    Asymmetric Policy on Border Management

    The management of state borders between Indonesia and PNG holds potential problems in the form of differences in socio-economic conditions between the two countries. While socially and culturally, people in the two countries have close kinship ties. This paper intends to recommend an asymmetric policy for managing national border areas, especially between Merauke and PNG. The conclusion of this study is that the adoption of the ChinaVietnam border management model can be applied in the management of the Indonesia-PNG border. This adoption is accompanied by several strategies in the form of: acceleration of construction of facilities and infrastructure, strengthening the socio-economic conditions of border communities, human resource development and strengthening of good institutional aspects.Peluncuran dana desa di Papua selalu meningkat setiap tahun yang diberikan oleh pemerintah, tetapi di sisi lain memunculkan masalah baru terkait pengelolaan keuangan dan transparansi. Studi ini bertujuan untuk memberikan pemikiran baru tentang transparansi keuangan dana desa yang cocok untuk wilayah Papua. Metode yang digunakan adalah studi literatur, dan hasil penelitian ini adalah transparansi yang harus dilakukan, yaitu penyediaan dan akses informasi yang jelas tentang perencanaan, prosedur pelaksanaan dan akuntabilitas (papan reklame, situs web, Facebook), adanya musyawarah yang melibatkan masyarakat dan masyarakat adat / Ketua Adat, Keterbukaan proses manajemen, Pengawasan dilakukan oleh seluruh komunitas, Transparansi informasi tentang dokumen pengelolaan keuangan Desa, sosialisasi dan pelatihan terus menerus dari pemerintah ke Desa untuk pengelolaan Dana Desa yang efektif, dan Bantuan Dana Desa sebenarnya direkrut dari orang-orang yang memiliki integritas diri yang sangat tinggi

    Call Me Caitlyn: Making and making over the 'authentic' transgender body in Anglo-American popular culture

    A conception of transgender identity as an ‘authentic’ gendered core ‘trapped’ within a mismatched corporeality, and made tangible through corporeal transformations, has attained unprecedented legibility in contemporary Anglo-American media. Whilst pop-cultural articulations of this discourse have received some scholarly attention, the question of why this 'wrong body' paradigm has solidified as the normative explanation for gender transition within the popular media remains underexplored. This paper argues that this discourse has attained cultural pre-eminence through its convergence with a broader media and commercial zeitgeist, in which corporeal alteration and maintenance are perceived as means of accessing one’s ‘authentic’ self. I analyse the media representations of two transgender celebrities: Caitlyn Jenner and Nadia Almada, alongside the reality TV show TRANSform Me, exploring how these women’s gender transitions have been discursively aligned with a cultural imperative for all women, cisgender or trans, to display their authentic femininity through bodily work. This demonstrates how established tropes of authenticity-via-bodily transformation, have enabled transgender to become culturally legible through the wrong body trope. Problematically, I argue, this process has worked to demarcate ideals of ‘acceptable’ transgender subjectivity: self-sufficient, normatively feminine, and eager to embrace the possibilities for happiness and social integration provided by the commercial domain

    Bone Marrow–generated Dendritic Cells Pulsed with Tumor Extracts or Tumor RNA Induce Antitumor Immunity against Central Nervous System Tumors

    Recent studies have shown that the brain is not a barrier to successful active immunotherapy that uses gene-modified autologous tumor cell vaccines. In this study, we compared the efficacy of two types of vaccines for the treatment of tumors within the central nervous system (CNS): dendritic cell (DC)-based vaccines pulsed with either tumor extract or tumor RNA, and cytokine gene–modified tumor vaccines. Using the B16/F10 murine melanoma (B16) as a model for CNS tumor, we show that vaccination with bone marrow–generated DCs, pulsed with either B16 cell extract or B16 total RNA, can induce specific cytotoxic T lymphocytes against B16 tumor cells. Both types of DC vaccines were able to protect animals from tumors located in the CNS. DC-based vaccines also led to prolonged survival in mice with tumors placed before the initiation of vaccine therapy. The DC-based vaccines were at least as effective, if not more so, as vaccines containing B16 tumor cells in which the granulocytic macrophage colony-stimulating factor gene had been modified. These data support the use of DC-based vaccines for the treatment of patients with CNS tumors

