The Mechanisms of Regulated Cell Death: Structural and Functional Proteomic Pathways Induced or Inhibited by a Specific Protein—A Narrative Review
[EN] Billions of cells die in us every hour, and our tissues do not shrink because there is a natural regulation where Cell Death (CD) is balanced with cell division. The process in which cells eliminate themselves in a controlled manner is called Programmed Cell Death (PCD). The PCD plays an important role during embryonic development, in maintaining homeostasis of the body’s tissues, and in the elimination of damaged cells, under a wide range of physiological and developmental stimuli. A multitude of protein mediators of PCD have been identified and signals have been found to utilize common pathways elucidating the proteins involved. This narrative review focuses on caspase-dependent and caspase-independent PCD pathways. Included are studies of caspase-dependent PCD such as Anoikis, Catastrophe Mitotic, Pyroptosis, Emperitosis, Parthanatos and Cornification, and Caspase-Independent PCD as Wallerian Degeneration, Ferroptosis, Paraptosis, Entosis, Methuosis, and Extracellular Trap Abnormal Condition (ETosis), as well as neutrophil extracellular trap abnormal condition (NETosis) and Eosinophil Extracellular Trap Abnormal Condition (EETosis). Understanding PCD from those reported in this review could shed substantial light on the processes of biological homeostasis. In addition, identifying specific proteins involved in these processes is mandatory to identify molecular biomarkers, as well as therapeutic targets. This knowledge could provide the ability to modulate the PCD response and could lead to new therapeutic interventions in a wide range of diseases.S
Synthesis and characterization of NaNiF3·3H2O: An unusual ordered variant of the ReO3 type
et al.A new hydrated sodium nickel fluoride with nominal composition NaNiF3·3H2O was synthesized using an aqueous solution route. Its structure was solved by means of ab initio methods from powder X-ray diffraction and neutron diffraction data. NaNiF3·3H2O crystallizes in the cubic crystal system, space group Pn3̅ with a = 7.91968(4) Å. The framework, derived from the ReO3 structure type, is built from NaX6 and NiX6 (X = O, F) corner-shared octahedra, in which F and O atoms are randomly distributed on a single anion site. The 2a × 2a × 2a superstructure arises from the strict alternate three-dimensional linking of NaX6 and NiX6 octahedra together with the simultaneous tilts of the octahedra from the cube axis (φ = 31.1°), with a significant participation of hydrogen bonding. NaNiF3·3H2O corresponds to a fully cation-ordered variant of the In(OH)3 structure, easily recognizable when formulated as NaNi(XH)6 (X = O, F). It constitutes one of the rare examples for the a+a+a+ tilting scheme with 1:1 cation ordering in perovskite-related compounds. The Curie-like magnetic behavior well-reflects the isolated paramagnetic Ni2+ centers without worth mentioning interactions. While X-ray and neutron diffraction data evidence Na/Ni order in combination with O/F disorder as a main feature of this fluoride, results from Raman and magic-angle spinning NMR spectroscopies support the existence of specific anion arrangements in isolated square windows identified in structural refinements. Mechanism of construction-destruction of macrostructural patterns in perthitic microcline as complex systems
[EN] Se han estudiado estructuras de feldespatos potásicos triclínicos (microclinas) intercrecidos con feldespatos sódicos en
texturas pertíticas procedentes de pegmatitas graníticas a diferentes escalas de observación, mediante microscopía de luz
transmitida (MLT), difracción de rayos-X (DRX), resonancia magnética nuclear de alta resolución del núcleo 27Al (RMN),
y mapas químicos de contenido en Na por microsonda electrónica (ME). Las pertitas pegmatíticas son sistemas complejos
en cuyos feldespatos potásicos se han identificado dos periodos genéticos independientes de recristalización durante el
enfriamiento geológico. En el primer periodo, en la estructura mineral se produce la construcción gradual de patrones
macroscópicos regulares pseudoperiódicos, como consecuencia del efecto combinado de un ordenamiento local no-ergódico
en la distribución Si/Al y de un efecto cooperativo global de la estructura del cristal. Desde las interfases incoherentes con
el feldespato sódico surgen frentes de transformación a modo de avalanchas que pueden coalescer longitudinalmente y
yuxtaponerse transversalmente para auto-ensamblarse, y finalmente evolucionar hasta formar dominios-macla (leyes Albita
y Periclina), como unidad cristalina de construcción de la macroestructura. Estos procesos de auto-organización se activan
cuando el sistema está abierto a la circulación de moléculas de agua catalizadoras para el ordenamiento de la red, al tiempo
que existe una estimulación externa por tensiones tectónicas de cizalla. Durante el segundo periodo se destruyen los patrones
estructurales creados previamente, por: (i) maclados de relajación y deformación, (ii) engrosamientos de dominios por
disolución-reprecipitación debidos a interacciones con fluidos acuosos a baja temperatura, (iii) reorientaciones de dominiosmacla
según ley Periclina hacia orientaciones de ley Albita.[ES] The structures of microcline perthites collected from granitic pegmatites have been studied at different observation scales
using optical microscopy (OM), X-ray diffraction (XRD), 27Al MASS-NMR spectroscopy, and electron microprobe (EMPA)
with Na mappings. Pegmatite perthites are identified as complex systems. During the cooling process, K-rich feldspars
display two separated genetic periods of recrystallization, each one with variable intensity along different samples. During
the first period, in the mineral structure a progressive construction of macrostructural pseudo-periodic patterns is produced.
