    SCH9, a gene of Saccharomyces cerevisiae that encodes a protein distinct from, but functionally and structurally related to, cAMP-dependent protein kinase catalytic subunits

    A new gene, SCH9, was isolated from Saccharomyces cerevisiae by its ability to complement a cdc25ts mutation. Sequence analysis indicates that it encodes a 90,000-dalton protein with a carboxy-terminal domain homologous to yeast and mammalian cAMP-dependent protein kinase catalytic subunits. In addition to suppressing loss of CDC25 function, multicopy plasmids containing SCH9 suppress the growth defects of strains lacking the RAS genes, the CYR1 gene, which encodes adenylyl cyclase, and the TPK genes, which encode the cAMP-dependent protein kinase catalytic subunits. Cells lacking SCH9 grow slowly and have a prolonged G1 phase of the cell cycle. This defect is suppressed by activation of the cAMP effector pathway. We propose that SCH9 encodes a protein kinase that is part of a growth control pathway which is at least partially redundant with the cAMP pathway

    Predictive maintenance has become important for avoiding unplanned downtime of modern vehicles. With increasing functionality the exchanged data between Electronic Control Units (ECU) grows simultaneously rapidly. A large number of in-vehicle signals are provided for monitoring an aging process. Various components of a vehicle age due to their usage. This component aging is only visible in a certain number of in-vehicle signals. In this work, we present a signal selection method for in-vehicle signals in order to determine relevant signals to monitor and predict powertrain component aging of vehicles. Our application considers the aging of powertrain components with respect to clogging of structural components. We measure the component aging process in certain time intervals. Owing to this, unevenly spaced time series data is preprocessed to generate comparable in-vehicle data. First, we aggregate the data in certain intervals. Thus, the dynamic in-vehicle database is reduced which enables us to analyze the signals more efficiently. Secondly, we implement machine learning algorithms to generate a digital model of the measured aging process. With the help of Local Interpretable Model-Agnostic Explanations (LIME) the model gets interpretable. This allows us to extract the most relevant signals and to reduce the amount of processed data. Our results show that a certain number of in-vehicle signals are sufficient for predicting the aging process of the considered structural component. Consequently, our approach allows to reduce data transmission of in-vehicle signals with the goal of predictive maintenance.Прогнозное техническое обслуживание является важным для предотвращения незапланированных простоев современных транспортных средств. С расширением функциональности одновременно происходит быстрый рост обмена данными между электронными блоками управления. Большое количество бортовых сигналов позволяет осуществлять мониторинг процесса старения. Старение компонентов автомобиля зависит от того, как они используются. Элементы старения выявляются благодаря наличию ряда бортовых сигналов. В данной статье предложен метод выбора бортовых сигналов с целью определения соответствующих для проведения мониторинга и прогнозирования старения компонентов силовой передачи транспортных средств. Процесс старения рассматривается на основе степени засорения конструктивных элементов. Измерение процесса старения компонентов осуществляется в определенные промежутки времени. Благодаря такому подходу данные, полученные в неравномерно распределенные промежутки времени, предварительно обрабатываются для формирования сравниваемых бортовых показателей. На первом этапе агрегируем данные в определенные интервалы. Тем самым динамическая база бортовых данных уменьшается, что позволяет более эффективно анализировать сигналы. Также используем алгоритм машинного обучения с целью создания цифровой модели для измерения процесса старения. С помощью методологии локальных интерпретируемых модельно-агностических объяснений модель становится интерпретируемой. Это позволяет извлекать наиболее релевантные сигналы и тем самым сокращать объем обрабатываемых данных. Полученные результаты показывают, что для прогнозирования процесса старения рассматриваемого структурного компонента достаточно определенного количества бортовых сигналов. Таким образом, предлагаемый подход позволяет сократить передачу данных бортовых сигналов для проведения профилактического обслуживания

    Numerical Modeling of Propellant Boil-Off in a Cryogenic Storage Tank

    A numerical model to predict boil-off of stored propellant in large spherical cryogenic tanks has been developed. Accurate prediction of tank boil-off rates for different thermal insulation systems was the goal of this collaboration effort. The Generalized Fluid System Simulation Program, integrating flow analysis and conjugate heat transfer for solving complex fluid system problems, was used to create the model. Calculation of tank boil-off rate requires simultaneous simulation of heat transfer processes among liquid propellant, vapor ullage space, and tank structure. The reference tank for the boil-off model was the 850,000 gallon liquid hydrogen tank at Launch Complex 39B (LC- 39B) at Kennedy Space Center, which is under study for future infrastructure improvements to support the Constellation program. The methodology employed in the numerical model was validated using a sub-scale model and tank. Experimental test data from a 1/15th scale version of the LC-39B tank using both liquid hydrogen and liquid nitrogen were used to anchor the analytical predictions of the sub-scale model. Favorable correlations between sub-scale model and experimental test data have provided confidence in full-scale tank boil-off predictions. These methods are now being used in the preliminary design for other cases including future launch vehicle

    Effects of flow alteration on Apple-ring Acacia (Faidherbia albida) stands, Middle Zambezi floodplains, Zimbabwe

