    High body adiposity drives glucose intolerance and increases cardiovascular risk in normoglycemic subjects

    Objective: We aimed to assess the utility of the 2 - hour oral glucose tolerance test (OGTT) value to discriminate between different cardiometabolic profiles and examine the role of body composition to predict the associated increased risk for glucose impairment, beta cell dysfunction and cardiovascular disease. Methods: Subjects with normal fasting glucose (NFG) completed a 2 - h OGTT and were categorized to the carbohydrate metabolism alterations (CMA) or contro l group based upon a 2 - h glucose threshold of 7.8 mmol l - 1 . Body composition, visceral adipose tissue, OGTT - based parameters and cardiovascular risk factors (CVRF) such as hypertension, dyslipidemia, obstructive sleep apnea, non - alcoholic fatty liver disea se and smoking status, were measured. Results: Subjects with CMA exhibited a significantly higher 1 - h postload glucose, greater decline in beta cell function and CVRF profile. After multivariate adjustment, excess of total body and visceral fat was associ ated with an increased risk of CMA, - cell dysfunction, CVRF and a lower whole - body insulin sensitivity. Conclusions: These data support the ethiopathogenic role of body and visceral fat in the development of glucose derangements and CVRF early on in the metabolic dysregulation process. Thus, body composition analysis and OGTT assessment performed in individuals with NFG enables a better identification of patients at risk of developing type 2 diabetes and cardiovascular disease

    Delocalized single-photon Dicke states and the Leggett- Garg inequality in solid state systems

    We show how to realize a single-photon Dicke state in a large one-dimensional array of two- level systems, and discuss how to test its quantum properties. Realization of single-photon Dicke states relies on the cooperative nature of the interaction between a field reservoir and an array of two-level-emitters. The resulting dynamics of the delocalized state can display Rabi-like oscillations when the number of two-level emitters exceeds several hundred. In this case the large array of emitters is essentially behaving like a mirror-less cavity. We outline how this might be realized using a multiple-quantum-well structure and discuss how the quantum nature of these oscillations could be tested with the Leggett-Garg inequality and its extensions.Comment: 29 pages, 5 figures, journal pape

    Novel insights into the cardio-protective effects of FGF21 in lean and obese rat hearts

    Aims: Fibroblast growth factor 21 (FGF21) is a hepatic metabolic regulator with pleotropic actions. Its plasma concentrations are increased in obesity and diabetes; states associated with an increased incidence of cardiovascular disease. We therefore investigated the direct effect of FGF21 on cardio-protection in obese and lean hearts in response to ischemia. Methods and Results: FGF21, FGF21-receptor 1 (FGFR1) and beta-Klotho (βKlotho) were expressed in rodent, human hearts and primary rat cardiomyocytes. Cardiac FGF21 was expressed and secreted (real time RT-PCR/western blot and ELISA) in an autocrine-paracrine manner, in response to obesity and hypoxia, involving FGFR1-βKlotho components. Cardiac-FGF21 expression and secretion were increased in response to global ischemia. In contrast βKlotho was reduced in obese hearts. In isolated adult rat cardiomyocytes, FGF21 activated PI3K/Akt (phosphatidylinositol 3-kinase/Akt), ERK1/2(extracellular signal-regulated kinase) and AMPK (AMP-activated protein kinase) pathways. In Langendorff perfused rat [adult male wild-type wistar] hearts, FGF21 administration induced significant cardio-protection and restoration of function following global ischemia. Inhibition of PI3K/Akt, AMPK, ERK1/2 and ROR-α (retinoic-acid receptor alpha) pathway led to significant decrease of FGF21 induced cardio-protection and restoration of cardiac function in response to global ischemia. More importantly, this cardio-protective response induced by FGF21 was reduced in obesity, although the cardiac expression profiles and circulating FGF21 levels were increased. Conclusion: In an ex vivo Langendorff system, we show that FGF21 induced cardiac protection and restoration of cardiac function involving autocrine-paracrine pathways, with reduced effect in obesity. Collectively, our findings provide novel insights into FGF21-induced cardiac effects in obesity and ischemia