    Authenticity, validation and sexualisation on Grindr: an analysis of trans women’s accounts

    The socio-historic sexualisation of transgender identities is reported to have disaffirming consequences for the broad trans community, and for trans women in particular. Given trans people’s increasing use of socio-sexual ‘hook-up’ apps, this paper looks at trans women’s talk of self/other identifications in relation to their regular use of Grindr. Eight semi-structured in-depth interviews were conducted with London-based women who identified as trans* in some way. A Foucauldian-informed discourse analysis highlights intersecting frames of trans authenticity, validation and sexualisation. Within these frames, trans women can be variously positioned in gendered and sexualised ways. Specifically, a discourse of trans authenticity is seen to involve the marking out of an identificatory truth that is situated in culturally acceptable and hence de-sexualised womanhood, while a competing discourse of trans validation involves an ambiguity and eroticism that can serve to reimagine this truth. Trans subjectivities can thus consist of a desire for authentic (gendered and non-sexualised) selfhood, on the one hand, and self-affirming ambiguity and sexualisation on the other. That trans women can construct ambivalent relationships with trans-sexualisation discourse highlights the limitation of anti-sexualisation advocacy and implications for supporting trans sexualities are considered

    Evidence for a Transport-Trap Mode of Drosophila melanogaster gurken mRNA Localization

    The Drosophila melanogaster gurken gene encodes a TGF alpha-like signaling molecule that is secreted from the oocyte during two distinct stages of oogenesis to define the coordinate axes of the follicle cell epithelium that surrounds the oocyte and its 15 anterior nurse cells. Because the gurken receptor is expressed throughout the epithelium, axial patterning requires region-specific secretion of Gurken protein, which in turn requires subcellular localization of gurken transcripts. The first stage of Gurken signaling induces anteroposterior pattern in the epithelium and requires the transport of gurken transcripts from nurse cells into the oocyte. The second stage of Gurken signaling induces dorsovental polarity in the epithelium and requires localization of gurken transcripts to the oocyte's anterodorsal corner. Previous studies, relying predominantly on real-time imaging of injected transcripts, indicated that anterodorsal localization involves transport of gurken transcripts to the oocyte's anterior cortex followed by transport to the anterodorsal corner, and anchoring. Such studies further indicated that a single RNA sequence element, the GLS, mediates both transport steps by facilitating association of gurken transcripts with a cytoplasmic dynein motor complex. Finally, it was proposed that the GLS somehow steers the motor complex toward that subset of microtubules that are nucleated around the oocyte nucleus, permitting directed transport to the anterodorsal corner. Here, we re-investigate the role of the GLS using a transgenic fly assay system that includes use of the endogenous gurken promoter and biological rescue as well as RNA localization assays. In contrast to previous reports, our studies indicate that the GLS is sufficient for anterior localization only. Our data support a model in which anterodorsal localization is brought about by repeated rounds of anterior transport, accompanied by specific trapping at the anterodorsal cortex. Our data further indicate that trapping at the anterodorsal corner requires at least one as-yet-unidentified gurken RLE

    This article considers how sex and gender – as conceptual categories and as a lived experience – are subject to contestation and renegotiation in the contemporary UK. Exploring gendered shifts through the lenses of identity and embodiment, the article captures key moments where certainties have been undone within feminist and transgender thought and activism. Yet such fissures resound with calls for a return to traditional understandings of the sexed body. The article pays particular attention to debates within feminism around transgender issues, and sketches out a climate of transgender moral panic whereby conservative thinkers and some feminist activists are joining forces with the aim of resurrecting gender binaries

