    GLUT1 expression in malignant tumors and its use as an immunodiagnostic marker

    OBJECTIVE: To analyze glucose transporter 1 expression patterns in malignant tumors of various cell types and evaluate their diagnostic value by immunohistochemistry. INTRODUCTION: Glucose is the major source of energy for cells, and glucose transporter 1 is the most common glucose transporter in humans. Glucose transporter 1 is aberrantly expressed in several tumor types. Studies have implicated glucose transporter 1 expression as a prognostic and diagnostic marker in tumors, primarily in conjunction with positron emission tomography scan data. METHODS: Immunohistochemistry for glucose transporter 1 was performed in tissue microarray slides, comprising 1955 samples of malignant neoplasm from different cell types. RESULTS: Sarcomas, lymphomas, melanomas and hepatoblastomas did not express glucose transporter 1. Fortyseven per cent of prostate adenocarcinomas were positive, as were 29% of thyroid, 10% of gastric and 5% of breast adenocarcinomas. Thirty-six per cent of squamous cell carcinomas of the head and neck were positive, as were 42% of uterine cervix squamous cell carcinomas. Glioblastomas and retinoblastomas showed membranous glucose transporter 1 staining in 18.6% and 9.4% of all cases, respectively. Squamous cell carcinomas displayed membranous expression, whereas adenocarcinomas showed cytoplasmic glucose transporter 1 expression. CONCLUSION: Glucose transporter 1 showed variable expression in various tumor types. Its absence in sarcomas, melanomas, hepatoblastomas and lymphomas suggests that other glucose transporters mediate the glycolytic pathway in these tumors. The data suggest that glucose transporter 1 is a valuable immunohistochemical marker that can be used to identify patients for evaluation by positron emission tomography scan.     Automatic 3D pulmonary nodule detection in CT images: a survey

    This work presents a systematic review of techniques for the 3D automatic detection of pulmonary nodules in computerized-tomography (CT) images. Its main goals are to analyze the latest technology being used for the development of computational diagnostic tools to assist in the acquisition, storage and, mainly, processing and analysis of the biomedical data. Also, this work identifies the progress made, so far, evaluates the challenges to be overcome and provides an analysis of future prospects. As far as the authors know, this is the first time that a review is devoted exclusively to automated 3D techniques for the detection of pulmonary nodules from lung CT images, which makes this work of noteworthy value. The research covered the published works in the Web of Science, PubMed, Science Direct and IEEEXplore up to December 2014. Each work found that referred to automated 3D segmentation of the lungs was individually analyzed to identify its objective, methodology and results. Based on the analysis of the selected works, several studies were seen to be useful for the construction of medical diagnostic aid tools. However, there are certain aspects that still require attention such as increasing algorithm sensitivity, reducing the number of false positives, improving and optimizing the algorithm detection of different kinds of nodules with different sizes and shapes and, finally, the ability to integrate with the Electronic Medical Record Systems and Picture Archiving and Communication Systems. Based on this analysis, we can say that further research is needed to develop current techniques and that new algorithms are needed to overcome the identified drawbacks

