    Some Observations on the Handling of Adams\u27 Platinum-Oxide Hydrogenation Catalyst

    Two problems are present in the measurement of the exact quantities of catalyst used in hydrogenations with platinum oxide. The first arises from the fact that platinum oxide is highly adsorbent and rapidly gains weight due to adsorption of atmospheric components when it is exposed to air. A procedure is described for making an approximate correction for this weight gain. The second problem is the identity of the actual catalytic material. The observations indicate that this is platinum metal, and it is shown that the platinum content varies with the batch of catalyst

    Wigner crystals in two-dimensional transition-metal dichalcogenides: Spin physics and readout

    Wigner crystals are prime candidates for the realization of regular electron lattices under minimal requirements on external control and electronics. However, several technical challenges have prevented their detailed experimental investigation and applications to date. We propose an implementation of two-dimensional electron lattices for quantum simulation of Ising spin systems based on self-assembled Wigner crystals in transition-metal dichalcogenides. We show that these semiconductors allow for minimally invasive all-optical detection schemes of charge ordering and total spin. For incident light with optimally chosen beam parameters and polarization, we predict a strong dependence of the transmitted and reflected signals on the underlying lattice periodicity, thus revealing the charge order inherent in Wigner crystals. At the same time, the selection rules in transition-metal dichalcogenides provide direct access to the spin degree of freedom via Faraday rotation measurements.Comment: 15 pages, 12 figure

    Evolutionary selection across the nuclear hormone receptor superfamily with a focus on the NR1I subfamily (vitamin D, pregnane X, and constitutive androstane receptors)

    BACKGROUND: The nuclear hormone receptor (NR) superfamily complement in humans is composed of 48 genes with diverse roles in metabolic homeostasis, development, and detoxification. In general, NRs are strongly conserved between vertebrate species, and few examples of molecular adaptation (positive selection) within this superfamily have been demonstrated. Previous studies utilizing two-species comparisons reveal strong purifying (negative) selection of most NR genes, with two possible exceptions being the ligand-binding domains (LBDs) of the pregnane X receptor (PXR, NR1I2) and the constitutive androstane receptor (CAR, NR1I3), two proteins involved in the regulation of toxic compound metabolism and elimination. The aim of this study was to apply detailed phylogenetic analysis using maximum likelihood methods to the entire complement of genes in the vertebrate NR superfamily. Analyses were carried out both across all vertebrates and limited to mammals and also separately for the two major domains of NRs, the DNA-binding domain (DBD) and LBD, in addition to the full-length sequences. Additional functional data is also reported for activation of PXR and the vitamin D receptor (VDR; NR1I1) to gain further insight into the evolution of the NR1I subfamily. RESULTS: The NR genes appear to be subject to strong purifying selection, particularly in the DBDs. Estimates of the ratio of the non-synonymous to synonymous nucleotide substitution rates (the ω ratio) revealed that only the PXR LBD had a sub-population of codons with an estimated ω ratio greater than 1. CAR was also unusual in showing high relative ω ratios in both the DBD and LBD, a finding that may relate to the recent appearance of the CAR gene (presumably by duplication of a pre-mammalian PXR gene) just prior to the evolution of mammals. Functional analyses of the NR1I subfamily show that human and zebrafish PXRs show similar activation by steroid hormones and early bile salts, properties not shared by sea lamprey, mouse, or human VDRs, or by Xenopus laevis PXRs. CONCLUSION: NR genes generally show strong sequence conservation and little evidence for positive selection. The main exceptions are PXR and CAR, genes that may have adapted to cross-species differences in toxic compound exposure

    Toponyms for centers of endemism in Madagascar

    A biogeographical model was proposed in 2006 to explain the centers of endemism and the importance of riparian forest of some watersheds as refuges or dispersal corridors during paleoclimatic oscillations. Here, we consider these geographical features highlighting their biological and socio-cultural importance. We explain the etymology or eponymy of the major rivers of the retreat - dispersal watersheds, i.e., the drainage basins of Bemarivo, Antainambalana, Mangoro, Manampatrana, Mananara South, Mandrare, Onilahy, Mangoky, Tsiribihina, Betsiboka, Maevarano, Sambirano, and Mahavavy North. We propose a toponymy for each of the 15 centers of endemism and highlight their peculiarities. We named the centers of endemism of Vohimarina, Masoala, Analanjirofo, Tanala, Manombo, Anosy, Ranopiso, Karimbola, Mikea, Menabe, Melaky, Sofia, Ampasindava, Ankify, and Ankarana. We illustrate each center of endemism with a flagship species and report on its natural and cultural histories, and conservation.RÉSUMÉUn modèle biogéographique a été proposé en 2006 pour expliquer les centres d’endémisme de la biodiversité et l’importance des ripisylves de certains bassins versants en tant que refuges ou couloirs de dispersion au cours des oscillations paléoclimatiques. Ici, nous considérons ces dispositifs géographiques en soulignant leur importance biologique et socio-culturelle. Dans un premier temps, nous expliquons la toponymie ou l’éponymie des grands fleuves des bassins refuges et de dispersion, à savoir les bassins de la Bemarivo, de l’Antainambalana, du Mangoro, de la Manampatrana, de la Mananara du Sud, du Mandrare, de l’Onilahy, du Mangoky, de la Tsiribihina, de la Betsiboka, de la Maevarano, du Sambirano et de la Mahavavy du Nord. Puis nous proposons une toponymie pour chacun des 17 centres et sous-centres d’endémisme en justifiant leurs particularités. Nous retenons ainsi les centres d’endémisme de Vohimarina, de l’Atsinanana (dont Masoala et Analanjirofo), Tanala, de Manombo, de l’Anosy, d’Ala maika (dont Ranopiso, Karimbola et Mikea), du Menabe, du Melaky, de la Sofia, d’Ampasindava, d’Ankify et de l’Ankarana. Nous illustrons chacun des centres d’endémisme avec une espèce symbolique et rapportons des aspects de son histoire naturelle et culturelle ainsi que de sa conservation

