The Hyphal-Associated Adhesin and Invasin Als3 of Candida albicans Mediates Iron Acquisition from Host Ferritin
Iron sequestration by host iron-binding proteins is an important mechanism of resistance to microbial infections. Inside oral epithelial cells, iron is stored within ferritin, and is therefore not usually accessible to pathogenic microbes. We observed that the ferritin concentration within oral epithelial cells was directly related to their susceptibility to damage by the human pathogenic fungus, Candida albicans. Thus, we hypothesized that host ferritin is used as an iron source by this organism. We found that C. albicans was able to grow on agar at physiological pH with ferritin as the sole source of iron, while the baker's yeast Saccharomyces cerevisiae could not. A screen of C. albicans mutants lacking components of each of the three known iron acquisition systems revealed that only the reductive pathway is involved in iron utilization from ferritin by this fungus. Additionally, C. albicans hyphae, but not yeast cells, bound ferritin, and this binding was crucial for iron acquisition from ferritin. Transcriptional profiling of wild-type and hyphal-defective C. albicans strains suggested that the C. albicans invasin-like protein Als3 is required for ferritin binding. Hyphae of an Îals3 null mutant had a strongly reduced ability to bind ferritin and these mutant cells grew poorly on agar plates with ferritin as the sole source of iron. Heterologous expression of Als3, but not Als1 or Als5, two closely related members of the Als protein family, allowed S. cerevisiae to bind ferritin. Immunocytochemical localization of ferritin in epithelial cells infected with C. albicans showed ferritin surrounding invading hyphae of the wild-type, but not the Îals3 mutant strain. This mutant was also unable to damage epithelial cells in vitro. Therefore, C. albicans can exploit iron from ferritin via morphology dependent binding through Als3, suggesting that this single protein has multiple virulence attributes
Radio source calibration for the VSA and other CMB instruments at around 30 GHz
Accurate calibration of data is essential for the current generation of CMB
experiments. Using data from the Very Small Array (VSA), we describe procedures
which will lead to an accuracy of 1 percent or better for experiments such as
the VSA and CBI. Particular attention is paid to the stability of the receiver
systems, the quality of the site and frequent observations of reference
sources. At 30 GHz the careful correction for atmospheric emission and
absorption is shown to be essential for achieving 1 percent precision. The
sources for which a 1 percent relative flux density calibration was achieved
included Cas A, Cyg A, Tau A and NGC7027 and the planets Venus, Jupiter and
Saturn. A flux density, or brightness temperature in the case of the planets,
was derived at 33 GHz relative to Jupiter which was adopted as the fundamental
calibrator. A spectral index at ~30 GHz is given for each. Cas A,Tau A, NGC7027
and Venus were examined for variability. Cas A was found to be decreasing at
percent per year over the period March 2001 to August 2004.
In the same period Tau A was decreasing at percent per year. A
survey of the published data showed that the planetary nebula NGC7027 decreased
at percent per year over the period 1967 to 2003. Venus showed
an insignificant ( percent) variation with Venusian illumination.
The integrated polarization of Tau A at 33 GHz was found to be

Analysis of a Method to Estimate Chlorophyll-a Concentration from Irradiance Measurements at Varying Depths
A model to estimate chlorophyll-a concentration and yellow substance absorption at 440 nm from irradiance measurements made at varying depths is examined. The derivation of the model, requiring irradiance measurements at three wavebands, is presented and tested on data collected in oligotrophic (with low chlorophyll concentrations) waters and in coastal waters (with both high and low chlorophyll concentrations). The results indicate excellent quantitative agreement with chlorophyll-a concentration and yellow substance absorption measurements. A sensitivity analysis of the model shows it to be highly sensitive to pressure sensor precision, the accuracy of the value used for the mean cosine of the downwelling radiance distribution, and the irradiance sensor measurement error. However, provided that these factors are taken into account, accurate estimates of chlorophyll-a concentrations in case I (phytoplankton-dominated) waters using a single (or multiple) irradiance sensor with three wavebands can be derived. This model can be applied to irradiance data from a variety of deployment methods including profilers, bottom-tethered moorings, subsurface drifters, and towed platforms
Two Rare Magnetic Cataclysmic Variables with Extreme Cyclotron Features Identified in the Sloan Digital Sky Survey
Two newly identified magnetic cataclysmic variables discovered in the Sloan
Digital Sky Survey (SDSS), SDSSJ155331.12+551614.5 and SDSSJ132411.57+032050.5,
have spectra showing highly prominent, narrow, strongly polarized cyclotron
humps with amplitudes that vary on orbital periods of 4.39 and 2.6 hrs,
respectively. In the former, the spacing of the humps indicates the 3rd and 4th
harmonics in a magnetic field of ~60 MG. The narrowness of the cyclotron
features and the lack of strong emission lines imply very low temperature
plasmas and very low accretion rates, so that the accreting area is heated by
particle collisions rather than accretion shocks. The detection of rare systems
like these exemplifies the ability of the SDSS to find the lowest accretion

