    Seasonal biodiversity and ecological studies on the epiphytic microalgae communities in polluted and unpolluted aquatic ecosystem at Assiut, Egypt

    A qualitative and quantitative study on epiphytic microalgae was carried out seasonally from November 2015 to August 2016 to follow up their community structures on aquatic macrophytes related to some physico-chemical properties of two polluted and unpolluted water bodies at Assiut, Egypt. A total of 169 species related to 64 genera of epiphytic microalgae were recorded. The most dominant algal group was Bacillariophyceae (43.2%), followed by Chlorophyceae (34.91%), Cyanophyceae (20.71%) and Euglenophyceae (1.18%). The total number of epiphytic algae fluctuated between 11.1 × 104 ind.g-1 plant dry wt. on Phragmites australis in summer at Nazlet Abdellah (polluted site) and 10.02 × 107 ind.g-1 plant dry wt. on Myriophyllum spicatum in winter at El-Wasta (unpolluted site). Some epiphytic microalgae were dominant as Pseudanabaena limnetica, Calothrix braunii, Scenedesmus acutus, and Ulnaria ulna. Others were specific on certain macrophytes as Aphanocapsa thermalis and Ulothrix sp., which grow on Phragmites australis, while Synechocystis minuscula attached itself on Myriophyllum spicatum. Analysis of PERMANOVA showed that the most important factors that induced the variation in epiphytic microalgae were the temporal variation and host plant. Water temperature, pH, nitrate, chloride, phosphate and total dissolved salts were the highest abiotic factors correlated with the variation in composition of epiphytic microalgae

    Long noncoding RNA metastasis-associated lung adenocarcinoma transcript 1 (MALAT1): A molecular predictor of poor survival in glioblastoma multiforme in Egyptian patients

    Background: Long noncoding RNAs (lncRNAs) are a recently discovered class of transcribed RNA molecules with a length of more than 200 nucleotides. Recent studies have shown that lncRNA metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) could play an important role in carcinogenesis and cancer progression in several types of malignancies.Objective: As little is known about the role and clinical significance of lncRNA MALAT1 in glioblastoma multiform (GBM) patients in Egyptian population, this study aimed to investigate the expressions of lncRNA-MALAT1 in human GBM samples and to correlate these expressions with the available clinicopathological features including patient survival data.Subjects and methods: The relative expression of MALAT1 was determined in 37 human glioblastoma formalin-fixed paraffin embedded (FFPE) tissue samples and 10 FFPE non-neoplastic brain tissues using quantitative reverse transcription polymerase chain reaction (qRT-PCR) technology.Results: The current results revealed that lncRNA MALAT1 expression was down-regulated in all tumor specimens compared to normal tissues. A receiver operating characteristic (ROC) curve analysis showed high diagnostic performance; area under curve (AUC) =0.925 ± 0.038 (P <0.001), 95% CI= 0.850–1.00, with 94.6% sensitivity, and 72.7% specificity. Lower MALAT1 expression was associated with poor prognosis; higher frequency of recurrence (P < 0.044), lower overall survival (P <0.005), and shorter disease-free survival (P < 0.004).Conclusion: Taken together, we could postulate that MALAT1 might have a tumor-suppressive function in GBM in Egyptian population and this specific type of lncRNAs may be included in the lists of both potential prognostic biomarkers and the future therapeutic targets for glioblastomas

    Effects of Grain Refining on Columnar-to-Equiaxed Transition in Aluminum Alloys

    The effects of grain refining in ultra-pure aluminum, commercially pure aluminum (1050), and Al-7%Si binary alloy were investigated, using different additions of Al-10%Ti, Al-5%Ti-1%B, and Al-4%B master alloys. Thermal analysis and metallography were used to assess the variations in microstructure resulting from these additions, at solidification rates of 0.8°C/s and ~10°C/s. The results revealed that addition of Al-4%B to ultra-pure aluminum forms AlB12 and AlB2 which have no grain-refining effect. Without grain refiner addition, the pure aluminum microstructure exhibits a mixture of columnar and equiaxed grains. Addition of 30ppm Ti is sufficient to promote equiaxed grains at ~10°C/s but requires addition of 1000 ppm B to obtain similar results at 0.8°C/s. Increasing the Si content to 7% reduces the initial grain size of pure aluminum from 2800 μm to ~1850 μm, and further to 450 μm with ddition of ~500ppm B. In commercial aluminum, the B reacts with traces of Ti forming Al3Ti and TiB2 phases which are active grain-refiners. In Al-7%Si, Ti reacts with Si forming (Al,Si)2Ti phase, which is a poor refining agent. This phenomenon is termed poisoning. No interaction between B and Si is observed in the commercial aluminum or Al-7%Si alloy when B is added

