    Peran Taman Bacaan Masyarakat (TBM) Rumah Tukik Dalam Meningkatkan Kreativitas Seni Anak dan Remaja di Kelurahan Bandulu Kecamatan Anyer Banten

    Tujuan dari penelitian ini adalah untuk mengetahui Peran Taman Bacaan Masyarakat (TBM) Rumah Tukik Dalam Meningkatan Kreativitas Seni Anak dan Remaja di Kelurahan Bandulu Kecamatan Anyer Banten. Penelitian ini menggunakan metode kualitatif dengan pendekatan deskriptif dalam penelitian ini untuk memperoleh data melalui wawancara, observasi, dan dokumentasi. Responden dalam wawancara ini berjumlah 10 orang, termasuk pengelola TBM, ketua TBM, dan pengunjung TBM. Adapun hal yang di teliti adalah (1) Peran TBM (2) Kreativitas seni anak dan remaja TBM (3) Hambatan-hambatan yang dihadapi. Hasil penelitian yang sudah di lakukan menunjukan bahwa TBM Rumah Tukik memiliki peran sebagai sumber pendidikan, informasi, dan hiburan bagi anak dan remaja daerah setempat melalui peran tersebut TBM Rumah Tukik menyediakan berbagai kegiatan kreativitas baik dalam bidang literasi dan kreativitas seni. Kegiatan yang sangat diminati anak dan remaja TBM Rumah Tukik salah satunya kegiatan kreativitas seni. Terbukti melalui kegiatan kreativitas seni anak dan remaja TBM aspek kreativitas meningkat khususnya adaptabilitas (adaptability), kelancaran (fluency), keaslian (authenticity), elaborasi (keterperincian), dan sensitivity (kepekaan). Namun, pada pelaksanaannya TBM dalam menjalankan fungsi dan tugasnya tidak sepenuhnya maksimal ada beberapa hambatan-hambatan yang dihadapi TBM Rumah Tukik yaitu: kesibukan anak-anak, kurangnya wawasan pengelola, serta sarana dan prasarana. Dengan demikian, TBM Rumah Tukik berperan sebagai sarana belajar serta pengembangan minat dan bakat bagi anak dan remaja dalam meningkatkan kreativitas seni mereka

    The conservation value of human-modified landscapes for the world's primates

    Land-use change pushes biodiversity into human-modified landscapes, where native ecosystems are surrounded by anthropic land covers (ALCs). Yet, the ability of species to use these emerging covers remains poorly understood. We quantified the use of ALCs by primates worldwide, and analyzed species' attributes that predict such use. Most species use secondary forests and tree plantations, while only few use human settlements. ALCs are used for foraging by at least 86 species with an important conservation outcome: those that tolerate heavily modified ALCs are 26% more likely to have stable or increasing populations than the global average for all primates. There is no phylogenetic signal in ALCs use. Compared to all primates on Earth, species using ALCs are less often threatened with extinction, but more often diurnal, medium or large-bodied, not strictly arboreal, and habitat generalists. These findings provide valuable quantitative information for improving management practices for primate conservation worldwide

    Type-1 Cannabinoid Receptors Reduce Membrane Fluidity of Capacitated Boar Sperm by Impairing Their Activation by Bicarbonate

    Background Mammalian spermatozoa acquire their full fertilizing ability (so called capacitation) within the female genital tract, where they are progressively exposed to inverse gradients of inhibiting and stimulating molecules. Methodology/Principal Findings In the present research, the effect on this process of anandamide, an endocannabinoid that can either activate or inhibit cannabinoid receptors depending on its concentration, and bicarbonate, an oviductal activatory molecule, was assessed, in order to study the role exerted by the type 1 cannabinoid receptor (CB1R) in the process of lipid membrane remodeling crucial to complete capacitation. To this aim, boar sperm were incubated in vitro under capacitating conditions (stimulated by bicarbonate) in the presence or in the absence of methanandamide (Met-AEA), a non-hydrolysable analogue of anandamide. The CB1R involvement was studied by using the specific inhibitor (SR141716) or mimicking its activation by adding a permeable cAMP analogue (8Br-cAMP). By an immunocytochemistry approach it was shown that the Met-AEA inhibits the bicarbonate-dependent translocation of CB1R from the post-equatorial to equatorial region of sperm head. In addition it was found that Met-AEA is able to prevent the bicarbonate-induced increase in membrane disorder and the cholesterol extraction, both preliminary to capacitation, acting through a CB1R-cAMP mediated pathway, as indicated by MC540 and filipin staining, EPR spectroscopy and biochemical analysis on whole membranes (CB1R activity) and on membrane enriched fraction (C/P content and anisotropy). Conclusions/Significance Altogether, these data demonstrate that the endocannabinoid system strongly inhibits the process of sperm capacitation, acting as membrane stabilizing agent, thus increasing the basic knowledge on capacitation-related signaling and potentially opening new perspectives in diagnostics and therapeutics of male infertility