CMB observations from the CBI and VSA: A comparison of coincident maps and parameter estimation methods
We present coincident observations of the Cosmic Microwave Background (CMB)
from the Very Small Array (VSA) and Cosmic Background Imager (CBI) telescopes.
The consistency of the full datasets is tested in the map plane and the Fourier
plane, prior to the usual compression of CMB data into flat bandpowers. Of the
three mosaics observed by each group, two are found to be in excellent
agreement. In the third mosaic, there is a 2 sigma discrepancy between the
correlation of the data and the level expected from Monte Carlo simulations.
This is shown to be consistent with increased phase calibration errors on VSA
data during summer observations. We also consider the parameter estimation
method of each group. The key difference is the use of the variance window
function in place of the bandpower window function, an approximation used by
the VSA group. A re-evaluation of the VSA parameter estimates, using bandpower

Estimating the bispectrum of the Very Small Array data
We estimate the bispectrum of the Very Small Array data from the compact and
extended configuration observations released in December 2002, and compare our
results to those obtained from Gaussian simulations. There is a slight excess
of large bispectrum values for two individual fields, but this does not appear
when the fields are combined. Given our expected level of residual point
sources, we do not expect these to be the source of the discrepancy. Using the
compact configuration data, we put an upper limit of 5400 on the value of f_NL,
the non-linear coupling parameter, at 95 per cent confidence. We test our
bispectrum estimator using non-Gaussian simulations with a known bispectrum,

Efficacy and tolerability of evolocumab vs. ezetimibe in patients with muscle-related statin intolerance: the GAUSS-3 randomized clinical trial
Importance: Muscle-related statin intolerance is reported by 5% to 20% of patients.
Objective: To identify patients with muscle symptoms confirmed by statin rechallenge and compare lipid-lowering efficacy for 2 nonstatin therapies, ezetimibe and evolocumab.
Design, Setting, and Participants: Two-stage randomized clinical trial including 511 adult patients with uncontrolled low-density lipoprotein cholesterol (LDL-C) levels and history of intolerance to 2 or more statins enrolled in 2013 and 2014 globally. Phase A used a 24-week crossover procedure with atorvastatin or placebo to identify patients having symptoms only with atorvastatin but not placebo. In phase B, after a 2-week washout, patients were randomized to ezetimibe or evolocumab for 24 weeks.
Interventions: Phase A: atorvastatin (20 mg) vs placebo. Phase B: randomization 2:1 to subcutaneous evolocumab (420 mg monthly) or oral ezetimibe (10 mg daily).
Main Outcome and Measures: Coprimary end points were the mean percent change in LDL-C level from baseline to the mean of weeks 22 and 24 levels and from baseline to week 24 levels.
Results: Of the 491 patients who entered phase A (mean age, 60.7 [SD, 10.2] years; 246 women [50.1%]; 170 with coronary heart disease [34.6%]; entry mean LDL-C level, 212.3 [SD, 67.9] mg/dL), muscle symptoms occurred in 209 of 491 (42.6%) while taking atorvastatin but not while taking placebo. Of these, 199 entered phase B, along with 19 who proceeded directly to phase B for elevated creatine kinase (Nâ=â218, with 73 randomized to ezetimibe and 145 to evolocumab; entry mean LDL-C level, 219.9 [SD, 72] mg/dL). For the mean of weeks 22 and 24, LDL-C level with ezetimibe was 183.0 mg/dL; mean percent LDL-C change, â16.7% (95% CI, â20.5% to â12.9%), absolute change, â31.0 mg/dL and with evolocumab was 103.6 mg/dL; mean percent change, â54.5% (95% CI, â57.2% to â51.8%); absolute change, â106.8 mg/dL (Pâ<â.001). LDL-C level at week 24 with ezetimibe was 181.5 mg/dL; mean percent change, â16.7% (95% CI, â20.8% to â12.5%); absolute change, â31.2 mg/dL and with evolocumab was 104.1 mg/dL; mean percent change, â52.8% (95% CI, â55.8% to â49.8%); absolute change, â102.9 mg/dL (Pâ<â.001). For the mean of weeks 22 and 24, between-group difference in LDL-C was â37.8%; absolute difference, â75.8 mg/dL. For week 24, between-group difference in LDL-C was â36.1%; absolute difference, â71.7 mg/dL. Muscle symptoms were reported in 28.8% of ezetimibe-treated patients and 20.7% of evolocumab-treated patients (log-rank Pâ=â.17). Active study drug was stopped for muscle symptoms in 5 of 73 ezetimibe-treated patients (6.8%) and 1 of 145 evolocumab-treated patients (0.7%).
Conclusions and Relevance: Among patients with statin intolerance related to muscle-related adverse effects, the use of evolocumab compared with ezetimibe resulted in a significantly greater reduction in LDL-C levels after 24 weeks. Further studies are needed to assess long-term efficacy and safety
