    Seasonal changes in nauplii and adults of Calanus hyperboreus (Copepoda) captured in sediment traps, Amundsen Gulf, Canadian Arctic

    AbstractArctic copepods were collected using time-series sediment traps in the Amundsen Gulf, Canadian Arctic, as part of the CASES (Canadian Arctic Shelf Exchange Study) program. Four sediment traps were deployed at three stations (200m depth for CA15, 200 and 400m depths for CA18, and 200m depth for CA20) from October 2003 to July 2004. We collected many copepod nauplii ranging in body length from 155 to 811μm, among which nauplii (mostly N1–2) of Calanus with a size of ∼190μm apparently increased in abundance from February to mid-March. Mature-stage adult females (AF) of Calanus hyperboreus were collected in the traps from February to March, and adult males of C. hyperboreus appeared from November to December at all stations. The likely spawning period of these AF coincided with the occurrence period of ∼190μm-sized nauplii. This finding suggests that these nauplii were derived from C. hyperboreus and that their breeding began at the beginning of November or December at the latest, continuing through April in the Amundsen Gulf

    Structural comparison of the free and DNA-bound forms of the purine repressor DNA-binding domain

    AbstractBackground: The purine repressor (PurR) regulates genes that encode enzymes for purine biosynthesis. PurR has a two domain structure with an N-terminal DNA-binding domain (DBD) and a C-terminal corepressor-binding domain (CBD). The three-dimensional structure of a ternary complex of PurR bound to both corepressor and a specific DNA sequence has recently been determined by X-ray crystallography.Results We have determined the solution structure of the PurR DBD by NMR. It contains three helices, with the first and second helices forming a helix-turn-helix motif. The tertiary structure of the three helices is very similar to that of the corresponding region in the ternary complex. The structure of the hinge helical region, however, which makes specific base contacts in the minor groove of DNA, is disordered in the DNA-free form.Conclusion The stable formation of PurR hinge helices requires PurR dimerization, which brings the hinge regions proximal to each other. The dimerization of the hinge helices is likely to be controled by the CBD dimerization interface, but is induced by specific-DNA binding

    Arctic in Rapid Transition: Priorities for the future of marine and coastal research in the Arctic.

    Understanding and responding to the rapidly occurring environmental changes in the Arctic over the past few decades require new approaches in science. This includes improved collaborations within the scientific community but also enhanced dialogue between scientists and societal stakeholders, especially with Arctic communities. As a contribution to the Third International Conference on Arctic Research Planning (ICARPIII), the Arctic in Rapid Transition (ART) network held an international workshop in France, in October 2014, in order to discuss high-priority requirements for future Arctic marine and coastal research from an early-career scientists (ECS) perspective. The discussion encompassed a variety of research fields, including topics of oceanographic conditions, sea-ice monitoring, marine biodiversity, land-ocean interactions, and geological reconstructions, as well as law and governance issues. Participants of the workshop strongly agreed on the need to enhance interdisciplinarity in order to collect comprehensive knowledge about the modern and past Arctic Ocean's geo-ecological dynamics. Such knowledge enables improved predictions of Arctic developments and provides the basis for elaborate decision-making on future actions under plausible environmental and climate scenarios in the high northern latitudes. Priority research sheets resulting from the workshop's discussions were distributed during the ICARPIII meetings in April 2015 in Japan, and are publicly available online

    Six legged robot (hexapod) has advantage over wheeled robot in its capability to walk over rough terrain. In this paper, hexapod mobility will be tested in order to measure its performance in walk through beam and stair. Behavior based architecture will be used in hexpod, so it can react quickly. Autonomous navigation application has been chosen here in order to prove that the architecture is running well. From simulation result, it can be seen that behavior based hexapod robot has good mobility (it can walk through obstacle that has 10 cm height) and it can accomplish its task to avoid the obstacles and find the light source

