    Genomic instability in human actinic keratosis and squamous cell carcinoma

    OBJECTIVE: To compare the repetitive DNA patterns of human actinic keratoses and squamous cell carcinomas to determine the genetic alterations that are associated with malignant transformation. INTRODUCTION: Cancer cells are prone to genomic instability, which is often due to DNA polymerase slippage during the replication of repetitive DNA and to mutations in the DNA repair genes. The progression of benign actinic keratoses to malignant squamous cell carcinomas has been proposed by several authors. MATERIAL AND METHODS: Eight actinic keratoses and 24 squamous cell carcinomas (SCC), which were pair-matched to adjacent skin tissues and/or leucocytes, were studied. The presence of microsatellite instability (MSI) and the loss of heterozygosity (LOH) in chromosomes 6 and 9 were investigated using nine PCR primer pairs. Random Amplified Polymorphic DNA patterns were also evaluated using eight primers. RESULTS: MSI was detected in two (D6S251, D9S50) of the eight actinic keratosis patients. Among the 8 patients who had squamous cell carcinoma-I and provided informative results, a single patient exhibited two LOH (D6S251, D9S287) and two instances of MSI (D9S180, D9S280). Two LOH and one example of MSI (D6S251) were detected in three out of the 10 patients with squamous cell carcinoma-II. Among the four patients with squamous cell carcinoma-III, one patient displayed three MSIs (D6S251, D6S252, and D9S180) and another patient exhibited an MSI (D9S280). The altered random amplified polymorphic DNA ranged from 70% actinic keratoses, 76% squamous cell carcinoma-I, and 90% squamous cell carcinoma-II, to 100% squamous cell carcinoma-III. DISCUSSION: The increased levels of alterations in the microsatellites, particularly in D6S251, and the random amplified polymorphic DNA fingerprints were statistically significant in squamous cell carcinomas, compared with actinic keratoses. CONCLUSION: The overall alterations that were observed in the repetitive DNA of actinic keratoses and squamous cell carcinomas indicate the presence of a spectrum of malignant progression

    Estimating a theoretical model of state banking competition using a dynamic panel: the Brazilian case

    In this paper we set up a model of regional banking competition based on Bresnahan (1982), Lau (1982) and Nakane (2002). The structural model is estimated using data from eight Brazilian states and a dynamic panel. The results show that on average the level of competition in the Brazilian banking system is high, even tough the null of perfect competition can be rejected at the usual significance levels. This result also prevails at the state level: Rio Grande do Sul, São Paulo, Rio de Janeiro, Pernambuco and Minas Gerais have high degree of competition.O presente artigo propõe um modelo regional de competição bancária baseado nas contribuições de Bresnahan (1982), Lau (1982) e Nakane (2002). O modelo estrutural formuladoé estimado utilizando as informações para oito estados brasileiros e um painel dinâmico. Os resultados mostram que, na média, o nível de competição no sistema bancário brasileiro é alto, embora a hipótese nula de competição perfeita possa ser rejeitada aos níveis usuais de significância. De uma forma geral, este padrão também prevalece nos estados: Rio Grande do Sul, São Paulo, Rio de Janeiro, Pernambuco e Minas Gerais têm elevado grau de competição

    Mechanical damages caused in banana Nanicão in the improvement process, packing and transport

    This study had as objective the evaluation of mechanical damages occurred in banana Nanicão during the improvement process, packing and distribution, identifying the probable critical points. The mechanical damages caused by transport, first cleaning; cleanness and sorting; preservation in the packing, transport, and mature were evaluated. The studied packing had been: torito wooden packing (18 kg), wood type ½ box, (13 kg) and cardboard (18 kg). The stage of preservation and transport of the fruits to the distribution center duplicated the light defects and quintupled the serious defects, causing rottenness after the acclimatization. The cardboard packing did not support the piling up and presented deformations, that resulted in the kneading the fruits of the inferior packing, causing a significant increase of the serious defects. The fruits conditioned in the involved packing of plastic bubble had presented an inferior number of serious damages when compared with the others packing, without the plastic.Este estudo teve como objetivo a avaliação de danos mecânicos ocorridos na banana Nanicão durante o processo de beneficiamento, transporte, embalagem e distribuição, identificando os prováveis pontos críticos. Avaliaram-se os danos mecânicos causados após o transporte, despistilagem e primeira despenca; limpeza e classificação; acondicionamento nas embalagens e transporte, e amadurecimento. As embalagens estudadas foram: embalagem de madeira torito (18 kg), madeira tipo ½ caixa (13 kg) e papelão (18 kg). Verificou-se que, na etapa de acondicionamento e transporte das frutas até o centro de distribuição, duplicaram os defeitos leves e os defeitos graves quintuplicaram, causando podridões após a climatização. A embalagem de papelão não suportou o empilhamento e apresentou deformações, que resultaram no amassamento das frutas que estavam nas embalagens inferiores e no aumento significativo dos defeitos graves.     Investigação no ensino de engenharia biomédica: dois casos de estudo

