The discovery of CRISPR in archaea and bacteria

CRISPR-Cas are self-/nonself-discriminating systems found in prokaryotic cells. They represent a remarkable example of molecular memory that is hereditarily transmitted. Their discovery can be considered as one of the first fruits of the systematic exploration of prokaryotic genomes. Although this genomic feature was serendipitously discovered in molecular biology studies, it was the availability of multiple complete genomes that shed light about their role as a genetic immune system. Here we tell the story of how this discovery originated and was slowly and painstakingly advanced to the point of understating the biological role of what initially was just an odd genomic feature.FJMM is funded by the Spanish Ministerio de Economía y Competitividad (BIO2014-53029P) and the European Commission/Instituto Nacional de Investigación y Tecnología Agraria y Alimentaria (291815 Era-Net ANIHWA). Several authors have affirmed the importance of exercising positive parenting in families whose children present educational, emotional, behavioural difficulties ... to reduce the repercussions of them in their parenting role. In this article, it aims to raise awareness about the real needs that arise in such family contexts, helping to improve their situation. An intervention project has been designed for these families. Due to justifying the program and detecting the real necessities of these contexts, a qualitative investigation was carried out using an interview as a method of collecting information Trabajo Fin de Grado Pedagogía US 2016/17 | Julia Rodriguez Valera 6 made on a 9-people sample part of the psycho-pedagogical center called Educadoss in Seville (Andalusia, Spain). Results have revealed a lack of formative and parental skills related with the emotional, behavioural and educational problems their children have, that complicate their educational practice. However, the prominent degree of family involvement is relevant. These results concur with other researches about the obstacles positive parental development has in such contexts and the necessities exposed. Evidences are provided to justify the project and to open new lines of interventions and researches related to the positive parenting.Universidad de Sevilla. Grado en Pedagogí

Characterization of a piezoelectric acoustic sensor fabricated for low-frequency applications: a comparative study of three methods

    Get PDF
Piezoelectric transducers are widely used for generating acoustic energy, and choosing the right radiating element is crucial for efficient energy conversion. In recent decades, numerous studies have been conducted to characterize ceramics based on their elastic, dielectric, and electromechanical properties, which have improved our understanding of their vibrational behavior and aided in the manufacturing of piezoelectric transducers for ultrasonic applications. However, most of these studies have focused on the characterization of ceramics and transducers using electrical impedance to obtain resonance and anti-resonance frequencies. Few studies have explored other important quantities such as acoustic sensitivity using the direct comparison method. In this work, we present a comprehensive study that covers the design, manufacturing, and experimental validation of a small-sized, easy-to-assemble piezoelectric acoustic sensor for low-frequency applications, using a soft ceramic PIC255 from PI Ceramic with a diameter of 10 mm and a thickness of 5 mm. We present two methods, analytical and numerical, for sensor design, followed by experimental validation, allowing for a direct comparison of measurements with simulated results. This work provides a useful evaluation and characterization tool for future applications of ultrasonic measurement systems. This workwas supported by the grants PID2020-116329GB-C22 and TED2021-129336B-I00 funded by MCIN/AEI/10.13039/501100011033 and by the European Union NextGenerationEU/PRTR. Light as an energy source in continuous cultures of bacteriorhodopsin-containing halobacteria

    Get PDF
The role of light as an energy source for slightly aereated cultures of halobac teria was studied, using continuous cultures with low nutrient concentrations and a low oxygen supply. A series of experiments were carried out with non illuminated and differently illuminated cultures and with ditferent oxygen transfer rates. Under low oxygen availability, light proved to be a decisively important energy source that allowed the populations to reach higher growth rates and much higher population densities. Oxygen inftuenced the growth over only a minimal level, below which neither the illuminated nor the dark cultures were atfected by the oxygen transfer rate. From these results, it appears that the bacteriorhodop sin-mediated energy supply could have a very important role for the ecology of halobacteria in their microaerophilic habitats. In the illuminated cultures, cells that originated purple colonies on plates appeared. These cells, which could be bacteriorhodopsin-constitutive mutants, are now being studied

BACKGROUND: The cataloging of marine prokaryotic DNA sequences is a fundamental aspect for bioprospecting and also for the development of evolutionary and speciation models. However, large amount of DNA sequences used to quantify prokaryotic biodiversity requires proper tools for storing, managing and analyzing these data for research purposes. DESCRIPTION: The Micro-Mar database has been created to collect DNA diversity information from marine prokaryotes for biogeographical and ecological analyses. The database currently includes 11874 sequences corresponding to high resolution taxonomic genes (16S rRNA, ITS and 23S rRNA) and many other genes including CDS of marine prokaryotes together with available biogeographical and ecological information. CONCLUSION: The database aims to integrate molecular data and taxonomic affiliation with biogeographical and ecological features that will allow to have a dynamic representation of the marine microbial diversity embedded in a user friendly web interface.
