    Numerical modelling and vibration serviceability assessment of a steel footbridge with a significant 3D dynamic behaviour

    This paper focuses on the vibration serviceability assessment and numerical modelling of an existing steel truss footbridge located in the outskirts of Castellón, Spain. The footbridge is rather slender and composed by a main span and two access ramps supported on three 4-arm piers of different heights. Due to the connection between the main span and the ramps at the top of the tall piers, longitudinal and lateral bending/torsion natural coupled modes of vibration coexist at low frequencies, with a relevant contribution of the piers and access ramps deformation. This leads to a significant three-dimensional and rather complex dynamic response under service conditions. With the aim of characterising the structural dynamic properties and assessing the level of vibrations induced by crossing pedestrians, two in-situ experimental test programmes are conducted. On the one hand, the structural response is measured during several hammer tests and the modal properties are identified and used to update a detailed 3D numerical model by means of a Genetic Algorithm. Due to the lack of information regarding the detailing of the piers foundations, two alternative models are analysed. The relevance of the pier-foundation system rotational stiffness is highlighted for the particular configuration. On the other hand, the footbridge main span response is recorded under different pedestrian activities: walking, running and vandal simulated actions. Finally, the vibration serviceability of the structure is assessed based on current codes and regulations

    Targeting SHIP-1 in Myeloid Cells Enhances Trained Immunity and Boosts Response to Infection

    beta-Glucan-induced trained immunity in myeloid cells leads to long-term protection against secondary infections. Although previous studies have characterized this phenomenon, strategies to boost trained immunity remain undefined. We found that beta-glucan-trained macrophages from mice with a myeloid-specific deletion of the phosphatase SHIP-1 (LysM Delta SHIP-1) showed enhanced proinflammatory cytokine production in response to lipopolysaccharide. Following beta-glucan training, SHIP-1-deficient macrophages exhibited increased phosphorylation of Akt and mTOR targets, correlating with augmented glycolytic metabolism. Enhanced training in the absence of SHIP-1 relied on histone methylation and acetylation. Trained LysM Delta SHIP-1 mice produced increased amounts of proinflammatory cytokines upon rechallenge in vivo and were better protected against Candida albicans infection compared with control littermates. Pharmacological inhibition of SHIP-1 enhanced trained immunity against Candida infection in mouse macrophages and human peripheral blood mononuclear cells. Our data establish proof of concept for improvement of trained immunity and a strategy to achieve it by targeting SHIP-1.We thank the members of the Immunobiology Lab for useful discussions.     Contribution of spoligotyping to the characterization of the population structure of Mycobacterium tuberculosis isolates in Portugal

    Tuberculosis is a major health problem in Portugal. To begin characterizing the population structure of Mycobacterium tuberculosis, spoligotyping was used for the systematic typing, through consecutive sampling, of patient isolates from the Amadora-Sintra area of Greater Lisbon. Distribution amongst major spoligotype families, including the Latin American Mediterranean (LAM), T, Haarlem and Beijing, was compared to that of the international spoligotype database SpolDB4 and to the European countries of traditional Portuguese immigration represented in SpolDB4. Spoligotypes from 665 isolates were analyzed and 97 shared international types (SITs) identified. In SpolDB4 Portugal is represented by part of the spoligotypes from this study explaining the reduced number of unidentified patterns. The importance of the LAM family, and especially of LAM1 and LAM9 sub-families that alone represented 38% of all the isolates in this study as compared to 8% relative to the European sub group, led us to believe that at least in this respect the population structure was closer to that of Africa and South America than to Europe. Spoligotypes characteristic of Portugal or Portuguese related settings were identified. These included SIT244 a T1 sub-family predominant in Portugal and Bangladesh, SIT64 a LAM 6 sub-family common to Portugal and Brazil, and SIT1106 a LAM 9 sub-family. These studies were the first in Portugal stressing the importance of monitoring the population structure of M. tuberculosis isolates, an important step towards gaining an understanding of tuberculosis and the dynamics of this disease

    The evolution of mobile bed tests: a step towards the future of coastal engineering

    Coastal Engineering still presents significant levels of uncertainty, much larger for sediment transport and morphodynamics than for the driving hydrodynamics. Because of that there is still a need for experimental research that addresses the water and sediment fluxes occurring at multiple scales in the near shore and for some of which there are still not universally accepted equations or closure sub-models. Large scale bed tests offer the possibility to obtain undistorted results under controlled conditions that may look at sediment transport and associated bed evolution under a variety of wave and mean water level conditions. The present limitations in conventional observation equipment preclude a clear advancement in knowledge or model calibration. However the new developments in opto-acoustic equipment should allow such an advancement to take place provided the new experimental equipment becomes more robust in parallel with a protocol for deployment and data processing. This paper will present the experimental approach to erosive and accretive beach dynamics, with emphasis on the accretive experiments. These accretive tests still present further uncertainties and sometimes cannot be explained with the present state of the art. Following this there is a presentation of the novel development of an acoustic bed form and suspended sediment imager, able to monitor bed forms near bed sediment transport and their corresponding dynamics. The next section deals with an acoustic high resolution concentration and velocity profiler that is able to infer even the elusive bed level, together with the near bed concentrated sediment transport and the details of fluxes on the stoss and lee sides of moving bed forms. This is followed by a discussion on the merits of novel optic techniques, using structured and unstructured light sources. There is also some remarks on new approaches. Illustrated by the use of ferro-fluids to obtain directly the shear stresses acting on a wall even under the presence of “some” sediment. The paper ends with some conclusions on the use of such mobile bed tests in present and future Coastal Engineering.Postprint (published version

