    Multivalent interaction and selectivities in selectin binding of functionalized gold colloids decorated with carbohydrate mimetics

    Colloidal gold particles with functionalized organic shells were applied as novel selectin binders. The ligand shell was terminated with different monocyclic carbohydrate mimetics as simplified analogs of the sLe(x) unit found in biological selectin ligands. The multivalent presentation of the sulfated selectin binding epitopes on the gold particles led to extremely high binding affinities towards L- and P-selectin and IC(50) values in the subnanomolar range. Depending on the ring size of the sulfated carbohydrate mimetic, its substitution pattern and its configuration, different selectivities for either L-selectin or P-selectin were obtained. These selectivities were not found for gold particles with simple acyclic sulfated alcohols, diols and triols in the ligand shell. In addition, the influence of the particle size and the thickness of the hydrophobic organic shell were systematically investigated

    Synthesis of tumor-associated MUC1-glycopeptides and their multivalent presentation by functionalized gold colloids

    The mucin MUC1 is a glycoprotein involved in fundamental biological processes, which can be found over-expressed and with a distinctly altered glycan pattern on epithelial tumor cells; thus it is a promising target structure in the quest for effective carbohydrate-based cancer vaccines and immunotherapeutics. Natural glycopeptide antigens indicate only a low immunogenicity and a T-cell independent immune response; however, this major drawback can be overcome by coupling of glycopeptide antigens multivalently to immunostimulating carrier platforms. In particular, gold nanoparticles are well suited as templates for the multivalent presentation of glycopeptide antigens, due to their remarkably high surface-to-volume ratio in combination with their high biostability. In this work the synthesis of novel MUC1-glycopeptide antigens and their coupling to gold nanoparticles of different sizes are presented. In addition, the development of a new dot-blot immunoassay to test the potential antigen-antibody binding is introduced

We report the block copolymer-assisted generation of locally regular chains of quasi-spherical gold nanoparticles with narrow particle diameter distribution by mild thermolysis of the non-polar gold precursor gold mesitylene inside the cylindrical nanopores of self-ordered anodic aluminium oxide (AAO). The block copolymer separates the gold mesitylene as well as the developing gold nanoparticles from the AAO pore walls so that surface nucleation and pinning of gold clusters are prevented. Growing quasi-spherical gold nanoparticles locally deform the polymer chains irreversibly adsorbed on the AAO pore walls, and the polymer chains are pushed into the space between the gold nanoparticles. The gold nanoparticles have, therefore, larger diameters and smaller specific surface than hypothetical pluglike gold entities with the same volume, the formation of which is suppressed.DFG, SPP 1165, NanodrÀhte und Nanoröhren: von kontrollierter Synthese zur Funktio

    Identifying determinants, pressures and trade-offs of crop residue use in mixed smallholder farms in Sub-Saharan Africa and South Asia

    <p>Crop residues (CR) have become a limited resource in mixed crop-livestock farms. As a result of the increasing demand and low availability of alternative resources, CR became an essential resource for household activities, especially for livestock keeping; a major livelihood element of smallholder farmers in the developing world. Farmers' decisions on CR use are determined by farmers' preferences, total crop production, availability of alternative resources and demand for CR. Interaction of these determinants can result in pressures and trade-offs of CR use. Determinants, pressures and trade-offs are shaped by the specific socio-economic and agro-ecological context of these mixed farms. The objective of this paper is to provide a comparative analysis of the determinants of CR use and to examine some options to cope with pressures and trade-offs in 12 study sites across Sub-Saharan Africa and South Asia. Drawing on socio-economic data at household and village level, we describe how cereal intensification and livestock feed demand influence use, pressures and trade-offs of CR use across study sites, specifically cereal residue. Our results show that in low cereal production and livestock feed demand sites, despite a low demand for CR and availability of alternative biomass, pressures and trade-offs of CR use are common particularly in the dry season. In sites with moderate cereal production, and low-moderate and moderate livestock feed demand, alternative biomass resources are scarce and most residues are fed to livestock or used to cover household needs. Subsequently, pressures and potential trade-offs are stronger. In sites with low cereal production and high livestock feed demand, pressures and trade-offs depend on the availability of better feed resources. Finally, sites with high cereal production and high livestock feed demand have been able to fulfil most of the demand for CR, limiting pressures and trade-offs. These patterns show that agricultural intensification, better management of communal resources and off-farm activities are plausible development pathways to overcome pressures and trade-offs of CR use. Although technologies can largely improve these trends, research and development should revisit past initiatives so as to develop innovative approaches to tackle the well-known problem of low agricultural production in many smallholder mixed systems, creating more sustainable futures.</p
