    Parsing demographic effects of canine parvovirus on a Minnesota wolf population

    We examined 35 years of relationships among wolf (Canis lupus) pup survival, population change and canine parvovirus (CPV) seroprevalence in northeastern Minnesota to determine when CPV exerted its strongest effects. Using correlation analysis of data from five periods of 7-years each from 1973 through 2007, we learned that the strongest effect of CPV on pup survival (r = -0.73) and on wolf population change (r = -0.92) was during 1987 to 1993. After that, little effect was documented despite a mean CPV seroprevalence from 1994 of 2007 of 70.8% compared with 52.6% during 1987 to 1993. We conclude that after CPV became endemic and produced its peak effect on the study population, that population developed enough immunity to withstand the disease

    Viruses and Drug Resistant Bacteria in Groundwater of Southeastern Minnesota

    ABSTRACT-Karst topography of soils in southeastern Minnesota is responsible for the formation of sinkholes, subsurface cracks, and underground rivers which may enhance the transportation of surface contaminants into groundwater. The present study was conducted to determine the presence of human pathogenic viruses, coliforms, fecal coliforms and coliphages in private rural wells of this area. The occurrence of drug resistance in bacteria isolated from groundwater also was studied. Coliform bacteria were detected at least once from 22 of the 26 sites sampled over 34 months. Water from 10 sites yielded drug-resistant indicator bacteria; 25 of 38 (65.8 percent) total coliforms and 9 of27 (33.3 percent) fecal coliforms tested were found to carry drug resistance. Human enteric viruses were detected by DNA hybridization and/or virus isolation techniques in nine samples from seven different sites, some in the absence of fecal coliforms. Of the 161 samples tested for coliphages, 13 samples from seven sites were found positive. On two occasions, coliphages were isolated from samples in which coliforms were absent. These findings indicate that potential public health problems exist in this region

    DEMOGRAPHIC EFFECTS OF CANINE PARVOVIRUS ON A FREE-RANGING WOLF POPULATION OVER 30 YEARS

    We followed the course of canine parvovirus (CPV) antibody prevalence in a subpopulation of wolves (Canis lupus) in northeastern Minnesota from 1973, when antibodies were first detected, through 2004. Annual early pup survival was reduced by 70%, and wolf population change was related to CPV antibody prevalence. In the greater Minnesota population of 3,000 wolves, pup survival was reduced by 40–60%. This reduction limited the Minnesota wolf population rate of increase to about 4% per year compared with increases of 16–58% in other populations. Because it is young wolves that disperse, reduced pup survival may have caused reduced dispersal and reduced recolonization of new range in Minnesota

    Phylogenetic analysis of Newcastle disease viruses isolated from waterfowl in the Upper Midwest Region of the United States

    <p>Abstract</p> <p>Background</p> <p>This study was conducted to characterize Newcastle disease virus (NDV) isolates obtained from waterfowl from the Upper Midwest region of the United States. A total of 43 NDVs were isolated by inoculation of cloacal samples in embryonated chicken eggs. These isolates were obtained from 24 mallards, seven American green-winged teals, six northern pintails, four blue-winged teals, and two wood ducks. Partial sequences of fusion gene were analyzed to determine the pathotypes and genotypes involved.</p> <p>Results</p> <p>Deduced amino acid sequence of the cleavage site of fusion (F) protein revealed that all isolates had avirulent motifs. Of the 43 isolates, 23 exhibited sequence motif of <sup>111</sup>GGKQGRL<sup>117 </sup>at the cleavage site, 19 exhibited <sup>111</sup>GEKQGRL<sup>117 </sup>while one isolate showed <sup>111</sup>GERQGRL<sup>117</sup>. Phylogenetic analysis based on comparison with different classes of NDVs revealed that all 43 isolates clustered with class II NDVs and none with class I NDVs. Within class II, five isolates were phylogenetically close to genotype I NDVs while the remaining 38 were close to genotype II.</p> <p>Conclusion</p> <p>We conclude that more than one genotype of NDV circulates in waterfowl in the Upper Midwest region of the US. Continuous surveillance may help better understand the epidemiology of NDVs maintained in wild bird populations and their relationship to NDVs in domestic poultry, if any.</p