    Macroscopic quantum resonators (MAQRO)

    Quantum physics challenges our understanding of the nature of physical reality and of space-time and suggests the necessity of radical revisions of their underlying concepts. Experimental tests of quantum phenomena involving massive macroscopic objects would provide novel insights into these fundamental questions. Making use of the unique environment provided by space, MAQRO aims at investigating this largely unexplored realm of macroscopic quantum physics. MAQRO has originally been proposed as a medium-sized fundamental-science space mission for the 2010 call of Cosmic Vision. MAQRO unites two experiments: DECIDE (DECoherence In Double-Slit Experiments) and CASE (Comparative Acceleration Sensing Experiment). The main scientific objective of MAQRO, which is addressed by the experiment DECIDE, is to test the predictions of quantum theory for quantum superpositions of macroscopic objects containing more than 10e8 atoms. Under these conditions, deviations due to various suggested alternative models to quantum theory would become visible. These models have been suggested to harmonize the paradoxical quantum phenomena both with the classical macroscopic world and with our notion of Minkowski space-time. The second scientific objective of MAQRO, which is addressed by the experiment CASE, is to demonstrate the performance of a novel type of inertial sensor based on optically trapped microspheres. CASE is a technology demonstrator that shows how the modular design of DECIDE allows to easily incorporate it with other missions that have compatible requirements in terms of spacecraft and orbit.     Exploring the Cosmic Web in the Sloan Digital Sky Survey Data Release Seven using the Local Dimension

    It is possible to visualize the Cosmic Web as an interconnected network of one-dimensional filaments, two-dimensional sheets and three-dimensional volume-filling structures which we refer to as clusters. We have used the Local Dimension D, which takes values D = 1, 2 and 3 for filaments, sheets and clusters, respectively, to analyse the Cosmic Web in a three-dimensional volume-limited galaxy sample from the Sloan Digital Sky Survey Data Release 7. The analysis was carried out separately using three different ranges of length-scales: 0.5-5, 1-10 and 5-50 h−1Mpch^{-1} {\rm Mpc}. We find that there is a progressive increase in the D values as we move to larger length-scales. At the smallest length-scale, the galaxies predominantly reside in filaments and sheets. There is a shift from filaments to sheets and clusters at larger scales. Filaments are completely absent at the largest length-scale (5-50 h−1Mpch^{-1} {\rm Mpc}). Considering the effect of the density environment on the Cosmic Web, we find that the filaments preferentially inhabit regions with a lower density environment as compared to sheets and clusters which prefer relatively higher density environments. A similar length-scale dependence and environment dependence was also found in a galaxy sample drawn from the Millennium Simulation which was analysed in exactly the same way as the actual data.Comment: 17 pages, 10 figures, published in MNRA

    Treatment of rats with a self-selected hyperlipidic diet, increases the lipid content of the main adipose tissue sites in a proportion similar to that of the lipids in the rest of organs and tissues

    Adipose tissue (AT) is distributed as large differentiated masses, and smaller depots covering vessels, and organs, as well as interspersed within them. The differences between types and size of cells makes AT one of the most disperse and complex organs. Lipid storage is partly shared by other tissues such as muscle and liver. We intended to obtain an approximate estimation of the size of lipid reserves stored outside the main fat depots. Both male and female rats were made overweight by 4-weeks feeding of a cafeteria diet. Total lipid content was analyzed in brain, liver, gastrocnemius muscle, four white AT sites: subcutaneous, perigonadal, retroperitoneal and mesenteric, two brown AT sites (interscapular and perirenal) and in a pool of the rest of organs and tissues (after discarding gut contents). Organ lipid content was estimated and tabulated for each individual rat. Food intake was measured daily. There was a surprisingly high proportion of lipid not accounted for by the main macroscopic AT sites, even when brain, liver and BAT main sites were discounted. Muscle contained about 8% of body lipids, liver 1-1.4%, four white AT sites lipid 28-63% of body lipid, and the rest of the body (including muscle) 38-44%. There was a good correlation between AT lipid and body lipid, but lipid in"other organs" was highly correlated too with body lipid. Brain lipid was not. Irrespective of dietary intake, accumulation of body fat was uniform both for the main lipid storage and handling organs: large masses of AT (but also liver, muscle), as well as in the"rest" of tissues. These storage sites, in specialized (adipose) or not-specialized (liver, muscle) tissues reacted in parallel against a hyperlipidic diet challenge. We postulate that body lipid stores are handled and regulated coordinately, with a more centralized and overall mechanisms than usually assumed