These regular patterns emerge due to a combined mechanism of non-ergodic Si/Al local ordering and a global cooperative
effect of the crystal structure. From the incoherent Na/K interfaces arise transformation fronts (avalanches) which can
coalesce longitudinally and juxtapose transversally to self-assemble into irregular domains. Finally, they can evolve up
to create regular twin-domains (Albite and Pericline laws) as the crystal unit of the macrostructural building. This selforganization
process is activated when the system is open to water circulation acting as a catalyst of Al/Si ordering in the
feldspar lattice linked with an external stimulation by shear tectonic stress. During the second period the structural patterns,
previously created, are destroyed by (i) relaxation and deformation twinning mechanisms, (ii) coarsening of twin-domains
produced by dissolution-precipitation due to aqueous fluids interactions at low temperature, (iii) reorientation of Periclinelaw

Caracterización estructural y espectroscópica de fibras cristalinas de Ce0.4Zr0.6O2 crecidas mediante el método de fusión zonal asistida por láser
[EN] A structural and spectroscopic characterization of crystalline rods of Ce0.4Zr0.6O2 grown by the laser floating zone (LFZ) method is presented. A precursor rod of Ce0.4Zr0.6O2 composition was sintered at 1500 ºC in air atmosphere and then processed by the LFZ technique with a CO2 laser. The processed material was characterized by XRD, SEM and Raman spectroscopy. In the as-grown, dark-color processed rod, the Raman spectrum evolves radially from a t’-like one, corresponding to Ce0.37Zr0.63O2 composition, at the edge of the rod, to a very broad and weak, cubic-like one, at the center. The degree of cerium reduction and oxygen non-stoichiometry were determined through measurements of the electronic Raman spectrum of Ce3+. A strong Ce3+ signal was found at the core of the rod, indicating strong reduction, whereas no Ce3+ signal was detected at the edge. To restore oxygen and Ce4+ content a section of the fiber was reoxidized at 620 ºC for 24 h. A very homogeneous spectrum was found in the reoxidized sample, that was assigned to a t’-phase of composition Ce0.42Zr0.58O2.[ES] Presentamos una caracterización estructural y espectroscópica de fibras cristalinas de Ce0.4Zr0.6O2 crecidas mediante fusión zonal asistida por láser (LFZ). Una barra del material precursor, de composición Ce0.4Zr0.6O2, fue sinterizada a 1500 ºC en atmósfera de aire y después procesada por LFZ con un láser de CO2. El material procesado fue caracterizado por difracción de rayos X, microscopía electrónica de barrido y espectroscopía Raman. En la fibra recién procesada, de color gris oscuro, el espectro Raman varía radialmente desde un espectro de tipo t’, correspondiente a una composición Ce0.37Zr0.63O2, en el borde de la fibra, hasta un espectro muy ensanchado y débil, de aspecto cúbico, en el centro. El grado de reducción del cerio y la no estequiometría del oxígeno se determinaron a través de las medidas del Raman electrónico del Ce3+. La señal de Ce3+ era muy intensa en el centro de la fibra, lo que indica una fuerte reducción, mientras que no se detectó en el borde. Para restablecer el contenido de oxígeno y de Ce4+ una parte de la fibra fue reoxidada a 620 ºC durante 24 h. Antibacterial and antifungal activity of ZnO containing glasses
A new family of non-toxic biocides based on low melting point (1250°C) transparent glasses with high content of ZnO (15–40wt%) belonging to the miscibility region of the B2O3-SiO2-Na2O-ZnO system has been developed. These glasses have shown an excellent biocide activity (logarithmic reduction >3) against Gram- (E. coli), Gram+ (S. aureus) and yeast (C. krusei); they are chemically stable in different media (distilled water, sea-like water, LB and DMEN media) as well as biocompatible. The cytotoxicity was evaluated by the Neutral Red Uptake using NIH-3T3 (mouse embryonic fibroblast cells) and the cell viability was >80%. These new glasses can be considered in several and important applications in the field of inorganic non-toxic biocide agents such as medical implants, surgical equipment, protective apparels in hospitals, water purifications systems, food packaging, food storages or textiles.This study has been supported by Consejo Superior de Investigaciones Cientificas (CSIC) project reference number 201360E012 and by the Spanish Ministry of Science and Innovation MAT2012-38645. Mussel shells from marine aquaculture act like ecosystem engineers: legacy effects on benthic communities
