Effects of Mesenchymal Stem Cell and Growth Factor Delivery on Cartilage Repair in a Mini-Pig Model
Objective We have recently shown that mesenchymal stem cells (MSCs) embedded in a hyaluronic acid (HA) hydrogel and exposed to chondrogenic factors (transforming growth factor-3 [TGF-3]) produce a cartilage-like tissue in vitro. The current objective was to determine if these same factors could be combined immediately prior to implantation to induce a superior healing response in vivo relative to the hydrogel alone. Design Trochlear chondral defects were created in Yucatan mini-pigs (6 months old). Treatment groups included an HA hydrogel alone and hydrogels containing allogeneic MSCs, TGF-3, or both. Six weeks after surgery, micro-computed tomography was used to quantitatively assess defect fill and subchondral bone remodeling. The quality of cartilage repair was assessed using the ICRS-II histological scoring system and immunohistochemistry for type II collagen. Results Treatment with TGF-3 led to a marked increase in positive staining for collagen type II within defects (P 0.05). Neither condition had an impact on other histological semiquantitative scores (P > 0.05), and inclusion of MSCs led to significantly less defect fill (P 0.05). Conclusions At this early healing time point, treatment with TGF-3 promoted the formation of collagen type II within the defect, while allogeneic MSCs had little benefit. Combination of TGF-3 and MSCs at the time of surgery did not produce a synergistic effect. An in vitro precultured construct made of these components may be required to enhance in vivo repair in this model system
The ALICE TPC, a large 3-dimensional tracking device with fast readout for ultra-high multiplicity events
The design, construction, and commissioning of the ALICE Time-Projection
Chamber (TPC) is described. It is the main device for pattern recognition,
tracking, and identification of charged particles in the ALICE experiment at
the CERN LHC. The TPC is cylindrical in shape with a volume close to 90 m^3 and
is operated in a 0.5 T solenoidal magnetic field parallel to its axis.
In this paper we describe in detail the design considerations for this
detector for operation in the extreme multiplicity environment of central
Pb--Pb collisions at LHC energy. The implementation of the resulting
requirements into hardware (field cage, read-out chambers, electronics),
infrastructure (gas and cooling system, laser-calibration system), and software
led to many technical innovations which are described along with a presentation
of all the major components of the detector, as currently realized. We also
report on the performance achieved after completion of the first round of
stand-alone calibration runs and demonstrate results close to those specified
in the TPC Technical Design Report.Comment: 55 pages, 82 figure
The Swedish Spine Register: development, design and utility
The Swedish Spine Register enables monitoring of surgical activities focusing on changes in trends over time, techniques utilized and outcome, when implemented in general clinical practice. Basic requirements for a prosperous register are unity within the profession, mainly patient-based documentation and a well functioning support system. This presentation focuses on the development and design of the register protocol, problems encountered and solutions found underway. Various examples on how the results can be presented and utilized are given as well as validation. Register data demonstrate significant gender differences in lumbar disc herniation surgery with females having more pain, lower quality of life and more pronounced disability preoperatively while improvement after surgery is similar between genders. Quality of life after surgery for degenerative disorders is significantly improved for disc herniation, stenosis, spondylolisthesis and disc degenerative disorders. Over the last 10 years, surgical treatment for spinal stenosis has increased gradually while disc herniation surgery decreases regarding yearly number of procedures. An added function to the register enables more complex prospective clinical studies to include register data together with data suitable for the individual study. A common core set of demographic, surgical and outcome parameters would enable comparisons of clinical studies within and between nations
Methane dynamics in the subarctic tundra : combining stable isotope analyses, plot- and ecosystem-scale flux measurements
Methane (CH4) fluxes were investigated in a subarctic Russian tundra site in a multi-approach study combining plot-scale data, ecosystem-scale eddy covariance (EC) measurements, and a fine-resolution land cover classification scheme for regional upscaling. The flux data as measured by the two independent techniques resulted in a seasonal (May-October 2008) cumulative CH4 emission of 2.4 (EC) and 3.7 gCH(4) m(-2) (manual chambers) for the source area representative of the footprint of the EC instruments. Upon upscaling for the entire study region of 98.6 km(2), the chamber measured flux data yielded a regional flux estimate of 6.7 gCH(4) m(-2) yr(-1). Our upscaling efforts accounted for the large spatial variability in the distribution of the various land cover types (LCTs) predominant at our study site. Wetlands with emissions ranging from 34 to 53 gCH(4) m(-2) yr(-1) were the most dominant CH4-emitting surfaces. Emissions from thermokarst lakes were an order of magnitude lower, while the rest of the landscape (mineral tundra) was a weak sink for atmospheric methane. Vascular plant cover was a key factor in explaining the spatial variability of CH4 emissions among wetland types, as indicated by the positive correlation of emissions with the leaf area index (LAI). As elucidated through a stable isotope analysis, the dominant CH4 release pathway from wetlands to the atmosphere was plant-mediated diffusion through aerenchyma, a process that discriminates against C-13-CH4. The CH4 released to the atmosphere was lighter than that in the surface porewater, and delta C-13 in the emitted CH4 correlated negatively with the vascular plant cover (LAI). The mean value of delta C-13 obtained here for the emitted CH4, 68.2 +/- 2.0 %, is within the range of values from other wetlands, thus reinforcing the use of inverse modelling tools to better constrain the CH4 budget. Based on the IPCC A1B emission scenario, a temperature increase of 6.1 degrees C relative to the present day has been predicted for the European Russian tundra by the end of the 21st Century. A regional warming of this magnitude will have profound effects on the permafrost distribution leading to considerable changes in the regional landscape with a potential for an increase in the areal extent of CH4-emitting wet surfaces.Peer reviewe
