    Humoral Immune Responses in Loggerhead (Caretta caretta) and Kemp\u27s Ridley (Lepidochelys kempii) Sea Turtles

    Sea turtles are a unique group of marine organisms that live in tropical and temperate waters across the globe. They are one of just a few groups of marine reptiles that currently inhabit oceanic environments, and therefore fill an important and distinct role in marine ecosystems. Though much research has been conducted on sea turtle nesting and ecology, comparatively little research has been done on sea turtle immunology, especially in turtles other than green sea turtles (Chelonia mydas). Using a simple technique, immunoglobulin Y (IgY), which is found in reptiles, birds, and amphibians, was purified. Using standard techniques, polyclonal antisera and monoclonal antibodies were made against two forms of IgY (known as IgY and IgY(ΔFc)) for both loggerhead (Caretta caretta) and Kemp\u27s ridley (Lepidochelys kempii) sea turtles. Following antibody production, serum samples from 60 loggerhead and 30 Kemp\u27s ridley individual turtles were examined to look for antibodies specific to 9 marine bacterial species (Escherichia coli, Vibrio cholera, V. parahaemolyticus, V. anguillarum, V. vulnificus, Mycobacterium marinum, Erysipelothrix rhusiopathiae, Streptococcus agalactiae, and Brevundimonas vesicularis). It was found that serum titer levels against these bacteria can vary significantly in regards to factors such as gender and year of serum collection. In addition, some correlations were found between total serum protein levels and serum titers against bacteria, and between straight carapace length and serum titers against bacteria. Measurements taken from these individual turtles (packed cell volume, glucose, and total protein) are similar to measurements seen in other wild, healthy turtles of the same species. This indicates that the bacterial data is not reflective of special or unusual circumstances (such as disease or injury), but is what can be expected from healthy, wild turtles from these populations. This is the first study of its kind to examine serum titers against marine bacteria, and therefore offers clues to the species of bacteria that sea turtles may be exposed to in their natural environments.     Exposure to Oil and Hypoxia Results In Alterations of Immune Transcriptional Patterns In Developing Sheepshead Minnows (\u3ci\u3eCyprinodon variegatus\u3c/i\u3e)

    The area and timing of the Deepwater Horizon oil spill highlight the need to study oil and hypoxia exposure in early life stage fishes. Though critical to health, little research has targeted the effect of oil and hypoxia exposure on developing immune systems. To this end, we exposed sheepshead minnows (Cyprinodon variegatus) at three early life stages: embryonic; post-hatch; and post-larval, to a high energy water accommodated fraction (HEWAF) of oil, hypoxia, or both for 48 hours. We performed RNAseq to understand how exposures alter expression of immune transcripts and pathways. Under control conditions, the embryonic to post-hatch comparison (first transition) had a greater number of significantly regulated immune pathways than the second transition (post-hatch to post-larval). The addition of oil had little effect in the first transition, however, hypoxia elicited changes in cellular and humoral immune responses. In the second transition, oil exposure significantly altered many immune pathways (43), and while hypoxia altered few pathways, it did induce a unique signature of generally suppressing immune pathways. These data suggest that timing of exposure to oil and/or hypoxia matters, and underscores the need to further investigate the impacts of multiple stressors on immune system development in early life stage fishes

    MINDS. JWST-MIRI Reveals a Dynamic Gas-Rich Inner Disk Inside the Cavity of SY Cha

    SY Cha is a T Tauri star surrounded by a protoplanetary disk with a large cavity seen in the millimeter continuum but has the spectral energy distribution (SED) of a full disk. Here we report the first results from JWST-MIRI Medium Resolution Spectrometer (MRS) observations taken as part of the MIRI mid-INfrared Disk Survey (MINDS) GTO Program. The much improved resolution and sensitivity of MIRI-MRS compared to Spitzer enables a robust analysis of the previously detected H2O, CO, HCN, and CO2 emission as well as a marginal detection of C2H2. We also report the first robust detection of mid-infrared OH and ro-vibrational CO emission in this source. The derived molecular column densities reveal the inner disk of SY Cha to be rich in both oxygen and carbon bearing molecules. This is in contrast to PDS 70, another protoplanetary disk with a large cavity observed with JWST, which displays much weaker line emission. In the SY Cha disk, the continuum, and potentially the line, flux varies substantially between the new JWST observations and archival Spitzer observations, indicative of a highly dynamic inner disk.Comment: 19 pages, 10 figures, 5 tables, accepted for publication in Ap