    The impounding of the Zambezi River by Kariba dam has regulated the river discharge of the Middle Zambezi river. This has been implicated in the failure of regeneration of Faidherbia albida in the downstream flood plain. This study aimed (1) to assess the effect of the altered flow regime of the river on the establishment and growth of F. albida on the Middle Zambezi floodplain and islands and (2) to test the potential of dendrochronology in detecting the age and long-term growth rates of F. albida in response to flow regime. Results indicated an uneven age distribution of F. albida stands on relatively ‘new islands’, dominated by young trees, while the floodplain and the ‘old island’ exhibited an even-aged stand but with a lack of regeneration, and a dying-off of older trees. The lack of F. albida establishment on the floodplain was linked to the decreased occurrence of flooding events, associated with a decrease in alluvial deposits, soil moisture and groundwater recharge. These effects may be enhanced by impact from browsers on regeneration of trees. Tree-ring analyses revealed the presence of distinct annual growth rings in F. albida and the applicability of dendrochronology for estimating F. albida population dynamics. The trees on the ‘new islands’ are younger and grow faster than those on the floodplain. Lack of competition and possible favourable moisture conditions suggest beneficial conditions for establishment and growth of the trees on the islands

    One of the tasks that recent philosophy of psychiatry has taken upon itself is to extend the range of understanding to some of those aspects of psychopathology that Jaspers deemed beyond its limits. Given the fundamental difficulties of offering a literal interpretation of the contents of primary delusions, a number of alternative strategies have been put forward including regarding them as abnormal versions of framework propositions described by Wittgenstein in On Certainty. But although framework propositions share some of the apparent epistemic features of primary delusions, their role in partially constituting the sense of inquiry rules out their role in helping to understand delusions

    Glass Bubbles Insulation for Liquid Hydrogen Storage Tanks

    A full-scale field application of glass bubbles insulation has been demonstrated in a 218,000 L liquid hydrogen storage tank. This work is the evolution of extensive materials testing, laboratory scale testing, and system studies leading to the use of glass bubbles insulation as a cost efficient and high performance alternative in cryogenic storage tanks of any size. The tank utilized is part of a rocket propulsion test complex at the NASA Stennis Space Center and is a 1960's vintage spherical double wall tank with an evacuated annulus. The original perlite that was removed from the annulus was in pristine condition and showed no signs of deterioration or compaction. Test results show a significant reduction in liquid hydrogen boiloff when compared to recent baseline data prior to removal of the perlite insulation. The data also validates the previous laboratory scale testing (1000 L) and full-scale numerical modeling (3,200,000 L) of boiloff in spherical cryogenic storage tanks. The performance of the tank will continue to be monitored during operation of the tank over the coming years. KEYWORDS: Glass bubble, perlite, insulation, liquid hydrogen, storage tank

    Delusions are often regarded as irrational beliefs, but their irrationality is not sufficient to explain what is pathological about them. In this paper we ask whether deluded subjects have the capacity to support the content of their delusions with reasons, that is, whether they can author their delusional states. The hypothesis that delusions are characterised by a failure of authorship, which is a dimension of self knowledge, deserves to be empirically tested because (a) it has the potential to account for the distinction between endorsing a delusion and endorsing a framework belief; (b) it contributes to a philosophical analysis of the relationship between rationality and self knowledge; and (c) it informs diagnosis and therapy in clinical psychiatry. However, authorship cannot provide a demarcation criterion between delusions and other irrational belief states

    Supplementary data and analysis for estimating walleye selectivity

    This document has been issued as VIMS Data Report 61 and provides data tables and results of exploratory analyses conducted as part of the complete data analysis for Myers et al. 2014 published in the Transactions of the American Fisheries Society. Estimates of size- and sex-specific selectivity of fishing gear are important for making informed management decisions. We distinguish between capture selectivity – the relative catchability of the components of the population – and harvest selectivity, which is the combined effects of capture selectivity and the decision to retain or release a fish of a given population component. We used short-term recaptures from three extensive tagging programs in Minnesota and Wisconsin to estimate directly the size- and sex- specific selectivity of angling for captured and for harvested walleye Sander vitreus, and of spear fishing for harvested walleye. Estimates were obtained using generalized linear models with an information-theoretic approach to determining the significance of individual and interactive effects of length and sex on selectivity. The primary conclusions of this research are presented in Myers et al. 2014. Residual analyses for the models presented in the manuscript, results of unpublished exploratory analyses, and the complete data set used to conduct the analyses are presented in this supplementary document. Through this data report, interested readers can repeat the analyses conducted in Myers et al. 2014, as well as see the results of additional analyses not presented in the primary publication

    Cloning and characterization of BCY1, a locus encoding a regulatory subunit of the cyclic AMP-dependent protein kinase in Saccharomyces cerevisiae

    We have cloned a gene (BCY1) from the yeast Saccharomyces cerevisiae that encodes a regulatory subunit of the cyclic AMP-dependent protein kinase. The encoded protein has a structural organization similar to that of the RI and RII regulatory subunits of the mammalian cyclic AMP-dependent protein kinase. Strains of S. cerevisiae with disrupted BCY1 genes do not display a cyclic AMP-dependent protein kinase in vitro, fail to grow on many carbon sources, and are exquisitely sensitive to heat shock and starvation