    Genotypes, Antibiotic Resistance, and ST-8 Genetic Clone in Campylobacter

    Rectal swabs from 155 sheep and 252 goats from Grenada were evaluated to determine the prevalence of Campylobacter spp., antibiotic resistance, and multilocus sequence types. Fifteen Campylobacter isolates were obtained (14 C. jejuni and 1 C. coli). The prevalence (3.7%) did not differ significantly between sheep (4.5%) and goats (3.2%). Among the seven antimicrobials tested, resistance was only detected for tetracycline (30.8%) and metronidazole (38.5%). Campylobacter isolates showed no significant difference between sheep and goats for type of antimicrobial resistance or percent of resistant isolates. Twelve of the isolates were successfully genotyped consisting of four recognized clonal complexes and three novel sequence types. Importantly, one isolate from one goat was identified as the C. jejuni sequence type-8, a zoonotic and tetracycline-resistant clone reported to be a highly virulent clone associated with ovine abortion in the USA. Although most samples were from comingled sheep and goat production units, there were no shared sequence types between these two host species. None of the sequence types identified in this study have previously been reported in poultry in Grenada, suggesting sheep- and goat-specific Campylobacter clones in Grenada. This is the first report of genotyping of Campylobacter isolates from sheep and goats in the Eastern Caribbean

    Hydroxychloroquine: A Comprehensive Review and Its Controversial Role in Coronavirus Disease 2019

    Hydroxychloroquine, initially used as an antimalarial, is used as an immunomodulatory and anti-inflammatory agent for the management of autoimmune and rheumatic diseases such as systemic lupus erythematosus. Lately, there has been interest in its potential efficacy against severe acute respiratory syndrome coronavirus 2, with several speculated mechanisms. The purpose of this review is to elaborate on the mechanisms surrounding hydroxychloroquine. The review is an in-depth analysis of the antimalarial, immunomodulatory, and antiviral mechanisms of hydroxychloroquine, with detailed and novel pictorial explanations. The mechanisms of hydroxychloroquine are related to potential cardiotoxic manifestations and demonstrate potential adverse effects when used for coronavirus disease 2019 (COVID-19). Finally, current literature associated with hydroxychloroquine and COVID-19 has been analyzed to interrelate the mechanisms, adverse effects, and use of hydroxychloroquine in the current pandemic. Currently, there is insufficient evidence about the efficacy and safety of hydroxychloroquine in COVID-19.KEY MESSAGES HCQ, initially an antimalarial agent, is used as an immunomodulatory agent for managing several autoimmune diseases, for which its efficacy is linked to inhibiting lysosomal antigen processing, MHC-II antigen presentation, and TLR functions. HCQ is generally well-tolerated although severe life-threatening adverse effects including cardiomyopathy and conduction defects have been reported. HCQ use in COVID-19 should be discouraged outside clinical trials under strict medical supervision

    Unusual optical quiescence of the classical BL Lac object PKS 0735+178 on intranight time-scale

    We present the result of our extensive intranight optical monitoring of the well-known low-energy peaked BL Lac (LBL) object PKS 0735+178. This long-term follow-up consists of R -band monitoring for a minimum duration of ∼4 hours, on 17 nights spanning 11 years (1998–2008). Using the CCD as an N-star photometer, a detection limit of around 1 per cent was attained for the intranight optical variability (INOV). Remarkably, an INOV amplitude of ≥3 per cent on hour-like time-scale was not observed on any of the 17 nights, even though the likelihood of a typical LBL showing such INOV levels in a single session of >4 hours duration is known to be high (∼50 per cent) . Our observations have thus established a peculiar long-term INOV quiescence of this radio-selected BL Lac object. Moreover, the access to unpublished optical monitoring data of similarly high sensitivity, acquired in another programme, has allowed us to confirm the same anomalous INOV quiescence of this LBL all the way back to 1989, the epoch of its historically largest radio outburst.