    Ecosystem function and particle flux dynamics across the Mackenzie Shelf (Beaufort Sea, Arctic Ocean): an integrative analysis of spatial variability and biophysical forcings

    A. Forest et al. -- 78 pages, 18 figures, 6 tablesA better understanding of how environmental changes affect organic matter fluxes in Arctic marine ecosystems is sorely needed. Here, we combine mooring times-series, ship-based measurements and remote-sensing to assess the variability and forcing factors of vertical fluxes of particulate organic carbon (POC) across the Mackenzie Shelf in 2009. We developed a geospatial model of these fluxes to proceed to an integrative analysis of their biophysical determinants in summer. Flux data were obtained with sediment traps and via a regional empirical algorithm applied to particle size-distributions (17 classes from 0.08–4.2 mm) measured by an Underwater Vision Profiler 5. Redundancy analyses and forward selection of abiotic/biotic parameters, linear trends, and spatial structures (i.e. principal coordinates of neighbor matrices, PCNM), were conducted to partition the variation of POC flux size-classes. Flux variability was explained at 69.5 % by the addition of a linear temporal trend, 7 significant PCNM and 9 biophysical variables. The interaction of all these factors explained 27.8 % of the variability. The first PCNM canonical axis (44.4 % of spatial variance) reflected a shelf-basin gradient controlled by bottom depth and ice concentration (p < 0.01), but a complex assemblage of fine-to-broad scale patterns was also identified. Among biophysical parameters, bacterial production and northeasterly wind (upwelling-favorable) were the two strongest explanatory variables (r2 cum. = 0.37), suggesting that bacteria were associated with sinking material, which was itself partly linked to upwelling-induced productivity. The second most important spatial structure corresponded actually to the two areas where shelf break upwelling is known to occur under easterlies. Copepod biomass was negatively correlated (p < 0.05) with vertical POC fluxes, implying that metazoans played a significant role in the regulation of export fluxes. The low fractal dimension of settling particles (1.26) and the high contribution (~94 %) of fast-sinking small aggregates (<1 mm; 20–30 m d−1) to the mass fluxes suggested that settling material across the region was overall fluffy, porous, and likely resulting from the aggregation of marine detritus, gel-like substances and ballast minerals. Our study demonstrates that vertical POC fluxes in Arctic shelf systems are spatially complex, sensitive to environmental forcings, and determined by both physicochemical mechanisms and food web functioning. In conclusion, we hypothesize that the incorporation of terrestrial matter into the Beaufort Sea food web could be catalyzed by bacteria via the incorporation of dissolved terrestrial carbon liberated through the photo-cleavage and/or hydrolysis of land-derived POC interweaved with marine aggregatesThis work would not have been possible without the professional and enthusiastic assistance of the officers and crew members of the CCGS Amundsen.     Anthropometry and body composition in ethnic Japanese and Caucasian adolescent girls: Considerations on ethnicity and menarche

    Objective: This study aimed to compare the various anthropometric and body composition parameters based on the ethnicity and the absence or presence of menarche.Design: A cross-sectional study with incomplete sampling, using the subject as the evaluation unit.Subjects: the final sample of 550 subjects was composed of 122 Japanese and 179 Caucasian premenarcheal adolescents, and 72 Japanese and 177 Caucasian postmenarcheal adolescents.Methods: the variables of body composition were measured through the following methods: bioelectrical impedance analysis, near-infrared interactance (NIR), Slaughter cutaneous skinfold equations and body mass index. Weight, height and sitting height were also evaluated.Results: the Japanese pre- and postmenarcheal girls presented lower weight and height values when compared with the Caucasian girls. in general, the Japanese premenarcheal girls presented less fat and fat-free mass than the premenarcheal Caucasian girls. This fact was demonstrated through NIR results. Conversely, the Japanese postmenarcheal adolescents accumulated more fat than their Caucasian counterparts. However, significant differences were solely encountered in the values of cutaneous skinfold percent body fat.     A Large Hadron Electron Collider at CERN