    Cervical intraepithelial neoplasia disease progression is associated with increased vaginal microbiome diversity

    Persistent infection with oncogenic Human Papillomavirus (HPV) is necessary for cervical carcinogenesis. Although evidence suggests that the vaginal microbiome plays a functional role in the persistence or regression of HPV infections, this has yet to be described in women with cervical intra-epithelial neoplasia (CIN). We hypothesised that increasing microbiome diversity is associated with increasing CIN severity. llumina MiSeq sequencing of 16S rRNA gene amplicons was used to characterise the vaginal microbiota of women with low-grade squamous intra-epithelial lesions (LSIL; n = 52), high-grade (HSIL; n = 92), invasive cervical cancer (ICC; n = 5) and healthy controls (n = 20). Hierarchical clustering analysis revealed an increased prevalence of microbiomes characterised by high-diversity and low levels of Lactobacillus spp. (community state type-CST IV) with increasing disease severity, irrespective of HPV status (Normal = 2/20,10%; LSIL = 11/52,21%; HSIL = 25/92,27%; ICC = 2/5,40%). Increasing disease severity was associated with decreasing relative abundance of Lactobacillus spp. The vaginal microbiome in HSIL was characterised by higher levels of Sneathia sanguinegens (P < 0.01), Anaerococcus tetradius (P < 0.05) and Peptostreptococcus anaerobius (P < 0.05) and lower levels of Lactobacillus jensenii (P < 0.01) compared to LSIL. Our results suggest advancing CIN disease severity is associated with increasing vaginal microbiota diversity and may be involved in regulating viral persistence and disease progression

    Examining the validity and consistency of the Adult Eating Behaviour Questionnaire-Español (AEBQ-Esp) and its relationship to BMI in a Mexican population

    Purpose: Appetitive traits in adults and their associations with weight can be measured using the Adult Eating Behaviour Questionnaire (AEBQ). The aim of this study was to confirm the factor structure of the Spanish AEBQ (AEBQ-Esp) in a Mexican sample and explore associations between the eight traits with body mass index (BMI). Method: A sample of 1023 adults, mean age of 36.8 ± 12.8 years, was recruited from Guadalajara, Mexico. Researchers weighed and measured participants, and they completed the AEBQ-Esp either online or in paper format and reported sociodemographic data. To test two alternative factor structures (eight factors including Hunger; seven factors excluding Hunger), confirmatory factor analysis (CFA) was used. Internal reliability was assessed using Cronbach’s alpha; test–retest reliability was assessed using intra-class correlation coefficients. Multivariate linear regressions were used to test for associations between the AEBQ subscales and BMI, adjusted for age, sex, format of AEBQ responses, education, marital and employment status. Results: A seven-factor structure was the best model fit using CFA, excluding the Hunger subscale but similar to the original AEBQ. Internal reliability was good for all subscales (Cronbach’s α = 0.70–0.86), and the intra-class correlation coefficient (0.70–0.91) reflected good test–retest reliability. In the fully adjusted models, Satiety Responsiveness [β = − 0.61; (− 1.01, − 0.21)] and Slowness in Eating [β = − 0.70; (− 1.01, − 0.39)] were negatively associated with BMI, and Emotional Over-Eating [β = 0.94; (0.62, 1.27)] was positively associated with BMI. Conclusions: The AEBQ-Esp (excluding Hunger) appears to be a valid and reliable psychometric questionnaire for measuring appetitive traits in a Mexican Spanish-speaking population. Some traits appear to be associated with BMI in adulthood and warrant further exploration. Level of evidence: Level III evidence obtained from well-designed cohort or case–control analytic studies.
