    Assessment of myocardial injury after reperfused infarction by T1ρ cardiovascular magnetic resonance.

    BackgroundThe evolution of T1ρ and of other endogenous contrast methods (T2, T1) in the first month after reperfused myocardial infarction (MI) is uncertain. We conducted a study of reperfused MI in pigs to serially monitor T1ρ, T2 and T1 relaxation, scar size and transmurality at 1 and 4 weeks post-MI.MethodsTen Yorkshire swine underwent 90 min of occlusion of the circumflex artery and reperfusion. T1ρ, T2 and native T1 maps and late gadolinium enhanced (LGE) cardiovascular magnetic resonance (CMR) data were collected at 1 week (n = 10) and 4 weeks (n = 5). Semi-automatic FWHM (full width half maximum) thresholding was used to assess scar size and transmurality and compared to histology. Relaxation times and contrast-to-noise ratio were compared in healthy and remote myocardium at 1 and 4 weeks. Linear regression and Bland-Altman was performed to compare infarct size and transmurality.ResultsRelaxation time differences between infarcted and remote myocardial tissue were ∆T1 (infarct-remote) = 421.3 ± 108.8 (1 week) and 480.0 ± 33.2 ms (4 week), ∆T1ρ = 68.1 ± 11.6 and 74.3 ± 14.2, and ∆T2 = 51.0 ± 10.1 and 59.2 ± 11.4 ms. Contrast-to-noise ratio was CNRT1 = 7.0 ± 3.5 (1 week) and 6.9 ± 2.4 (4 week), CNRT1ρ = 12.0 ± 6.2 and 12.3 ± 3.2, and CNRT2 = 8.0 ± 3.6 and 10.3 ± 5.8. Infarct size was not significantly different for T1ρ, T1 and T2 compared to LGE (p = 0.14) and significantly decreased from 1 to 4 weeks (p < 0.01). Individual infarct size changes were ∆T1ρ = -3.8%, ∆T1 = -3.5% and ∆LGE = -2.8% from 1 - 4 weeks, but there was no observed change in infarct size for T2 or histologically.ConclusionsT1ρ was highly correlated with alterations left ventricle (LV) pathology at 1 and 4 weeks post-MI and therefore it may be a useful method endogenous contrast imaging of infarction

    Tuning Design Parameters of ICAM-1-Targeted 3DNA Nanocarriers to Optimize Pulmonary Targeting Depending on Drug Type

    3DNA holds promise as a carrier for drugs that can be intercalated into its core or linked to surface arms. Coupling 3DNA to an antibody targeting intercellular adhesion molecule 1 (ICAM-1) results in high lung-specific biodistributions in vivo. While the role of individual parameters on ICAM-1 targeting has been studied for other nanocarriers, it has never been examined for 3DNA or in a manner capable of revealing the hierarchic interplay among said parameters. In this study, we used 2-layer vs. 4-layer anti-ICAM 3DNA and radiotracing to examine biodistribution in mice. We found that, below saturating conditions and within the ranges tested, the density of targeting antibodies on 3DNA is the most relevant parameter driving lung targeting over liver clearance, compared to the number of antibodies per carrier, total antibody dose, 3DNA dose, 3DNA size, or the administered concentration, which influenced the dose in organs but not the lung specific-over-liver clearance ratio. Data predicts that lung-specific delivery of intercalating (core loaded) drugs can be tuned using this biodistribution pattern, while that of arm-linked (surface loaded) drugs requires a careful parametric balance because increasing anti-ICAM density reduces the number of 3DNA arms available for drug loading

    Using LiDAR to assess the effect of fire and floods on upland peat bogs, Waterfall Gully, Mount Lofty Ranges, South Australia

    A flood exceeding the 100 year average recurrence interval in November 2005 led to the failure of an upland peat bog in Waterfall Gully. The area is prone to severe bushfire and flood events and the control dam at the base of First Falls was filled with sediment sourced from Wilson Bog. A resistant quartzite bar at Fourth Falls has formed a natural constriction point against which burnt logs and debris have collected following previous fire events forming a natural dam resulting in sediment/peat accumulation upstream. The failure of the bog was inevitable as the vegetative material in the log-jam progressively weakened and rotted. Intense flooding triggered the failure but it was augmented by the build up of a critical mass of sediment upstream of the restriction point. The downstream force of the flood waters and the weight of the saturated bog sediments was enough to overcome the basal frictional forces resulting in slumping and headward erosion. LiDAR data clearly shows an erosion channel scoured out by the flood. Approximately 5000 m3 of sediment (-10,100 tonnes) was washed downstream. LiDAR coupled with a tri-spectral scanner has the capacity to identify other upland peat bogs due to their high NDVI value and assess their stability on steep slopes or narrow valleys. Fire is another risk to the stability of these bogs as it has the potential to remove binding vegetation and expose unconsolidated sediments to erosion during subsequent rain events. Groundsurface and vegetation surface DEM\u27s generated from LiDAR combined with NDVI maps derived from a tri-spectral scanner provide an ideal tool to monitor and assess the risk of slumping in other upland peat bogs

