    Microstructural evolution of AISI304 stainless steel in the Steckel Mill rolling process

    Includes bibliographical references (leaves [105]-110).The microstructural evolution of AISI304 stainless steel in the Steckel mill rolling process is investigated. This study includes the analysis of mill logs, microstructural examination of the mill product, deformation simulations and post deformation heat treatments. The mill logs from industry contains information about various process variables such as temperature, roll speed, dimensions of the mill strip and forces applied to it during the hot mill rolling process. The strain, strain rates and stresses on the mill strip can be calculated from the mill logs. An understanding of the metallurgical changes during rolling process can be gained by analysing the mean flow stress trends that evolve during rolling. Microstructural examination of the strip in different regions allows us to evaluate the property variations in the strip. This was done with microhardness testing, conventional optical microscopy and electron backscatter diffraction. The middle section of the strip demonstrated full recrystallization whereas the head and tail sections demonstrated no signs of recrystallization. The property differences through thickness proved to be negligible. Laboratory simulation was done in uniaxial compression testing on a Cam Plastometer. It was found that the temperature has a profound influence on the flow stress and the microstructure. The strain rates experienced in hot rolling does not have a significant effect on the flow stress and no measurable effect on the hardness. Heat treatments were done on the deformed uniaxial compression samples. The results of these heat treatments were analysed by two different methods: to deform the sample again after the heat treatment and to compare the yield stress from the first and second deformation and to measure the changes in room temperature hardness with the heat treatment time. The latter led to the development of a time to 50% recrystallization equation that allows the prediction of a direct annealing time for complete softening at the conclusion of the hot rolling process

    A compact, high resolution tracker for cosmic ray muon scattering tomography using semiconductor sensors

The tracker contains silicon strip sensors with an 80 μm pitch, precision mechanics and integrated cooling. The electronic readout of the sensors is performed by a scalable, inexpensive, flexible, FPGA-based system, which is demonstrated to achieve an event rate of 30 kHz. The tracker performance is compared with a Geant4 simulation. A scattering angle resolution compatible with 1.5 mrad at the 4 GeV average cosmic ray muon energy is demonstrated. Images of plastic, iron and lead samples are obtained using an Angle Statistics Reconstruction algorithm. The images demonstrate good contrast between low and high atomic number materials

    Fate and transport of volatile organic compounds in glacial till and groundwater at an industrial site in Northern Ireland

    Volatile organic compound (VOC) contamination of subsurface geological material and groundwater was discovered on the Nortel Monkstown industrial site, Belfast, Northern Ireland. The objectives of this study were to (1) investigate the characteristics of the geological material and its influences on contaminated groundwater flow across the site using borehole logs and hydrological evaluations, and (2) identify the contaminants and examine their distribution in the subsurface geological material and groundwater using chemical analysis. This report focuses on the eastern car park (ECP) which was a former storage area associated with trichloroethene (TCE) degreasing operations. This is where the greatest amount of volatile organic compounds (VOCs), particularly TCE, were detected. The study site is on a complex deposit of clayey glacial till with discontinuous coarser grained lenses, mainly silts, sands and gravel, which occur at 0.45-7.82 m below ground level (bgl). The lenses overall form an elongated formation that acts as a small unconfined shallow aquifer. There is a continuous low permeable stiff clayey till layer beneath the lenses that performs as an aquitard to the groundwater. Highest concentrations of VOCs, mainly TCE, in the geological material and groundwater are in these coarser lenses at similar to 4.5-7 m bgl. Highest TCE measurements at 390,000 mu g L-1 for groundwater and at 39,000 mu g kg(-1) at 5.7 m for geological material were in borehole GA19 in the coarse lens zone. It is assumed that TCE gained entrance to the subsurface near this borehole where the clayey till was thin to absent above coarse lenses which provided little retardation to the vertical migration of this dense non-aqueous phase liquid (DNAPL) into the groundwater. However, TCE is present in low concentrations in the geological material overlying the coarse lens zone. Additionally, VOCs appear to be associated with poorly drained layers and in peat < 3.0 m bgl in the ECP. Some indication of natural attenuation as VOCs degradation products vinyl chloride (VC) and dichloromethane (DCM) also occur on the site