    Involvement of Complexin 2 in Docking, Locking and Unlocking of Different SNARE Complexes during Sperm Capacitation and Induced Acrosomal Exocytosis

    Acrosomal exocytosis (AE) is an intracellular multipoint fusion reaction of the sperm plasma membrane (PM) with the outer acrosomal membrane (OAM). This unique exocytotic event enables the penetration of the sperm through the zona pellucida of the oocyte. We previously observed a stable docking of OAM to the PM brought about by the formation of the trans-SNARE complex (syntaxin 1B, SNAP 23 and VAMP 3). By using electron microscopy, immunochemistry and immunofluorescence techniques in combination with functional studies and proteomic approaches, we here demonstrate that calcium ionophore-induced AE results in the formation of unilamellar hybrid membrane vesicles containing a mixture of components originating from the two fused membranes. These mixed vesicles (MV) do not contain the earlier reported trimeric SNARE complex but instead possess a novel trimeric SNARE complex that contained syntaxin 3, SNAP 23 and VAMP 2, with an additional SNARE interacting protein, complexin 2. Our data indicate that the earlier reported raft and capacitation-dependent docking phenomenon between the PM and OAM allows a specific rearrangement of molecules between the two docked membranes and is involved in (1) recruiting SNAREs and complexin 2 in the newly formed lipid-ordered microdomains, (2) the assembly of a fusion-driving SNARE complex which executes Ca2+-dependent AE, (3) the disassembly of the earlier reported docking SNARE complex, (4) the recruitment of secondary zona binding proteins at the zona interacting sperm surface. The possibility to study separate and dynamic interactions between SNARE proteins, complexin and Ca2+ which are all involved in AE make sperm an ideal model for studying exocytosis

    Expression of Tas1 Taste Receptors in Mammalian Spermatozoa: Functional Role of Tas1r1 in Regulating Basal Ca2+ and cAMP Concentrations in Spermatozoa

    Background: During their transit through the female genital tract, sperm have to recognize and discriminate numerous chemical compounds. However, our current knowledge of the molecular identity of appropriate chemosensory receptor proteins in sperm is still rudimentary. Considering that members of the Tas1r family of taste receptors are able to discriminate between a broad diversity of hydrophilic chemosensory substances, the expression of taste receptors in mammalian spermatozoa was examined. Methodology/Principal Findings: The present manuscript documents that Tas1r1 and Tas1r3, which form the functional receptor for monosodium glutamate (umami) in taste buds on the tongue, are expressed in murine and human spermatozoa, where their localization is restricted to distinct segments of the flagellum and the acrosomal cap of the sperm head. Employing a Tas1r1-deficient mCherry reporter mouse strain, we found that Tas1r1 gene deletion resulted in spermatogenic abnormalities. In addition, a significant increase in spontaneous acrosomal reaction was observed in Tas1r1 null mutant sperm whereas acrosomal secretion triggered by isolated zona pellucida or the Ca2+ ionophore A23187 was not different from wild-type spermatozoa. Remarkably, cytosolic Ca2+ levels in freshly isolated Tas1r1-deficient sperm were significantly higher compared to wild-type cells. Moreover, a significantly higher basal cAMP concentration was detected in freshly isolated Tas1r1-deficient epididymal spermatozoa, whereas upon inhibition of phosphodiesterase or sperm capacitation, the amount of cAMP was not different between both genotypes. Conclusions/Significance: Since Ca2+ and cAMP control fundamental processes during the sequential process of fertilization, we propose that the identified taste receptors and coupled signaling cascades keep sperm in a chronically quiescent state until they arrive in the vicinity of the egg - either by constitutive receptor activity and/or by tonic receptor activation by gradients of diverse chemical compounds in different compartments of the female reproductive tract

    This chapter on Asian urbanism begins by examining how Asian urbanism can be seen as both actually existing and imagined, taking into consideration the ways in which Asian urbanism has entailed the use of successful Asian cities as reference points for other cities in the Global South on the one hand, and how such referencing practices often entail the rendering of Asian urbanism as imagined models and ideologies that are detached from the realities of the receiving end of the model transfer on the other. The ensuing section examines how Asian urbanism can be situated in the context of state-society relations, with a particular emphasis on the role of the Asian states that exhibited developmental and/or authoritarian orientations in the late twentieth century. The penultimate section explores the socio-spatiality of Asian urbanism, summarising some salient characteristics of Asian urbanism. The final section concludes with an emphasis on the need of avoiding Asian exceptionalism, and also of having a pluralistic perspective on Asian urbanism