    Desde os anos oitenta que os avanços das ciências básicas e das ciências da engenharia têm dado um impulso sem precedentes à investigação médica, o que criou as condições para a afirmação da Engenharia Biomédica como ramo autónomo da Engenharia. Este novo ramo da Engenharia impôs-se em domínios que incluem a imagiologia biomédica, a bioinformática, a biotecnologia, a biomecânica, os biomateriais, a engenharia de tecidos, próteses e orgãos artificiais, a análise e modelação de sinais fisiológicos e a gestão de sistemas de saúde. Já no século XXI, o volume de saber acumulado e a motivação para acelerar desenvolvimentos científicos e tecnológicos, levou à criação e organização nas universidades de uma oferta de cursos de graduação e pós-graduação em Engenharia Biomédica. Apesar do espectro muito largo da Engenharia Biomédica e dos cursos criados, é possível, mesmo ao nível de Mestrado pós-Bologna (licenciatura pré-Bologna), dar uma formação avançada que permita aos alunos realizar trabalhos de investigação complexos e com impacto clínico. Neste artigo apresentamos dois destes casos de sucesso, representativos do papel da investigação ao longo do ensino bem actual da Engenharia Biomédica. No primeiro, intitulado “Caracterização Tridimensional da Placa de Ateroma da Bifurcação Carotídea com Ultrasonografia 3D”, é apresentado um método inovador de diagnóstico da Aterosclerose, baseado na reconstrução e caracterização tridimensional da lesão aterosclerótica ao nível da bifurcação carotídea. O segundo trabalho, intitulado “Reconstrução da Frente de Onda e Simulação da Acuidade Visual no Estudo do Impacto das Aberrações Ópticas em Olhos Submetidos a Cirurgia”, apresenta uma ferramenta computacional que gera informação adicional sobre a medição da frente de onda obtida com aparelhos comerciais, permitindo estudar o impacto de aberrações ópticas na acuidade visual de diferentes olhos. Este artigo está organizado em quatro partes. A primeira parte é uma nota histórica introdutória à Engenharia Biomédica em geral. A segunda parte apresenta o que do ponto de vista de ensino superior em Engenharia Biomédica, ao nível de Mestrado Integrado de 1º e 2º ciclos, pode em geral viabilizar a fácil inserção dos estudantes na Investigação, conduzindo-os de imediato a resultados de investigação concretos. A terceira parte constitui a parte central deste artigo onde se apresentam dois exemplos ilustrativos do sucesso da formação de 5 anos em Engenharia Biomédica e da inserção da investigação, desde muito cedo, nessa formação.     Antidepressant effects of a single dose of ayahuasca in patients with recurrent depression: a preliminary report

    Objectives: Ayahuasca (AYA), a natural psychedelic brew prepared from Amazonian plants and rich in dimethyltryptamine (DMT) and harmine, causes effects of subjective well-being and may therefore have antidepressant actions. This study sought to evaluate the effects of a single dose of AYA in six volunteers with a current depressive episode. Methods: Open-label trial conducted in an inpatient psychiatric unit. Results: Statistically significant reductions of up to 82% in depressive scores were observed between baseline and 1, 7, and 21 days after AYA administration, as measured on the Hamilton Rating Scale for Depression (HAM-D), the Montgomery-Åsberg Depression Rating Scale (MADRS), and the Anxious-Depression subscale of the Brief Psychiatric Rating Scale (BPRS). AYA administration resulted in nonsignificant changes in Young Mania Rating Scale (YMRS) scores and in the thinking disorder subscale of the BPRS, suggesting that AYA does not induce episodes of mania and/or hypomania in patients with mood disorders and that modifications in thought content, which could indicate psychedelic effects, are not essential for mood improvement. Conclusions: These results suggest that AYA has fast-acting anxiolytic and antidepressant effects in patients with a depressive disorder

    uncovering differences during Ehrlichia canis infection

    The tick vector Rhipicephalus sanguineus is established as a complex of closely related species with high veterinary-medical significance, in which the presence of different genetic, morphological, and biological traits has resulted in the recognition of different lineages within taxa. One of the most striking differences in the "temperate" and "tropical" lineages of R. sanguineus (s.l.) is the vector competence to Ehrlichia canis, suggesting that these ticks tolerate and react differently to pathogen infection. The present study addresses the SG and MG proteome of the R. sanguineus tropical and temperate lineages and compares their proteomic profile during E. canis infection. Batches of nymphs from the two lineages were allowed to feed on naïve and experimentally E. canis infected dogs and after molting, adults were dissected, and salivary glands and midgut tissues separated. Samples were screened for the presence of E. canis before proteomic analyses. The representation of the proteins identified in infected and non-infected tissues of each lineage was compared and gene ontology used for protein classification. Results highlight important differences in those proteomic profiles that added to previous reported genetic, biological, behavioral, and morphological differences, strengthening the hypothesis of the existence of two different species. Comparing infected and non-infected tissues, the results show that, while in midgut tissues the response to E. canis infection is similar in the salivary glands, the two lineages show a different pattern of protein representation. Focusing on the proteins found only in the infected condition, the data suggests that the cement cone produced during tick feeding may be implicated in pathogen infection. This study adds useful information to the debate on the controversial R. sanguineus systematic status, to the discussion related with the different vectorial competence occurring between the two lineages and identifies potential targets for efficient tick and tick-borne disease control.publishersversionpublishe

    Academic success in schools in the Algarve: when leadership is part of the solution and not of the problem

    This paper presents the results of a case study conducted among school principals in the southern region of Portugal, located in urban, suburban and rural settings. The five cases that comprise this study were subjected to a final rating of “Very Good” in the external evaluation process of schools, whose data were collected through semi-structured interviews and documentary analysis (regulations and educational project, reports of the external evaluation of responsibility of Inspeção Geral de Educação e Ciência de Portugal). The data collected in this exploratory study were systematized and analyzed according to the following indicators: the students’ academic success, the teachers’ professional development, the organizational development and the identifying features of schools principals. Overall, we cannot affirm the existence of differences according to geographical area, although some particularities which proved to be interesting and indicative of good leadership practices could be highlighted.info:eu-repo/semantics/publishedVersio