    Hierarchical Hough all-sky search for periodic gravitational waves in LIGO S5 data

    We describe a new pipeline used to analyze the data from the fifth science run (S5) of the LIGO detectors to search for continuous gravitational waves from isolated spinning neutron stars. The method employed is based on the Hough transform, which is a semi-coherent, computationally efficient, and robust pattern recognition technique. The Hough transform is used to find signals in the time-frequency plane of the data whose frequency evolution fits the pattern produced by the Doppler shift imposed on the signal by the Earth's motion and the pulsar's spin-down during the observation period. The main differences with respect to previous Hough all-sky searches are described. These differences include the use of a two-step hierarchical Hough search, analysis of coincidences among the candidates produced in the first and second year of S5, and veto strategies based on a χ2\chi^2 test.Comment: 7 pages, 2 figures, Amaldi08 proceedings, submitted to JPC

    Model Channel Ion Currents in NaCl - SPC/E Solution with Applied-Field Molecular Dynamics

    Using periodic boundary conditions and a constant applied field, we have simulated current flow through an 8.125 Angstrom internal diameter, rigid, atomistic channel with polar walls in a rigid membrane using explicit ions and SPC/E water. Channel and bath currents were computed from ten 10-ns trajectories for each of 10 different conditions of concentration and applied voltage. An electric field was applied uniformly throughout the system to all mobile atoms. On average, the resultant net electric field falls primarily across the membrane channel, as expected for two conductive baths separated by a membrane capacitance. The channel is rarely occupied by more than one ion. Current-voltage relations are concentration-dependent and superlinear at high concentrations.Comment: Accepted for publication in Biophysical Journa

    Recovery Signals of Rhodoliths Beds since Bottom Trawling Ban in the SCI Menorca Channel (Western Mediterranean)

    One of the objectives of the LIFE IP INTEMARES project is to assess the impact of bottom trawling on the vulnerable benthic habitats of the circalittoral bottoms of the Menorca Channel (western Mediterranean), designated a Site of Community Importance (SCI) within the Natura 2000 network. The present study compares the epibenthic communities of four areas, subjected to different bottom trawl fishing intensity levels. The assignment of fishing effort levels was based on the fishing effort distribution in the area calculated from Vessel Monitoring System (VMS) data and the existence of two Fishing Protected Zones in the Menorca Channel. Biological samples were collected from 39 beam trawl stations, sampled during a scientific survey on April 2019. We compare the diversity, composition, and density of the epibenthic flora and fauna, together with the rhodoliths coverage and the morphology of the main species of rhodoliths of four areas subjected to different levels of bottom trawl fishing effort, including one that has never been impacted by trawling. Our results have shown negative impacts of bottom trawling on rhodoliths beds and the first signals of their recovery in areas recently closed to this fishery, which indicate that this is an effective measure for the conservation of this habitat of special interest and must be included in the management plan required to declare the Menorca Channel as a Special Area of Conservation.En prens

    Clinico-pathological characteristics and outcomes of patients with early-onset colorectal cancer

    [Background]: The rising incidence of colorectal cancer (CRC) among young patients is alarming. We aim to characterize the clinico-pathological features and outcomes of patients with early-onset CRC (EOCRC). [Methods]: We included all of the patients with pathologically confirmed diagnosis of CRC at Hospital Universitario La Paz from October 2016 to September 2020. EOCRC age cut-off was 50 years. All statistical analyses were carried out using SPSS v.25. [Results]: A total of 1152 patients were diagnosed with CRC, fifty-nine (5,1%) of them were After a median follow-up of 24 months, 279 patients have died. Median overall survival (OS) was not reached in either group (p = 0,06). Three-year OS was 80% (95%CI: 73-87) and 67 (95%CI: 65-69) in the younger and older group, respectively. In patients with localized disease that underwent surgery or other antineoplastic treatment ( n = 856), 159 events for disease-free survival (DFS) were observed. Median DFS was [Conclusions]: Patients with EOCRC are diagnosed at a more advanced stage and display distinct biological features (more prevalence of dMMR and WT tumors among others). Studies focusing on screening in this population and deeper molecular profiling are needed

    Quantum state preparation and macroscopic entanglement in gravitational-wave detectors

    Long-baseline laser-interferometer gravitational-wave detectors are operating at a factor of 10 (in amplitude) above the standard quantum limit (SQL) within a broad frequency band. Such a low classical noise budget has already allowed the creation of a controlled 2.7 kg macroscopic oscillator with an effective eigenfrequency of 150 Hz and an occupation number of 200. This result, along with the prospect for further improvements, heralds the new possibility of experimentally probing macroscopic quantum mechanics (MQM) - quantum mechanical behavior of objects in the realm of everyday experience - using gravitational-wave detectors. In this paper, we provide the mathematical foundation for the first step of a MQM experiment: the preparation of a macroscopic test mass into a nearly minimum-Heisenberg-limited Gaussian quantum state, which is possible if the interferometer's classical noise beats the SQL in a broad frequency band. Our formalism, based on Wiener filtering, allows a straightforward conversion from the classical noise budget of a laser interferometer, in terms of noise spectra, into the strategy for quantum state preparation, and the quality of the prepared state. Using this formalism, we consider how Gaussian entanglement can be built among two macroscopic test masses, and the performance of the planned Advanced LIGO interferometers in quantum-state preparation
