    Methodological considerations in the analysis of fecal glucocorticoid metabolites in tufted capuchins (Cebus apella)

    Analysis of fecal glucocorticoid (GC) metabolites has recently become the standard method to monitor adrenocortical activity in primates noninvasively. However, given variation in the production, metabolism, and excretion of GCs across species and even between sexes, there are no standard methods that are universally applicable. In particular, it is important to validate assays intended to measure GC production, test extraction and storage procedures, and consider the time course of GC metabolite excretion relative to the production and circulation of the native hormones. This study examines these four methodological aspects of fecal GC metabolite analysis in tufted capuchins (Cebus apella). Specifically, we conducted an adrenocorticotrophic hormone (ACTH) challenge on one male and one female capuchin to test the validity of four GC enzyme immunoassays (EIAs) and document the time course characterizing GC me- tabolite excretion in this species. In addition, we compare a common field-friendly technique for extracting fecal GC metabolites to an established laboratory extraction methodology and test for effects of storing “field extracts” for up to 1 yr. Results suggest that a corticosterone EIA is most sensitive to changes in GC production, provides reliable measures when extracted according to the field method, and measures GC metabolites which remain highly stable after even 12 mo of storage. Further, the time course of GC metabolite excretion is shorter than that described yet for any primate taxa. These results provide guidelines for studies of GCs in tufted capuchins, and underscore the importance of validating methods for fecal hormone analysis for each species of interest

    Common marmoset (Callithrix jacchus) personality, subjective well-being, hair cortisol level and AVPR1a, OPRM1, and DAT genotypes

    We studied personality, subjective well-being, and hair cortisol level, in common marmosets Callithrix jacchus, a small, cooperatively breeding New World monkey, by examining their associations with one another and genotypes. Subjects were 68 males and 9 females that lived in the RIKEN Center for Life Science Technologies. Personality and subjective well-being were assessed by keeper ratings on two questionnaires, hair samples were obtained to assay cortisol level and buccal swabs were used to assess AVPR1a, OPRM1 and DAT genotypes. Three personality domains—Dominance, Sociability, and Neuroticism—were identified. Consistent with findings in other species, Sociability and Neuroticism were related to higher and lower subjective well-being, respectively. Sociability was also associated with higher hair cortisol levels. The personality domains and hair cortisol levels were heritable and associated with genotypes: the short form of AVPR1a was associated with lower Neuroticism and the AA genotype of the A111T SNP of OPRM1 was related to lower Dominance, lower Neuroticism, and higher hair cortisol level. Some genetic associations were not in directions that one would expect given findings in other species. These findings provide insights into the proximate and ultimate bases of personality in common marmosets, other primates and humans

    Urinary C-Peptide of Insulin as a Non-Invasive Marker of Nutritional Status: Some Practicalities

    Nutritional status is a critical element of many aspects of animal ecology, but has proven difficult to measure non-invasively in studies of free-ranging animals. Urinary C-peptide of insulin (UCP), a small polypeptide cleaved in an equimolar ratio from proinsulin when the body converts it to insulin, offers great promise in this regard, and recent studies of several non-human primate species have utilized it with encouraging results. Despite this, there are a number of unresolved issues related to the collection, processing, storage and transport of samples. These include: contamination of samples on collection (most commonly by dirt or faeces), short-term storage before returning to a field station, differences in processing and long-term storage methods (blotting onto filter paper, freezing, lyophilizing), and for frozen samples, transportation while keeping samples frozen. Such issues have been investigated for urine samples in particular with respect to their effects on steroid hormone metabolites, but there has been little investigation of their effects on UCP measurement. We collected samples from captive macaques, and undertook a series of experiments where we systematically manipulated samples and tested the effects on subsequent UCP measurements. We show that contamination of urine samples by faeces led to a decrease in UCP levels by >90%, but that contamination with dirt did not have substantial effects. Short-term storage (up to 12 hours) of samples on ice did not affect UCP levels significantly, but medium-term storage (up to 78 hours) did. Freezing and lyophilization for long-term storage did not affect UCP levels, but blotting onto filter paper did. A transportation simulation showed that transporting frozen samples packed in ice and insulated should be acceptable, but only if it can be completed within a period of a few days and if freeze-thaw can be avoided. We use our data to make practical recommendations for fieldworkers

    Bonobo personality traits are heritable and associated with vasopressin receptor gene 1a variation

    Despite being closely related, bonobos and chimpanzees show remarkable behavioral differences, the proximate origins of which remain unknown. This study examined the link between behavioral variation and variation in the vasopressin 1a receptor gene (Avpr1a) in bonobos. Chimpanzees are polymorphic for a ~360 bp deletion (DupB), which includes a microsatellite (RS3) in the 5′ promoter region of Avpr1a. In chimpanzees, the DupB deletion has been linked to lower sociability, lower social sensitivity, and higher anxiety. Chimpanzees and bonobos differ on these traits, leading some to believe that the absence of the DupB deletion in bonobos may be partly responsible for these differences, and to the prediction that similar associations between Avpr1a genotypes and personality traits should be present in bonobos. We identified bonobo personality dimensions using behavioral measures (Sociability(B), Boldness(B), Openness(B), Activity(B)) and trait ratings (Assertiveness(R), Conscientiousness(R), Openness(R), Agreeableness(R), Attentiveness(R), Extraversion(R)). In the present study we found that all 10 dimensions have nonzero heritabilities, indicating there is a genetic basis to personality, and that bonobos homozygous for shorter RS3 alleles were lower in Attentiveness(R) and higher in Openness(B). These results suggest that variations in Avpr1a genotypes explain both within and between species differences in personality traits of bonobos and chimpanzees

