Condicionantes Práticas e Instrução de Processo

A presente dissertação propõe um projecto de arquitectura de beneficiação e ampliação do Museu Ferroviário de Macinhata do Vouga. Pretende-se abranger um largo espectro de elementos da prática profissional da arquitectura, tentando (dentro do possível) corresponder a uma série de expectativas e condicionantes criadas tanto pela entidade contratante como pela legislação aplicável. Este pretensão ganha real dimensão pela complexidade das relações entre as entidades envolvidas na gestão do museu: Câmara Municipal de Águeda (CMA), Comboios de Portugal (CP), Fundação Museu Nacional Ferroviário (FMNF) e Rede Ferroviária Nacional (REFER), que em 2015 incorpora, por fusão a EP - Estradas de Portugal, passando a denominar-se Infraestruturas de Portugal (IP). Tentar-se-á desmistificar, “desconstruindo” o processo, a consciência generalizada de que os projectos públicos de arquitectura são sempre dotados de grande liberdade criativa, técnica e orçamental. Pese embora o enredo e a trama supracitados, o projecto tentará não deixar de cumprir o seu propósito. Servir o museu através da arquitectura e dinamizar economicamente e socialmente a região do baixo Vouga. O legado da linha ferroviária é de tamanha importância para Macinhata do Vouga que as actuais instalações são insuficientes para promover adequadamente todo o espólio existente. Nesse sentido o projecto prevê uma ampliação significativa do museu, bem como a requalificação do existente. Nesta ampliação e requalificação, o museu será dotado com as melhorias inerentes a um projecto de arquitectura contemporâneo. Não esquecendo a importância da preservação da história e do grande espólio ferroviário existente, este projecto é uma oportunidade para que este legado chegue em melhores condições e a mais gente. Para isso será inevitável requalificar e melhorar o existente, ampliar e abrir portas ao futuro, e atrair mais visitantes com mais conteúdo e diferentes formas.This dissertation proposes an architectural project for the improvement and expansion of the Macinhata do Vouga’s Railway Museum. It is intended to cover a wide spectrum of elements of professional practice in architecture, trying (as far as possible) to meet a series of expectations and conditions created by both the contracting entity and the applicable legislation. This will gains real dimension due to the complexity of the relations between the entities involved in the management of the museum: Águeda City Council, Trains of Portugal, National Railway Museum Foundation and National Railway Grid, which in 2015 incorporates, by merger, Roads of Portugal, changing its name to Infrastructures of Portugal. Attempts will be made to demystify, “deconstructing” the process, the widespread awareness that public architectural projects are always endowed with great creative, technical and budgetary freedom. Despite the aforementioned plot, the project will try not to fail to fulfill its true purpose. Serve the museum through architecture and boost the region of Baixo Vouga economically and socially. The legacy of the railway line is of such importance to Macinhata do Vouga that the current facilities are insufficient to adequately promote the entire existing estate. In this sense, the project foresees a significant expansion of the museum, as well as the requalification of the existing one. In this expansion and requalification, the museum will be endowed with the improvements inherent to a contemporary architectural project. Not forgetting the importance of preserving history and the great existing railway assets, this project is an opportunity for this legacy to arrive in better conditions and to more people. For that, it will be inevitable to requalify and improve the existing, expand and open doors to the future, and attract more visitors with more content and different forms

As Artes no Xadrez da Globalização: Heranças e Desafios no Início de um Novo Milénio

Com o tema presente, As Artes no Xadrez da Globalização: Heranças e Desafios no início de um novo Milénio, pretendemos auscultar qual o lugar das Artes Visuais na contemporaneidade, o que passa, necessariamente, pela procura de um (re)entendimento do próprio conceito de arte. Assim, utilizámos como metodologia o estudo de grandes exposições internacionais, procurando entender quais as constantes / diferenças dos seus modelos expositivos. Ocupámo-nos, nomeadamente, da Documenta de Kassel (2012), da Bienal de Veneza (2013), da Manifesta e da Bienal de São Paulo (2014), tendo visitado três destas exposições nas edições específicas acima anotadas.With this theme, The Arts in the Chess of Globalisation: Legacies and Challenges at the Beginning of a New Millennium, we aimed to understand which is the place of the Visual Arts in contemporary times, a task implying a (re)understanding of the own concept of art. In that sense, we used as methodology the study of some large international exhibitions, trying to understand the constants and differences of their exhibition models. We studied the Documenta of Kassel (2012), the Venice Biennale (2013), the Manifest and the Bienal de São Paulo (2014), and we visited three of these exhibitions in the specific years noted above

Intervention Project - Taking Care with Art: So you'll never forget me!

Introdução: Cuidar-te com Arte: Para que nunca me esqueças! - estudo de caso com perspetiva orientada para a prática, executado na Unidade de Cuidados Continuados Integrados Bento XVI em Fátima, Portugal. Este estudo apresentou-se como um projeto socioeducativo e artístico, implementado junto de indivíduos institucionalizados com perturbação neurocognitiva (PNC major ou ligeira) e dos seus cuidadores informais. Objetivo: O objetivo foi o de compreender qual o papel das práticas artísticas enquanto instrumento de intervenção no restabelecimento de vínculos e laços afetivos entre estas famílias. Métodos: Ao longo de seis sessões de expressão artística, ocorridas quinzenalmente, identificaram-se as potencialidades de linguagens artísticas enquanto linguagens alternativas, no desenvolvimento de uma compreensão empática, procurando compreender o contributo das práticas artísticas na facilitação da expressão de sentimentos por parte dos indivíduos com PNC e no minimizar do sentimento de frustração vivenciado pelos cuidadores informais na comunicação com estes indivíduos. Resultados: Por meio desta intervenção concluiu-se que as linguagens artísticas, contribuem fortemente para a aproximação e (re)criação de laços entre os indivíduos com PNC e os seus cuidadores, na medida em que permitem a estes (re)descobrir capacidades nos seus familiares, que consideravam perdidas. Este tipo de projetos socioeducativos e artísticos possibilita igualmente, atenuar a carga negativa associada a perturbações neurocognitivas. Evaluation of skin absorption of drugs from topical and transdermal formulations

