    Proximate composition and amino acid profile of European eel skin: influence of body weight

A eliminación destes residuos supón un importante inconveniente na industria de transformación de peixe. A pel grosa da anguía considérase un residuo na industria de transformación de peixe e na casa e adoita converterse en produtos de baixo valor no mercado. Os obxectivos deste estudo foron avaliar as variacións da composición química xeral e do contido de aminoácidos da pel de anguía en relación co peso corporal. O contido de humidade diminuíu e o contido de lípidos aumentou co peso das anguías. A pel da anguía tiña un alto contido proteico, que non variou significativamente co peso da anguía. A glicina foi o aminoácido máis abundante. O aminoácido esencial predominante era a leucina. As anguías máis pequenas contiñan niveis máis altos de treonina, valina, isoleucina, leucina, lisina, ácido glutámico e tirosina que os outros grupos. As anguías máis grandes contiñan niveis máis altos de metionina, hidroxiprolina, glicina, arginina, alanina e prolina que os outros grupos. Os índices de proteínas mostraron que a pel das anguías máis pequenas era de maior calidade nutricional.Fish processing industries worldwide are discarding million kilograms of fish waste per year. The disposal of these wastes is an important handicap in fish processing industry. The thick eel skin is considered a waste product in the fish processing industry and in the home and is usually converted into low market-value products. The aims of this study were to evaluate the variations in the general chemical composition and amino acid content of eel skin in relation to body weight. The moisture content decreased and the lipid content increased with the weight of the eels. Eel skin had a high protein content, which did not vary significantly with eel weight. Glycine was the most abundant amino acid. The predominant essential amino acid was leucine. The smaller eels contained higher levels of threonine, valine, isoleucine, leucine, lysine, glutamic acid and tyrosine than the other groups. The larger eels contained higher levels of methionine, hydroxyproline, glycine, arginine, alanine and proline than the other groups. The protein indices showed that the skin from smaller eels was of higher nutritional quality.Xunta de Galicia | Ref. ED431E 2018/0

    Osteogenic effects of simvastatin-loaded mesoporous titania thin films

    The use of statins in the field of bone regeneration is under current investigation due to the existing demand for non-toxic anabolic agents capable of enhancing bone formation in cases of substantial loss. Simvastatin, a coenzyme currently prescribed in clinics to inhibit cholesterol biosynthesis, has been proven to promote osteogenic differentiation by stimulating bone formation and inhibiting osteoclasts activity. We present the loading of simvastatin in mesoporous TiO2 thin films toward combining the pro-osteogenic properties of this molecule with the demonstrated bioactivity of titania. TiO2 thin films processing and characterization were carried out, as well as evaluation of MC3T3-E1 pre-osteoblasts viability when directly incubated with different concentrations of simvastatin, followed by the analysis of osteogenic activity promoted by simvastatin upon loading in the thin films. The accessible porosity of 36% quantified on the 95 ± 5 nm thick mesoporous thin films, together with pore diameters of 5.5 nm, necks between pores of 2.8 nm and interpore distances of 12 ± 2 nm allow the loading of the simvastatin molecule, as confirmed by FTIR spectroscopy. Simvastatin was found to promote MC3T3-E1 pre-osteoblasts viability at concentrations ≤0.01 g l−1, with a cytotoxicity threshold of 0.05 g l−1. We additionally found that film loadings with 0.001 g l−1 simvastatin promotes statistically higher MC3T3-E1 pre-osteoblast proliferation whereas a higher concentration of 0.01 g l−1 leads to statistically higher osteogenic activity (ALP synthesis), after 21 days of incubation, as compared to unloaded films. These results demonstrate the potential of simvastatin local administration based on bioactive mesoporous thin films to promote pro-osteogenic properties. By focusing this strategy on the coating of metallic prostheses, the supply of simvastatin to the target tissue can be favored and risks of systemic side effects will be reduced while enhancing the osteointegration of the implants.Fil: Lopez Alvarez, Miriam.     Assessment of a Novel Automatic Real-Time PCR Assay on the Cobas 4800 Analyzer as a Screening Platform for Hepatitis C Virus Genotyping in Clinical Practice : Comparison with Massive Sequencing

