    Design effects for multiple design samples

    In some situations the sample design of a survey is rather complex, consisting of fundamentally different designs in different domains. The design effect for estimates based upon the total sample is a weighted sum of the domainspecific design effects. We derive these weights under an appropriate model and illustrate their use with data from the European Social Survey (ESS)

    Chemical warfare between leafcutter ant symbionts and a co-evolved pathogen

    Acromyrmex leafcutter ants form a mutually beneficial symbiosis with the fungus Leucoagaricus gongylophorus and with Pseudonocardia bacteria. Both are vertically transmitted and actively maintained by the ants. The fungus garden is manured with freshly cut leaves and provides the sole food for the ant larvae, while Pseudonocardia cultures are reared on the ant-cuticle and make antifungal metabolites to help protect the cultivar against disease. If left unchecked, specialized parasitic Escovopsis fungi can overrun the fungus-garden and lead to colony collapse. We report that Escovopsis upregulates the production of two specialized metabolites when it infects the cultivar. These compounds inhibit Pseudonocardia and one, shearinine D, also reduces worker behavioral defences and is ultimately lethal when it accumulates in ant tissues. Our results are consistent with an active evolutionary arms race between Pseudonocardia and Escovopsis, which modifies both bacterial and behavioral defences such that colony collapse is unavoidable once Escovopsis infections escalate

    A previously unreported 26-membered polyene macrocyclic lactam, sceliphrolactam, was Isolated from an actinomycete, Streptomyces sp., associated with the mud dauber, Sceliphron caementarium. Sceliphrolactam's structure was determined by 1D- and 2D-NMR, MS, UV, and IR spectral analysis. Sceliphrolactam displays antifungal activity against amphotericin B-resistant Candida albicans (MIC = 4 mu g/mL, 8.3 mu M).N

    Politics at the Cutting Edge: Intergovernmental Policy Innovation in the Affordable Care Act

    In the eight years since the passage of the Patient Protection and Affordable Care Act (ACA), state governments have remained critical sites of contention over the law. Intense partisan conflict over ACA implementation has raised questions about traditional theories of intergovernmental relations, which posit that federal–state cooperation depends largely on policy design. Yet, few studies have examined how partisanship, as well as other important factors, shape state policy innovations under the ACA. This article examines the ACA’s State Innovation Models (SIM) initiative. SIM is specifically geared towards incentivizing states to experiment with new models of payment and delivery that can improve health outcomes and/or reduce health-care costs. Drawing on a combination of quantitative and qualitative evidence, we find that states’ participation in SIM is shaped by partisanship, administrative capacity, and state policy legacies. Our findings have implications for future efforts at intergovernmental health reforms

    Reduction of PTEN protein and loss of epidermal growth factor receptor gene mutation in lung cancer with natural resistance to gefitinib (IRESSA)

    Gefitinib (IRESSA), an epidermal growth factor receptor (EGFR) tyrosine kinase (TK) inhibitor, has antitumour activity in the advanced non-small-cell lung cancer (NSCLC) setting. However, in chemotherapy-naïve patients with advanced NSCLC, the addition of gefitinib to standard chemotherapy regimens failed to increase survival. These results suggest the need for improved patient selection and combination rationales for targeted therapies. We have identified subpopulations of an adenocarcinoma cell line that are naturally resistant to gefitinib, and have analysed the cDNA expression profiles, genomic status of EGFR gene and the effect of gefitinib on signalling pathways in these cell lines in order to identify key mechanisms for naturally acquired resistance to gefitinib. Gefitinib-resistant subpopulations demonstrated increased Akt phosphorylation (not inhibited by gefitinib), reduced PTEN protein expression and loss of the EGFR gene mutation when compared with parental cell lines. These differences in Akt and PTEN protein expression were not evident from the cDNA array profiles. These data suggests that (1) the EGFR gene mutation may be possibly lost in some cancer cells with other additional mechanisms for activating Akt, (2) reintroduction of PTEN or pharmacological downregulation of the constitutive PI3K–Akt-pathway activity may be an attractive therapeutic strategy in cancers with gefitinib resistance

    Physiological quality of soybean and wheat seeds produced with alternative potassium sources

    O uso de fontes não convencionais para fornecimento de K às plantas tem sido amplamente estudado, mas os efeitos de materiais alternativos na qualidade fisiológica das sementes não são conhecidos. Este estudo teve como objetivo avaliar a qualidade fisiológica de sementes de soja e trigo em função da aplicação fontes de potássio em uma sucessão de culturas. O delineamento experimental foi o de blocos ao acaso com quatro repetições. Os tratamentos constaram de três fontes de K (KCl, rocha alcalina e fonolito moído, com 58%, 11% e 8.42% de K2O, respectivamente) aplicados em quatro doses (0, 25, 50 e 100 kg K2O ha-1). As doses de potássio foram aplicadas na soja e seu efeito residual foi avaliado na cultura do trigo, cultivado em sucessão. Logo após a colheita, as sementes de soja e trigo foram avaliadas pelos testes de teor de água, massa de sementes, germinação, primeira contagem, condutividade elétrica, comprimento de plântulas e massa da matéria seca de plântulas. Plantas de soja adubadas com fontes alternativas para fornecimento de K produzem sementes com maior massa e menor permeabilidade de membranas comparado às com KCl; maior qualidade fisiológica de sementes de soja e massa de sementes de trigo são obtidas com maiores doses de K2O independente da fonte.The use of unconventional sources of K for plants has been widely studied, but the effects of alternative materials on physiological seed quality are still relatively unknown. The objective of this study was to evaluate the physiological quality of soybean and wheat seeds after using different potassium sources in a crop succession. The experimental design was a completely randomized block with four replications. Treatments consisted of three K sources (KCl, alkaline rock and ground phonolite, with 58%, 11% and 8.42% of K2O, respectively) applied in four doses (0, 25, 50 and 100 kg K2O ha-1). Potassium doses were applied in soybean and their residual effects were evaluated on the following wheat crop. Soybean and wheat seeds were evaluated immediately after harvesting by tests for moisture content, seed weight, germination, first count, electrical conductivity, seedling length and seedling dry matter. Soybean plants fertilized with alternative sources of K produced heavier seeds with a lower coat permeability compared to KCl; the physiological quality of soybean seeds and the weight of wheat seeds increase due to higher K2O doses, independently of their source.UNESP FCA Department of Plant Production/AgricultureUNESP FCA Department of Plant Production/Agricultur

    Microbiomes of ant castes implicate new microbial roles in the fungus-growing ant Trachymyrmex septentrionalis

    Fungus-growing ants employ several defenses against diseases, including disease-suppressing microbial biofilms on their integument and in fungal gardens. Here, we compare the phenology of microbiomes in natural nests of the temperate fungus-growing ant Trachymyrmex septentrionalis using culture-dependent isolations and culture-independent 16S-amplicon 454-sequencing. 454-sequencing revealed diverse actinobacteria associated with ants, including most prominently Solirubrobacter (12.2–30.9% of sequence reads), Pseudonocardia (3.5–42.0%), and Microlunatus (0.4–10.8%). Bacterial abundances remained relatively constant in monthly surveys throughout the annual active period (late winter to late summer), except Pseudonocardia abundance declined in females during the reproductive phase. Pseudonocardia species found on ants are phylogenetically different from those in gardens and soil, indicating ecological separation of these Pseudonocardia types. Because the pathogen Escovopsis is not known to infect gardens of T. septentrionalis, the ant-associated microbes do not seem to function in Escovopsis suppression, but could protect against ant diseases, help in nest sanitation, or serve unknown functions