    This document provides a brief overview of the recently published report on the design of the Large Hadron Electron Collider (LHeC), which comprises its physics programme, accelerator physics, technology and main detector concepts. The LHeC exploits and develops challenging, though principally existing, accelerator and detector technologies. This summary is complemented by brief illustrations of some of the highlights of the physics programme, which relies on a vastly extended kinematic range, luminosity and unprecedented precision in deep inelastic scattering. Illustrations are provided regarding high precision QCD, new physics (Higgs, SUSY) and electron-ion physics. The LHeC is designed to run synchronously with the LHC in the twenties and to achieve an integrated luminosity of O(100) fb1^{-1}. It will become the cleanest high resolution microscope of mankind and will substantially extend as well as complement the investigation of the physics of the TeV energy scale, which has been enabled by the LHC

    The repertoire of ICE in prokaryotes underscores the unity, diversity, and ubiquity of conjugation

    Horizontal gene transfer shapes the genomes of prokaryotes by allowing rapid acquisition of novel adaptive functions. Conjugation allows the broadest range and the highest gene transfer input per transfer event. While conjugative plasmids have been studied for decades, the number and diversity of integrative conjugative elements (ICE) in prokaryotes remained unknown. We defined a large set of protein profiles of the conjugation machinery to scan over 1,000 genomes of prokaryotes. We found 682 putative conjugative systems among all major phylogenetic clades and showed that ICEs are the most abundant conjugative elements in prokaryotes. Nearly half of the genomes contain a type IV secretion system (T4SS), with larger genomes encoding more conjugative systems. Surprisingly, almost half of the chromosomal T4SS lack co-localized relaxases and, consequently, might be devoted to protein transport instead of conjugation. This class of elements is preponderant among small genomes, is less commonly associated with integrases, and is rarer in plasmids. ICEs and conjugative plasmids in proteobacteria have different preferences for each type of T4SS, but all types exist in both chromosomes and plasmids. Mobilizable elements outnumber self-conjugative elements in both ICEs and plasmids, which suggests an extensive use of T4SS in trans. Our evolutionary analysis indicates that switch of plasmids to and from ICEs were frequent and that extant elements began to differentiate only relatively recently. According to the present results, ICEs are the most abundant conjugative elements in practically all prokaryotic clades and might be far more frequently domesticated into non-conjugative protein transport systems than previously thought. While conjugative plasmids and ICEs have different means of genomic stabilization, their mechanisms of mobility by conjugation show strikingly conserved patterns, arguing for a unitary view of conjugation in shaping the genomes of prokaryotes by horizontal gene transfer

    Association between actual weight status, perceived weight and depressive, anxious symptoms in Chinese adolescents: a cross-sectional study

    <p>Abstract</p> <p>Backgroud</p> <p>The purpose of this study was to describe actual measured weight and perceived weight and to explore associations with depressive, anxiety symptoms in school adolescents in China.</p> <p>Methods</p> <p>A sample of 1144 Chinese adolescents was randomly selected from four schools in Wuhan, China, including 665 boys and 479 girls with ages ranging between 10 and 17 years. Actual measured weight and height and perceived weight status were compared to anxiety and depressive symptoms measured using the revised Self-Rating Anxiety Scale and Children's Depression Inventory. A general linear model was used to compare differences in psychological symptoms among the teenagers with different measured and perceived weights.</p> <p>Results</p> <p>When compared with standardized weight tables (WHO age- and gender-specific body mass index (BMI) cutoffs (2007 reference)), girls were more likely to misperceive themselves as overweight, whereas more boys misclassified their weight status as underweight. The adolescents who perceived themselves as overweight were more likely to experience depressive and anxiety symptoms (except girls) than those who perceived themselves as normal and/or underweight. However, no significant association was found between depressive and anxiety symptoms actual measured weight status.</p> <p>Conclusions</p> <p>Perceived weight status, but not the actual weight status, was associated with psychological symptoms.</p
