    <i>Melav2</i>, an <i>elav</i>-like gene, is essential for spermatid differentiation in the flatworm <i>Macrostomum lignano</i>

    BackgroundFailure of sperm differentiation is one of the major causes of male sterility. During spermiogenesis, spermatids undergo a complex metamorphosis, including chromatin condensation and cell elongation. Although the resulting sperm morphology and property can vary depending on the species, these processes are fundamental in many organisms. Studying genes involved in such processes can thus provide important information for a better understanding of spermatogenesis, which might be universally applied to many other organisms.ResultsIn a screen for genes that have gonad-specific expression we isolated an elav-like gene, melav2, from Macrostomum lignano, containing the three RNA recognition motifs characteristic of elav-like genes. We found that melav2 mRNA was expressed exclusively in the testis, as opposed to the known elav genes, which are expressed in the nervous system. The RNAi phenotype of melav2 was characterized by an aberrant spermatid morphology, where sperm elongation often failed, and an empty seminal vesicle. Melav2 RNAi treated worms were thus male-sterile. Further analysis revealed that in melav2 RNAi treated worms precocious chromatin condensation occurred during spermatid differentiation, resulting in an abnormally tightly condensed chromatin and large vacuoles in round spermatids. In addition, immunostaining using an early-spermatid specific antibody revealed that melav2 RNAi treated worms had a larger amount of signal positive cells, suggesting that many cells failed the transition from early spermatid stage.ConclusionWe characterize a new function for elav-like genes, showing that melav2 plays a crucial role during spermatid differentiation, especially in the regulation of chromatin condensation and/or cell elongation

    Co-navigating the Complexities of School Reform: The Establishment and On-going Maintenance of Relational Trust in School Reform Efforts

    The purpose of this research was to examine the ways in which the principal and literacy coach collectively developed and maintained relational trust in order to establish school literacy reform efforts. Drawing from a larger set of data, we employed qualitative methods to explore interviews and surveys from the principals and literacy coaches at two different schools who were able to implement literacy reform for several consecutive years. The relational trust established between the coach and principal enabled them to co-navigate issues that might have otherwise impeded literacy reform efforts in their school. Acting together, the principal and the coach were able to communicate a common vision for literacy reform, which resulted in increased implementation of the reform framework in their schools

    The 2011 Mw 7.1 Van (Eastern Turkey) earthquake

    [1] We use interferometric synthetic aperture radar (InSAR), body wave seismology, satellite imagery, and field observations to constrain the fault parameters of the Mw 7.1 2011 Van (Eastern Turkey) reverse-slip earthquake, in the Turkish-Iranian plateau. Distributed slip models from elastic dislocation modeling of the InSAR surface displacements from ENVISAT and COSMO-SkyMed interferograms indicate up to 9 m of reverse and oblique slip on a pair of en echelon NW 40 °–54 ° dipping fault planes which have surface extensions projecting to just 10 km north of the city of Van. The slip remained buried and is relatively deep, with a centroid depth of 14 km, and the rupture reaching only within 8–9 km of the surface, consistent with the lack of significant ground rupture. The up-dip extension of this modeled WSW striking fault plane coincides with field observations of weak ground deformation seen on the western of the two fault segments and has a dip consistent with that seen at the surface in fault gouge exposed in Quaternary sediments. No significant coseismic slip is found in the upper 8 km of the crust above the main slip patches, except for a small region on the eastern segment potentially resulting from the Mw 5.9 aftershock on the same day. We perform extensive resolution tests on the data to confirm the robustness of the observed slip deficit in the shallow crust. We resolve a steep gradient in displacement at the point where the planes of the two fault segments ends are inferred to abut at depth, possibly exerting some structural control on rupture extent

    Nucleotide excision repair leaves a mark on chromatin: DNA damage detection in nucleosomes

    Global genome nucleotide excision repair (GG-NER) eliminates a broad spectrum of DNA lesions from genomic DNA. Genomic DNA is tightly wrapped around histones creating a barrier for DNA repair proteins to access DNA lesions buried in nucleosomal DNA. The DNA-damage sensors XPC and DDB2 recognize DNA lesions in nucleosomal DNA and initiate repair. The emerging view is that a tight interplay between XPC and DDB2 is regulated by post-translational modifications on the damage sensors themselves as well as on chromatin containing DNA lesions. The choreography between XPC and DDB2, their interconnection with post-translational modifications such as ubiquitylation, SUMOylation, methylation, poly(ADP-ribos)ylation, acetylation, and the functional links with chromatin remodelling activities regulate not only the initial recognition of DNA lesions in nucleosomes, but also the downstream recruitment and necessary displacement of GG-NER factors as repair progresses.     “Having all of your internal resources exhausted beyond measure and being left with no clean-up crew”: defining autistic burnout

    Background: Although autistic adults often discuss experiencing ‘‘autistic burnout’’ and attribute serious negative outcomes to it, the concept is almost completely absent from the academic and clinical literature. Methods: We used a community-based participatory research approach to conduct a thematic analysis of 19 interviews and 19 public Internet sources to understand and characterize autistic burnout. Interview participants were autistic adults who identified as having been professionally diagnosed with an autism spectrum condition. We conducted a thematic analysis, using a hybrid inductive–deductive approach, at semantic and latent levels, through a critical paradigm. We addressed trustworthiness through multiple coders, peer debriefing, and examination of contradictions. Results: Autistic adults described the primary characteristics of autistic burnout as chronic exhaustion, loss of skills, and reduced tolerance to stimulus. They described burnout as happening because of life stressors that added to the cumulative load they experienced, and barriers to support that created an inability to obtain relief from the load. These pressures caused expectations to outweigh abilities resulting in autistic burnout. Autistic adults described negative impacts on their health, capacity for independent living, and quality of life, including suicidal behavior. They also discussed a lack of empathy from neurotypical people and described acceptance and social support, time off/reduced expectations, and doing things in an autistic way/unmasking as associated in their experiences with recovery from autistic burnout. Conclusions: Autistic burnout appears to be a phenomenon distinct from occupational burnout or clinical depression. Better understanding autistic burnout could lead to ways to recognize, relieve, or prevent it, including highlighting the potential dangers of teaching autistic people to mask or camouflage their autistic traits, and including burnout education in suicide prevention programs. These findings highlight the need to reduce discrimination and stigma related to autism and disability

    Parasite Component Community of Smalltooth Sawfish Off Florida: Diversity, Conservation Concerns, and Research Applications

    Compared with that of other charismatic elasmobranchs, the component community of metazoan parasites infecting endangered smalltooth sawfish Pristis pectinata is exceedingly poorly characterized: adults of Dermophthirioides pristidis and Neoheterocotyle inpristi (ectoparasitic flatworms of skin and gill, respectively) were the only confirmed parasites prior to the description, based on specimens reported herein, of Mycteronastes caalusi. Our opportune and directed parasitological examinations of 290 smalltooth sawfish (277 live inspections; 13 necropsies; 671 to 2640 mm stretch total length) in south Florida coastal waters revealed at least 8 species of Platyhelminthes, 9 of Arthropoda, 4 of Annelida, and 1 of Nematoda. This collection includes representatives of an undescribed species of Aporocotylidae (Digenea) and myriad new host records, considerably updating and advancing our understanding of smalltooth sawfish symbionts. We also confirm that D. pristidis and N. inpristi are extant and propose D. pristidis as a reliable biological tag. Some of these parasites are evidently highly host-specific and so vulnerable to extinction

Successful development of the parenting role may be negatively influenced by the Neonatal Intensive Care Unit (NICU) environment and nursing practices.
