    Improving chronological control for environmental sequences from the last glacial period

    Recognition of palaeoclimatic instability in the Greenland ice cores has spurred researchers to identify corresponding evidence in other terrestrial records from the last glacial stage. Such evidence is critical for establishing how much environmental stress precipitated Neanderthal and Late Pleistocene megafaunal extinctions, although a need for improved chronology has been consistently highlighted. In formerly glaciated and periglaciated areas of northern Europe, palaeoenvironmental sequences are frequently discontinuous. These often yield high-resolution proxy-based quantitative palaeotemperature estimates but can be hard to date, due to difficulties in removing contamination from biological samples at the limits of the radiocarbon technique (c.30-50kya). Here we demonstrate, for the first time using samples with independent age control, that different radiocarbon pretreatments can generate different age data and that gentler, less effective treatments applied to avoid sample loss may not yield reliable age-estimates. We advocate alternative harsher pretreatment using a strong acid-base-acid protocol. This provides an acceptable balance between contamination removal and excessive sample loss and generates more accurate ages, significantly enhancing our ability to detect and understand the impacts of palaeoclimatic instability in the terrestrial record of the last glacial

    The Dutch chronic lower limb-threatening ischemia registry (THRILLER): A study protocol for popliteal and infrapopliteal endovascular interventions

    INTRODUCTION: Chronic limb-threatening ischemia (CLTI) is the end stage of peripheral arterial disease (PAD) and is associated with high amputation rates, mortality and disease-related health care costs. In infrapopliteal arterial disease (IPAD), endovascular revascularization should be considered for the majority of anatomical and clinical subgroups of CLTI. However, a gap of high-quality evidence exists in this field. The aim of the Dutch Chronic Lower Limb-Threatening Ischemia Registry (THRILLER) is to collect real world data on popliteal and infrapopliteal endovascular interventions. METHODS: THRILLER is a clinician-driven, prospective, multicenter, observational registry including all consecutive patients that undergo a popliteal or infrapopliteal endovascular intervention in seven Dutch hospitals. We estimate that THRILLER will include 400-500 interventions annually. Standardized follow-up visits with wound monitoring, toe pressure measurement and duplex ultrasonography will be scheduled at 6-8 weeks and 12 months after the intervention. The independent primary endpoints are primary patency, limb salvage and amputation free survival. Patients must give informed consent before participation and will be included according to predefined reporting standards. A data log of patients who meet the inclusion criteria but are not included in the registry will be maintained. We intend to conduct the first interim analysis two years after the start of inclusion. The results will be published in a scientific journal. DISCUSSION: Despite innovations in medical therapy and revascularization techniques, patients with CLTI undergoing endovascular revascularization still have a moderate prognosis. Previous prospective cohort studies were hampered by small sample sizes or heterogeneous reporting. Randomized controlled trials (RCTs) have high costs, potential conflicts of interest and give a limited reflection of daily practice. THRILLER aims to provide the largest prospective well phenotyped up-to-date dataset on treatment outcomes in CLTI patients to answer multiple underexplored research questions regarding diagnostics, medication, patient selection, treatment strategies and post intervention follow-up

    Transcriptome profiling of the rice blast fungus during invasive plant infection and in vitro stresses

    <p>Abstract</p> <p>Background</p> <p>Rice blast is the most threatening disease to cultivated rice. <it>Magnaporthe oryzae</it>, its causal agent, is likely to encounter environmental challenges during invasive growth in its host plants that require shifts in gene expression to establish a compatible interaction. Here, we tested the hypothesis that gene expression patterns during <it>in planta </it>invasive growth are similar to <it>in vitro </it>stress conditions, such as nutrient limitation, temperature up shift and oxidative stress, and determined which condition most closely mimicked that of <it>in planta </it>invasive growth. Gene expression data were collected from these <it>in vitro </it>experiments and compared to fungal gene expression during the invasive growth phase at 72 hours post-inoculation in compatible interactions on two grass hosts, rice and barley.</p> <p>Results</p> <p>We identified 4,973 genes that were differentially expressed in at least one of the <it>in planta </it>and <it>in vitro </it>stress conditions when compared to fungal mycelia grown in complete medium, which was used as reference. From those genes, 1,909 showed similar expression patterns between at least one of the <it>in vitro </it>stresses and rice and/or barley. Hierarchical clustering of these 1,909 genes showed three major clusters in which <it>in planta </it>conditions closely grouped with the nutrient starvation conditions. Out of these 1,909 genes, 55 genes and 129 genes were induced and repressed in all treatments, respectively. Functional categorization of the 55 induced genes revealed that most were either related to carbon metabolism, membrane proteins, or were involved in oxidoreduction reactions. The 129 repressed genes showed putative roles in vesicle trafficking, signal transduction, nitrogen metabolism, or molecular transport.</p> <p>Conclusions</p> <p>These findings suggest that <it>M. oryzae </it>is likely primarily coping with nutrient-limited environments at the invasive growth stage 72 hours post-inoculation, and not with oxidative or temperature stresses.</p

    Early Ipswichian (last interglacial) sea level rise in the channel region : Stone Point Site of Special Scientific Interest, Hampshire, England

    Constraining the speed of sea level rise at the start of an interglacial is important to understanding the size of the ‘window of opportunity’ available for hominin migration. This is particularly important during the last interglacial when there is no evidence for significant hominin occupation anywhere in Britain. There are very few finer grained fossiliferous sequences in the Channel region that can be used to constrain sea level rise and they are preserved only to the north of the Channel, in England. Of these, the sequence at Stone Point SSSI is by far the most complete. Data from this sequence has been previously reported, and discussed at a Quaternary Research Association Field Meeting, where a number of further questions were raised that necessitated further data generation. In this paper, we report new data from this sequence – thin section analysis, isotopic determinations on ostracod shells, new Optical Stimulated Luminescence ages and Amino Acid Recem analyses. These show early sea level rise in this sequence, starting during the pre-temperate vegetation zone IpI, but no early warming. The implications of this almost certainly last interglacial sequence for the human colonisation of Britain and our understanding of the stratigraphic relationship of interglacial estuarine deposits with their related fluvial terrace sequences is explored

    The design of a double ended interferometer (DEI)

    At NMISA the SI unit for length is realised by an iodine stabilised He-Ne laser, an optical measurement. In industry most measurements are performed by mechanical probing. Gauge blocks are the link between the optical measurements and the mechanical measurements, and are therefore critical in disseminating traceability. The gauge block length is currently determined by wringing the gauge block onto a platen. This is laborious, requires skill and causes contact errors. Since around 1943, there have been efforts to build a double ended interferometer where gauge block length can be determined without wringing it onto a platen. While there has been a lot of progress in building DEIs, to date, there is still no commercially available DEI. We present a collaboration project between the National Metrology Institute of South Africa (NMISA), the National Institute of Standards (NIS Egypt) and Stellenbosch University (SUN) in South Africa to build a double ended interferometer for use at the respective National Metrology Institutes. We investigate improvements to the interferometric calibration of gauge blocks and recent developments. The different systems currently in use in the national metrology laboratories are described and various designs are investigated. Lastly the expected outcome of the project will be discussed
