    Tracking the origin of invasive Rosa rubiginosa populations in Argentina

    The exact geographic origin of invasive species populations is rarely known; however, such knowledge is vital to understanding species’ invasion success, spread, and evolution as well as for assessing any biological control options. We investigated the shrub Rosa rubiginosa L., focusing on the presumed European origin of invasive populations in Argentina. We analyzed eight polymorphic microsatellite loci among 102 native (European) and 29 invasive (mainly central Argentinean and Patagonian) populations. Genetic diversity in the invasive range was clearly lower than in the native range, possibly because of a low number of introductions. Contrary to earlier hypotheses, the interpretation of principal coordinate analysis results and Jaccard dissimilarities contradicts the idea of the Argentinean populations having a Spanish origin. Instead, we found a close similarity between Argentinean samples and those from Germany, the Czech Republic, and Austria. We therefore assume that these neighboring countries are the most probable source regions for the Argentinean populations, which in some cases may also have arrived via Chile. According to historic information, emigrants from these regions may have introduced R. rubiginosa to South America in the nineteenth century on at least two occasions, either for food or as rootstock material for propagating living fences.Fil: Hirsch, Heidi.     <p>Abstract</p> <p>Background</p> <p>Multiple hybridization events gave rise to pentaploid dogroses which can reproduce sexually despite their uneven ploidy level by the unique canina meiosis. Two homologous chromosome sets are involved in bivalent formation and are transmitted by the haploid pollen grains and the tetraploid egg cells. In addition the egg cells contain three sets of univalent chromosomes which are excluded from recombination. In this study we investigated whether differential behavior of chromosomes as bivalents or univalents is reflected by sequence divergence or transcription intensity between homeologous alleles of two single copy genes (<it>LEAFY</it>, <it>cGAPDH</it>) and one ribosomal DNA locus (<it>nrITS</it>).</p> <p>Results</p> <p>We detected a maximum number of four different alleles of all investigated loci in pentaploid dogroses and identified the respective allele with two copies, which is presumably located on bivalent forming chromosomes. For the alleles of the ribosomal DNA locus and <it>cGAPDH </it>only slight, if any, differential transcription was determined, whereas the <it>LEAFY </it>alleles with one copy were found to be significantly stronger expressed than the <it>LEAFY </it>allele with two copies. Moreover, we found for the three marker genes that all alleles have been under similar regimes of purifying selection.</p> <p>Conclusions</p> <p>Analyses of both molecular sequence evolution and expression patterns did not support the hypothesis that unique alleles probably located on non-recombining chromosomes are less functional than duplicate alleles presumably located on recombining chromosomes.</p

    Combination treatment with an ETA‐receptor blocker and an ACE inhibitor is not superior to the respective monotherapies in attenuating chronic transplant vasculopathy in different aorta allotransplantation rat models

    Background. The effect of the specific endothelin A (ETA)‐receptor antagonist LU 302146 (LU) was assessed in a normotensive model of chronic transplant vasculopathy, i.e. orthotopic allotransplantation of the infrarenal abdominal aorta from spontaneously hypertensive‐to‐Wistar-Kyoto (SHR‐to‐WKY) rats. A second experimental setting was used to confirm the results in a different model, which to some extent may also address the issue of blood pressure (BP) in transplant vasculopathy, i.e. orthotopic allotransplantation of infrarenal abdominal aorta from WKY‐to‐SHR rats. Untreated sham‐operated and isografted WKY and SHR served as controls. Allotransplanted animals treated with the angiotensin‐converting enzyme (ACE) inhibitor trandolapril served as positive treatment controls. Methods. Rats were randomized to receive standard diet or a diet designed to deliver either 30 mg LU/kg bw/day, 0.3 mg/kg bw/day trandolapril or a combination of both. The duration of either experiment was 8 weeks. BP was measured by tail plethysmography. Results. Treatment with LU did not affect systolic BP in either experimental setting. In contrast, trandolapril and combination treatment significantly reduced systolic BP in SHRs. The increase in aortic wall thickness (given in mm) was abrogated to a similar extent in the three treatment groups as compared with untreated allotransplanted animals in either experimental setting (e.g. WKY sham‐operated 0.084±0.013, P<0.05 vs treatment groups; WKY isotransplanted 0.100±0.010, P<0.05 vs treatment groups; WKY allotransplanted 0.289±0.077, P<0.05 vs all groups; WKY allotransplanted+trandolapril 0.185±0.025; WKY allotransplanted+LU 301246 0.192±0.049; WKY allotransplanted+LU 301246+trandolapril 0.190±0.041). This was due to an attenuation of the increase of intima and media thickness. Treatment with LU and trandolapril were similarly effective in attenuating the increase of the number of proliferating cell nuclear antigen (PCNA)‐positive cells in the intima. Again, combination treatment did not confer additional benefit. In contrast, trandolapril was more effective than LU in attenuating the increase in the number of PCNA‐positive cells in the media. Trandolapril or combination treatment, but not LU, attenuated transforming growth factor‐β expression in aortic allografts. Conclusions. The ETA‐receptor blockade abrogates allograft vasculopathy in two different aorta allotransplantation models to a similar extent as ACE inhibition even in the absence of concomitant immunosuppression. At least in SHRs the effect of ETA‐receptor blockade is independent of BP. This finding is consistent with the notion that ETA‐receptor mediated events play a partly BP‐independent role in the genesis of chronic transplant vasculopath

    Untangling the hedge: Genetic diversity in clonally and sexually transmitted genomes of European wild roses, Rosa L.