Ecosystem engineers are organisms that cause changes in the physical state of biotic and abiotic structures that modulate the availability of resources to other species, thus affecting biochemical cycles. Molluscs, especially bivalves such as mussels, are widespread in coastal environments and they are excellent ecosystem engineers because of the durability of their shells, which add complexity and heterogeneity to benthic environments. The presence of mussel farms favours the accumulation of shells in benthic environments and may influence surrounding bare sediments, with potential legacy effects on benthic communities. We studied the effects of the accumulation of mussel shells at finfish farms and mussel farms by experimentally comparing bare sediment and sediment with fragmented shells in terms of the abundance of the most relevant faunal groups, specifically polychaete families as well as physical–chemical variables in sediment water samples, specifically organic matter (OM), redox potential, and acid-volatile sulphides (AVS) NH4+ and PO43−. The experiment was replicated under two environmental conditions over a period of 35 days: eutrophic muddy sediments and oligotrophic sandy sediments. The OM and AVS values were significantly higher in the eutrophic sediment with mussel shells. Only NH4+ was positively affected by the mussel shells in the oligotrophic conditions. Differences between the two environments were observed, and the effect of the mussel shells on the polychaete assemblages was more significant in the oligotrophic conditions. Mussel shell accumulations affected the structure of benthic assemblages by modifying their heterogeneity and complexity, which suggests that the presence of mussel farms above bare sediment may affect ecosystem functioning. Aquaculture has potentially negative or positive effects that must be addressed on a large scale, considering the increased input of organic matter and also the simultaneous presence of mussel shell waste, both of which alter the surrounding environment. Therapeutic concentrations of varenicline increases exocytotic release of catecholamines from human and rat adrenal chromaffin cells in the presence of nicotine
Cardiovascular side effects of varenicline and a case report of a hypertensive crisis in a varenicline-prescribed patient with pheochromocytoma have been reported. The goal of the present study was to determine whether such side effects might derive, in part, from increased exocytosis of secretory vesicles and subsequent catecholamine release triggered by varenicline in human chromaffin cells of the adrenal gland. In this study, we performed electrophysiological plasma membrane capacitance and carbon fiber amperometry experiments to evaluate the effect of varenicline on exocytosis and catecholamine release, respectively, at concentrations reached during varenicline therapy (100 nM). Experiments were conducted in the absence or presence of nicotine, at plasma concentrations achieved right after smoking (250 nM) or steady-state concentrations (110 nM), in chromaffin cells of the adrenal gland obtained from human organ donors. Cells were stimulated with short pulses (10 ms) of acetylcholine (ACh; 300 μM) applied at 0.2 Hz, in order to closer mimic the physiological situation at the splanchnic nerve-chromaffin cell synapse. In addition, rat chromaffin cells were used to compare the effects obtained in cells from a more readily available species. Varenicline increased the exocytosis of secretory vesicles in human and rat chromaffin cells in the presence of nicotine, effects that were not due to an increase of plasma membrane capacitance or currents triggered by the nicotinic agonists alone. BARD1 Pathogenic Variants are Associated with Triple-Negative Breast Cancer in a Spanish Hereditary Breast and Ovarian Cancer Cohort
