    Geminivirus replication protein decreases PCNA sumoylation at two acceptor sites

    Geminiviruses are plant viruses with circular, single-stranded DNA (ssDNA) genomes that infect a broad range of plants causing substantial crop diseases worldwide. They replicate in nuclei of infected cells by using host DNA replication machinery and an essential protein encoded in their genome designated Rep (replication-associated protein). This multifunctional protein induces the accumulation of the host factors involved in replication and it is capable of interacting with a lot of plant proteins including PCNA (Proliferating Cell Nuclear Antigen), a processivity factor that coordinates a wide range of processes involved in maintenance, duplication and transmission of the genome, and the sumoylation enzyme that conjugates SUMO to target proteins (SUMO-conjugating enzyme- SCE). PCNA modification by SUMO, and also ubiquitin, has long been known to be of key importance for determining how DNA damage is processed by the replisome and for maintenance of overall genome integrity. In yeast, PCNA sumoylation has been associated to DNA repair involving homologous recombination (HR). Previously, we reported that Rep ectopic expression does not result in broad changes in the sumoylation pattern of plant cells, but it modifies the sumoylation state of selected host proteins. In this work, we show, using a reconstituted sumoylation system in Escherichia coli, that tomato PCNA is sumoylated at two residues, K254 and K164, and that co-expression of the Rep protein suppresses PCNA sumoylation at these lysines.     Stimulated Raman Scattering for All Optical Switches

    We theoretically and experimentally investigate an all optical switch based on stimulated Raman scattering in optical fibers. The experimental setup consists of a Raman circuit of two stages connected in series through a bandpass filter. In the first stage, we have a saturated amplifier, in this stage the pump pulses are saturated when pump and signal are launched to the input or the pump pulses remain without saturation when pump only is launched at the input. The second stage works as the Raman amplifier; for this stage amplification is directly dependent on the pump power entering from the first stage. For the case when pump pulse only is launched at the input pass to the second stage without saturation and amplifies the signal entering in the second stage, very intense signal pulses appear at the output of this stage. For the case when both pump and signal pulses are launched to the input, the pump pulse is saturated in the first stage and the filter rejected the amplified signal, so that only low power pump enters the second stage and consequently no signal pulses appear at the output. We show that the contrast can be improved when using fibers with normal and anomalous dispersion connected in series in the first stage. The best contrast (the ratio of energies) obtained was 15 dB at 6 W pump peak power

    Hydrothermal–microwave processing for starch extraction from mexican avocado seeds: operational conditions and characterization

    Avocado seeds are an agroindustrial residue widely produced in Mexico that are causing various environmental problems due to their accumulation. The evaluation of avocado residues to recover biopolymers by microwave-assisted extraction (MAE) and the characterization of avocado starch properties were studied in the present work. A central-composite design was used to optimize the MAE process. Moreover, a comparison was performed between MAE non-isothermal mode (NO–ISO) and conventional extraction. Starch optimization by MAE was obtained at 161.09 °C for 56.23 min with an extraction yield of 49.52% ± 0.69%, while with NO–ISO at 161 °C was obtained 45.75% ± 2.18%. Conventional extraction was 39.04% ± 2.22%. Compared with conventional starch, MAE starch showed similar proprieties and molecular spectra. In contrast, MAE starch showed high solubility, low water absorption capacity, a non-granular structure with small particle size (<2 μm) and polydispersity of fragments at different sizes of polymers. Therefore, MAE is a viable technology to extract the starch, and avocado seed can be considered an excellent starch source for the development of novel functional foods, contributing to promoting sustainability across the food chain.N/Ainfo:eu-repo/semantics/publishedVersio

    In vivo effects of romidepsin on T-Cell activation, apoptosis and function in the BCN02 HIV-1 kick&Kill clinical trial

    Romidepsin (RMD) is a well-characterized histone deacetylase inhibitor approved for the treatment of cutaneous T-cell lymphoma. in vitro and in vivo studies have demonstrated that it is able to induce HIV-1 gene expression in latently infected CD4+ T cells from HIV-1+ individuals on suppressive antiretroviral therapy. However, in vitro experiments suggested that RMD could also impair T-cell functionality, particularly of activated T cells. Thus, the usefulness of RMD in HIV-1 kick&kill strategies, that aim to enhance the immune system elimination of infected cells after inducing HIV-1 viral reactivation, may be limited. In order to address whether the in vitro observations are replicated in vivo, we determined the effects of RMD on the total and HIV-1-specific T-cell populations in longitudinal samples from the BCN02 kick&kill clinical trial (NCT02616874). BCN02 was a proof-of-concept study in 15 early treated HIV-1+ individuals that combined MVA.HIVconsv vaccination with three weekly infusions of RMD given as a latency reversing agent. Our results show that RMD induced a transient increase in the frequency of apoptotic T cells and an enhanced activation of vaccine-induced T cells. Although RMD reduced the number of vaccine-elicited T cells secreting multiple cytokines, viral suppressive capacity of CD8+ T cells was preserved over the RMD treatment. These observations have important implications for the design of effective kick&kill strategies for the HIV-1 cure

    Producción de aromáticos por despolimerización reductiva de lignina sobre catalizadores carbonosos producidos a partir de lignosulfonato sódico.