    HEPATOPROTECTIVE AND HEPATOTHERAPUTIC EFFECTS OF PROPOLIS AGAINST D-GALACTOSAMINE/LIPOPOLYSACCHARIDE-INDUCED LIVER DAMAGE IN RATS

    Objective: The aim of the present study was to investigate the potential hepatoprotective and hepatotherapeutic activities of propolis against D-galactosamine and lipopolysaccharide (D-GaIN/LPS)-induced hepatotoxicity in rats.Methods: Hepatotoxicity was induced in rats by intra peritoneal injection of GalN (300 mg/kg) and LPS (30 μg/kg). In the hepatoprotection experiment, propolis was administered orally for 10 days before induction of hepatoxicity. In another experiment (hepatotherapy), propolis was dosed immediately after GalN/LPS injection.Results: Injection of GalN/LPS to rats induced hepatic damage that was manifested by a significant increase in the activities of aminotransferases, alkaline phosphatase, lactate dehydrogenase and levels of tumor necrosis factor-alpha (TNF-α) and total bilirubin in serum. Liver homogenate of intoxicated animals had the lower content of reduced glutathione with increased levels of the hepatic malondialdehyde and caspase-3 enzyme. Histological data presented marked damage in liver sections of intoxicated rats. Oral dosing of propolis before or once immediately after intoxication reversed these altered parameters near to normal values.Conclusion: Liver apoptotic events such as DNA fragmentation and increased caspase-3 activity observed during intoxication were prevented by pre and post- propolis treatment. These results suggest that propolis could afford significant protection and therapy in alleviation of hepatotoxicity.Â

    Clinical intestinal transplantation: New perspectives and immunologic considerations

    Background: Although tacrolimus-based immunosuppression has made intestinal transplantation feasible, the risk of the requisite chronic high- dose treatment has inhibited the widespread use of these procedures. We have examined our 1990-1997 experience to determine whether immunomodulatory strategies to improve outlook could be added to drug treatment. Study Design: Ninety-eight consecutive patients (59 children, 39 adults) with a panoply of indications received 104 allografts under tacrolimus-based immunosuppression: intestine only (n = 37); liver and intestine (n = 50); or multivisceral (n = 17). Of the last 42 patients, 20 received unmodified adjunct donor bone marrow cells; the other 22 were contemporaneous control patients. Results: With a mean followup of 32 ± 26 months (range, 1-86 months), 12 recipients (3 intestine only, 9 composite grafts) are alive with good nutrition beyond the 5-year milestone. Forty-seven (48%) of the total group survive bearing grafts that provide full (91%) or partial (9%) nutrition. Actuarial patient survival at 1 and 5 years (72% and 48%, respectively) was similar with isolated intestinal and composite graft recipients, but the loss rate of grafts from rejection was highest with intestine alone. The best results were in patients between 2 and 18 years of age (68% at 5 years). Adjunct bone marrow did not significantly affect the incidence of graft rejection, B-cell lymphoma, or the rate or severity of graft-versus-host disease. Conclusions: These results demonstrate that longterm rehabilitation similar to that with the other kinds of organ allografts is achievable with all three kinds of intestinal transplant procedures, that the morbidity and mortality is still too high for their widespread application, and that the liver is significantly but marginally protective of concomitantly engrafted intestine. Although none of the endpoints were markedly altered by donor leukocyte augmentation (and chimerism) with bone marrow, establishment of the safety of this adjunct procedure opens the way to further immune modulation strategies that can be added to the augmentation protocol

    Design, synthesis and antitumor evaluation of novel pyrazolopyrimidines and pyrazoloquinazolines

    A series of N-aryl-7-aryl-pyrazolo[1,5-a]pyrimidines 18a–u and N-aryl-pyrazolo[1,5-a] quinazolines 25a–c were designed and synthesized via the reaction of 5-aminopyrazoles 11a–c with enaminones 12a–g or 19, respectively. The new compounds were screened for their in vitro antitumor activity toward liver (HepG-2) and breast (MCF-7) human cancer cells using 3-[4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide MTT assay. From the results, it was found that all compounds showed dose-dependent cytotoxic activities against both HepG-2 and MCF-7 cells. Two compounds 18o and 18a were selected for further investigations. Cell cycle analysis of liver (HepG-2) cells treated with 18o and breast (MCF-7) cells treated with 18a showed cell cycle arrest at G2/M phase and pro-apoptotic activity as indicated by annexin V-FITC staining.publishedVersio