Only a small fraction of hereditary breast and/or ovarian cancer (HBOC) cases are caused by germline variants in the high-penetrance breast cancer 1 and 2 genes (BRCA1 and BRCA2). BRCA1-associated ring domain 1 (BARD1), nuclear partner of BRCA1, has been suggested as a potential HBOC risk gene, although its prevalence and penetrance are variable according to populations and type of tumor. We aimed to investigate the prevalence of BARD1 truncating variants in a cohort of patients with clinical suspicion of HBOC. A comprehensive BARD1 screening by multigene panel analysis was performed in 4015 unrelated patients according to our regional guidelines for genetic testing in hereditary cancer. In addition, 51,202 Genome Aggregation Database (gnomAD) non-Finnish, non-cancer European individuals were used as a control population. In our patient cohort, we identified 19 patients with heterozygous BARD1 truncating variants (0.47%), whereas the frequency observed in the gnomAD controls was 0.12%. We found a statistically significant association of truncating BARD1 variants with overall risk (odds ratio (OR) = 3.78; CI = 2.10-6.48; p = 1.16 x 10(-5)). This association remained significant in the hereditary breast cancer (HBC) group (OR = 4.18; CI = 2.10-7.70; p = 5.45 x 10(-5)). Furthermore, deleterious BARD1 variants were enriched among triple-negative BC patients (OR = 5.40; CI = 1.77-18.15; p = 0.001) compared to other BC subtypes. Our results support the role of BARD1 as a moderate penetrance BC predisposing gene and highlight a stronger association with triple-negative tumors
Home parenteral nutrition registry in Spain for the year 2010: NADYA-SENPE group
Objetivos: Comunicar los datos del Registro del Grupo
NADYA SENPE de Nutrición Parenteral Domiciliaria
NPD en España del año 2010.
Material y métodos: Estudio descriptivo de la base de
datos del Registro de ámbito nacional de NPD del grupo
NADYA-SENPE (1 de enero de 2010 al 31 de diciembre de
2010). Para el cálculo de prevalencias se utilizó los últimos
datos publicados por el Instituto Nacional de Estadística.
Resultados: Se registraron 148 pacientes procedentes de
23 hospitales, 86 mujeres (58,11%) y 9 niños (6,08%). La
edad media de los 139 pacientes adultos fue de 53,06 ±
15,41 años. La duración media de la NPD fue de 316,97
días/paciente. El diagnóstico más frecuente en los niños
(menores de 14 años) fue intestino corto traumático con 5
casos (55,55%) y en los adultos la neoplasia en tratamiento
paliativo 29 (19,59%). El motivo de la indicación de la NPD
fue el síndrome de intestino corto en 74 ocasiones (47%).
La vía de acceso más frecuentemente registrada fue el catéter
tunelizado en 36 (22,78%) casos seguido del reservorio
en 13 (8,23%) y otras vías en 3 ocasiones (1,90%). Se registraron
23 infecciones relacionadas con el catéter (82,14%),
lo que representa 0,49/1000 días de NP y todas ellas ocurrieron
en los adultos. A lo largo del año finalizaron 24 episodios
de NPD, la causa más frecuente fue el paso a la vía
oral en 12 episodios (50%). Se registró que los pacientes
tenían una actividad normal en 70 episodios de NPD
(44,30%) con una total autonomía en 88 de episodios
(55,69%). Se identificaron 39 (24,68%) posibles candidatos
para trasplante intestinal.
Conclusiones: El número de pacientes registrados es
discretamente inferior al del año anterior, aunque el
número de hospitales participantes es el mismo. La complicación
más frecuente sigue siendo la infección relacionada
con el catéter aunque ha disminuido su incidencia
respecto a años anteriores, presentándose la tasa más
baja desde la creación del registro.
Las diferencias en la participación en el registro observadas
por Comunidades Autónomas lleva a plantear el
desarrollo de estrategias de implementación del registro.
Se observa un aumento progresivo de la duración de los
días de NPD a lo largo de los años que hace pensar en la
cronicidad de algunos pacientes, pero nos obliga a estudiar
la existencia de un posible factor de confusión, en el
caso de que existiera un olvido de cierre de algún episodio
por lo que se hace necesario actualizar el registro con sistemas
de alertas periódicas que faciliten la revisión de los
pacientes incluidos y optimice la validez del registroObjectives: To report the Group Registry NADYASENPE
data about home parenteral nutrition (HPN) in
Spain in 2010.