    El incremento de la producción de lignina en la industria de la producción de pasta de celulosa y en las biorefinerías lignocelulósicas, junto con la creciente demanda de procesos con cero producción de residuos en el marco de la bioeconomía circular, hace necesario desarrollar nuevas metodologías para la valorización de la lignina. Este trabajo propone su doble valorización mediante su conversión en catalizadores basados en carbono y la aplicación de éstos en su despolimerización reductiva para la producción de monómeros aromáticos de alto valor añadido. Los carbones activados empleados como soporte catalítico fueron preparados por activación química de un lignosulfonato de sodio con H3PO4 en relación 3/1 (masa agente activante/precursor carbonoso) y activados a 500 °C. El carbón activado fue impregnado con distintas cantidades de sales precursoras de níquel, molibdeno y cobalto y sometido a tratamiento térmico a 800 °C, produciendo un catalizador de níquel y dos catalizadores bimetálicos de Ni-Mo, y Co-Mo. La despolimerización reductiva de lignina organosolv se llevó a cabo empleando estos catalizadores en un reactor discontinuo agitado a 350 °C y 100 bar de presión inicial de H2 durante 4 horas. En todas las reacciones se obtuvo una fase gaseosa, una líquida orgánica, una líquida acuosa y una sólida, las cuales fueron caracterizadas a través de diferentes técnicas. El catalizador carbonoso con Ni y Mo fue el más activo, despolimerizando en gran medida la lignina, y mostrando una moderada actividad en la hidrodesoxigenación de los correspondientes monoméros aromáticos en condiciones suaves. La fase líquida orgánica obtenida presentó un alto rendimiento hacia los monoméros y elevada selectividad hacia aromáticos oxigenados de alto valor económico, como los alquilfenoles. Además, en las pruebas de reacción utilizando como materia prima otra lignina de alto contenido de azufre, el catalizador mantuvo su actividad.Universidad de Málaga.     The regionalization of framing issues for producers Livestock Group of Validation and Technology Transfer (GGAVATT) from San Jose Miahuatlán Veracruz, Mexico, under the current production of native grass prairie with mainly during the months of higher temperature have a poor performance and thus milk production declines which promotes a great use of concentrated feeds which causes an increase in production cost and less use to them. For this purpose we made a diagnosis of current conditions GGAVATT production, yielding information that helps us source and compared to determine precisely what the effect of applying the proposed alternative. We propose an alternative implementation of pastures with improved pastures needed to produce a greater amount of forage per unit area especially in the colder months.La regionalización de la problemática encuadrando a los productores del Grupo Ganadero de Validación y Transferencia de Tecnología (GGAVATT) de San José Miahuatlán Veracruz, México, bajo la actual situación de producción de praderas básicamente con gramas nativas que durante los meses de menor frío tienen un pobre desempeño y por ende la producción de leche se deteriora lo cual promueve una gran utilización de alimentos concentrados lo que provoca un incremento en el costo de producción y una menor utilidad para estos. Para dicho propósito se realizó un diagnóstico de las condiciones actuales de producción del GGAVATT, arrojando información que nos sirve de partida y de comparación para determinar con precisión cuál es el efecto de aplicar la alternativa propuesta. Se plantea como alternativa la implantación de praderas con pastos mejorados que permitan producir una mayor cantidad de forrajes por unidad de superficie sobre todo en los meses de menor temperatura

    Standardized Outcome Measurement for Patients With Coronary Artery Disease: Consensus From the International Consortium for Health Outcomes Measurement (ICHOM)

    Coronary artery disease (CAD) outcomes consistently improve when they are routinely measured and provided back to physicians and hospitals. The International Consortium for Health Outcomes Measurement (ICHOM) established a Working Group to define a standard set of outcome measures and risk factors of CAD care. Members were drawn from 4 continents and 6 countries. Using a modified Delphi method, the Group defined who should be tracked, what should be measured, and when such measurements should be performed. Thirteen specific outcomes were chosen, including acute complications occurring within 30 days of acute myocardial infarction, coronary artery bypass grafting surgery, or percutaneous coronary intervention; and longitudinal outcomes for up to 5 years for patient‐reported health status (Seattle Angina Questionnaire [SAQ‐7], elements of Rose Dyspnea Score, and Patient Health Questionnaire [PHQ‐2]), cardiovascular hospital admissions, cardiovascular procedures, renal failure, and mortality. Baseline demographic, cardiovascular disease, and comorbidity information is included to improve the interpretability of comparisons

    Diálogos sobre transdisciplina: los investigadores y su objeto de estudio

    A la transdisciplinariedad se le ha definido como “una feliz transgresión de las fronteras entre las disciplinas” y es en este tono en que se presenta esta obra, que recopila las experiencias y reflexiones, las discusiones y propuestas de una veintena de investigadores y académicos que hablan sobre o desde la transdisciplina acerca de los temas de su interés o especialidad. La aproximación se da desde perspectivas académicas diversas y se adereza con expresiones estéticas que van desde la poesía hasta la pintura, a través de las cuales se busca ofrecer un espacio a las rutas posibles y limitaciones connaturales de acceder a la realidad para construir conocimiento “de frontera”, “en las fronteras”. Los abordajes son fruto de la exploración, filiación, encantos y desencantos por parte de los autores con la entidad de su búsqueda, quienes buscan contestar, entre otras, las siguientes cuestiones: ¿Cómo establecer un acercamiento transdisciplinar al objeto de estudio? ¿Qué hace a un objeto de estudio transdisciplinar? ¿Cómo impacta la transdisciplinariedad la identidad del académico?
