CRISPR-Cas9: A Revolutionary Tool for Cancer Modelling

The cancer-modelling field is now experiencing a conversion with the recent emergence of the RNA-programmable CRISPR-Cas9 system, a flexible methodology to produce essentially any desired modification in the genome. Cancer is a multistep process that involves many genetic mutations and other genome rearrangements. Despite their importance, it is difficult to recapitulate the degree of genetic complexity found in patient tumors. The CRISPR-Cas9 system for genome editing has been proven as a robust technology that makes it possible to generate cellular and animal models that recapitulate those cooperative alterations rapidly and at low cost. In this review, we will discuss the innovative applications of the CRISPR-Cas9 system to generate new models, providing a new way to interrogate the development and progression of cancers.This work was supported by FIS project no PI14/01884 to Sandra Rodriguez-Perales from the Spanish plan nacional I+d+, Instituto de Salud Carlos III FEDER. Functional characterization of two enhancers located downstream FOXP2

Background: Mutations in the coding region of FOXP2 are known to cause speech and language impairment. However, it is not clear how dysregulation of the gene contributes to language deficit. Interestingly, microdeletions of the region downstream the gene have been associated with cognitive deficits. Methods: Here, we investigate changes in FOXP2 expression in the SK-N-MC neuroblastoma human cell line after deletion by CRISPR-Cas9 of two enhancers located downstream of the gene. Results: Deletion of any of these two functional enhancers downregulates FOXP2, but also upregulates the closest 3′ gene MDFIC. Because this effect is not statistically significant in a HEK 293 cell line, derived from the human kidney, both enhancers might confer a tissue specific regulation to both genes. We have also found that the deletion of any of these enhancers downregulates six well-known FOXP2 target genes in the SK-N-MC cell line. Proyecto de investigación recobros y crisis financiera del sistema de salud en Colombia (2008-2014) Estudio del caso Saludcoop EPS

La presente investigación sobre la crisis financiera que afecta al Sistema General de Seguridad Social en Salud en Colombia ahonda en las características del modelo de seguridad social en salud que se implementó a partir de la Ley 100 de 1993 y el desequilibrio financiero generado a raíz de la falta de controles administrativos por parte del Ministerio de Salud que fue aprovechado por las Empresas Promotoras de Salud, llevando al sistema a un grave crisis que aún persiste, siendo una de las causas principales los recobros excesivos por altos costos de medicamentos y tratamientos médicos contribuyendo a debilitar el Sistema de Salud. En los últimos días, la Fiscalía General de la Nación ha citado a Carlos Palacino, otrora Presidente del conglomerado Saludcoop EPS para imputarle cargos por los delitos de enriquecimiento ilícito y por desviar los recursos de la salud de los afiliados a Saludcoop hacia patrimonios particulares, en lo que constituye el escándalo de corrupción más grave de la última década en Colombia. Los hechos que dieron lugar a la investigación penal se remontan hacia unos seis años atrás, cuando empiezan las investigaciones de la Contraloría General de la República y de la Procuraduría General de la Nación, ante graves denuncias de corrupción que se formularon por parte del Senador Jorge Enrique Robledo y por el columnista de la Revista Semana Daniel Coronell, en donde señalaban a Palacino de enriquecerse a costa de los recursos con los cuales se debía atender a los pacientes de la que llegó a ser la EPS más grande del país en solo 10 años de haber sido creada. Para el año 2011 el Presidente de la República anunció ante el país que gracias a la labor coordinada de las autoridades se logró desmantelar un cartel de la corrupción en materia de recobros al Fosyga. El Senador Jorge Enrique Robledo ha denunciado en debate de control político adelantado en el Senado en agosto de 2016, como ya lo ha venido haciendo en distintos escenarios, que el Ministerio de Salud no ha tomado acciones efectivas para frenar la corrupción en el sistema de salud, que a cambio de defender a los pacientes las medidas que se toman buscan proteger las 10 inversiones de las EPS y ha indicado que por culpa del Ministerio –ante la liquidación de Saludcoop EPS- se han perdido 1,4 billones de pesos que esa EPS se apropió en forma irregular según fallo proferido por la Contraloría General de la Nación que quedó en firme desde el año 2014.The present investigation on the financial crisis that affects the General System of Social Security in Health in Colombia delves into the characteristics of the model of social security in health that was implemented from the Law 100 of 1993 and the financial imbalance generated as a result of the lack of administrative controls by the Ministry of Health that was taken advantage of by Health Promoting Companies, leading the system to a serious crisis that still persists, one of the main causes being excessive recoveries due to high costs of medicines and medical treatments contributing to weaken the Health System. Sistema de gestión de inventarios y los tiempos de producción en periodo de pandemia COVID-19 Trupal S.A., Trujillo, 2020