    The unequivocal identification of hepatitis C virus (HCV) subtypes 1a/1b and genotypes 2 to 6 is required for optimizing the effectiveness of interferon-free, direct-acting antiviral therapies. We compared the performance of a new real-time HCV genotyping assay used on the Cobas 4800 system (C4800) with that of high-resolution HCV subtyping (HRCS). In total, 502 samples were used, including 184 samples from chronic HCV patients (from routine laboratory activity during April 2016), 5 stored samples with double HCV genotype infections for testing the limitations of the method, and 313 samples from a screening protocol implemented in our hospital (from May to August 2016) based on the new method to further determine its genotyping accuracy. A total of 282 samples, including 171 from April 2016 (the 13 remaining had too low of a viral load for HRCS), 5 selected with double infections, and 106 from screening, were analyzed by both methods, and 220 were analyzed only by the C4800. The C4800 correctly subtyped 125 of 126 1a/1b samples, and the 1 remaining sample was reported as genotype 1. The C4800 correctly genotyped 38 of 45 non-1a/1b samples (classified by HRCS), and it reported the remaining 7 samples as indeterminate. One hundred two of 106 non-1a/1b genotype samples that were identified using the C4800 for screening were confirmed by HRCS. In the 4 remaining samples, 3 were correctly reported as genotype 1 (without defining the subtype) and 1 was reported as indeterminate. None of the samples were misgenotyped. Four of 7 samples with double HCV infections were correctly genotyped by the C4800. Excluding the 5 selected double-infected samples, the C4800 showed 95.7% concordant results for genotyping HCVs 2 to 6 and 1a/1b subtyping, and 99.2% concordance for subtyping 1a/1b single infections in clinical samples. To improve laboratory workflow, we propose using the C4800 as a first-line test for HCV genotyping and 1a/1b classification, followed by transferring non-1a/1b samples to a center where HRCS is available, if further characterization is needed

    CARMENES input catalog of M dwarfs: VII. New rotation periods for the survey stars and their correlations with stellar activity

    Abridged: We measured photometric and spectroscopic ProtP_{\rm rot} for a large sample of nearby bright M dwarfs with spectral types from M0 to M9, as part of our continual effort to fully characterize the Guaranteed Time Observation programme stars of the CARMENES survey. We determine ProtP_{\rm rot} for 129 stars. Combined with the literature, we tabulate ProtP_{\rm rot} for 261 stars, or 75% of our sample. We evaluate the plausibility of all periods available for this sample by comparing them with activity signatures and checking for consistency between multiple measurements. We find that 166 of these stars have independent evidence that confirmed their ProtP_{\rm rot}. There are inconsistencies in 27 periods, which we classify as debated. A further 68 periods are identified as provisional detections that could benefit from independent verification. We provide an empirical relation for the ProtP_{\rm rot} uncertainty as a function of the ProtP_{\rm rot} value, based on the dispersion of the measurements. We show that published formal errors seem to be often underestimated for periods 10\gtrsim 10 d. We highlight the importance of independent verification on ProtP_{\rm rot} measurements, especially for inactive M dwarfs. We examine rotation-activity relations with emission in X-rays, Hα\alpha, Ca II H & K, and surface magnetic field strengths. We find overall agreement with previous works, as well as tentative differences in the partially versus fully convective subsamples. We show ProtP_{\rm rot} as a function of stellar mass, age, and galactic kinematics. With the notable exception of three transiting planet systems and TZ Ari, all known planet hosts in this sample have Prot15P_{\rm rot} \gtrsim 15 d. This indicates that important limitations need to be overcome before the radial velocity technique can be routinely used to detect and study planets around young and active stars.Comment: Accepted for publication in A&

    Non-productive angiogenesis disassembles Aß plaque-associated blood vessels

    The human Alzheimer’s disease (AD) brain accumulates angiogenic markers but paradoxically, the cerebral microvasculature is reduced around Aß plaques. Here we demonstrate that angiogenesis is started near Aß plaques in both AD mouse models and human AD samples. However, endothelial cells express the molecular signature of non-productive angiogenesis (NPA) and accumulate, around Aß plaques, a tip cell marker and IB4 reactive vascular anomalies with reduced NOTCH activity. Notably, NPA induction by endothelial loss of presenilin, whose mutations cause familial AD and which activity has been shown to decrease with age, produced a similar vascular phenotype in the absence of Aß pathology. We also show that Aß plaque-associated NPA locally disassembles blood vessels, leaving behind vascular scars, and that microglial phagocytosis contributes to the local loss of endothelial cells. These results define the role of NPA and microglia in local blood vessel disassembly and highlight the vascular component of presenilin loss of function in AD

    Overview of recent TJ-II stellarator results

    The main results obtained in the TJ-II stellarator in the last two years are reported. The most important topics investigated have been modelling and validation of impurity transport, validation of gyrokinetic simulations, turbulence characterisation, effect of magnetic configuration on transport, fuelling with pellet injection, fast particles and liquid metal plasma facing components. As regards impurity transport research, a number of working lines exploring several recently discovered effects have been developed: the effect of tangential drifts on stellarator neoclassical transport, the impurity flux driven by electric fields tangent to magnetic surfaces and attempts of experimental validation with Doppler reflectometry of the variation of the radial electric field on the flux surface. Concerning gyrokinetic simulations, two validation activities have been performed, the comparison with measurements of zonal flow relaxation in pellet-induced fast transients and the comparison with experimental poloidal variation of fluctuations amplitude. The impact of radial electric fields on turbulence spreading in the edge and scrape-off layer has been also experimentally characterized using a 2D Langmuir probe array. Another remarkable piece of work has been the investigation of the radial propagation of small temperature perturbations using transfer entropy. Research on the physics and modelling of plasma core fuelling with pellet and tracer-encapsulated solid-pellet injection has produced also relevant results. Neutral beam injection driven Alfvénic activity and its possible control by electron cyclotron current drive has been examined as well in TJ-II.
