    First Report of an OXA-23 Carbapenemase-Producing Acinetobacter baumannii Clinical Isolate Related to Tn2006 in Spain

    A carbapenem-resistant Acinetobacter baumannii clinical isolate belonging to European clone II and sequence type 2 was recov-ered from a patient in the Son Espases hospital in Mallorca, Spain. Genetic analysis showed the presence of the bla OXA-23 gene in association with the widely disseminated transposon Tn2006. This is the first reported identification of A. baumannii carrying bla OXA-23 in Spain

    C4-C5 fused pyrazol-3-amines: when the degree of unsaturation and electronic characteristics of the fused ring controls regioselectivity in Ullmann and acylation reactions

    Pyrazol-3-amine is a scaffold present in a large number of compounds with a wide range of biological activities and, in many cases, the heterocycle is C4-C5 fused to a second ring. Among the different reactions used for the decoration of the pyrazole ring, Ullmann and acylation have been widely applied. However, there is some confusion in the literature regarding the regioselectivity of such reactions (substitution at N1 or N2 of the pyrazole ring) and no predictive rule has been so far established. As a part of our work on 3-amino-pyrazolo[3,4-b]pyridones 13, we have studied the regioselectivity of such reactions in different C4-C5 fused pyrazol-3-amines. As a rule of thumb, the Ullmann and acylation reactions take place, predominantly, at the NH and non-protonated nitrogen atom of the pyrazole ring respectively, of the most stable initial tautomer (1H- or 2H-pyrazole), which can be easily predicted by using DFT calculations

    Mission and system architecture for an operational network of earth observation satellite nodes

    Nowadays, constellations and distributed networks of satellites are emerging as clear development trends in the space system market to enable augmentation, enhancement, and possibilities of new applications for future Earth Observation (EO) missions. While the adoption of these satellite architectures is gaining momentum for the attaining of ever more stringent application requirements and stakeholder needs, the efforts to analyze their benefits and suitability, and to assess their impact for future programmes remains as an open challenge to the EO community. In this context, this paper presents the mission and system architecture conceived during the Horizon 2020 ONION project, a European Union research activity that proposes a systematic approach to the optimization of EO space infrastructures. In particular, ONION addressed the design of complementary assets that progressively supplement current programs and took part in the exploration of needs and implementation of architectures for the Copernicus Space Component for EO. Among several use cases considered, the ONION project focused on proposing system architectures to provide improved revisit time, data latency and image resolution for a demanding application scenario of interest: Marine Weather Forecast (MWF). A set of promising system architectures has been subject of a comprehensive assessment, based on mission analysis expertise and detailed simulation for evaluating several key parameters such as revisit time and data latency of each measurement of interest, on-board memory evolution and power budget of each satellite of the constellation, ground station contacts and inter-satellite links. The architectures are built with several heterogeneous satellite nodes distributed in different orbital planes. Each platform can embark different instrument sets, which provide the required measurements for each use case. A detailed mission analysis has then been performed to the selected architecture for the MWF use case, including a refined data flow analysis to optimize system resources; a refined power budget analysis; a delta-V and a fuel budget analysis considering all the possible phases of the mission. This includes from the correction of launcher injection errors and acquisition of nominal satellite position inside the constellation, orbit maintenance to control altitude, collision avoidance to avoid collision with space debris objects and end-of-life (EOL) disposal to comply with EOL guidelines. The relevance of the system architecture selected for the MWF has been evaluated for three use cases of interest (Arctic sea-ice monitoring, maritime fishery pressure and aquaculture, agricultural hydric stress) to show the versatility and the feasibility of the chosen architecture to be adapted for other EO applications.This project has received funding from the European Union's Horizon 2020 research and innovation programme under grant agreement No 687490

    Aberrant gene expression in mucosa adjacent to tumor reveals a molecular crosstalk in colon cancer

    Background: A colorectal tumor is not an isolated entity growing in a restricted location of the body. The patient's gut environment constitutes the framework where the tumor evolves and this relationship promotes and includes a complex and tight correlation of the tumor with inflammation, blood vessels formation, nutrition, and gut microbiome composition. The tumor influence in the environment could both promote an anti-tumor or a pro-tumor response. Methods: A set of 98 paired adjacent mucosa and tumor tissues from colorectal cancer (CRC) patients and 50 colon mucosa from healthy donors (246 samples in total) were included in this work. RNA extracted from each sample was hybridized in Affymetrix chips Human Genome U219. Functional relationships between genes were inferred by means of systems biology using both transcriptional regulation networks (ARACNe algorithm) and protein-protein interaction networks (BIANA software). Results: Here we report a transcriptomic analysis revealing a number of genes activated in adjacent mucosa from CRC patients, not activated in mucosa from healthy donors. A functional analysis of these genes suggested that this active reaction of the adjacent mucosa was related to the presence of the tumor. Transcriptional and protein-interaction networks were used to further elucidate this response of normal gut in front of the tumor, revealing a crosstalk between proteins secreted by the tumor and receptors activated in the adjacent colon tissue; and vice versa. Remarkably, Slit family of proteins activated ROBO receptors in tumor whereas tumor-secreted proteins transduced a cellular signal finally activating AP-1 in adjacent tissue. Conclusions: The systems-level approach provides new insights into the micro-ecology of colorectal tumorogenesis. Disrupting this intricate molecular network of cell-cell communication and pro-inflammatory microenvironment could be a therapeutic target in CRC patient