La presente investigación titulada “Sistema de gestión de inventarios y los tiempos de producción en periodo de pandemia COVID-19 Trupal S.A., Trujillo, 2020”, se basa en las teorías del sistema de gestión de inventarios y los tiempos de producción; se utilizó una metodología descriptiva y una muestra compuesta por todos los Sku utilizados en el proceso productivo del papel. Para el cual se utilizó los instrumentos de: análisis ABC, VED, un diagrama DAP, un estudio de tiempos con los valores de Westinghouse y suplementos OIT y una matriz Amef, se obtuvieron resultados del cálculo del costo de mantener inventarios en el almacén de insumos que es S/ 3,738.71 y del almacén de materias primas es S/ 841.41, adicionalmente el análisis de costos indico que el costo de inventarios sin modelo del CMI es de S/ 75,237.68; asimismo el costo con modelo del CMI es de S/ 25,941.89, con una variación del 66%. Lo que permite concluir que la mejora y el manejo de un óptimo sistema de gestión de inventarios influyen en los tiempos de producción de la empresa Trupal S.A

Plan de marketing como herramienta de gestión para hoteles boutique ubicados en el centro histórico de Cartagena, afiliados a Cotelco

La investigación apuntará al diseño estratégico de un Plan de Marketing para los Hoteles Boutique Cartagena que le permita tener un pronóstico lo más acertado posible del futuro y rentabilidad de la empresa, identificando los riesgos en su fase inicial para prevenirlos con buenas alternativas de solución, logrando de esta manera convertir el plan en una herramienta clave y fundamental que guie a sus empresarios de manera eficiente al crecimiento y éxito de la misma. Su objetivo apuntó a formular un Plan de Marketing Estratégico como herramienta de gestión para los Hoteles Boutique del centro histórico de Cartagena de Indias afiliados a Cotelco, a través de un análisis situacional interno y externo, para la creación y ejecución de sus propios planes de mejoramiento de los servicios e imágenes corporativas. Método: La metodología utilizada es de carácter descriptiva-a o nalítica, ya que el objeto del estudio está orientado a la caracterización del sector turístico y hotelero de Cartagena, dándole prioridad a las condiciones actuales y potenciales de los hoteles boutique. El diseño investigativo utilizado fue el exploratorio descriptivo. El método de muestreo utilizado fue el no probalístico a juicio para poblaciones finitas. La población objeto de estudio está compuesta por los Hoteles Boutiques del Centro Histórico de Cartagena de los cuales se tomó el 80% de la muestra que corresponde a 17 establecimientos. Se tomaron dos tipos de universo: un universo documental y un universo de campo. De las técnicas de recolección de la información se utilizó la entrevista, aplicada a los administradores y representantes de los 17 hoteles Boutique, la cual se complementó con la observación de campo e información de clientes. Como fuentes secundarias se utilizaron instructivos, documentos, manuales, libros y textos especializados que facilitaron sus administradores y/o representantes. Además de la información consultada en la WEB con datos referente al tema. Para procesar la información se utilizó el método estadístico, y el análisis deductivo; partiendo de lo general, aceptado como válido, hacia aplicaciones particulares. Después de realizada esta investigación se puedo comprobar que los hoteles Boutique del centro histórico de Cartagena, manejan ciertas estrategias para dirigir y gestionar eficazmente, los recursos internos de cada hotel, pero carecen de un plan de marketing definido, de los hoteles que conformaron la muestra que si tiene su propio plan de marketing por escrito son el hotel monterrey, hotel charlestón y el hotel Santa Clara. Los hoteles boutique son la respuesta a una oferta de servicio de hospedaje cinco estrellas en la ciudad de Cartagena, ya que ésta cuenta con infraestructura hotelera necesaria para satisfacer las exigencias de empresarios de alta categoría. De acuerdo con los resultados de los indicadores financieros que evalúan la rentabilidad económica del proyecto, se ratifica la viabilidad de este al tener un alto Valor Presente Neto y una muy buena Tasa de Retorno. El proyecto de Hoteles boutique presenta una buena proyección internacional, ya que los mismos huéspedes extranjeros generar un marketing viral que permitirá aumentar el número de empresarios provenientes del exterior que visten estos hoteles.Incluye bibliografía, listas de tablas, figuras, cuadros y anexo

Effectiveness of Sprayed Bioactive Fruit Extracts in Counteracting Protein Oxidation in Lamb Cutlets Subjected to a High-Oxygen MAP

