    External stimuli help restore post-partum ovarian activity in Pelibuey sheep

    Post-partum anestrus is a problem on farms, and its duration depends on the frequency and intensity of suckling which affects reproduction and production efficiency to become a determining economic factor. The aim of this study was to determine the post-partum reproductive response in ewe to a "male effect" with an ovulation induction protocol of five days using progesterone and the application of a metabolic restorative (MR; Metabolase ®). One hundred and twenty females were randomly assigned to one of four treatments: T1: Continuous suckling (CS; n = 29), T2: CS + MR (n = 29), T3: CS + Male Effect (ME; n = 32), and T4: CS + MR + ME. The percentage of females in ovulation, weight changes among females and lambs, the onset of estrus, calving, fecundity, and prolificacy were also determined. The ovulation percentage was higher in CS + ME and CS + MR + ME (75.0 and 73.3%) than in the other treatments. Weight changes in females and lambs were different among periods. The onset of estrus was similar for CS and CS + MR (25.9 ± 1.9 and 25.7 ± 0.7 h, respectively). The calving percentage was higher for CS + MR (86.2%) than other treatments.     The IBdata Web System for Biological Collections: Design Focused on Usability

    The software design process must put users at the core of the process to enable them to meet their specific objectives effectively, efficiently, and successfully. Thus, a software design for a computing system to consult biological collections guided by the concept of usability will result in an effective and efficient biodiversity informatics tool. Here, we introduce IBdata, a web system to consult biological collections, developed using a design approach based on the architecture of three layers: database, business rules, and user interface. The user interface design was guided by the concept of usability focused on four core concepts: simplicity, adaptability, guide the user through the journey, and feedback. The IBdata web system that we developed is composed of three modules (query, capture and editing, and administration), permitting it to query a database with about 1.7 million specimen records. Biodiversity data query systems must be effective and efficient and should meet the user’s expectations. Software design methodologies play a central role in achieving these goals, and, in this context, interface design techniques that put the user at the core of development are valuable, as in the development of the IBdata web system

    Administration of royal jelly in estrus synchronization protocols for wool and hair sheep

    Objetive: To know the most representative results of the use of royal jelly in reproductive protocols in wool and hair sheep,Desing/methodology/approach: A review of studies referenced in scientific databases published in the livestock sector was carried out. Results: In sheep, the administration of royal jelly in conjunction with reproductive management protocols improves the response to oestrus timing, oestrus onset time and duration, number of large follicles, ovulatory rate, and gestation rate. Study limitations/implications: Royal jelly is a substance that has beneficial effects on sheep reproductive variables, however the cost can be a limitation for its incorporation into the synchronization protocols, in addition to being necessary to elucidate the active metabolites that exert the action and the most effective route of administration.Findings/conclusions: Royal jelly can be an alternative to be incorporated in the estrous synchronization programs in sheep in order to replace some hormonal and without reducing reproductive variables.Objective: To assess the most relevant results on the usage of royal jelly in reproductive protocols of wool and hair sheep.     A Mitocentric View of the Main Bacterial and Parasitic Infectious Diseases in the Pediatric Population

    Infectious diseases occur worldwide with great frequency in both adults and children. Both infections and their treatments trigger mitochondrial interactions at multiple levels: (i) incorporation of damaged or mutated proteins to the complexes of the electron transport chain, (ii) mitochondrial genome (depletion, deletions, and point mutations) and mitochondrial dynamics (fusion and fission), (iii) membrane potential, (iv) apoptotic regulation, (v) generation of reactive oxygen species, among others. Such alterations may result in serious adverse clinical events with great impact on children’s quality of life, even resulting in death. As such, bacterial agents are frequently associated with loss of mitochondrial membrane potential and cytochrome c release, ultimately leading to mitochondrial apoptosis by activation of caspases-3 and -9. Using Rayyan QCRI software for systematic reviews, we explore the association between mitochondrial alterations and pediatric infections including (i) bacterial: M. tuberculosis, E. cloacae, P. mirabilis, E. coli, S. enterica, S. aureus, S. pneumoniae, N. meningitidis and (ii) parasitic: P. falciparum. We analyze how these pediatric infections and their treatments may lead to mitochondrial deterioration in this especially vulnerable population, with the intention of improving both the understanding of these diseases and their management in clinical practice

    Long-term follow-up of certolizumab pegol in uveitis due to immune-mediated inflammatory diseases: multicentre study of 80 patients

    ObjectivesTo evaluate effectiveness and safety of certolizumab pegol (CZP) in uveitis due to immune-mediated inflammatory diseases (IMID).MethodsMulticentre study of CZP-treated patients with IMID uveitis refractory to conventional immunosuppressant. Effectiveness was assessed through the following ocular parameters: best-corrected visual acuity, anterior chamber cells, vitritis, macular thickness and retinal vasculitis. These variables were compared between the baseline, and first week, first, third, sixth months, first and second year.ResultsWe studied 80 (33 men/47 women) patients (111 affected eyes) with a mean age of 41.6 +/- 11.7 years. The IMID included were: spondyloarthritis (n=43), Behcet's disease (n=10), psoriatic arthritis (n=8), Crohn's disease (n=4), sarcoidosis (n=2), juvenile idiopathic arthritis (n=1), reactive arthritis (n=1), rheumatoid arthritis (n=1), relapsing polychondritis (n=1),ConclusionsCZP seems to be effective and safe in uveitis related to different IMID, even in patients refractory to previous biological drugs

