    The Art of Polyfoam Puppetry

    The construction of polyfoam puppets for theatrical productions is a highly specialized art form and requires a process that is often learned via mentorships and apprenticeships. Limited learning materials and resources presently exist or are publicly available on the techniques for producing these complex and genre specific puppets. This project investigated and visually documented the creative process and craftsmanship in the creation of polyfoam puppets. Working collaboratively with an undergraduate student of film and digital media, theatre Faculty Designers created an instructional video presentation of the step by step stages of polyfoam puppetry construction.     The lipidome of Crithidia fasiculata and its plasticity

    We would like to thank the Engineering and Physical Sciences Research Council, University of St. Andrews, and the EPSRC Centre for Doctoral Training in Critical Resource Catalysis (CRITICAT) for financial support [Ph.D. studentship to MC; Grant code: EP/L016419/1].Crithidia fasiculata belongs to the trypanosomatidae order of protozoan parasites, bearing close relation to other kinetoplastid parasites such as Trypanosoma brucei and Leishmania spp. As an early diverging lineage of eukaryotes, the study of kinetoplastid parasites has provided unique insights into alternative mechanisms to traditional eukaryotic metabolic pathways. Crithidia are a monogenetic parasite for mosquito species and have two distinct lifecycle stages both taking place in the mosquito gut. These consist of a motile choanomastigote form and an immotile amastigote form morphologically similar to amastigotes in Leishmania. Owing to their close relation to Leishmania, Crithidia are a growing research tool, with continuing interest in its use as a model organism for kinetoplastid research with the added benefit that they are non-pathogenic to humans and can be grown with no special equipment or requirements for biological containment. Although comparatively little research has taken place on Crithidia, similarities to other kinetoplast species has been shown in terms of energy metabolism and genetics. Crithidia also show similarities to kinetoplastids in their production of the monosaccharide D-arabinopyranose similar to Leishmania, which is incorporated into a lipoarabinogalactan a major cell surface GPI-anchored molecule. Additionally, Crithidia have been used as a eukaryotic expression system to express proteins from other kinetoplastids and potentially other eukaryotes including human proteins allowing various co- and post-translational protein modifications to the recombinant proteins. Despite the obvious usefulness and potential of this organism very little is known about its lipid metabolism. Here we describe a detailed lipidomic analyses and demonstrate the possible placidity of Crithidia’s lipid metabolis. This could have important implications for biotechnology approaches and how other kinetoplastids interact with, and scavenge nutrients from their hosts.Publisher PDFPeer reviewe

    Cambrian age Welsh slate: a global heritage stone resource from the United Kingdom

    Slate from the Cambrian succession of North Wales is a well-known source of building products from the United Kingdom and is here advocated as a suitable “Global Heritage Stone Resource”. Its first recorded use was in the Roman period in Wales, and subsequently from the sixteenth century throughout the British Isles. During the 16th and 17th centuries several small companies worked the slate belt from Bethesda to the Nantlle valley but in the mid-18th these were gradually taken over or amalgamated and three large operations came to dominate the industry: Penrhyn, Dinorwic, and the Moel Tryfan to Dorothea group of quarries. From the late eighteenth century production expanded rapidly supplying markets worldwide especially to northern Europe and the British colonies. Slate has been used in all its forms but most notably as roofing slates in the construction of buildings at all levels in society and for buildings of the highest historical and architectural importance. Modernisation of the industry has enabled Cambrian Welsh Slate to continue to be quarried today in an environmentally sensitive manner by Welsh Slate Ltd

    Cet article démontre que “The Cousins” est sans nul doute l’une des meilleures nouvelles d’Elizabeth Spencer car c’est dans cette longue histoire qu’elle tisse le plus habilement le passé dans le présent et la réalité interne de son narrateur dans son environnement extérieur. Elle décrit des décennies et des milliers de kilomètres de territoire dans ses quarante-trois pages de sorte que la nouvelle se lit, comme l’ont noté plusieurs commentateurs, plus comme un roman que comme une nouvelle. Les préoccupations caractéristiques de Spencer (l’évolution individuelle dans un environnement familial étouffant, le choc qui surgit de la rencontre entre le connu et l’inconnu, les mystères souvent douloureux de l’intimité, la prégnance du passé dans le présent) se retrouvent toutes dans “The Cousins” avec une subtilité remarquable et même avec un certain mystère. L’histoire est indéniablement le produit de la maturité de Spencer, comme en témoigne l’imbrication sans faille des réalités extérieures et intérieures, des vies passées et présentes, de la sensibilité masculine et féminine, et de la narration. Ce texte confirme le statut de Spencer comme l’un des maîtres incontestés de la nouvelle

    A General Route for Post-Translational Cyclization of mRNA Display Libraries

    Cyclic peptides are attractive scaffolds for the design of conformationally constrained molecular therapeutics. Previously, biological display libraries could only be cyclized via disulfide bonds, which are labile and can be reduced in an intracellular environment. In this paper, we construct high diversity, covalently cyclized mRNA display libraries (>10^(13) sequences) and analyze the cyclization reaction using MALDI-TOF MS and unnatural amino acid incorporation. Our route allows the extent of cyclization to be evaluated quantitatively and is broadly applicable to a variety of cyclization chemistries

    Predictive intelligence for a rail traffic management system

    As the demands on terrestrial transport systems increase, there is a growing need for greater efficiencies. More intelligent mobility and ultimately autonomous transport assets will deliver these efficiencies through the evolution of cooperative intelligent transport system (C-ITS) technology. Central to this evolution will be the capability to accurately and precisely position assets within their environment and relative to each other to predefined and regulated standards. The core of modern positioning and navigation methods are the global navigation satellite systems (GNSS) (e.g. GPS, Galileo, GLONASS and BeiDou). These systems rely on line of sight radio frequency signals, which are vulnerable to obstruction and/or interference (e.g. multipath and/or non-line of sight reception). As a result, the position accuracy is degraded and therefore GNSS would greatly benefit from a priori intelligence that predicts where and when obscuration or interference will occur. Similarly, a real time assessment of where and when GNSS signal reception will be restored and the location of the satellites in the sky will aid in restoring satellite lock. This paper describes a computer vision system that utilises 360o images to derive a priori intelligence to predict changes in the environment that may threaten position and navigation integrity