High-oxygen packaging atmosphere (High-Ox-MAP) promotes meat protein oxidation and leads to texture deterioration. This study was conceived to assess the extent to which sprayed fruit extracts could inhibit the oxidative damage to proteins in lamb cutlets subjected to High-Ox-MAP (10 days/4 °C) and subsequent roasting (10 min/180 °C). Extracts from oaknut (Quercus ilex subsp. ballota; QI), rose hips (Rosa canina L.; RC), common hawthorn (Crataegus monogyna Jacq.; CM) and strawberry tree (Arbutus unedo L.; AU) were characterized for bioactive compounds (phenolic subclasses, tocopherols and ascorbic acid) and in vitro bioactivities. While the four fruits showed relevant antioxidant potential, CM had the highest phenolics and tocopherol content and that was reflected in efficient antiradical activity. The in vitro activity of this fruit to inhibit meat protein oxidation was, however, lower than that displayed by the other fruits. Taking the results altogether, CM was also found to be most efficient in protecting lamb cutlets from lipid oxidation. All fruits were able to inhibit thiols oxidation except RC, which seemed to reduce protein thiols. Among fruits, QI was the most efficient in protecting lamb cutlets against protein carbonylation as a plausible involvement of ellagitannins. The inhibition of protein oxidation by QI was reflected in significantly lower instrumental hardness in cooked lamb cutlets. Spraying lamb cutlets with extracts from QI, RC and CM improved consumers’ purchase intention after chilled storage. Sucrose-responsive intercommunicated janus nanoparticles network

[EN] Inspired by biological systems, the development of artificial nanoscale materials that communicate over a short distance is still at its early stages. This work shows a new example of a cooperating system with intercommunicated devices at the nanoscale. The system is based on the new sucrose-responsive Janus gold-mesoporous silica (Janus Au-MS) nanoparticles network with two enzyme-powered nanodevices. These nanodevices involve two enzymatic processes based on invertase and glucose oxidase, which are anchored on the Au surfaces of different Janus Au-MS nanoparticles, and N-acetyl-L-cysteine and [Ru(bpy)(3)](2+) loaded as chemical messengers, respectively. Sucrose acts as the INPUT, triggering the sequential delivery of two different cargoes through the enzymatic control. Hypoxia signaling and cholesterol/steroidogenic acute regulatory protein 1 axis: interplay and role in alcohol and non-alcohol-related liver diseases

Metabolic zonation in the liver carries out the maintenance of organ and body homeostasis. Hypoxia is an inherent physiological feature of the liver and contributes to the zonal properties of the hepatic parenchyma. As a master regulator of hypoxia, the transcription factor hypoxia-inducing factor (HIF) is stabilized primarily by oxygen availability, and it is thought to contribute to steatohepatitis due to alcohol-related (ASH) and non-alcohol-related liver disease (NASH). Cholesterol has emerged as an important player in both diseases, and hypoxia increases hepatic cholesterol levels. Steroidogenic acute regulatory protein 1 (STARD1) is a mitochondrial outer membrane protein that transfers cholesterol to mitochondrial inner membrane for metabolic processing and acts as the rate-limiting step in the alternative pathway of bile acid synthesis in hepatocytes. STARD1 expression increases in ASH and NASH and determines the accumulation of cholesterol in mitochondria, which impacts the physico-chemical mitochondrial membranes properties and as a consequence impairs the activity of specific mitochondrial solute carriers, such as the 2-oxoglutarate carrier (2-OGC), limiting the exchange between cytosolic glutathione and mitochondrial 2-oxoglutarate (2-OG). Although HIF-1 is stabilized in hypoxia largely due to the requirement of prolylhydroxylases (PHDs) for oxygen to signal HIF degradation, PHDs are also dependent on 2-OG, and therefore it is conceivable that impairment of 2-OGC by STARD1-mediated cholesterol accumulation may contribute to HIF-1 stabilization due in part to decreased availability of cytosolic 2-OG. Comportamiento sísmico de edificaciones de tapia pisada reforzadas con marcos de madera y viga de coronación en concreto

Earth construction is present all over the world. Many of these constructions are in seismic regions and have a high seismic vulnerability. To reduce this vulnerability and prevent collapse this paper proposes an integrated strategy consisting in an external reinforcement with wooden elements in both directions and on both sides of the walls (rammed earth) and a rigid diaphragm by means of an upper concrete beam. This beam is proposed to be built with a smooth transition between the earth and the concrete to improve compatibility. The results of shake table tests on a 1:6 scaled model and its unreinforced counterpart show a significant improvement in the global behaviour of the construction (The results of the tests showed a reduction in the displacements up to 80% in the reinforced models), achieving the main objective of preventing collapse while the structure remains almost intact.La construcción en tierra está presente en todo el mundo. Muchas de estas construcciones se encuentran en regiones sísmicas y tienen una vulnerabilidad alta. Con el fin de reducir esta vulnerabilidad se propone una estrategia integrada que consiste en un refuerzo externo con elementos de madera en ambas direcciones y en ambos lados de los muros (en tapia pisada) simultáneamente con la implementación de un diafragma rígido por medio de una viga de coronación en concreto. Se propone que la viga se construya con una transición entre la tierra y el concreto para mejorar la compatibilidad. Los resultados de ensayos en mesa vibratoria en un modelo a escala 1:6 y su contraparte no reforzada muestran una mejora significativa en el comportamiento global de la construcción (reducción en los desplazamientos hasta del 80% en los modelos reforzados), logrando el objetivo de prevenir el colapso, manteniendo la integridad de la estructura