    Clinicopathological evaluation of chronic traumatic encephalopathy in players of American football

    IMPORTANCE: Players of American football may be at increased risk of long-term neurological conditions, particularly chronic traumatic encephalopathy (CTE). OBJECTIVE: To determine the neuropathological and clinical features of deceased football players with CTE. DESIGN, SETTING, AND PARTICIPANTS: Case series of 202 football players whose brains were donated for research. Neuropathological evaluations and retrospective telephone clinical assessments (including head trauma history) with informants were performed blinded. Online questionnaires ascertained athletic and military history. EXPOSURES: Participation in American football at any level of play. MAIN OUTCOMES AND MEASURES: Neuropathological diagnoses of neurodegenerative diseases, including CTE, based on defined diagnostic criteria; CTE neuropathological severity (stages I to IV or dichotomized into mild [stages I and II] and severe [stages III and IV]); informant-reported athletic history and, for players who died in 2014 or later, clinical presentation, including behavior, mood, and cognitive symptoms and dementia. RESULTS: Among 202 deceased former football players (median age at death, 66 years [interquartile range, 47-76 years]), CTE was neuropathologically diagnosed in 177 players (87%; median age at death, 67 years [interquartile range, 52-77 years]; mean years of football participation, 15.1 [SD, 5.2]), including 0 of 2 pre–high school, 3 of 14 high school (21%), 48 of 53 college (91%), 9 of 14 semiprofessional (64%), 7 of 8 Canadian Football League (88%), and 110 of 111 National Football League (99%) players. Neuropathological severity of CTE was distributed across the highest level of play, with all 3 former high school players having mild pathology and the majority of former college (27 [56%]), semiprofessional (5 [56%]), and professional (101 [86%]) players having severe pathology. Among 27 participants with mild CTE pathology, 26 (96%) had behavioral or mood symptoms or both, 23 (85%) had cognitive symptoms, and 9 (33%) had signs of dementia. Among 84 participants with severe CTE pathology, 75 (89%) had behavioral or mood symptoms or both, 80 (95%) had cognitive symptoms, and 71 (85%) had signs of dementia.     The Origin and Kinematics of Cold Gas in Galactic Winds: Insight from Numerical Simulations

    We study the origin of Na I absorbing gas in ultraluminous infrared galaxies motivated by the recent observations by Martin of extremely superthermal linewidths in this cool gas. We model the effects of repeated supernova explosions driving supershells in the central regions of molecular disks with M_d=10^10 M_\sun, using cylindrically symmetric gas dynamical simulations run with ZEUS-3D. The shocked swept-up shells quickly cool and fragment by Rayleigh-Taylor instability as they accelerate out of the dense, stratified disks. The numerical resolution of the cooling and compression at the shock fronts determines the peak shell density, and so the speed of Rayleigh-Taylor fragmentation. We identify cooled shells and shell fragments as Na I absorbing gas and study its kinematics. We find that simulations with a numerical resolution of \le 0.2 pc produce multiple Rayleigh-Taylor fragmented shells in a given line of sight. We suggest that the observed wide Na I absorption lines, = 320 \pm 120 km s^-1 are produced by these multiple fragmented shells traveling at different velocities. We also suggest that some shell fragments can be accelerated above the observed average terminal velocity of 750 km s^-1 by the same energy-driven wind with an instantaneous starburst of \sim 10^9 M_\sun. The bulk of mass is traveling with the observed average shell velocity 330 \pm 100 km s^-1. Our results show that an energy-driven bubble causing Rayleigh-Taylor instabilities can explain the kinematics of cool gas seen in the Na I observations without invoking additional physics relying primarily on momentum conservation, such as entrainment of gas by Kelvin-Helmholtz instabilities, ram pressure driving of cold clouds by a hot wind, or radiation pressure acting on dust.
