    Actinorhizal nitrogen fixing nodules: infection process, molecular biology and genomics

    Actinorhizal hosts are non-leguminous perennial plants belonging to 8 angiosperm families. They are capable of forming root nodules as a result of infection by a nitrogen-fixing actinomycete called Frankia. Actinorhizal nodules consist of multiple lobes, each of which represents a modified lateral root with infected cells in the expanded cortex. This article summarizes the most recent knowledge about this original symbiotic process. The infection process is described both at cytological and molecular levels. The use of transgenic Casuarinaceae for studying in actinorhizal nodules the regulation of several symbiotic promoters from legumes is also discussed. With progress in plant genome sequencing, comparative genomics in legumes and actinorhizal plants should contribute to the understanding of the evolutionary history of nitrogen-fixing symbioses.     Biased sex ratios in Western Europe populations of little bustard (Tetrax tetrax) as a potential warning signal of unbalanced mortalities

    Adult sex ratios (ASRs) have proved to correlate with population trends, which make them potential useful indicators of a species’ population trajectory and conservation status. We analysed ASRs and proportion of juveniles in flocks of an endangered steppe bird, the Little Bustard Tetrax tetrax, using surveys made during the non-breeding period in seven areas within its Western European range (one in Portugal, four in Spain, and two in France). We found overall male-biased ASRs, as all the seven surveyed areas showed a male-biased ASR mean value. Five areas were below the threshold median value (female sex ratio = 0.4) considered to be consistent with an increased probability of extinction, according to earlier population viability analyses for the species. We also found a significant positive correlation between female ratio and the proportion of young individuals in the non-breeding flocks surveyed. Our results (strongly male-biased ASRs) support the hypothesis that the viability of Little Bustard populations in Western Europe is threatened by an excess of female mortality, something that should be quantified in the future, and emphasise the value of monitoring sex ratio as a population viability indicator in species where monitoring survival is difficult to achieve.This paper is a contribution to the REMEDINAL 3 (S2013/MAE-2719) network which funded a post-doc contract for ESD.     Identification of precursor transcripts for 6 novel miRNAs expands the diversity on the genomic organisation and expression of miRNA genes in rice

    <p>Abstract</p> <p>Background</p> <p>The plant miRNAs represent an important class of endogenous small RNAs that guide cleavage of an mRNA target or repress its translation to control development and adaptation to stresses. MiRNAs are nuclear-encoded genes transcribed by RNA polymerase II, producing a primary precursor that is subsequently processed by DCL1 an RNase III Dicer-like protein.</p> <p>In rice hundreds of miRNAs have been described or predicted, but little is known on their genes and precursors which are important criteria to distinguish them from siRNAs. Here we develop a combination of experimental approaches to detect novel miRNAs in rice, identify their precursor transcripts and genes and predict or validate their mRNA targets.</p> <p>Results</p> <p>We produced four cDNA libraries from small RNA fractions extracted from distinct rice tissues. By <it>in silico </it>analysis we selected 6 potential novel miRNAs, and confirmed that their expression requires OsDCL1. We predicted their targets and used 5'RACE to validate cleavage for three of them, targeting a PPR, an SPX domain protein and a GT-like transcription factor respectively.</p> <p>In addition, we identified precursor transcripts for the 6 miRNAs expressed in rice, showing that these precursors can be efficiently processed using a transient expression assay in transfected <it>Nicotiana benthamiana </it>leaves. Most interestingly, we describe two precursors producing tandem miRNAs, but in distinct arrays. We focus on one of them encoding osa-miR159a.2, a novel miRNA produced from the same stem-loop structure encoding the conserved osa-miR159a.1. We show that this dual osa-miR159a.2-osa-miR159a.1 structure is conserved in distant rice species and maize. Finally we show that the predicted mRNA target of osa-miR159a.2 encoding a GT-like transcription factor is cleaved <it>in vivo </it>at the expected site.</p> <p>Conclusion</p> <p>The combination of approaches developed here identified six novel miRNAs expressed in rice which can be clearly distinguished from siRNAs. Importantly, we show that two miRNAs can be produced from a single precursor, either from tandem stem-loops or tandemly arrayed in a single stem-loop. This suggests that processing of these precursors could be an important regulatory step to produce one or more functional miRNAs in plants and perhaps coordinate cleavage of distinct targets in the same plant tissue.</p

    Extracorporeal Membrane Oxygenation for Severe Acute Respiratory Distress Syndrome associated with COVID-19: An Emulated Target Trial Analysis.

    RATIONALE: Whether COVID patients may benefit from extracorporeal membrane oxygenation (ECMO) compared with conventional invasive mechanical ventilation (IMV) remains unknown. OBJECTIVES: To estimate the effect of ECMO on 90-Day mortality vs IMV only Methods: Among 4,244 critically ill adult patients with COVID-19 included in a multicenter cohort study, we emulated a target trial comparing the treatment strategies of initiating ECMO vs. no ECMO within 7 days of IMV in patients with severe acute respiratory distress syndrome (PaO2/FiO2 <80 or PaCO2 ≥60 mmHg). We controlled for confounding using a multivariable Cox model based on predefined variables. MAIN RESULTS: 1,235 patients met the full eligibility criteria for the emulated trial, among whom 164 patients initiated ECMO. The ECMO strategy had a higher survival probability at Day-7 from the onset of eligibility criteria (87% vs 83%, risk difference: 4%, 95% CI 0;9%) which decreased during follow-up (survival at Day-90: 63% vs 65%, risk difference: -2%, 95% CI -10;5%). However, ECMO was associated with higher survival when performed in high-volume ECMO centers or in regions where a specific ECMO network organization was set up to handle high demand, and when initiated within the first 4 days of MV and in profoundly hypoxemic patients. CONCLUSIONS: In an emulated trial based on a nationwide COVID-19 cohort, we found differential survival over time of an ECMO compared with a no-ECMO strategy. However, ECMO was consistently associated with better outcomes when performed in high-volume centers and in regions with ECMO capacities specifically organized to handle high demand.     De l’inscription à un MOOC à la certification académique Le cas de la collection de MOOC « compétences du numérique et C2i Niveau 1 »

    Dans le présent article, nous montrons comment a été standardisé un dispositif de formation à distance interuniversitaire par collection de MOOC (Massive Open Online Course), qui amène à une certification académique. Nous présentons d’abord la mise en oeuvre et l’évaluation d’un dispositif de formation : la collection de MOOC en compétences du numérique sur la plateforme nationale FUN (France Université Numérique). Puis, nous décrivons la mise en oeuvre du processus de certification permettant aux inscrits au MOOC de passer la certification C2i1 (Certificat Informatique et Internet niveau 1) académique délivrée par les universités françaises. Cette approche a été accompagnée expérimentalement et les résultats obtenus ont montré la possibilité de créer des parcours amenant à la délivrance de véritables certifications reconnues académiquement pour un public non étudiant
