    As pastilhas TERMOPAR nas aulas experimentais de termologia

    Diante da situação atual da educação brasileira, principalmente na área das ciências naturais e suas tecnologias, mais pontualmente em física, é inquestionável a necessidade de novas abordagens, novas práticas capazes de levar a uma potencial aprendizagem significativa. Neste trabalho, após discutir a atual crise na educação e o aparecimento de novas metodologias, enfatizamos a importância da experimentação no ensino de física e como essa prática é justificada pela teoria sócio-histórica de Vygotsck. Em seguida, sugerimos como inovação, o uso das partilhas termopar nas atividades experimentais de termologia, que além de tornarem esses experimentos mais seguros e baratos, ainda torna conhecido um dispositivo presente em diversos aparelhos de nosso dia a dia, como bebedouros, por exemplo; permitindo também o aprofundamento em vários conceitos de termologia e eletricidade, a partir do conhecimento de um efeito já conhecido desde o século XIX, mas ainda ignorado pela maioria dos professores de física. Após apresentar as pastilhas e descrever umas práticas que foram muito bem sucedidas, deixaremos uma apostila para auxiliar os professores que resolvam utilizá-las.Coordenação de Aperfeiçoamento de Pessoal de Nível SuperiorDue to current situation of brasilian education in Natural Sciences area and their Technologies, more punctually in phisics, it is unquestionable the need for new approaches, new practices able to lead to a true meaningfull learning. In this work, after discussing their current crisis in education and the emergence of new methodologies, emphasizes the importance of experimentation in teaching of phisics and how this practice is justified by the theory of the Vygotsck. And then I suggest as innovation the use of thermocouples in thermology experimental activities, which in addiction make these experiments safer and cheaper and still, becomes a known device present in various devices in our daily life like water fouintains, for example; also allowing the deepening in various concepts from thermology and electricity, from the knowledge of an effect that has been known since the nineteen century, but still ignored by most physics teachers. After presenting the thermocouples and describe their practices were very successful, I leave a handout to support teachers who want to use them

    Novel aminoquinoline-based solvatochromic fluorescence probe : interaction with albumin, lysozyme and characterization of amyloid fibrils

    A novel aminoquinoline derivative (AQ) was synthesized and applied as a solvatochromic fluorescent probe to study proteins and their alterations. AQ is not fluorescent in aqueous solution but has its fluorescence quantum yield significantly increased upon binding to albumin. The generation of an induced circular dichroism signal in AQ confirmed the complexation. The Job's plot method revealed an 1:1 stoichiometry for the host-guest complex. The binding constant was determined by AQ fluorescence increase (2.7 × 105 mol−1 L) and by protein intrinsic fluorescence quenching (5.1 × 105 mol−1 L). The displacement of AQ from albumin by warfarin and ibuprofen showed that Sudlow's drug site-I is the preferential binding site. By applying the Bilot-Kawski solvatochromic model to the spectral shifts of fifteen solvents, the microenvironment dielectric constant at albumin site-I was estimated (ε = 14.8). In agreement, the average fluorescence lifetime of AQ complexed with albumin (6.11 ns) was close to dichloromethane (6.53 ns) and acetone (6.34 ns), which have dielectric constants of 8.9 and 21.0, respectively. Albumin was thermically treated to formation of amyloid fibril aggregates. AQ was able to differentiate the altered and native protein. Sodium dodecyl sulfate-induced aggregation of lysozyme to amyloid fibril was also efficiently detected by the AQ fluorescence increase. AQ was as efficient as the chromogenic bromocresol purple in the quantitative analysis of albumin.