    Procalcitonin for diagnosis of infection and guide to antibiotic decisions: past, present and future

    There are a number of limitations to using conventional diagnostic markers for patients with clinical suspicion of infection. As a consequence, unnecessary and prolonged exposure to antimicrobial agents adversely affect patient outcomes, while inappropriate antibiotic therapy increases antibiotic resistance. A growing body of evidence supports the use of procalcitonin (PCT) to improve diagnosis of bacterial infections and to guide antibiotic therapy. For patients with upper and lower respiratory tract infection, post-operative infections and for severe sepsis patients in the intensive care unit, randomized-controlled trials have shown a benefit of using PCT algorithms to guide decisions about initiation and/or discontinuation of antibiotic therapy. For some other types of infections, observational studies have shown promising first results, but further intervention studies are needed before use of PCT in clinical routine can be recommended. The aim of this review is to summarize the current evidence for PCT in different infections and clinical settings, and discuss the reliability of this marker when used with validated diagnostic algorithms

    Predicting mortality with pneumonia severity scores: importance of model recalibration to local settings

    In patients with community-acquired pneumonia (CAP) prediction rules based on individual predicted mortalities are frequently used to support decision-making for in-patient vs. outpatient management. We studied the accuracy and the need for recalibration of three risk prediction scores in a tertiary-care University hospital emergency-department setting in Switzerland. We pooled data from patients with CAP enrolled in two randomized controlled trials. We compared expected mortality from the original pneumonia severity index (PSI), CURB65 and CRB65 scores against observed mortality (calibration) and recalibrated the scores by fitting the intercept α and the calibration slope β from our calibration model. Each of the original models underestimated the observed 30-day mortality of 11%, in 371 patients admitted to the emergency department with CAP (8·4%, 5·5% and 5·0% for the PSI, CURB65 and CRB65 scores, respectively). In particular, we observed a relevant mortality within the low risk classes of the original models (2·6%, 5·3%, and 3·7% for PSI classes I-III, CURB65 classes 0-1, and CRB65 class 0, respectively). Recalibration of the original risk models corrected the miscalibration. After recalibration, however, only PSI class I was sensitive enough to identify patients with a low risk (i.e. <1%) for mortality suitable for outpatient management. In our tertiary-care setting with mostly referred in-patients, CAP risk scores substantially underestimated observed mortalities misclassifying patients with relevant risks of death suitable for outpatient management. Prior to the implementation of CAP risk scores in the clinical setting, the need for recalibration and the accuracy of low-risk re-classification should be studied in order to adhere with discharge guidelines and guarantee patients' safet

    Genetic Variation in Cell Death Genes and Risk of Non-Hodgkin Lymphoma

    Background Non-Hodgkin lymphomas are a heterogeneous group of solid tumours that constitute the 5th highest cause of cancer mortality in the United States and Canada. Poor control of cell death in lymphocytes can lead to autoimmune disease or cancer, making genes involved in programmed cell death of lymphocytes logical candidate genes for lymphoma susceptibility. Materials and Methods We tested for genetic association with NHL and NHL subtypes, of SNPs in lymphocyte cell death genes using an established population-based study. 17 candidate genes were chosen based on biological function, with 123 SNPs tested. These included tagSNPs from HapMap and novel SNPs discovered by re-sequencing 47 cases in genes for which SNP representation was judged to be low. The main analysis, which estimated odds ratios by fitting data to an additive logistic regression model, used European ancestry samples that passed quality control measures (569 cases and 547 controls). A two-tiered approach for multiple testing correction was used: correction for number of tests within each gene by permutation-based methodology, followed by correction for the number of genes tested using the false discovery rate. Results Variant rs928883, near miR-155, showed an association (OR per A-allele: 2.80 [95% CI: 1.63–4.82]; pF = 0.027) with marginal zone lymphoma that is significant after correction for multiple testing. Conclusions This is the first reported association between a germline polymorphism at a miRNA locus and lymphoma

    Characterization of growth and metabolism of the haloalkaliphile Natronomonas pharaonis

    Natronomonas pharaonis is an archaeon adapted to two extreme conditions: high salt concentration and alkaline pH. It has become one of the model organisms for the study of extremophilic life. Here, we present a genome-scale, manually curated metabolic reconstruction for the microorganism. The reconstruction itself represents a knowledge base of the haloalkaliphile's metabolism and, as such, would greatly assist further investigations on archaeal pathways. In addition, we experimentally determined several parameters relevant to growth, including a characterization of the biomass composition and a quantification of carbon and oxygen consumption. Using the metabolic reconstruction and the experimental data, we formulated a constraints-based model which we used to analyze the behavior of the archaeon when grown on a single carbon source. Results of the analysis include the finding that Natronomonas pharaonis, when grown aerobically on acetate, uses a carbon to oxygen consumption ratio that is theoretically near-optimal with respect to growth and energy production. This supports the hypothesis that, under simple conditions, the microorganism optimizes its metabolism with respect to the two objectives. We also found that the archaeon has a very low carbon efficiency of only about 35%. This inefficiency is probably due to a very low P/O ratio as well as to the other difficulties posed by its extreme environment
