationship within stakeholders as a mean to make elderly citizens’ urban living more sustainable in terms of participating, learning and expressing actively