    Non-invasive lung cancer diagnosis by detection of GATA6 and NKX2-1 isoforms in exhaled breath condensate

Early LC diagnosis is crucial to reduce the high case fatality rate of this disease. In this case–control study, we developed an accurate LC diagnosis test using retrospectively collected formalin-fixed paraffin-embedded (FFPE) human lung tissues and prospectively collected exhaled breath condensates (EBCs). Following international guidelines for diagnostic methods with clinical application, reproducible standard operating procedures (SOP) were established for every step comprising our LC diagnosis method. We analyzed the expression of distinct mRNAs expressed from GATA6 and NKX2-1, key regulators of lung development. The Em/Ad expression ratios of GATA6 and NKX2-1 detected in EBCs were combined using linear kernel support vector machines (SVM) into the LC score, which can be used for LC detection. LC score-based diagnosis achieved a high performance in an independent validation cohort. We propose our method as a non-invasive, accurate, and low-price option to complement the success of computed tomography imaging (CT) and chest X-ray (CXR) for LC diagnosis

    TOMO-ETNA experiment at Etna volcano: Activities on land

    In the present paper we describe the on-land field operations integrated in the TOMO-ETNA experiment carried out in June-November 2014 at Mt. Etna volcano and surrounding areas. This terrestrial campaign consists in the deployment of 90 short-period portable three-component seismic stations, 17 broadband seismometers and the coordination with 133 permanent seismic station belonging to Italy’s Istituto Nazionale di Geofisica e Vulcanologia (INGV). This temporary seismic network recorded active and passive seismic sources. Active seismic sources were generated by an array of air-guns mounted in the Spanish oceanographic vessel “Sarmiento de Gamboa” with a power capacity of up to 5200 cubic inches. In total more than 26,000 shots were fired and more than 450 local and regional earthquakes were recorded. We describe the whole technical procedure followed to guarantee the success of this complex seismic experiment. We started with the description of the location of the potential safety places to deploy the portable network and the products derived from this search (a large document including full characterization of the sites, owners and indication of how to arrive to them). A full technical description of the seismometers and seismic sources is presented. We show how the portable seismic network was deployed, maintained and recovered in different stages. The large international collaboration of this experiment is reflected in the participation of more than 75 researchers, technicians and students from different institutions and countries in the on-land activities.     Predicting protein linkages in bacteria: Which method is best depends on task

    <p>Abstract</p> <p>Background</p> <p>Applications of computational methods for predicting protein functional linkages are increasing. In recent years, several bacteria-specific methods for predicting linkages have been developed. The four major genomic context methods are: Gene cluster, Gene neighbor, Rosetta Stone, and Phylogenetic profiles. These methods have been shown to be powerful tools and this paper provides guidelines for when each method is appropriate by exploring different features of each method and potential improvements offered by their combination. We also review many previous treatments of these prediction methods, use the latest available annotations, and offer a number of new observations.</p> <p>Results</p> <p>Using <it>Escherichia coli </it>K12 and <it>Bacillus subtilis</it>, linkage predictions made by each of these methods were evaluated against three benchmarks: functional categories defined by COG and KEGG, known pathways listed in EcoCyc, and known operons listed in RegulonDB. Each evaluated method had strengths and weaknesses, with no one method dominating all aspects of predictive ability studied. For functional categories, as previous studies have shown, the Rosetta Stone method was individually best at detecting linkages and predicting functions among proteins with shared KEGG categories while the Phylogenetic profile method was best for linkage detection and function prediction among proteins with common COG functions. Differences in performance under COG versus KEGG may be attributable to the presence of paralogs. Better function prediction was observed when using a weighted combination of linkages based on reliability versus using a simple unweighted union of the linkage sets. For pathway reconstruction, 99 complete metabolic pathways in <it>E. coli </it>K12 (out of the 209 known, non-trivial pathways) and 193 pathways with 50% of their proteins were covered by linkages from at least one method. Gene neighbor was most effective individually on pathway reconstruction, with 48 complete pathways reconstructed. For operon prediction, Gene cluster predicted completely 59% of the known operons in <it>E. coli </it>K12 and 88% (333/418)in <it>B. subtilis</it>. Comparing two versions of the <it>E. coli </it>K12 operon database, many of the unannotated predictions in the earlier version were updated to true predictions in the later version. Using only linkages found by both Gene Cluster and Gene Neighbor improved the precision of operon predictions. Additionally, as previous studies have shown, combining features based on intergenic region and protein function improved the specificity of operon prediction.</p> <p>Conclusion</p> <p>A common problem for computational methods is the generation of a large number of false positives that might be caused by an incomplete source of validation. By comparing two versions of a database, we demonstrated the dramatic differences on reported results. We used several benchmarks on which we have shown the comparative effectiveness of each prediction method, as well as provided guidelines as to which method is most appropriate for a given prediction task.</p