    While European wild roses are abundant and widely distributed, their morphological taxonomy is complicated and ambiguous. In particular, the polyploid Rosa section Caninae (dogroses) is characterised by its unusual meiosis, causing simultaneous clonal and sexual transmission of sub-genomes. This hemisexual reproduction, which often co-occurs with vegetative reproduction, defies the standard definition of species boundaries. We analysed seven highly polymorphic microsatellite loci, scored for over 2 600 Rosa samples of differing ploidy, collected across Europe within three independent research projects. Based on their morphology, these samples had been identified as belonging to 21 dogrose and five other native rose species. We quantified the degree of clonality within species and at individual sampling sites. We then compared the genetic structure within our data to current rose morpho-systematics and searched for hemisexually co-inherited sets of alleles at individual loci. We found considerably fewer copies of identical multi-locus genotypes in dogroses than in roses with regular meiosis, with some variation recorded among species. While clonality showed no detectable geographic pattern, some genotypes appeared to be more widespread. Microsatellite data confirmed the current classification of subsections, but they did not support most of the generally accepted dogrose microspecies. Under canina meiosis, we found co-inherited sets of alleles as expected, but could not distinguish between sexually and clonally inherited sub-genomes, with only some of the detected allele combinations being lineage-specific

    Die Sömmerung von Karpfenteichen war lange Zeit fester Bestandteil der Karpfenteichwirtschaft. Im Zusammenhang mit sinkenden Wasserdargeboten rückt die Sömmerung erneut in den Blick. Der Bericht beschreibt die Ergebnisse von Versuchen zur Teichsömmerung. Er richtet sich an alle interessierte Teichwirte. Die Sömmerung kann heute aus hydrologischen, betriebswirtschaftlichen oder seuchenbiologischen Gründen durchaus wieder sinnvoll sein. Durch die Sömmerung von Karpfenteichen entstehen als Nebeneffekt naturschutzfachlich hochwertigste Habitate. Auf Sömmerungsflächen entwickelt sich eine besonders schützenswerter Flora und Fauna.     Homoarginine, heart failure, and sudden cardiac death in haemodialysis patients

    Aims Sudden cardiac death (SCD) is a major contributor to the excess mortality of patients on maintenance dialysis. Homoarginine deficiency may lead to decreased nitric oxide availability and endothelial dysfunction. Based on this rationale we assessed whether homoarginine deficiency is a risk factor for SCD in dialysis patients. Methods and results This study examined the association of homoarginine with cardiovascular outcomes in 1255 diabetic haemodialysis patients from the German diabetes and dialysis study. During a median of 4 years of follow-up, hazard ratios (HR) (95% CI) for reaching the following pre-specified, adjudicated endpoints were determined: SCD, myocardial infarction, stroke, death due to heart failure, and combined cardiovascular events. There was a strong association of low homoarginine concentrations with the presence of congestive heart failure and left ventricular hypertrophy as well as increased levels of brain natriuretic peptide. Per unit decrease in homoarginine, the risk of SCD increased three-fold (HR 3.1, 95% CI 2.0–4.9), attenuating slightly in multivariate models (HR 2.4; 95% CI 1.5–3.9). Patients in the lowest homoarginine quintile experienced a more than two-fold increased risk of SCD, and more than three-fold increased risk of heart failure death than patients in the highest quintile, which accounted for the high incidence of combined cardiovascular events. Low homoarginine showed a trend towards increased risk of stroke, however, myocardial infarction was not meaningfully affected. Conclusion Low homoarginine is a strong risk factor for SCD and death due to heart failure in haemodialysis patients. Further studies are needed to elucidate the underlying mechanisms, offering the potential to develop new interventional strategies

    Spin-locking in low-frequency reaction yield detected magnetic resonance

    The purported effects of weak magnetic fields on various biological systems from animal magnetoreception to human health have generated widespread interest and sparked much controversy in the past decade. To date the only well established mechanism by which the rates and yields of chemical reactions are known to be influenced by magnetic fields is the radical pair mechanism, based on the spin-dependent reactivity of radical pairs. A diagnostic test for the operation of the radical pair mechanism was proposed by Henbest et al. [J. Am. Chem. Soc., 2004, 126, 8102] based on the combined effects of weak static magnetic fields and radiofrequency oscillating fields in a reaction yield detected magnetic resonance experiment. Here we investigate the effects on radical pair reactions of applying relatively strong oscillating fields, both parallel and perpendicular to the static field. We demonstrate the importance of understanding the effect of the strength of the radiofrequency oscillating field; our experiments demonstrate that there is an optimal oscillating field strength above which the observed signal decreases in intensity and eventually inverts. We establish the correlation between the onset of this effect and the hyperfine structure of the radicals involved, and identify the existence of ‘overtone’ type features appearing at multiples of the expected resonance field positio

    Chemical Magnetoreception: Bird Cryptochrome 1a Is Excited by Blue Light and Forms Long-Lived Radical-Pairs

    Cryptochromes (Cry) have been suggested to form the basis of light-dependent magnetic compass orientation in birds. However, to function as magnetic compass sensors, the cryptochromes of migratory birds must possess a number of key biophysical characteristics. Most importantly, absorption of blue light must produce radical pairs with lifetimes longer than about a microsecond. Cryptochrome 1a (gwCry1a) and the photolyase-homology-region of Cry1 (gwCry1-PHR) from the migratory garden warbler were recombinantly expressed and purified from a baculovirus/Sf9 cell expression system. Transient absorption measurements show that these flavoproteins are indeed excited by light in the blue spectral range leading to the formation of radicals with millisecond lifetimes. These biophysical characteristics suggest that gwCry1a is ideally suited as a primary light-mediated, radical-pair-based magnetic compass receptor