    Sperm competition-induced plasticity in the speed of spermatogenesis

    Background: Sperm competition between rival ejaculates over the fertilization of ova typically selects for the production of large numbers of sperm. An obvious way to increase sperm production is to increase testis size, and most empirical work has focussed on this parameter. Adaptive plasticity in sperm production rate could also arise due to variation in the speed with which each spermatozoon is produced, but whether animals can respond to relevant environmental conditions by modulating the kinetics of spermatogenesis in this way has not been experimentally investigated. Results: Here we demonstrate that the simultaneously hermaphroditic flatworm Macrostomum lignano exhibits substantial plasticity in the speed of spermatogenesis, depending on the social context: worms raised under higher levels of sperm competition produce sperm faster. Conclusions: Our findings overturn the prevailing view that the speed of spermatogenesis is a static property of a genotype, and demonstrate the profound impact that social environmental conditions can exert upon a key developmental process. We thus identify, to our knowledge, a novel mechanism through which sperm production rate is maximised under sperm competition

    Accounting for refrigeration heat exchange in energy performance simulations of large food retail buildings

    Heat exchange between chilled food storage and conditioned spaces in large food retail stores is not currently required as part of design stage regulatory compliance energy performance models. Existing work has identified that this exchange has a significant impact on store energy demand and subsequently leads to unrealistic assessment of building performance. Research presented in this article uses whole building dynamic thermal simulation models that are calibrated against real store performance data, quantifying the impact of the refrigeration driven heat exchange. Proxy refrigerated units are used to simulate the impact of these units for the sales floor areas. A methodology is presented that allows these models to be simplified with the aim of calculating a realistic process heat exchange for refrigeration and including this in thermal simulation models; a protocol for the measurement of chilled sales areas and their inclusion in the building models is also proposed. It is intended that this modelling approach and the calculated process heat exchange inputs can be used to improve the dynamic thermal simulation of large food retail stores, reduce gaps between predicted and actual performance and provide more representative inputs for design stage and regulatory compliance energy calculations

    Assessment of a novel, capsid-modified adenovirus with an improved vascular gene transfer profile

    <p>Background: Cardiovascular disorders, including coronary artery bypass graft failure and in-stent restenosis remain significant opportunities for the advancement of novel therapeutics that target neointimal hyperplasia, a characteristic of both pathologies. Gene therapy may provide a successful approach to improve the clinical outcome of these conditions, but would benefit from the development of more efficient vectors for vascular gene delivery. The aim of this study was to assess whether a novel genetically engineered Adenovirus could be utilised to produce enhanced levels of vascular gene expression.</p> <p>Methods: Vascular transduction capacity was assessed in primary human saphenous vein smooth muscle and endothelial cells using vectors expressing the LacZ reporter gene. The therapeutic capacity of the vectors was compared by measuring smooth muscle cell metabolic activity and migration following infection with vectors that over-express the candidate therapeutic gene tissue inhibitor of matrix metalloproteinase-3 (TIMP-3).</p> <p>Results: Compared to Adenovirus serotype 5 (Ad5), the novel vector Ad5T*F35++ demonstrated improved binding and transduction of human vascular cells. Ad5T*F35++ mediated expression of TIMP-3 reduced smooth muscle cell metabolic activity and migration in vitro. We also demonstrated that in human serum samples pre-existing neutralising antibodies to Ad5T*F35++ were less prevalent than Ad5 neutralising antibodies.</p> <p>Conclusions: We have developed a novel vector with improved vascular transduction and improved resistance to human serum neutralisation. This may provide a novel vector platform for human vascular gene transfer.</p&gt

    In vitro and in vivo evaluation of human adenovirus type 49 as a vector for therapeutic applications

    The human adenovirus phylogenetic tree is split across seven species (A–G). Species D adenoviruses offer potential advantages for gene therapy applications, with low rates of pre-exist-ing immunity detected across screened populations. However, many aspects of the basic virology of species D—such as their cellular tropism, receptor usage, and in vivo biodistribution profile— remain unknown. Here, we have characterized human adenovirus type 49 (HAdV-D49)—a rela-tively understudied species D member. We report that HAdV-D49 does not appear to use a single pathway to gain cell entry, but appears able to interact with various surface molecules for entry. As such, HAdV-D49 can transduce a broad range of cell types in vitro, with variable engagement of blood coagulation FX. Interestingly, when comparing in vivo biodistribution to adenovirus type 5, HAdV-D49 vectors show reduced liver targeting, whilst maintaining transduction of lung and spleen. Overall, this presents HAdV-D49 as a robust viral vector platform for ex vivo manipulation of human cells, and for in vivo applications where the therapeutic goal is to target the lung or gain access to immune cells in the spleen, whilst avoiding liver interactions, such as intravascular vaccine applications