    How Contemporary Human Reproductive Behaviors Influence the Role of Fertility-Related Genes: The Example of the P53 Gene

    Studies on human fertility genes have identified numerous risk/protective alleles involved in the occurrence of reproductive system diseases causing infertility or subfertility. Investigations we carried out in populations at natural fertility seem to suggest that the clinical relevance that some fertility genes are now acquiring depends on their interaction with contemporary reproductive behaviors (birth control, delayed childbearing, and spacing birth order, among others). In recent years, a new physiological role in human fertility regulation has emerged for the tumor- suppressor p53 gene (P53), and the P53 Arg72Pro polymorphism has been associated with recurrent implantation failure in humans. To lend support to our previous observations, we examined the impact of Arg72Pro polymorphism on fertility in two samples of Italian women not selected for impaired fertility but collected from populations with different (premodern and modern) reproductive behaviors. Among the women at near-natural fertility (n = 98), the P53 genotypes were not associated with different reproductive efficiency, whereas among those with modern reproductive behaviors (n = 68), the P53 genotypes were associated with different mean numbers of children [Pro/Pro = 0.75<Pro/Arg = 1.7<Arg/Arg = 2, (p = 0.056)] and a significant negative relationship between the number of children and P53 Pro allele frequencies (p = 0.028) was observed. These results are consistent with those of clinical studies reporting an association between the P53 Pro allele and recurrent implantation failure. By combining these findings with previous ones, we suggest here that some common variants of fertility genes may have become “detrimental” following exposure to modern reproductive patterns and might therefore be associated with reduced reproductive success. Set within an evolutionary framework, this change could lead to the selection of a set of gene variants fitter to current reproductive behaviors as the shift to later child-bearing age in developed countries

    MINDS. Abundant water and varying C/O across the disk of Sz 98 as seen by JWST/MIRI

    MIRI/MRS on board the JWST allows us to probe the inner regions of protoplanetary disks. Here we examine the disk around the classical T Tauri star Sz 98, which has an unusually large dust disk in the millimetre with a compact core. We focus on the H2_2O emission through both its ro-vibrational and pure rotational emission. Furthermore, we compare our chemical findings with those obtained for the outer disk from Atacama Large Millimeter/submillimeter Array (ALMA) observations. In order to model the molecular features in the spectrum, the continuum was subtracted and LTE slab models were fitted. The spectrum was divided into different wavelength regions corresponding to H2_2O lines of different excitation conditions, and the slab model fits were performed individually per region. We confidently detect CO, H2_2O, OH, CO2_2, and HCN in the emitting layers. The isotopologue H218^{18}_2O is not detected. Additionally, no other organics, including C2_2H2_2, are detected. This indicates that the C/O ratio could be substantially below unity, in contrast with the outer disk. The H2_2O emission traces a large radial disk surface region, as evidenced by the gradually changing excitation temperatures and emitting radii. The OH and CO2_2 emission are relatively weak. It is likely that H2_2O is not significantly photodissociated; either due to self-shielding against the stellar irradiation, or UV-shielding from small dust particles. The relative emitting strength of the different identified molecular features point towards UV-shielding of H2_2O in the inner disk of Sz 98, with a thin layer of OH on top. The majority of the organic molecules are either hidden below the dust continuum, or not present. In general, the inferred composition points to a sub-solar C/O ratio (<0.5) in the inner disk, in contrast with the larger than unity C/O ratio in the gas in the outer disk found with ALMA.Comment: Submitted to A&A on May 25 2023. 18 pages, 11 figure

    Increase in serotype 19A prevalence and amoxicillin non-susceptibility among paediatric Streptococcus pneumoniae isolates from middle ear fluid in a passive laboratory-based surveillance in Spain, 1997-2009