The skin barrier function has been attributed to the stratum corneum and represents a major challenge in clinical practice pertaining to cutaneous administration of drugs. Despite this, a large number of bioactive compounds have been successfully administered via cutaneous administration because of advances in the design of topical and transdermal formulations. In vitro and in vivo evaluations of these novel drug delivery systems are necessary to characterize their quality and efficacy. This review covers the most well-known methods for assessing the cutaneous absorption of drugs as an auxiliary tool for pharmaceutical formulation scientists in the design of drug delivery systems. In vitro methods as skin permeation assays using Franz-type diffusion cells, cutaneous retention and tape-stripping methods to study the cutaneous penetration of drugs, and in vivo evaluations as pre-clinical pharmacokinetic studies in animal models are discussed. Alternative approaches to cutaneous microdialysis are also covered. Recent advances in research on skin absorption of drugs and the effect of skin absorption enhancers, as investigated using confocal laser scanning microscopy, Raman confocal microscopy, and attenuated total reflectance Fourier-transform infrared spectroscopy, are reviewed

Correlação litoestratigráfica entre depósitos plaistocênicos na região brazos-trinity, golfo do méxico. / Lithostratigraphic correlation between plaistocenic deposits in the brazos-trinity region, gulf of mexico.

A correlação litoestratigráfica é de grande importância para determinar regiões com prováveis acumulações de petróleo e gás, além do que, para a investigação de estruturas geológicas em subsuperfície seja no ambiente continental ou marinho, relacionado texturas das rochas, com suas fácies e perfis geofísicos de poços. Neste artigo focamos no estudo de perfis geofísicos de 3 poços de sondagem na região de golfo do México, atribuído interpretações a dados de propriedades físicas, no caso a resposta da radiação de raio gama adjacentes aos poços. A correlação entre os poços é utilizada para fazer uma estimativa visual do ambiente com o objetivo de identificar estruturas propícias para a exploração de recursos naturais. Os resultados mostram que os níveis de profundidade podem ser utilizados para o mapeamento e delineamento dos contatos entre camadas com continuidade lateral. Devido a natureza ambígua de interpretação de perfis de raio gama, utilizamos conjuntamente informações de litologia para cada poço, para aumentar a qualidade do resultado da interpretação. A partir de tais informações foi possível fazer uma correlação litológica dos poços, entre contraste de argila/lama e arenito/areia, esses contatos foram nitidamente identificados nos perfis de raios gama, demonstrando características propensas à presença de hidrocarbonetos. Fazendo uma estimativa visual dessas estruturas, ficou evidente que possível fazer uma interpretação robusta, no que se refere às mudanças de litologias e profundidades adjacentes.

Triggering of protection mechanism against Phoneutria nigriventer spider venom in the brain

In experiments with rats, P. nigriventer venom (PNV) temporarily disrupts the properties of the BBB by affecting both the transcellular and the paracellular route. However, it is unclear how cells and/or proteins participate in the transient opening of the BBB. The present study demonstrates that PNV is a substrate for the multidrug resistance protein-1 (MRP1) in cultured astrocyte and endothelial cells (HUVEC) and increases mrp1 and cx43 and down-regulates glut1 mRNA transcripts in cultured astrocytes. The inhibition of nNOS by 7-nitroindazole suggests that NO derived from nNOS mediates some of these effects by either accentuating or opposing the effects of PNV. In vivo, MRP1, GLUT1 and Cx43 protein expression is increased differentially in the hippocampus and cerebellum, indicating region-related modulation of effects. PNV contains a plethora of Ca(2+), K(+) and Na(+) channel-acting neurotoxins that interfere with glutamate handling. It is suggested that the findings of the present study are the result of a complex interaction of signaling pathways, one of which is the NO, which regulates BBB-associated proteins in response to PNV interference on ions physiology. The present study provides additional insight into PNV-induced BBB dysfunction and shows that a protective mechanism is activated against the venom. Knocking Down Low Molecular Weight Protein Tyrosine Phosphatase (LMW-PTP) Reverts Chemoresistance through Inactivation of Src and Bcr-Abl Proteins

The development of multidrug resistance (MDR) limits the efficacy of continuous chemotherapeutic treatment in chronic myelogenous leukemia (CML). Low molecular weight protein tyrosine phosphatase (LMW-PTP) is up-regulated in several cancers and has been associated to poor prognosis. This prompted us to investigate the involvement of LMW-PTP in MDR. In this study, we investigated the role of LMW-PTP in a chemoresistant CML cell line, Lucena-1. Our results showed that LMW-PTP is highly expressed and 7-fold more active in Lucena-1 cells compared to K562 cells, the non-resistant cell line. Knocking down LMW-PTP in Lucena-1 cells reverted chemoresistance to vincristine and imatinib mesylate, followed by a decrease of Src and Bcr-Abl phosphorylation at the activating sites, inactivating both kinases. On the other hand, overexpression of LMW-PTP in K562 cells led to chemoresistance to vincristine. Our findings describe, for the first time, that LMW-PTP cooperates with MDR phenotype, at least in part, through maintaining Src and Bcr-Abl kinases in more active statuses.