    Risk factors for stress and mental disorders in pharmacists and pharmacist aides

    Introduction: Mental disorders and work-related stress may result from situations related to the work process and its poor organization. Among the most affected by mental disorders are the professionals that integrate health services, including pharmacists and pharmacy assistants. The role of the pharmacist and the pharmaceutical care suffered major changes during the twentieth century and culminate in their current powers that, associated with the fact of being in the interface between the prescription and dispensing of medications, with a responsibility to reduce treatment risks and consequently involved in legal and medical/social issues, daily submit the professionals to stressor factors. Objective: To identify the most prevalent risk factors for stress and mental disorders related to work of pharmacist and pharmacist’ aides. Methodology: a literature review, using the keywords “Pharmacy / Pharmacist ‘Aides”, “Mental Disorders”, “Stress” and “Burnout” in the databases: PubMed, SciELO, BVS and Science Direct. Results / Discussion: There were twelve articles, with seven of them stress factors related to the profession and five specifically mental disorders. Among the first, six of them are international literature, and the only national paper is from Bastos et al. (2010), that expresses possible stressors using a series of interviews with community pharmacists in Rio de Janeiro. Mental disorders are covered only undergraduate students in pharmacy, as quoted by Hunt et al. (2013), which show cases of depression and obsessive-compulsive disorder related and sleep disorders and anhedonia. Conclusion: The most prevalent stressor factors found were: long work hours, frequent interruptions and insufficient number of workers. Introdução: Os transtornos mentais e o estresse relacionados ao trabalho podem ser provenientes de situações inerentes ao processo de trabalho e sua organização. Dentre os profissionais mais afetados estão aqueles que integram os serviços de saúde, envolvendo farmacêuticos e auxiliares de farmácia. O papel do farmacêutico e, consequentemente, a assistência farmacêutica, sofreram profundas mudanças durante o século XX que culminaram em suas atuais atribuições que, associadas ao fato de estarem na interface entre a prescrição e a dispensação de medicamentos com a responsabilidade de diminuir riscos terapêuticos, envolvidos em questões de cunho legal e clínico/social, submetem diariamente os profissionais a fatores estressores. Objetivo: Identificar os fatores de risco mais prevalentes para estresse e transtornos mentais relacionados ao trabalho em farmacêuticos e auxiliares de farmácia. Metodologia: Realizada uma revisão bibliográfica, utilizando os descritores “Farmacêuticos/ Auxiliares de farmácia” (“Pharmacist / Pharmacist’ Aides”), “Transtorno mental” (“Mental Disorders”), “Estresse” (“Stress”) e “Esgotamento profissional” (“Burnout”) nas bases de dados: PubMed, ScieLo, BVS e Science Direct. Resultados/Discussão: Foram encontrados doze artigos: sete abordavam fatores estressores relativos à profissão; cinco abordavam especificamente transtornos mentais. Dentre os primeiros, seis deles são de literatura internacional, sendo que o único artigo nacional é o de Bastos et al. (2010), que expressa possíveis fatores estressores após análise de uma série de entrevistas com farmacêuticos comunitários do Rio de Janeiro. Os transtornos de origem mental são abordados em estudos com alunos de graduação em farmácia (Hunt et al., 2013) que mostram casos de depressão e transtorno obsessivo-compulsivo relacionados a distúrbios do sono e anedonia. Conclusão: Os fatores mais prevalentes encontrados foram: extensa jornada de trabalho, número insuficiente de profissionais no ambiente laboral e diversas interrupções durante turno de trabalho

    Metal-free synthesis of indanes by iodine(III)-mediated ring contraction of 1, 2-dihydronaphthalenes

    A metal-free protocol was developed to synthesize indanes by ring contraction of 1, 2-dihydronaphthalenes promoted by PhI(OH)OTs (HTIB or Koser's reagent). This oxidative rearrangement can be performed in several solvents (MeOH, CH3CN, 2 , 2, 2-trifluoroethanol (TFE), 1 , 1, 1, 3, 3, 3-hexafluoroisopropanol (HFIP), and a 1:4 mixture of TFE:CH2Cl2) under mild conditions.     Metallochlorophylls of magnesium, copper and zinc: evaluation of the influence of the first coordination sphere on their solvatochromism and aggregation properties

    In this study the role of different metal centers (magnesium, zinc and copper) on the enhancement of the hydrophilic character of metallochlorophylls, was evaluated. The solvatochromism as well as the aggregation process for these compounds in water/ethanol mixtures at different volume ratios were evaluated using Fluorescence, and Resonant Light Scattering (RLS) measurements, aiming to characterize the behavior of these compounds. Independently on the studied metallochlorophyll, the presence of at least 60% of water results in a considerable increase in the fluorescence emission, probably a direct consequence of a lower aggregation of these compounds, which is confirmed by the results from RLS measurements. Additionally, the results suggest that magnesium and zinc chlorophyll should be promising phototherapeutic agents for Photodynamic Therapy.No presente estudo, o papel de diferentes centros metálicos (magnésio, zinco e cobre) sobre o caráter hidrofílico de metaloclorofilas foi avaliado. O solvatocromismo, assim como o processo de agregação para esses compostos, em misturas água/etanol foram avaliados usando medidas de fluorescência e de espalhamento de luz ressonante (ELR), visando caracterizar o comportamento desses compostos. Independente da metaloclorofila estudada, a presença de ao menos 60% de água resulta em incremento considerável na emissão de fluorescência apresentada por esses compostos. Isso, provavelmente, é uma conseqüência direta de uma menor agregação, o que é confirmado pelos resultados de ELR.     The step of incorporation of Bacillus coagulans GBI-30 6086 into “requeijão cremoso” processed cheese does not affect metabolic homeostasis of rats