    BACKGROUND: Conjugate vaccines, such as the 7-valent conjugate vaccine (PCV7), alter serotype nasopharyngeal carriage, potentially increasing cases of otitis media by non-vaccine serotypes. METHODS: All paediatric middle ear fluid (MEF) isolates received in the Spanish Reference Laboratory for Pneumococci through a passive, laboratory-based surveillance system from January 1997 to June 2009 were analysed. Data from 1997 to 2000 were pooled as pre-vaccination period. Trends over time were explored by linear regression analysis. RESULTS: A total of 2,077 isolates were analysed: 855 belonging to PCV7 serotypes, 466 to serotype 19A, 215 to serotype 3, 89 to serotype 6A and 452 to other serotypes ( 35% isolates) since PCV7 strains represented < 11% of total clinical isolates. CONCLUSIONS: In contrast to reports on invasive pneumococcal strains, in MEF isolates the reduction in the prevalence of PCV7 serotypes was not associated with decreases in penicillin/erythromycin non-susceptibility. The high prevalence of serotype 19A among paediatric MEF isolates and the amoxicillin non-susceptibility found in this serotype are worrisome since amoxicillin is the most common antibiotic used in the treatment of acute otitis media. These data suggest that non-PCV7 serotypes (mainly serotype 19A followed by serotypes 3 and 6A) are important etiological agents of acute otitis media and support the added value of the broader coverage of the new 13-valent conjugate vaccine.This study was supported in part by an unrestricted grant from Pfizer S.A., Madrid, Spain and PRISM-AG, Madrid, Spain.     Background Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage.     Development of Social Variation in Reproductive Schedules: A Study from an English Urban Area

    Background: There is striking social variation in the timing of the onset of childbearing in contemporary England, with the mean age at first motherhood about 8 years earlier in the most deprived compared to the least deprived neighbourhoods. However, relatively little is known about how these social differences in reproductive schedule develop in childhood. Methodology/Principal Findings: We studied the development of differences in reproductive schedules, using a crosssectional survey over 1000 school students aged 9–15 in the metropolitan borough of North Tyneside. Students from more deprived neighbourhoods had earlier ideal ages for parenthood than those from more affluent ones, and these differences were fully apparent by age 11. We found evidence consistent with three mechanisms playing a role in maintaining the socioeconomic gradient. These were: vertical intergenerational transmission (students whose own parents were younger at their birth wanted children younger); oblique intergenerational transmission (students in neighbourhoods where parents were younger in general wanted children earlier); and low parental investment (students who did not feel emotionally supported by their own parents wanted children at a younger age). Conclusions/Significance: Our results shed some light on the proximate factors which may be involved in maintaining early childbearing in disadvantaged communities. They help understand why educational initiatives aimed at adolescents tend to have no effect, whereas improving the well-being of poor families with young children may do so. Our results also sugges

    MINDS. The detection of 13^{13}CO2_{2} with JWST-MIRI indicates abundant CO2_{2} in a protoplanetary disk

    We present JWST-MIRI MRS spectra of the protoplanetary disk around the low-mass T Tauri star GW Lup from the MIRI mid-INfrared Disk Survey (MINDS) GTO program. Emission from 12^{12}CO2_{2}, 13^{13}CO2_{2}, H2_{2}O, HCN, C2_{2}H2_{2}, and OH is identified with 13^{13}CO2_{2} being detected for the first time in a protoplanetary disk. We characterize the chemical and physical conditions in the inner few au of the GW Lup disk using these molecules as probes. The spectral resolution of JWST-MIRI MRS paired with high signal-to-noise data is essential to identify these species and determine their column densities and temperatures. The QQ-branches of these molecules, including those of hot-bands, are particularly sensitive to temperature and column density. We find that the 12^{12}CO2_{2} emission in the GW Lup disk is coming from optically thick emission at a temperature of ∌\sim400 K. 13^{13}CO2_{2} is optically thinner and based on a lower temperature of ∌\sim325 K, may be tracing deeper into the disk and/or a larger emitting radius than 12^{12}CO2_{2}. The derived NCO2N_{\rm{CO_{2}}}/NH2ON_{\rm{H_{2}O}} ratio is orders of magnitude higher than previously derived for GW Lup and other targets based on \textit{Spitzer}-IRS data. This high column density ratio may be due to an inner cavity with a radius in between the H2_{2}O and CO2_{2} snowlines and/or an overall lower disk temperature. This paper demonstrates the unique ability of JWST to probe inner disk structures and chemistry through weak, previously unseen molecular features.Comment: 15 pages, 10 figures. Accepted to ApJ