Material and methods: A descriptive study of the database
of the national registry of HPN of NADYA-SENPE
(December 10, 2009 to December 10, 2010). For the calculation
of prevalence the latest data published by the Institute
National Statistics Office (01/01/2009) was used.
Results: There were registered 148 patients from 23
hospitals, 86 women (58.11%) and 9 children (6.08%).
The average age of the 139 patients older than 14 years
was 53.06 ± 15.41 years. The average duration of HPN
was 316.97 days/patient. The most common diagnosis in
those younger than 14 years was short bowel traumatic
with 5 cases (55.55%) and in those older than 14 years,
palliative care cancer with 29 cases (19.59%). The reason
for the indication for HPN was short bowel syndrome in
74 cases (47%). The access via most frequently recorded
was tunneled catheter in 36 cases (22.78%) followed by
implanted port-catheters in 13 cases (8.23%) and other
pathways in 3 cases (1.90%). There were 23 catheterrelated
infections (82.14%) which represented 0.49 /1,000
days of PN, all of which occurred in cases older than 14
years. During the year 24 episodes of HPN ended, the
most frequent cause was the transition to oral nutrition in
12 episodes (50%). It was reported that patients had a
normal activity in 70 episodes of HPN (44.30%) with complete
autonomy in 88 episodes (55.69%). Some patients 39
(24.68%) were potential candidates for intestinal transplantation.
Conclusions: The number of registered patients is
slightly lower than the previous year, although the number
of participating hospitals is the same. The most frequent
complication remains catheter-related infection
but its incidence has decreased from previous years, presenting
the lowest rate since the creation of the record.
Differences in participation in the registry observed in
the Autonomous Communities causes the development of
implementation strategies. There is a gradual increase in
day length of HPN over the years, which suggests the
chronic treatments of some patients and obliges to study
the existence of a possible confounding factor, in case
there is an oversight of closing an episode. Therefore, it is
necessary to update the registry with warning systems
that facilitate periodic review of the patients and optimize
the validity of registratio
Follow-Up Study Confirms the Presence of Gastric Cancer DNA Methylation Hallmarks in High-Risk Precursor Lesions
Intestinal metaplasia confers an increased risk of progression to gastric cancer. However, some intestinal metaplasia patients do not develop cancer. The development of robust molecular biomarkers to stratify patients with advanced gastric precursor lesions at risk of cancer progression will contribute to guiding programs for prevention. Starting from a genome-wide methylation study, we have simplified the detection method regarding candidate-methylation tests to improve their applicability in the clinical environment. We identified CpG methylation at the ZNF793 and RPRM promoters as a common event in intestinal metaplasia and intestinal forms of gastric cancer. Furthermore, we also showed that Helicobacter pylori infection influences DNA methylation in early precursor lesions but not in intestinal metaplasia, suggesting that therapeutic strategies to prevent epigenome reprogramming toward a cancer signature need to be adopted early in the precursor cascade. To adopt prevention strategies in gastric cancer, it is imperative to develop robust biomarkers with acceptable costs and feasibility in clinical practice to stratified populations according to risk scores. With this aim, we applied an unbiased genome-wide CpG methylation approach to a discovery cohort composed of gastric cancer (n = 24), and non-malignant precursor lesions (n = 64). Then, candidate-methylation approaches were performed in a validation cohort of precursor lesions obtained from an observational longitudinal study (n = 264), with a 12-year follow-up to identify repression or progression cases. H. pylori stratification and histology were considered to determine their influence on the methylation dynamics. As a result, we ascertained that intestinal metaplasia partially recapitulates patterns of aberrant methylation of intestinal type of gastric cancer, independently of the H. pylori status. Two epigenetically regulated genes in cancer, RPRM and ZNF793, consistently showed increased methylation in intestinal metaplasia with respect to earlier precursor lesions. In summary, our result supports the need to investigate the practical utilities of the quantification of DNA methylation in candidate genes as a marker for disease progression. In addition, the H. pylori-dependent methylation in intestinal metaplasia suggests that pharmacological treatments aimed at H. pylori eradication in the late stages of precursor lesions do not prevent epigenome reprogramming toward a cancer signature