    Social personality traits in chimpanzees: temporal stability and structure of behaviourally assessed personality traits in three captive populations

    Animals of many species show consistency in behaviour across time and contexts that differs from other individuals' behaviour in the same population. Such ‘personality’ affects fitness and has therefore become an increasingly relevant research topic in biology. However, consistent variation in social behaviour is understudied. In socially living species, behaviour occurs in a social environment and social interactions have a significant influence on individual fitness. This study addressed personality in social behaviour of 75 captive chimpanzees in three zoos by coding observed behaviour. Fifteen behavioural variables were significantly repeatable (range 0.21–0.93) in at least two of the three zoos. The behaviours showed considerable long-term stability across 3 years, which did not differ from the short-term repeatability. The repeatable behaviours were then analysed with factor analyses. They formed five independent factors, three of which consisted of social traits and were labelled ‘sociability’, ‘positive affect’ and ‘equitability’. The two non-social behaviour factors were labelled ‘anxiety’ and ‘activity’. The factor scores were analysed for sex and population differences. Males had higher factor scores in all traits except ‘sociability’. The factor scores differed also between the zoos, implying considerable external effects in trait expression. The results show that chimpanzees show personality in a broad range of social and non-social behaviours. The study highlights the importance of assessing personality in the social behaviour, especially in cohesive social species, as only then can we understand the consequences of personality in socially living species

    A meta-analysis of correlated behaviours with implications for behavioural syndromes: Mean effect size, publication bias, phylogenetic effects and the role of mediator variables

    In evolutionary and behavioural ecology, increasing attention is being paid to the fact that functionally distinct behaviours are often not independent from each other. Such phenomenon is labelled as behavioural syndrome and is usually demonstrated by phenotypic correlations between behaviours like activity, exploration, aggression and risk-taking across individuals in a population. However, published studies disagree on the strength, and even on the existence of such relationships. To make general inferences from this mixed evidence, we quantitatively reviewed the literature using modern meta-analytic approaches. Based on a large dataset, we investigated the overall relationship between behaviours that are expected to form a syndrome and tested which factors can mediate heterogeneities in study outcomes. The average strength of the phenotypic correlation between behaviours was weak; we found no effect of the phylogeny of species but did observe significant publication bias. However, even accounting for this bias, the mean effect size was positive and statistically different from zero (r = 0. 198). Effect sizes showed considerable heterogeneity within species, implying a role for population-specific adaptation to environmental factors and/or between-study differences in research design. There was a significant positive association between absolute effect size and repeatability of behaviours, suggesting that within-individual variation of behavioural traits can set up an upper limit for the strength of the detected phenotypic correlations. Moreover, spatial overlap between the contexts in which different behaviours were assayed increased the magnitude of the association. The small effect size for the focal relationship implies that a huge sample size would be required to demonstrate a correlation between behaviours with sufficient statistical power, which is fulfilled only in very few studies. This suggests that behavioural syndromes often remain undetected and unpublished. Collectively, our meta-analysis revealed a number of points that might be worth to consider in the future study of behavioural syndromes. © 2012 The Author(s).Peer Reviewe

    Impulsivity and Suicidality in Adolescent Inpatients

    Suicide is the second leading cause of death among adolescents, and impulsivity has emerged as a promising marker of risk. The present study tested whether distinct domains of impulsivity are differentially associated with suicide ideation, plans, and attempts. Adolescents (n = 381; boys = 106, girls = 275) aged 13–19 years (M = 15.62, SD = 1.41) were recruited from an acute, residential treatment program. Within 48 h of admission to the hospital, participants were administered structured clinical interviews assessing mental health disorders and suicidality. Following these interviews, participants completed self-report questionnaires assessing symptom severity and impulsivity. Consistent with past research, an exploratory factor analysis of our 90-item impulsivity instrument resulted in a three-factor solution: Pervasive Influence of Feelings, Feelings Trigger Action, and Lack of Follow-Through. Concurrent analysis of these factors confirmed hypotheses of unique associations with suicide ideation and attempts in the past month. Specifically, whereas Pervasive Influence of Feelings (i.e., tendency for emotions to shape thoughts about the self and the future) is uniquely associated with greater suicidal ideation, Feelings Trigger Action (i.e., impulsive behavioral reactivity to emotions) is uniquely associated with the occurrence of suicide attempts, even after controlling for current psychiatric diagnoses and symptoms. Exploratory gender analyses revealed that these effects were significant in female but not male adolescents. These findings provide new insight about how specific domains of impulsivity differentially increase risk for suicide ideation and attempts. Implications for early identification and prevention of youth suicide are discussed