    EFFECT OF POULTRY MANURE AND NITROGEN BIO-FERTILIZER (NITROBEN) ON GROWTH, YIELD AND CHEMICAL CONTENTS OF SQUASH PLANTS

    Two field experiments were carried out during the two successive seasons of 2005 and 2006 at Banha (Qalubia Governorate) to study the effect of three rates of poultry manures (50, 75 and 100 N unit/fed.) and the biofertilizer nitrogen (nitroben) on growth, yield, quality and chemical composi-tion of squash. Obtained data showed that using 100 N unit/fed. from poultry manure gave the highest vegetative growth characters, yield and quality. Application 100 N unit/fed. from poultry manure also increased the nitrogen percentage and heavy metals in squash tissues. In addition, using nitroben biofertilizer gave the highest vegetative growth characters, yield, quality and nitrogen per-centage. On the contrary, nitroben biofertilizer reduced the heavy metals in squash tissue

    Isolation and characterisation of human gingival margin-derived STRO-1/MACS+ and MACS− cell populations

    Recently, gingival margin-derived stem/progenitor cells isolated via STRO-1/magnetic activated cell sorting (MACS) showed remarkable periodontal regenerative potential in vivo. As a second-stage investigation, the present study's aim was to perform in vitro characterisation and comparison of the stem/progenitor cell characteristics of sorted STRO-1-positive (MACS+) and STRO-1-negative (MACS−) cell populations from the human free gingival margin. Cells were isolated from the free gingiva using a minimally invasive technique and were magnetically sorted using anti-STRO-1 antibodies. Subsequently, the MACS+ and MACS− cell fractions were characterized by flow cytometry for expression of CD14, CD34, CD45, CD73, CD90, CD105, CD146/MUC18 and STRO-1. Colony-forming unit (CFU) and multilineage differentiation potential were assayed for both cell fractions. Mineralisation marker expression was examined using real-time polymerase chain reaction (PCR). MACS+ and MACS− cell fractions showed plastic adherence. MACS+ cells, in contrast to MACS− cells, showed all of the predefined mesenchymal stem/progenitor cell characteristics and a significantly higher number of CFUs (P<0.01). More than 95% of MACS+ cells expressed CD105, CD90 and CD73; lacked the haematopoietic markers CD45, CD34 and CD14, and expressed STRO-1 and CD146/MUC18. MACS− cells showed a different surface marker expression profile, with almost no expression of CD14 or STRO-1, and more than 95% of these cells expressed CD73, CD90 and CD146/MUC18, as well as the haematopoietic markers CD34 and CD45 and CD105. MACS+ cells could be differentiated along osteoblastic, adipocytic and chondroblastic lineages. In contrast, MACS− cells demonstrated slight osteogenic potential. Unstimulated MACS+ cells showed significantly higher expression of collagen I (P<0.05) and collagen III (P<0.01), whereas MACS− cells demonstrated higher expression of osteonectin (P<0.05; Mann–Whitney). The present study is the first to compare gingival MACS+ and MACS− cell populations demonstrating that MACS+ cells, in contrast to MACS− cells, harbour stem/progenitor cell characteristics. This study also validates the effectiveness of the STRO-1/MACS+ technique for the isolation of gingival stem/progenitor cells.     Cognitive Functioning in a Pilot Sample of Childhood Cancer Patients in Egypt

    Abstract A subset of cancer survivors experience cognitive deficits that can last for many years after the completion of chemotherapy. The etiology of this problem is largely unknown, so the present study aimed to assess cognitive functioning in childhood patients with cancer and to investigate the proposed disposing factors including variables related to disease, treatment, and some socio-demographic characteristics. In a case control study parents of 67 cancer patients aged 8-12 years, completed the parent proxy report of PedsQL™ 3.0 Cognitive Functioning Scale (Arabic versions), as well as a separate sheet for socio-demographic data. Control group consisted of 37 healthy subjects from the same age group were subjected to the same methodology for comparison. All patients under the study have successfully accomplished their treatment protocol and were in complete remission during the evaluation. Hematological malignancies represented 70.1% of the patients sample, with the highest proportion for ALL (52.2%). Brain tumors represented 40% of the solid malignancies (29.9% of the study patients). Cognitive functioning score was significantly lower in the solid group (69.6±37.3) compared to the hematologic group (85.1±22.2) (t = 2.1, p =0.038). Cognitive functioning score was also lower in solid group versus control subjects (p =0.047), while it showed no significant difference between hematological malignancies and control group. Older age at diagnosis, urban residence, illiterate mothers, higher duration of treatment as well as long duration of hospital admission were associated with a lower cognitive score in the solid tumors group compared to hematological group