    Dairy product consumption is a common habit in Brazil. These products present a good matrix for probiotic incorporation. Thus, in this study the feasibility of producing a probiotic "requeijao cremoso" incorporated with Bacillus coagulans GBI-30 6086 in three different steps and its metabolic effect in an animal model for 2 weeks has been evaluated. Wistar adult health rats were randomized into one to five groups (n = 8 for each group): Control (C); "requeijao cremoso" without probiotic (RC); probiotic inoculated in the milk before pasteurization at 65 degrees C/30 min (RPP); "requeijao cremoso" inoculated before the fusion step and consequently exposed to 90 degrees C/5 min (RPF); and "requeijao cremoso" inoculated after fusion step, i.e., once the product temperature reached 50 degrees C (RPAF). At the end of treatment, analysis of molecular markers of proteins of stress and antioxidant system, HSP 25, 60, 70 and 90, SOD and catalase were performed in the animals' muscles by Western Blot technique. The HSP25, HSP90 and catalase levels of C, RPP, RPF, and RPAF were similar, indicating that the homeostasis remained unchanged. The incorporation of B. coagulans GBI-30 6086 in the "requeijao cremoso" was shown to be stable and the microorganism remained viable in all steps tested.     Conserved and highly expressed tRNA derived fragments in zebrafish

    Background: Small non-coding RNAs (sncRNAs) are a class of transcripts implicated in several eukaryotic regulatory mechanisms, namely gene silencing and chromatin regulation. Despite significant progress in their identification by next generation sequencing (NGS) we are still far from understanding their full diversity and functional repertoire. Results: Here we report the identification of tRNA derived fragments (tRFs) by NGS of the sncRNA fraction of zebrafish. The tRFs identified are 18–30 nt long, are derived from specific 5′ and 3′ processing of mature tRNAs and are differentially expressed during development and in differentiated tissues, suggesting that they are likely produced by specific processing rather than random degradation of tRNAs. We further show that a highly expressed tRF (5′tRF-ProCGG) is cleaved in vitro by Dicer and has silencing ability, indicating that it can enter the RNAi pathway. A computational analysis of zebrafish tRFs shows that they are conserved among vertebrates and mining of publicly available datasets reveals that some 5′tRFs are differentially expressed in disease conditions, namely during infection and colorectal cancer. Conclusions: tRFs constitute a class of conserved regulatory RNAs in vertebrates and may be involved in mechanisms of genome regulation and in some diseases.     Content of inorganic solutes in lettuce grown with brackish water in different hydroponic systems

    A B S T R A C T The objective of this study was to evaluate the growth and accumulation of ions in lettuce grown in different hydroponic systems and recirculation frequencies. The experimental design was randomized blocks with 8 treatments and 4 replicates. The evaluated hydroponic systems were Nutrient Flow Technique (NFT) and an adapted Deep Flow Technique (DFT), the latter with recirculation frequencies of 0.25, 2 and 4 h. Both systems used fresh water and brackish water. Plant growth, accumulation of inorganic solutes (Na + , K + , Cl -and NO 3 -) and the correlation between dry matter production and Na + /K + and Cl -/NO 3 -were evaluated. The salinity of the water used to prepare the nutrient solution caused decrease in growth and K + and NO 3 -levels, and increased contents of Na + and Cl -in the plants. When using fresh water the highest dry matter production was obtained in the NFT system. In case of brackish water the adapted DFT system increased the production, in relation to NFT system (at same recirculation frequency: 0.25 h). It was found that the choice of the type of hydroponic system and recirculation interval for the cultivation of lettuce depends on the quality of the water used to prepare the nutrient solution. Teor de solutos inorgânicos em alface cultivada com água salobra em diferentes sistemas hidropônicos . A salinidade da água utilizada no preparo da solução nutritiva proporcionou diminuição no crescimento e nos teores de K + e NO 3 -e aumentou os teores de Na + e Cl -nas plantas. Quando foi empregada água doce, o sistema NFT proporcionou maior produção de massa de matéria seca. No caso de água salobra o sistema DFT adaptado aumentou a produção, em relação ao sistema NFT (na mesma frequência de recirculação: 0,25 h). Verificou-se que a escolha do sistema hidropônico e do intervalo de recirculação para o cultivo da alface depende da qualidade da água utilizada no preparo da solução nutritiva. R E S U M