    An Allograft Glioma Model Reveals the Dependence of Aquaporin-4 Expression on the Brain Microenvironment

    Aquaporin-4 (AQP4), the main water channel of the brain, is highly expressed in animal glioma and human glioblastoma in situ. In contrast, most cultivated glioma cell lines don’t express AQP4, and primary cell cultures of human glioblastoma lose it during the first passages. Accordingly, in C6 cells and RG2 cells, two glioma cell lines of the rat, and in SMA mouse glioma cell lines, we found no AQP4 expression. We confirmed an AQP4 loss in primary human glioblastoma cell cultures after a few passages. RG-2 glioma cells if grafted into the brain developed AQP4 expression. This led us consider the possibility of AQP4 expression depends on brain microenvironment. In previous studies, we observed that the typical morphological conformation of AQP4 as orthogonal arrays of particles (OAP) depended on the extracellular matrix component agrin. In this study, we showed for the first time implanted AQP4 negative glioma cells in animal brain or flank to express AQP4 specifically in the intracerebral gliomas but neither in the extracranial nor in the flank gliomas. AQP4 expression in intracerebral gliomas went along with an OAP loss, compared to normal brain tissue. AQP4 staining in vivo normally is polarized in the astrocytic endfoot membranes at the glia limitans superficialis and perivascularis, but in C6 and RG2 tumors the AQP4 staining is redistributed over the whole glioma cell as in human glioblastoma. In contrast, primary rat or mouse astrocytes in culture did not lose their ability to express AQP4, and they were able to form few OAPs

    Transphobic ‘honour’-based abuse : a conceptual tool

    This paper proposes that an understanding of transphobic ‘honour’-based abuse can be employed as a conceptual tool to explore trans people’s experiences of familial abuse. This conception has evolved by connecting a sociology of shame, Goffman's work on stigma and 'honour'-based ideology. The discussion draws upon findings of a qualitative study which explored trans people's experiences of domestic violence and abuse. Narrative interviews were undertaken with fifteen trans people who had either experienced abuse or whose perceptions were informed experientially through their support of others. Transcripts were analysed using the Listening Guide. Findings indicate that trans people can experience abuse as a result of a family's perceptions of shame and stigma. This paper offers a novel way of conceptualising trans people's experiences of family-based abuse, but it also holds potential for understanding other relational contexts, for example, those of intimate partnerships

    Ganglioside composition and histology of a spontaneous metastatic brain tumour in the VM mouse

    Glycosphingolipid abnormalities have long been implicated in tumour malignancy and metastasis. Gangliosides are a family of sialic acid-containing glycosphingolipids that modulate cell–cell and cell–matrix interactions. Histology and ganglioside composition were examined in a natural brain tumour of the VM mouse strain. The tumour is distinguished from other metastatic tumour models because it arose spontaneously and metastasizes to several organs including brain and spinal cord after subcutaneous inoculation of tumour tissue in the flank. By electron microscopy, the tumour consisted of cells (15 to 20 μm in diameter) that had slightly indented nuclei and scant cytoplasm. The presence of smooth membranes with an absence of junctional complexes was a characteristic ultrastructural feature. No positive immunostaining was found for glial or neuronal markers. The total ganglioside sialic acid content of the subcutaneously grown tumour was low (12.6 ± 0.9 μg per 100 mg dry wt, n= 6 separate tumours) and about 70% of this was in the form of N-glycolylneuraminic acid. In contrast, the ganglioside content of the cultured VM tumour cells was high (248.4 ± 4.4 μg, n= 3) and consisted almost exclusively of N-acetylneuraminic acid. The ganglioside pattern of the tumour grown subcutaneously was complex, while GM3, GM2, GM1, and GD1a were the major gangliosides in the cultured tumour cells. This tumour will be a useful natural model for evaluating the role of gangliosides and other glycolipids in tumour cell invasion and metastasis. © 2001 Cancer Research Campaign http://www.bjcancer.co