    [Background]: The rising incidence of colorectal cancer (CRC) among young patients is alarming. We aim to characterize the clinico-pathological features and outcomes of patients with early-onset CRC (EOCRC). [Methods]: We included all of the patients with pathologically confirmed diagnosis of CRC at Hospital Universitario La Paz from October 2016 to September 2020. EOCRC age cut-off was 50 years. All statistical analyses were carried out using SPSS v.25. [Results]: A total of 1152 patients were diagnosed with CRC, fifty-nine (5,1%) of them were After a median follow-up of 24 months, 279 patients have died. Median overall survival (OS) was not reached in either group (p = 0,06). Three-year OS was 80% (95%CI: 73-87) and 67 (95%CI: 65-69) in the younger and older group, respectively. In patients with localized disease that underwent surgery or other antineoplastic treatment ( n = 856), 159 events for disease-free survival (DFS) were observed. In those patients with metastatic EOCRC with mutational status assessed (n ¼23), no difference in OS according to RAS was observed (p ¼ 0,55).[Conclusions]: Patients with EOCRC are diagnosed at a more advanced stage and display distinct biological features (more prevalence of dMMR and WT tumors among others).     Architectural optimization framework for earth-observing heterogeneous constellations : marine weather forecast case

    Earth observation satellite programs are currently facing, for some applications, the need to deliver hourly revisit times, subkilometric spatial resolutions, and near-real-time data access times. These stringent requirements, combined with the consolidation of small-satellite platforms and novel distributed architecture approaches, are stressing the need to study the design of new, heterogeneous, and heavily networked satellite systems that can potentially replace or complement traditional space assets. In this context, this paper presents partial results from ONION, a research project devoted to studying distributed satellite systems and their architecting characteristics. A design-oriented framework that allows selecting optimal architectures for the given user needs is presented in this paper. The framework has been used in the study of a strategic use-case and its results are hereby presented. From an initial design space of 5586 potential architectures, the framework has been able to preselect 28 candidate designs by an exhaustive analysis of their performance and by quantifying their quality attributes. This very exploration of architectures and the characteristics of the solution space are presented in this paper along with the selected solution and the results of a detailed performance analysis.Postprint (author's final draft

    Economic impact of pressure ulcers. An integrative review

    Las lesiones por presión son un importante problema de salud con una gran repercusión epidemiológica y un gran impacto a nivel de salud y calidad de vida que genera importantes costes para las personas, instituciones y sistemas de salud. A pesar de la importancia económica del problema de las lesiones por presión, prácticamente no existe información sistematizada acerca de las diferentes dimensiones de su coste. Para cubrir esta falta de información se planteó la realización de una revisión integrativa acerca de la dimensión económica del problema de las lesiones por presión. Se han identificado y analizado 89 documentos con información económica acerca del problema de las lesiones por presión. La información se ha sistematizado basándose en los siguientes apartados: tiempo necesario para la cicatrización, impacto en las estancias hospitalarias, costes relacionados con la seguridad de los pacientes, impacto en los grupos relacionados de diagnóstico, coste total, coste por episodio, por tipo de tratamiento, de las complicaciones, de las demandas legales, de los años de vida ganados ajustados a calidad de vida, en pacientes lesionados medulares y con otras condiciones, y los costes de medidas preventivas.Pressure Lesions are an important health problem with a great epidemiological impact that affects the health status and quality of life producing important expenses for persons, healthcare institutions and health systems. There is not available systematized information about the different cost components of pressure lesions although the important economic dimension of such problem. In order to cover this lack of systematized information we performed an integrative review about the economic dimension of pressure lesions. We have identified and analyzed 89 documents with economic information about pressure lesions problem. Information has been systematized according with the next categories: time for healing, cost of additional hospital stays, costs related with safety of patients, DRG costs, total cost of treatment, episode’s cost, by type of treatment, related with complications, QALYs, costs in spinal cord patients and in patients with other conditions and global prevention costs

    Gibberellin A1 Metabolism Contributes to the Control of Photoperiod-Mediated Tuberization in Potato

    Some potato species require a short-day (SD) photoperiod for tuberization, a process that is negatively affected by gibberellins (GAs). Here we report the isolation of StGA3ox2, a gene encoding a GA 3-oxidase, whose expression is increased in the aerial parts and is repressed in the stolons after transfer of photoperiod-dependent potato plants to SD conditions. Over-expression of StGA3ox2 under control of constitutive or leaf-specific promoters results in taller plants which, in contrast to StGA20ox1 over-expressers previously reported, tuberize earlier under SD conditions than the controls. By contrast, StGA3ox2 tuber-specific over-expression results in non-elongated plants with slightly delayed tuber induction. Together, our experiments support that StGA3ox2 expression and gibberellin metabolism significantly contribute to the tuberization time in strictly photoperiod-dependent potato plants
